Objective To observe the effect and study intervenient action of Kuijieling Granules (KG) in treating ulcerative colitis (UC) during the active period by the clinical experiment. Methods The patients of active UC (damp-heat syndrome) were randomly divided into 2 groups (KG+SASP group and SASP group) to observe pathological changes of the mucosa,syndrome effect and main symptoms scores,and the protein expression of TLR4,CD14 and NF-?B p65 were detected by immunohistochemistry. Results KG+SASP group had better effect than SASP group in syndrome and mucosa (P0.05). There were outstanding differences among the pathologic grade of UC about IA of TLR4,CD14 and NF-?B p65. Objective To observe the change of gastro-intestinal hormone levels about syndrome type of stagnation of liver qi, deficiency of spleen and stomach, liver qi stagnation and spleen qi deficiency in patients with irritable bowel syndrome (IBS), and to probe into the correlation between TCM syndrome type and gastro-intestinal hormone levels. Objective To establish the model of VX2 soft tissue xenograft in rabbit, and choose the best time for PET-CT imaging. To analyze the result of the imaging with semi-quantitative, and evaluate the feasibility of the study with VX2 soft-tissue tumor model in rabbit for PET-CT imaging. Methods 20 Japanese white rabbits were successfully implanted with VX2 tumors in thighs by the injection of tissue mass suspension.Then they were scanned with PET-CT after the diameter of the tumor is >2 cm. Results The diameter of the tumor>2 cm at 14 d post implant. The average volume of tumor was (114.57±9.87) cm3.The average SUVmax of tumor's and normal tissue's in the other thigh was 5.85±0.92 and 0.22±0.09 (t=27.03, P<0.01). SUVmax was not associated with tumor volume (r=0.02, P=0.93).Conclusion The imaging of VX2 soft tissue xenograft is clear by clinic PET-CT, and it is suitable for the study of PET-CT.PET-CT imaging performed at 14 d post implantation had the highest imaging quality.
5.Use creatinine reduction ratio to predict the graft function recovery after kidney transplantation

Objeetive To discuss the correlation of creatinine reduction ratio(CRR2)from posttransplant day 1 to day 2 and early graft function recovery status after kidney transplantation. Methods Clinical data of 80 patients after renal transplantation from Jan 2005 to Mar 2007 were retrospectively analyzed.Patients were divided into three groups according to the post-operative serum creatinine level:53 patients within IGF group[cereatinine<265.2 μmol/L by post-operative day(POD)no.5],14 patients within SGF group(creatinine>265.2 μmol/L on POD no.5,but no need for dialysis),and 13 patients within DGF group(need for dialysis in the first week post-transplant).Then the value and 99%CI of CRRz of these three groups were calculated. Results The value of CRR2 of IGF,SGF and DGF was(46.8±14.6)%,(25.6±13.5)%and(0.7±17.7)%respectively.And CRR2 99%CI of IGF,SGF and DGF was 41%-52%,15%-36%and-14%-16 0A respectively.There was significant difference in the value of CRR2 among IGF,SGF and DGF group.So a criteria for early diagnosis of IGF,SGF and DGF by CRR2 99%C1 was established:IGF(CRR2≥40%),SGF (15%<CRR2<40%)and DGF(CRR2≤15%). Conclusion CRR2 has a good correlation with early graft function recovery after kidney transplantation,and can be used to predict the occurrence of SGF and DGF.
6.GAT-1 and GAD65 mRNA expressions in different brain regions in dogs at brain propofol uptake equilibrium.

OBJECTIVETo investigate the expressions of gamma aminobutyric acid transporter 1 (GAT-1) and glutamate decarboxylase 65 (GAD65) mRNA in different brain regions at brain propofol uptake equilibrium in dogs.
METHODSEighteen 12- to 18-month-old healthy hybrid dogs were randomized equally into control group (group C), low dose group (group L), and high dose group (group H). In groups L and H, anesthesia was administered by intravenous injection of 5.5 and 7.0 mg/kg propofol followed by propofol infusion at a constant rate of 55 and 70 mg·kg(-1)·h(-1) for 50 min, respectively. Blood samples were taken from the internal carotid artery and jugular vein to measure plasma propofol concentrations, and the brain tissues of the hypothalamus, sub thalamus, dorsal thalamus, hippocampus, pons, parietal lobe and frontal lobe were examined for GAT-1 and GAD65 mRNA expressions using quantitative real-time PCR.
RESULTSIn groups L and H, propofol infusion at a constant rate for 50 min resulted in comparable plasma propofol concentrations between the internal carotid artery and jugular vein (P>0.05), but the concentrations differed significantly between the two groups (P<0.01). GAT-1 mRNA levels in the hypothalamus and hippocampus were significantly higher in groups L and H than in group C (P<0.05 and P<0.01), but comparable between the former two groups. The variations of GAT-1 mRNA levels between the hypothalamus and hippocampus were similar in both group L [(61.26∓7.17)% and (79.34∓39.95)%, P>0.05] and group H [(74.64∓19.63)% and (97.12∓32.31)%, P>0.05]. GAT-1 mRNA levels in other brain regions showed no significant difference among the 3 groups. GAD65 mRNA levels were similar between group L and group H, but both significantly higher than that in group C (P<0.01). GAD65 mRNA in other brain regions had no significant difference among the 3 groups.
CONCLUSIONGAT-1 mRNA in the hypothalamus and hippocampus and GAD65 mRNA in the dorsal thalamus are upregulated when propofol uptake reaches an equilibrium in the brain of dogs.

