1.Identification and analysis of membrane estrogen receptor in inters titial cells of Cajal

Objective:To detect and analyze the membrane estrogen receptor(mER)in primary cultured interstitial cells of Cajal(ICC).Methods:Interstitial Cells of Cajal(ICC)'s surface binding sites for 17-?estradiol(E2)were detected by cell-impermeant ligand using confocal microscopy.Radioligand binding assay and Scatchard software were used to analyzed the characteristics of mER.Results:Immunofluorescence shows the staining pattern of nonfixed,nonpermeabilized ICC incubated with E2BSAFITC.The radioligand binding assay were analyzed by Scatchard software,The Bmax of mER was 45.75 fmol/mg protein and the kD was 0.717 3 nmol/L.Conclusion:A form of the estrogen receptor is present within the cell membrane of ICC and maybe capable of mediating rapid effect of estrogen.
2.Influence of trimetazidine combined with atorvastatin on heart function and the BNP level in patients with coronary heart failure

Objective To investigate the influence of trimetazidine combined with atorvastatin on heart func-tion and the BNP level in patients with cornary heart failure.Methods The clinical data of 400 patients with cornary heart disease were retrospectively analyzed.194 patients in the control group were treated with conventional treatment, 206 patients in the observation group were treated with trimetazidine combined with atorvastatin on the basis of the control group.The treatment effect,cardiac function and BNP level were observed and compared.Results The effec-tive rate of the observation group was 91.26%,which was significantly higer than 76.29% of the control group (χ2 =16.667,P <0.05).After treatment,the cardiac index,stroke volume,cardiac output,left ventricular ejection fraction and BNP level of the two groups were significantly higher than before treatment,and those in the observation group were significantly higher than the control group(t =2.505,2.851,3.308,3.452,all P <0.05).Conclusion Trime-tazidine combined with atorvastatin in treatment of cornary heart failure has good curative effect,it can reduce the serum level of BNP,improve cardiac function significantly,which is of great value for clinical use.
3.Identification and characterization of class 1 integron among E. coli from healthy students' enteric strains

Objective To explore the distribution and characterization of class 1 integrons in E.coli from healthy feces,and to elucidate the status of gene-cassettes.Methods Routine method was used to isolate E.coli,antibiotics susceptibility was tested by the disk diffusion method;class 1 integron was detected by PCR assay;PCR products were sequenced and analyzed.Results Of 97 samples,76 isolates were identified,and 25 isolates were multiple-drug resistant.The antibiogram was sulfamethoxazole-trimethoprim,ampicillin,streptomycin,tetracycline,erythromycin.14 of 25 isolates carried class 1 integrons,and the size of integrons differed from 1 800 bp(10 strains) to 750 bp(4 strains).The sequenced PCR product demonstrated that the 1 800 bp integron laboured aadA1-dfrA14-orf gene cassette conferred the resistance to sulfamethoxazole-timethoprim,streptomycin and aminoglycoside;the 750 bp integron laboured dfrA14 gene cassette conferred the resistance to sulfamethoxazole-trimethoprim.Conclusion The different kinds of class 1 integrons exist in E.coli from the healthy students,and determine the multiple-resistant antibiotics.
4.Treatment and prevention of deep vein thrombosis after stroke

Owing to the advanced age,limb hemiplegia,dehydration,and vessel wall injury,stroke may be easy to cause venous thromboembolism (VTE).VTE mainly include deep vein thrombosis (DVT) and pulmonary embolism (PE).DVT refers to abnormal blood clotting in the veins and impedes venous return.The dislodgement of emboli from the vessel wall can form a thrombotic embolism,and cause PE,myocardial infarction,and stroke; it can not only prolong hospitalization,but also increase the mortality.This article reviews the incidence,risk factors,treatment,and prevention of DVT after a stroke.
5.Research progress of early onset gestational diabetes mellitus

