OBJECTIVE:To establish the quality standard for Qiwei kangti powders.METHODS:Radix Astragali,Angelica sinensis,Salvia miltiorrhiza were identified by TLC.The content of tanshinone Ⅱ A(the chief content in Salvia milti-orrhiza) was determined by HPLC.RESULTS:The TLC spots developed were fairly specific.Tanshinone Ⅱ A showed a good linear relationship at a range of 5.44~32.64 ?g?mL-1(r=0.999 8),and its average recovery was 99.54%(RSD=0.955%,n=9).CONCLUSION:The established quality standard is applicable for the quality control of Qiwei kangti powders.
2.Advances in research on animal models of chronic rejection after lung transplantation

The most important cause of late mortality after lung transplantation is obliterative bronchiolitis ( OB) . It is clear that a good animal model is indispensable to further unravel and clarify the pathogenesis of bronchiolitis obliterans syndrome ( BOS) .Many animal models have been developed to study BOS, however, so far, none of these models truly mimics the human condition.In recent years mouse models of orthotopic lung transplantation have been established, which provide potential possibilities for further studies of OB/BOS after lung transplantation.The aim of this article was to review the pros and cons of those animal models, and discuss the possible approaches to establish animal models of chronic rejec-tion after lung transplantation.
3.Study on application effect of paclitaxel combined with carboplatin neoadjuvant chemotherapy in patients with advanced cervical carcinoma

Objective To explore the application effect of paclitaxel combined with carboplatin neoadjuvant chemotherapy in patients with advanced cervical carcinoma.Methods Forty patients with stage Ⅱa and above cervical cancer who were treated between August 2015 and August 2016 were selected as the research objectives and all patients were diagnosed by pathological examination.They were divided randomly into experimental group(n=20)and control group(n=20).Patients in the experimental group were treated with intravenous chemotherapy before operation.This scheme included paclitaxel and carboplatin.It included 2 cycles,and the interval between the 2 cycles was 3 weeks.Extensive hysterectomy and pelvic lymphadenectomy were made in 14 days after chemotherapy.The patients in the control group were treated only with extensive hysterectomy and pelvic lymphadenectomy.The efficacy of neoadjuvant chemotherapy was evaluated according to WHO criteria,and observed the operation condition.Adverse drug reactions were evaluated according to WHO anti-cancer drug adverse reaction grade.The quality of life of the two groups was evaluated by the mos item short from health survey(SF-36).Results 20 patients completed chemotherapy,CR5 cases,PR11 cases,SD3 cases,PD1 cases,and the total effective rate(CR+PR)was 80%.At the same time,The paracervical tissue becomed soft in varying degrees,the gap widened,the invasive lesions were controlled in different degrees.Adverse reactions were mild and relieved without special treatment.There was no kidney and heart function damage.All cases were operated successfully after chemotherapy.There were no significant difference in operation time and blood loss between the two groups.The rate of lymph node metastasis,the infiltration rate of uterine tissue and the residual rate of tumor tissue in the vaginal margin were lower than those in the control group(P<0.01).The SF-36 scores of the two groups were higher than those before treatment,but the score of the experimental group was higher significantly than that of the control group,and the difference was statistically significant(P<0.01).Conclusion Paclitaxel combined with carboplatin in neoadjuvant chemotherapy of cervical cancer can reduce the tumor volume,increase the chance of surgery,has less toxic side effects,lower metastatic rate and improve the quality of life of patients.It is worthy of further clinical research and recommendation.
4.Determination of paeoniflorin,paeonol and ?-cyperone in Baixiangdan Capsule by HPLC

AIM: To establish the analytical methods for determing paeoniflorin,paeonol and ?-cyperone in Baixiangdan Capsule(extract of Radix Paeoniae alba,extract of paeonol,volatile oil of Rhizoma Cyperi). METHODS: The contents of paeoniflorin,paeonol and ?-cyperone in the capsule were determined by HPLC respectively. RESULTS: The linear range of paeoniflorin was within 0.6125.508 ?g(r=(0.999 3)) and the average recovery was 98.16%(RSD=1.18%,n=5).The linear range of paeonol was 0.150 4 ?g-1.353 6 ?g(r=(0.999 8)) and the average recovery was 98.16%(RSD=1.52%,n=5).The linear range of ?-cyperone was(0.271)-2.439 ?g(r=(0.999 9)) and the average recovery was 98.02%(RSD=1.58%,n=5). CONCLUSION: The method is reliable,accurate,feasible and reproducible and can be applied to the quality control of Baixiangdan Capsule.

The 2016 conference on iodine and thyroid disease is aimed to establish a platform for sharing the achievement on fundamental,preventive and clinical research of iodine and thyroid disease,for exchanging information between producer and consumer on iodized salt,iodine additive and iodine determination,for communication on policy and measures concerning iodine deficiency disorders (IDD) prevention and control,for promoting the exchange of experience between international and domestic scholars in IDD prevention and control and the clinical,fundamental research update and achievement,and for discussing the key and difficult problems.After communication and discussion,the participants at the 2016 conference drafted a consensus on iodine and thyroid disease.Through incorporating the current IDD prevention and control schedule and task,this article describes,elaborates and explains the key items of the consensus with expectation of a better utilization and application of the consensus in the future in IDD prevention and control.
6.A novel rat model of atherosclerosis induced by inflammation and the therapeutic effect of Rb1

