1.The immunological character of corneal epithelial cells

Objective:To investigate the capability of corneal cells to stimulate lymphocyte proliferation and the expression of costimulatory molecules on primary human corneal epithelial cells.Methods:Human corneal epithelial cells were cocultivated with peripheral blood mononuclear cells(PBMC). Expression of HLA class Ⅱ antigens, CD40, CD154, CD80 and CD86 was measured by immunohistochemical staining,and the expression of such molecules above on rejected corneal grafts was contrastingly assessed. Activation of lymphocytes was determined by measuring upregulation of CD69 by FACS analysis.Results:HLAⅡ expression was detectable on human primary corneal epithelial cells,and furthermore, upregualtion of costimulatory molecules CD40 and CD80 were also found after induction with ?-interferon. Both of HLA and CD40 were expressed on rejected corneal grafts. T lymphocytes were activated by epithelial cells in the coculture system.Conclusion:Human corneal epithelial cells are involved in rejection process of the corneal transplantation. The immunological reaction is triggered by epithelial cells,and endothelial cells suffer as the targets.
2.Clone of S gene in occult hepatitis B virus infection and construction of prokaryotic expression vector

Objective To construct the clone vector of S gene in occult hepatitis B virus infection.To analyse its mutations and to construct its prokaryotic expression vector.Methods To amplify the HBV S gene with polymerase chain reaction(PCR),recombinate the S gene and vector PUC18,sub-clone the recombination and construct its prokaryotic expression vector with pThiohis B. Results There were two samples negative with positive HBV DNA from 64 samples with negative hepatitis B surface antigen.The clone vector and prokaryotic fusion protein expression vector of an occult HBV infection′s S gene were successfully constructed.The S gene was not changed by sequencing analysis.Conclusion The fluorescence quantitive polymerase chain reaction (FQ-PCR) can examine low-copies in occult hepatitis B virus infection.Determining quantitivly HBV DNA of recombination can identify the recombination and display multiplication state intuitirely.It is very successful in constructing the clone vector and prokaryotic fusion protein expression vector.
3.Human retinal pigment epithelial cells and T-lymphocyte activation

Objective To investigate the expression of Human leucocyte antigen(HLA)-DP,-DQ,-DR and CD40 in human retinal pigment epithelial(RPE)cells,to determine their molecule expression in immune response process,and their abilities to stimulate T lymphocyte activation. Methods Human RPE cells were cultured with or without(-IFN respectively.Expression of HLA-DP,-DQ,-DR and CD40 was measured by immunohistochemical staining.Meanwhile,peripheral blood mononuelear cells(PBMC)were co-cultured with RPE cells in vitro,and then the expression of activated lymphocytes CD69 was measured by fluorescence activated eell sorter(FACS). Results Expression of HLA-DP,-DQ,-DR and CD40 antigen were enhanced by γ-interferon inducement.Increasing amount of CD69 positive Iymphocytes were found in the co-culture system of RPE cells and PBMC. Conclusion T-lymphocytes in the peripheral blood were activated by human RPE cells which is antigen presenting cells with immunological characteristics potential.

6.Comparison of Effects of Oxycodone Hydrochloride and Sufentanil on Anesthesia Recovery in Neurosurgical Operation

Objective To observe the recovery efficiency of sufentanil and oxycodone hydrochloride in neurosurgical operation.Methods Total of 120 patients scheduled for microvascular decompression under general anesthesia were randomly divided into 4 groups to receive 0.03 mg·kg-1 oxycodone (group H1), 0.05 mg·kg-1 oxycodone (group H2), 0.08 mg·kg-1 oxycodone (group H3), and 0.08 μg·kg-1 sufentanil (group S).Each patient accepted 4 ng·mL-1 of remifentanil and 5-6 mg·kg-1·h-1 of propofol after induction to maintain bispectral index (BIS) value at 40-60.On starting to suture dura mater, oxycodone or sufentanil was given in various doses through intravenous route.The BIS value, the amount consumption of propofol, anesthesia and operation duration, and the drug delivery time of each group were recorded.The time of spontaneous breath recovery, awaking and extubation, verbal rating score (VRS) and sedation score (SS) at 10 min, 20 min, 30 min after operation as well as side-effects were also recorded.Results The VRS scores had no statistic difference among the groups at 10 min after operation, while the average scores of H1 were higher than those of H2 and H3 at 20, 30 min after operation.No statistic difference of the SS scores was showed among 4 groups at 10 min after operation, but it was lower in group H1 than those in group H3, and lower in group S than those in H2 at 20 min after operation;group H3 had a significantly higher SS scores than H1, H2 and S at 30 min after operation.Conclusion Oycodone hydrochloride 0.05 mg·kg-1 or sufentanil 0.08 μg·kg-1 can both provide satisfactory recovery quality in neurosurgical operation.