7.Application of QCC activities in the efficiency improvements of the inventory management of oral tablets in ward pharmacy

Objective To enhance oral tablets ward pharmacy inventory efficiency and improve the comprehensive quality of the pharmacist by QCC activities .Method Main factors to extend the ward pharmacy tablet counting time were analyzed through collecting the previous ward pharmacy tablet count time and account matching rate .According to the main problems , the strategies were found out to be implemented ,identify and evaluate the implementation result .Result In view of the oral medicine ward pharmacy inventory of the reasons for the low efficiency ,following measures should be taken :reducing oral drug dispensing station varieties ,finishing cargo ,strengthening personnel training ,changing inventory counting method and optimi-zation of inventory materials and re-enacted drugs location code and so on .Ward pharmacy tablet counting time reduce from 6 .5 hours to 3 hours (reducing by 53 .85% ) .Conclusion The QCC activities could significantly improve the oral medicine ward pharmacy inventory efficiency and comprehensive quality of pharmacists .
8.Comparison of platelet rich fibrin and acellular dermal matrix in repairing rabbits′ oral mucosal defects

Objective:
To investigate the effects of platelet-rich fibrin (PRF) and acellular dermal matrix (ADM) on the repair of oral mucosal defects and to provide the basis for soft tissue growth in oral implant operations.
Methods:
Thirty-six healthy male Japanese big ear rabbits were randomly divided into the PRF group, ADM group, Autograft group (autologous connective tissue transplantation group) and Control group (blank control group); each group contained nine rabbits. Between the midline and the hard palate maxillary incisors, in an 8-mm location preparation and a 10-mm standard mucosa defect, the ADM group, PRF and Autograft group were implanted with ADM, autologous PRF and autologous cornification mucosa, respectively, whereas the control group had wound gauze compression processing at 7, 14, and 21 days to determine the wound healing rate in the area selected by HE staining. The inflammatory grade and average epithelial thickness were observed, and the results were statistically analyzed.
Results :
Compared with the control group, the PRF, ADM and Autograft groups had significantly advanced wound healing (P < 0.05). The wound healing degree in the PRF group was similar to that of the ADM group at all time points (P > 0.05). The wound healing degree in the PRF and ADM groups was lower than that of the Autograft group at each time point (P < 0.05). HE staining results showed that compared with the control group, the levels of inflammation in the PRF group, ADM group and Autograft group were reduced, and the difference was statistically significant (P < 0.05). Nevertheless, there was no significant difference between the PRF, ADM and Autograft groups (P > 0.05). The epithelial thickness in the ADM group was similar to that in the Autograft group (P > 0.05). The epithelial thickness in the ADM group was higher than that in the PRF group at 7 d and 14 d (P < 0.05), but there was no significant difference at 21 d (P > 0.05).
Conclusion
PRF and ADM have similar healing effects in repairing oral mucosa defects, and they can be used as soft tissue augmentation materials instead of connective tissue transplantation.
9.Improved primary culture and primary identification of human pituitary adenoma cells

Objective To explore an improving primary culture method of human pituitary adenoma cells.Methods Thirty-six pituitary adenoma specimens,collected from excision and conformed by pathology in our hospital from November 2014 to June 2015,were used.Based on the former experience from papers and ourselves,our primary culture methods of several different kinds of human pituitary adenomas were improved.Purification and passage culture of cells were performed in the following experiments.CCK-8 assay was used to detect the multiplication capacity of pituitary adenoma cells;immunofluorescent staining and Western blotting were used to observe the cytokeratin and vimentin protein expressions.Results By using the improve culture methods,tumor cells proliferated in suspension with good status and had the tendency of aggregation in culture medium.The proliferation test showed that the tumor cells enjoyed proliferation ability in vitro;tumor cells could pass over five generations.The growth curve of pituitary adenoma cells showed S pattern.Immunofluorescent staining and Western blotting indicated negative cytokeratin and vimentin protein expressions,showing that the cells were from epithelium.Conclusion Each type of pituitary adenoma cells has characteristics of suspended growth and tendency of gathering by using this improved method.
10.Efficacy of endolymphatic sac-mastoid shunt surgery for intractable Meniere's disease.

OBJECTIVETo investigate the effect and mechanism of endolymphatic sac-mastoid shunt surgery for intractable Meniere's disease of different stages according to hearing level.
METHODSData from 240 patients diagnosed with unilateral MD strictly meeting the criteria issued by Chinese Academy of Otolaryngology-Head and Neck Surgery Committee (2006) from January 1983 to January 2012 were analyzed in this study. Endolymphatic sac-mastoid shunt surgery was performed in affected ear for each patient. The evaluation of therapy followed the guidelines issued by Chinese Academy of Otolaryngology Head and Neck Surgery Committe. Vertigo control and auditory function were measured in two-year's follow-up.
RESULTSAccording to the preoperative staging of hearing, among these 240 patients, there were 12 cases in stage I (with an average hearing threshold < 25 dB), 130 cases in stage II (with an average hearing threshold of 25-40 dB), 85 in stage III (41-70 dB) and 13 cases in stage IV(with an average hearing threshold > 70 dB). Overall control rate of vertigo was 77.9% (187/240) in two-year's follow-up, with total control 49.2% (118/240) and substantial control 28.7% (69/240). The hearing was improved in 25.0% (60/240) of cases, no change in 59.2% (142/240) of cases, and worse in 15.8% (38/240) of cases. According to different stages, vertigo control rate was 83.3% (10/12) in stage I, 82.3% (107/130) in stage II, 75.3% (64/85) in stage III and 46.2% (6/13) in stage IV. Vertigo control rate of stage IV patients was significantly lower than that of stage II and III patients (χ(2) = 9.318 and 4.692, P < 0.05), while vertigo control rate of stage I, II, III patients had no significant difference with each other (P > 0.05).
CONCLUSIONEndolymphatic sac-mastoid shunt operation is an effective method in the treatment of intractable Meniere's disease, but the effect was poor in stage IV patients.