Gestational diabetes refers to the first occurrence or discovery of different degrees of abnormal glucose tolerance during pregnancy, which is one of the most common complications during pregnancy. As the incidence rate increased year by year, the complications of mother and child increased significantly. In recent years, gestational diabetes mellitus has been paid more and more attention. Most of the current researches are about the diagnosis and treatment of gestational diabetes 24 weeks and later, and the theoretical basis for the screening method and treatment of early gestational diabetes is still lacking. With the further understanding of early gestational diabetes mellitus and its adverse maternal and infant pregnancy outcomes, it is imperative to actively carry out the research on the screening and treatment of early gestational diabetes mellitus.
6. Multi-detector CT features of benign and malignant pulmonary focal ground-glass opacity nodules: A binary logistic regression analysis

Objective: To evaluate the different multi-detector computed tomography features of benign and malignant pulmonary focal ground-glass opacity (fGGO) nodules. Methods: The demographic data(sex and age), nodule location, size, and multi-detector CT (MDCT) features (shape, margin, interface, internal characteristics, and adjacent structures) of 82 fGGO patients were retrospectively analyzed. Differences between benign and malignant fGGO were analyzed by using chisquare test, Fisher exact test, and Mann-Whitney U test. Binary logistic regression analysis was also performed to draw a regression equation. Results: The 82 fGGO patients included 61 with malignant and 21 with benign nodules. No statistical differences were found between benign and malignant fGGO nodules in terms of demographic data, nodule location, or nodule size. Binary logistic regression analysis showed that lobulation, interface, and pleural indentation were the important features of malignant nodules, with the corresponding odds ratio (OR) being 8.122, 3.139, and 9.076, respectively. The sensitivity, specificity, and accuracy of the obtained equation were 93.4%, 66.7%, and 86.6%, respectively. Conclusion: An fGGO nodule with lobulation, well-defined but coarse interface, and pleural indentation in its MDCT image, is more likely to be malignant.
7. Influence of lung inflation on arterial spin labeling signal in MR perfusion imaging of human lungs

Objective: To investigate the influence of lung inflation on MR perfusion imaging of human lung using an arterial spin labeling sequence called flow sensitive alternating inversion recovery (FAIR). Methods: Coronal perfusion-weighted images were obtained at different respiratory phases from 10 healthy volunteers on a 1.5T whole body scanner (GE medical system) using FAIR sequence. The changes of tagging efficiency of pulmonary parenchyma (ΔSI%), pulmonary blood flow and area of the scanning slice of different respiratory phases were analyzed. Results: (1) Significant difference was found in ΔSI% between different respiratory phases(right lung P=0.021 5, left lung P=0.008 4), with that at end expiration greater than that at end inspiration. (2) Significant difference was also found in pulmonary blood flow at different respiratory phases (right lung P= 8. 92 X 10-5, left lung P=0.000 2), with that at end expiration higher than that at end inspiration. (3) The areas of the scanning slice were also significantly different at different respiratory phases (right lung P=2.94 X 10-5, left lung P=0.000 5), with that at end inspiration larger than that at end expiration. Conclusion: Pulmonary blood flow during expiration is higher than that during inspiration, which might be due to the decreased lung volume and increased vascular density during expiration.
8.Analysis of Usage of Antibiotics and Bacterial Drug Resistance in Infected Infants of Pediatrics in Changchun