The effect of diethylcarbamazine (DEC), an inhibitor of leukotrienes (LTS) synthesis or indomethacin, an inhibitor of prostaglandins (PGs) synthesis was investigated in chronic hypoxic pulmonary hypertension and reactivity of pulmonary vessels in different strains of rats. [Objective]This paper will pitch-in TCM psychosomatic theory(from Huangdi Neijing),exploring the disorders of depression with the metabolic syndrome and pathogenesis of traditional Chinese medicine,so as to provide effective ideas for the treatment of depression with metabolic syndrome.[Methods]:Through expansion on traditional chinese medicine's basic understanding of depression with metabolic syndrome,utilizing basic TCM viscera-state theory, from the causes of this disease,key factors,pathogenesis transformation three aspects to discuss the pathogenesis of the disease. [Results]By discussing the heart,liver,spleen function of zang-fu organs and their mutual relations in the role and impact of the disease development process, this paper thinks of the nature of disease as asthenia in origin and asthenia in superficiality,which locates in spleen,has association with heart and liver, phlegm, qi and blood stasis is the disease of pathological product of pathological factors to promote the development of the disease.Kidney would be involved in it with long course of disease.The therapy could focus on tonifying spleen,nourishing heart and soothing liver,with methods of promoting the circulation of qi,removing blood stasis and reducing phlegm,meanwhile looking out to protect kidney.[Conclusion]Disorder of depression and pathogenesis of TCM of metabolic syndrome,to guide the clinical treatment of the disease and the theory of traditional Chinese medicine development has practical significance.
10.Recognition of environmental estrogen-like compounds based on fluorescent imaging analysis of ERα-EGFP nuclear granules formation

Objective To establish a quick method to probe the environmental estrogen-like compounds via fluorescent imaging analysis in high content analysis (HCA) technology.Methods HCA assays were performed to quantitatively in-vestigate the effect of collected environmental pollutants , including bisphenol A , nonylphenol , 1,2-phenylenediamine , 4-aminophenol, resorcinol, 3-aminophenol, 1,4-phenylenediamine and 2,5-diaminotoluene on nuclear granules formation ( Foci-formation) of estrogen receptor α( ERα)-EGFR( enhanced green fluorescent protein ) fusion-protein, which was se-lected as a research model in this study .The results were confirmed by the ERαtranscriptional activity by luciferase as-says.Results Compounds 1,2-phenylenediamine, 4-aminophenol, resorcinol, 3-aminophenol, 1,4-phenylenediamine and 2,5-diaminotoluene sulfate could not induce the ERα-EGFR nuclear granule formation.17-β-Estradiol, bisphenol A, or nonylphenol enhanced ERα-EGFR nuclear granule formation in a dose-dependent manner .The EC50 value was (4.17 ± 0.41) nmol/L, (1.48 ±1.79) μmol/L,or (3.70 ±0.78) μmol/L, respectively.The minimum detectable concentration was 1 nmol/L (17-β-estradiol)and 300 nmol/L (bisphenol A, nonylphenol).In luciferase tests, 17-β-estradiol, bisphe-nol A, or nonylphenol increased ERαtranscriptional activity in a dose-dependent manner ,and the EC50 value was (4.46 ±0.56) nmol/L, (2.31 ±0.21) μmol/L, or (6.60 ±0.94) μmol/L, respectively.The minimum detectable concentration was 3 nmol/L (17-β-estradiol), 300 nmol/L (bisphenol A),and 1 μmol/L (nonylphenol).Conclusion As an efficient method for ERαagonist identification , HCA assays based on the cell image phenotypes analysis can be used in quick recog -nition of environmental compounds with ER agonist-like activity.In all experimental compounds , only bisphenol A and non-ylphenol have a clear ER agonist-like activity .
