2.Endoscopic nasobiliary drainage in the treatment of bile leakage after laparoscopic cholecystectomy

Objective To study the efficacy of endoscopic retrograde cholangio-pancreatography (ERCP) and endoscopic nasobiliary drainage (ENBD) in the diagnosis and treatment of bile leakage after laparoscopic cholecystectomy (LC).Methods The clinical data of 9 patients with bile leakage after LC were analyzed retrospectively.Results The procedure of ERCP was successful in all 9 patients.Bile leakage was treated successfully by ENBD in 8 patients with no further complication,and 1 patient was treated surgically because of biliary ductal injury.Conclusions ERCP is recommended as a safe and efficacious option to detect and treat bile leakage after LC.ENBD combined with percutaneous drainage of abdominal collections can cure bile leakage after LC.
3.Electromyographic Analysis of Muscle Activities of Knee Joint in Patients with Patellofemoral Pain Syndrome

Objective To analyze electromyographic (EMG) activities of vastus medialis obliquus (VMO) and vastus lateralis (VL) in patients with patellofemoral pain syndrome (PFPS) before and after an exercise program.Methods 26 subjects with PFPS were randomly divided into the EMG biofeedback plus exercise group (group A) and exercise only group (group B) with 13 cases in each group. All patients in two groups were trained with family exercise program, but the patients of the group A used a EMG biofeedback while training. The relative activities of VMO and VL of all patients in two groups were assessed with the EMG apparatus for a continuous 6 hours period before and 8 weeks after training. At the same time the intensity of the knee pain was also assessed.Results There was no statistics difference in VMO/VL EMG ratio of the group B ( P>0.05), whereas the group A had significantly higher VMO/VL EMG ratio ( P<0.05).Conclusion The EMG biofeedback apparatus used in home exercise program of PFPS patients can improve the recruitment of VMO.
4.Effects of long-term sleep deprivation on mitochondria stress in locus coeruleus and the tyrosine hydroxylasic projection in mice.

OBJECTIVETo observe the changes of mitochondria stress in locus coeruleus and the tyrosine hydroxylasic projection after long-term sleep deprivation.
METHODSSleep deprivation mice model was set up by employing "novel environments" method. The expression of NAD -dependent deacetylase Sirtuin type 3 (SIRT3), which regulates mitochondrial energy production and oxidative stress, and heat shock protein 60 (HSP60), a major biomarker of mitochondrial stress, and the tyrosine hydroxylasic projection from locus coeruleus were analyzed after a 5-day sleep deprivation.
RESULTSCompared to the control group, the expression of SIRT3 in locus coeruleus was significantly decreased in respouse to long-term sleep deprivation, while the expression of HSP60 was significantly increased. In addition, relative to control group, pereentage area of the tyrosine hydroxylasic projection to anterior cingulate cortex was substantial decreased in long-term sleep deprivation group.
CONCLUSIONLong-term sleep deprivation induced the decreased level of SIRT3 expression and the elevation of mitochondrial stress in locus coenileus, which may further lead to the loss of tyrosine hydroxylasic projection in mice.

5.The flap combining peroneus iongus muscle with nutrient vessels of nerve repairing the chronic achilles tendon rupture with the skin defect

Objective To explore the treatment methods and clinical result with the flap combining peroneus longus muscle supplying with the nutrient vessels of sural nerve to repaire the chronic achilles tendon rupture with skin defect.Methods After anatomic investigating,Analyzing 6 cases.Excising peroneus lon- gus muscle combining with the foot external flap,supplying with the pediele of the nutrient vessels of sural nerve to repaire the chronic achilles tendon rupture with skin defect,among them,the size of flap was about 6.0 cm?5.5 cm~16.5 cm?11.0 cm,the defect length of achilles tendon 2.0~7.5 cm.Results All ca- ses observed 4 to 16 months,all eases skin and achilles tendon were survived completely,the flap skin appear- ence was good,none ulcer.1 cases part necrosis,1 eases rerupture,Both healed after changed dressings.Ac- cording to Arner-Lindholm criteria to test the efficacy the result were excellent in 3 patients,good in 2,poor in 1.Conclusion It is an ideal way using the flap combining peroneus logus muscle with the nutrient vessels of sural nerve repairing the chronic achilles tendon rupture with skin defect.The advantage is operting conven- iently,transferred adjaeently,repairing the skin and tendon defect simultaneously,healing fastly and anti-bac- teria strongly,biomeehanies property and function is similar to the heel,it can fit the heel and skin for repai- ring.

7.Long-term efficacy of surgical treatment of slow transit constipation with pelvic floor hernia and rectal mucosal prolapse

Objective To evaluate the long-term therapeutic efficacy of subtotal colectomy,ascending colon-rectum anastomosis, pelvic floor hernia repair, functional rectal suspension, and uterine suspension surgery for slow transit constipation with pelvic floor hernia and rectal mucosal prolapse.Methods From June 2007 to May 2008, 35 patients with intractable constipation caused by slow colonic transit combined with pelvic floor hernia and rectal mucosal prolapse underwent subtotal colectomy and ascending colon-rectum anastomosis, pelvic floor hernia repair, functional rectal suspension, and uterine suspension surgery. Postoperative defecation, complications, quality of life, and degree of satisfaction were followed-up. Results The average follow-up period was two years. At one month after the operation, the average defecation frequency was five times (2 -8 times) a day, with a semi-liquid stool consistency; After two years the frequency was twice ( 1 -3 times) a day, with solid stool consistency. Of the 35 patients, 19 were satisfied with the surgical efficacy, and 16 were very satisfied. All the patients' quality of life improved significantly. Conclusions For patients suffering from slow transit constipation with pelvic floor hernia and rectal mucosal prolapse subtotal colectomy, ascending colon-rectum anastomosis, pelvic floor hernia repair,functional rectal suspension, and uterine suspension surgery has satisfactory results.
8.Preparation and bioactivity assay of mIL-4-SA fusion protein

Purpose To prepare streptavidin-tagged mouse interleukin-4(mIL-4-SA)bifunctional fusion protein and to study on its bioactivity.Methods The mIL-4 gene was cloned by RT-PCR and cloned into pET21 vector to get mIL-4-SA-pET21 expression plasmid.The mIL-4-SA fusion protein was expressed in BL21 (DE3)host bacteria and purified through the Ni-NTA affinity chromatography and refolded by dilution and dialysis.The effect of mIL-4-SA fusion protein on mouse thymocytes proliferation was evaluated by MTY.Flow cytometric analysis was performed to detect the mIL-4-SA fusion protein on the biotinylated B16F10 tumor cells.Results The mIL-4-SA-pET21 vector was successful by constructed and the mIL-4-SA fusion protein was expressed in BL21(DE3)at about 35%of total bacterial proteins.The purity of mIL-4-SA Was about 95% through Ni-NTA.The mIL-4-SA fusion protein exhibited bifunctional activities,i.e.,stimulative effect for mouse thymocyte proliferation and SA-mediated high-affinity binding to biotinylated cell surfaces(anchoring modified rate Was about 96.69%).Conclusion The mIL-4-SA fusion protein was expected to be developed for the treatment of tumors.
