Objective To explore the significance of Survivin and PTEN in gastric cancer development and correlation between Survivin and PTEN.Methods Immunohistochemical methods were applied to detect the expres.sion of Survivin and PTEN in 105 pathological specimens of gastric cancer, in comparison to that in 98 negative gastric mucosa tissues. Results The positive rate of Survivin expression was 82.9% (87/105) in tissue specimens of gastric cancer and 6.1% (6/98) in tissue specimens of non-gastric cancer; The difference between them was statistically sig nificant (P <0.01).The positive rates of PTEN expression were 34.3% (36/105) in tissue specimens of gastric canc-er and 99.0% (97/98) in tissue specimens of non-gastric cancer; The difference between them was statistically signifi-cant ( P <0.01).The expression of Survivin and PTEN in tissues of gastric cancer had nothing to do with the gender of patients ( P > 0.05 ) , but was correlated with the degree of tumor cell differentiation, depth of invasion, lymph node me-tastasis, distant organ metastasis and TNM staging(P <0.05).Analysis by Spearman showed that Survivin was nega-tively correlated with PTEN( r = -0.23 ,P < 0.05 ).Conclusion The incidence of gastric cancer was related to the abnormally high level of Survivin expression and absence of PTEN expression; The expression of Survivin and PTEN in tissues of gastric cancer was unrelated with the gender of patients, but related to the degree of tumor cell differentia-tion, depth of invasion, lymph node metastasis, distant organ metastasis and TNM staging. The expression of Survivin was negatively correlated with the expression of PTEN.

Objective To explore the application and efficacy of laparoscopic radical operation for rectal cancer after preoperative short-time radiotherapy. Methods The clinical data of 108 patients with Dukes B and C rectal cancer were analyzed retrospectively.Thirty-five patients underwent laparoscopic radical operation after preoperative short-time radiotherapy, meanwhile 30 patients underwent laparoscopic radical operation, and 43 patients underwent open operation, both later groups without preoperative radiotherapy. Results There were not significant differences in preoperative general condition, tumor size and stage, pathological type, site of operation, and mode of operation between the 3 groups. But there was a higher rate of radical resection and sphincter preservation in the laparoscopic operation plus radiotherapy group than in the other groups(P

Objective To explore the locating, parameter measurement and 3D display of nucleus accumbens in human brain in terms of digital anatomy .Methods The raw data of the head specimen of a 45-year-old male adult with 0.5mm as the section spacing was collected by using digital milling machine .Three hundreds images of continual cross sections containing brain were chosen and the segmentation of the caudate nucleus , putamen and nucleus accumbens was accomplished with Photoshop CS .The nucleus accumbens on the images of continual coronal section reconstruction were distinguished according to Harvard Medical School ’ s segment method to calculate the volume of nucleus accumbens and collect the correlative location information .The caudate nucleus , putamen and nucleus accumbens were 3D visualize with the software of Amira 3.1.1.Results The nucleus accumbens , the adjoining structure and the lesion target of nucleus accumbens were all clearly visible .The left nucleus accumbens volume was 972.5mm3 , and the right was 830.6mm3 .The 3D coordinate value was the left ( -11.0, 24.4, 1.3) and the right (9.3, 23.9, 1.7).Conclusion The digital anatomy of nucleus accumbens can distinctly display the nucleus accumbens , form and confirm it ’ s volume, location and adjoining area , which is useful to clinician .

OBJECTIVETo detect the CDH1 gene methylation of suspension cells in intraoperative abdominal lavage fluid from colorectal cancer patients, and to examine its association with clinicopathology and prognosis.
METHODSReal-time methylation-specific polymerase chain reaction (qMSP) was used to investigate the methylation status of the CDH1 gene promoter 5'-CpG islands from intraoperative abdominal lavage fluid in 102 patients with colorectal cancer. The associations between methylation of CDH1 genes and clinicopathologic features and prognosis were investigated.
RESULTSAmong the 102 colorectal cancer patients, aberrant methylation of CDH1 gene was detected in 47 patients. Significant associations were found between CDH1 methylation status and tumor size, growth pattern, differentiation, distant metastasis, and clinical staging (all P<0.05). The median progression-free survival was 25.98 months for CDH1 methylation group and 41.36 months for non-methylated group, and the difference was statistically significant (P<0.01). Cox model analysis revealed that CDH1 methylation status in intraoperative peritoneal lavage fluid was an independent factor associated with postoperative survival in colorectal cancer patients (50.23% vs. 86.51%, P=0.001).
CONCLUSIONSColorectal cancer patients with aberrant methylation of 5'-CpG of CDH1 gene promoter of suspension cells in abdominal lavage have higher malignancy, more metastasis and worse prognosis.

Journal of Clinical Otorhinolaryngology Head and Neck Surgery 2013;27(19):1076-1079
OBJECTIVE:
To investigate the extracellular release of high mobility group box 1 (HMGB1) in laryngeal Hep-2 carcinoma cells induced by hypoxia and its possible mechanism.
METHOD:
The changes of HMGB1 concentration in the culture medium as well as HMGB1 protein and mRNA expression in Hep-2 cells were investigated after the cells were cultured with 1% O2 for different durations. Inhibitory effects of MAPK pathway inhibitors (PD98059. SP600125, and SB202190) and nuclear NF-kappaB pathway inhibitor (PDTC) with various concentrations on extracellular HMGB1 release were observed in hypoxia-induced Hep-2 cells. The HMGB1 concentration and HMGB1 protein expression were measured by enzyme-linked immunosorbent assay (ELISA) and western blot, respectively. The HMGB1 mRNA expression was determined by real-time quantitative PCR(RT-PCR).
RESULT:
The HMGB1 concentration in the culture medium and the HMGB1 protein expression in Hep-2 cells increased after the cells were subjected to hypoxia culture for 12 h in a time-dependent manner. The level of HMGB1 mRNA expression in Hep-2 cells increased after the cells were induced by hypoxia for 6h PD98059 and SP600125 with 20 micromol/ L and PDTC with 50 mg/L partly inhibited extracellular release of HMGB1 in hypoxia-cultured Hcp-2 cells.
CONCLUSION
Hypoxia induces laryngeal carcinoma cells to release HMGH1. which may be related to MAPK/NF-kappaB signaling pathway.