Objective To investigate the usage of antibiotics in pediatrics patients in Changchun,and detect the bacterial drug resistance of main pathogens for direction of the appropriate clinical choice of drug antibiotics.Methods A total of 900 cases of infectious patients were retrospectively investigated in antibiotic usage in the ⅢA hospital in Changchun from January 2003 to November 2005 and the data were statistics analyzed.The bacterium were isolated from all kinds of specimen of children in hospital patients from December 2004 to November 2005,according to the method of "Clinical Medicine Test Handbook".All bacterial isolated from bacterial susceptibility tes-(ting) was done with Kirby-Bauey method.Tests were performed according to the guidelines of National Committee for Clinical Laboratory Standards(NCCLS)(2004).Results Antibiotics were administered in all of the cases.Four categories and 11 kinds of antibiotics were used.Usage of the first six antibiotics were as followed: ceftezole(22.3%),cefoperazone-sulbactam(21.6%),amoxicillin clavulanic acid((19.6%),)penicilin(17.2%),azithromycin(16.40%),oxacillin(10.1%).The rate of antibiotic use was 60.6% for solo use,70.7% for bigeminy use,81.7% for intravenously guttae,18.3% for peroral administration.Eight hundred and eighteen strains of pathogens were isolated.Gram-positive coccus,gram-negative accounted for 48.0% and 52.0%,respectively.All bacterium isolated showed a high resistance against penicillins and cephalosporins.Drug resistance rate of Gram-positive bacteria to penicillin was more than 50.0%,CNS,S.aureus,E.faecalis and S.pneumonic to erythromycin and cphalosporins was over 71.4% and 35.7%.The antibiotic remaining the most active against Gram-positive bacteria was vancomycin.The drug resistance rate against imipenem and ciprofloxacin were lower than(25.0%.)The antibiotic resistance rate of Gram-negative bacteria to ampicillin,cephalosporins and aztreonam were more than 83.0%,(40.0%) and 32.0%,to piperacillin and amoxicillin-clavulanic acid were morn than 68.9%,except P.aeruginosa(33.3%).The susceptibility of Gram-negative bacteria was imipenem(
9.Clinical application of superficial temporal artery island flap in the reconstruction of eyelid and periorbital defect

Serious defect of eyelid and periorbital tissue affects the appearance of patients and closure-eye function.Application of superficial temporal artery island flap to repair the defected tissue is considered to be an available method to reconstruct eyelid and periorbital tissue.ObjectiveThis study aims to investigate the indication,safety and effectivity of reconstructing the serious defect of eyelid and periorbital tissue by forward flow flap.MethodsEighteen eyes of 18 cases with serious eyelid and periorbital tissue defect were reconstructed by covering the defect zone with superficial temporal artery to form forward flow flap.The flap was made along the distribution of superficial temporal artery with the size over the tissue detect area and transferred to lesion via subcutaneous tunnel.The follow-up time was 6 months.Survival status of flap,plastic condition of lesion and surgical treatment were discussed.ResultsAmong 18 cases with eyelid and periorbital defect,the flaps were survived in 16 cases with the satisfactory repairing appearance,showing nice color,soft texture and normal sensation after plastic surgery.Reconstructed eyelids closed well.One case showed the temporary obstruction of venous refluence in early stage after operation and remained distal epidermal necrosis.Reoperation was performed in 6 patients because of flap hypertrophy and showed a successful outcome.One patients occurred the failure of plastic flap due to hypertrophy and underwent free-flap transplantation in 3 months following the first surgery and obtained a ultimately flap survival.ConclusionThe superficial temporal artery forward flow flap offers a stable and reliable blood supplying and provides adequate tissue of excellent color and texture match.It is an ideal and available flap for the reconstruction of serious eyelid and periorbital tissue defect.
10.Effects of cyclophosphamide on pharmacokinetics of buthionine sulfoximine in Walker-256 tumor-bearing rats

AIM To study the effects of cyclophosphamide (CTX) on the pharmacokinetics of buthionine sulfoximine (BSO)in Walker 256 tumor bearing rats. METHODS Walker 256 tumor bearing rats were treated ip for 4 days with saline or CTX in saline (20 mg?kg -1 ), then received iv BSO 200 mg?kg -1 . BSO concentration in rat plasma was determined by a reverse phase HPLC with fluorescence detection after precolumn derivatization with o phthaldialdehyde. Compartment model and pharmacokinetic parameters were determined by 3P87 software processed on a computer. RESULTS A single intravenous dose of BSO 200 mg?kg -1 was eliminated from plasma in a two compartment manner in tumor bearing rats. The pharmacokinetic parameters of BSO were as follows: In tumor bearing control rats, T 1/2? =(11 1?2 4) min, T 1/2? =(65?14) min, CLs=(12 8?1 3) ml?min -1 ?kg -1 , AUC=(262?26) mg?L -1 ?h; in tumor bearing CTX treated rats, T 1/2? =(8 2?1 8) min, T 1/2? =(42?3) min, CLs=(13 4?1 9) ml?min -1 ?kg -1 ,AUC=(252?35) mg ?L -1 ?h. CONCLUSION There is no significant difference between the parameters of tumor bearing control and CTX treated rats except T 1/2? .