Objective To establish a novel atherosclerosis model by inflammation in rats and investigate the anti-atherosclerotic effect of Rb1.Methods Healthy male SD rats were randomly divided into three groups, namely the control group, model group (using zymosan A to induce inflammation) and Rb1-treated group (12 rats in each group).The rats were administered liquid paraffin (i.p.), zymosan A (20 mg/kg, i.p., once every 4 days) or zymosan A and Rb1 (40 mg/kg, i.p., once daily), respectively.All animals were fed a high-fat diet for 10 weeks.At scheduled time points, pathological changes in the aorta were observed using Sudan IV staining and transmission electron microscopy.White blood cell count was used to assess the inflammation.The expression of NFκB, TNFα, IL6 was evaluated by real time PCR, im-munohistochemistry and ELISA, respectively.Results Typical atherosclerotic changes such as fatty streaks, plaque, foam cells in the rats following zymosan A induction were alleviated by Rb1 treatment.In the Rb1-treated group, there was a markedly decreased expression of NFκB, TNFα, and IL6.Conclusion The model of atherosclerosis can be established by inflammation based on high-fat diet in rats.Rb1 inhibits atherosclerosis through anti-inflammatory effect.
7.Blockade of CLC-3 chloride channel inhibited the viability and invasion of colorectal cancer cells

Objective:To examine the expression of CLC-3 in colorectal tissues and the effect of CLC-3 on the viability and invasion of colorectal cancer (CRC) SW480 and SW620 cells. Methods:The mRNA levels of CLC-3 in CRC cell lines were determined by RT-PCR. CLC-3 expression was inhibited by adding DIDS or NPPB to the CRC cells. Subsequently, cell viability and invasion were assessed by CCK-8 assay and Transwell assay, respectively. In addition, the effects of DIDS and NPPB on the Wnt orβ-catenin signaling pathways were de-termined by Western blot analysis. Results:The mRNA level of CLC-3 was remarkably increased in the CRC tissues compared with that in normal colorectal tissues (P<0.05) and was positively correlated with the T stage of CRC. The blockade of CLC-3 inhibited the viability and invasion of CRC cells (P<0.05). The expression ofβ-catenin, C-myc, cyclin D1, Ki-67, and survivin were evidently reduced by the in-hibition of CLC-3 (P<0.05). Conclusion:The inhibition of CLC-3 decreases the cell viability and invasion of CRC cells by reducing the ex-pression of the proteins related to the Wnt orβ-catenin signaling pathway.
8.The effects of compound of paeonol and PNS on expressions of collagenⅠandⅢprotein and mRNA in rat model of acute myocardial infarction

Objective To study the effect of paeonol (PAE) and panax notoginseny saponins (PNS) on the expressions of collagenⅠandⅢprotein and mRNA in rats with acute myocardial infarction (AMI), and to explore the molecular mecha?nism of improving myocardial fibrosis. Methods The rat model of AMI was made using the left anterior descending coro?nary branch ligation.According to the intervention rats were divided into model group, PAE (8 mg/kg) group, PNS (40 mg/kg) group, PAE (4 mg/kg)+PNS (20 mg/kg) group, PAE (8 mg/kg)+PNS (40 mg/kg) group and captopril positive control group (10 mg/kg). Sham operation group, only wear line without ligation. The left ventricular mass index (LVMI) was detected after treatment for 28 d. Masson staining was used to observe changes of myocardial fibrosis. Western blot assay and RT-PCR technique were used to detect protein and mRNA expression levels of collagenⅠandⅢ. Results The values of LVMI were increased in model group compared with those of sham operation group and treatment groups. Compared with PAE group and PNS group, values of LVMI were significantly decreased in PAE (4 mg/kg)+PNS (20 mg/kg) group and PAE (8 mg/kg)+PNS (40 mg/kg) group. There was a more significant decrease in formula high dose group (P < 0.01). The model group showed pathological change. There were different degrees of improvement in pathological structure in all treatment groups, more sig?nificant improvement was found in formula low dose group, formula high dose group and captopril positive control group. There were different degrees of increase in expressions of collagenⅠandⅢprotein and mRNA in model group compared with those of sham operation group and treatment groups. Compared with PAE group and PNS group, the protein and mRNA expression levels of collagenⅠandⅢwere significantly decreased in formula low dose group and formula high dose group,more significant decreased was found in formula high dose group (P<0.05). Conclusion Compound of paeonol and PNS can improve myocardial fibrosis in myocardial infarction rats, which may be related with reduced expressions of collagenⅠandⅢprotein and mRNA.
9.The role of serum procalcitonin in etiology diagnosis of sepsis in children

Objective To investigate the serum procalcitonin (PCT) levels in sepsis caused by the bacteria,virus and mycoplasma and explore the role of PCT in etiology diagnosis of sepsis in children.Methods Three hundreds and thirty critically ill children with sepsis caused by bacteria,virus and mycoplasma admitted in PICU of Hunan Children' s Hospital from Feb 1,2011 to Sep 1,2012 were reviewed and analyzed.The PCT levels were measured at admission and day 3.The differences in accidence of sepsis caused by bacteria,viruses and mycoplasma according to different serum PCT levels were analyzed.The differences of PCT levels at admission and day 3 in sepsic children caused by bacteria,viruses and mycoplasma were analyzed.Results The level of serum PCT in sepsis caused by bacterial infection were distinctly increased,caused by virus and mycoplasma infections was not obvious but the increases of serum PCT [0.71 (8.14)ng/ml,0.15 (1.68) ng/ml,0.28 (1.89) ng/ml].According to various PCT levels(0.05 ~ ng/ml,0.5 ~ng/ml,2 ~ ng/ml,10 ~ 300 ng/ml),the differences of accidence of sepsis caused by bacteria,virus and mycoplasma were also statistically significant(x2 =84.50,P < 0.01).The PCT level of septic children caused by bacterial infection in day 3 was significantly decreased compared with that at admission [0.32 (5.68) ng/ml vs 0.71 (8.14) ng/ml] (U =19.34,P <0.05).Conclusion PCT plays a certain role in etiology diagnosis of sepsis in children.The increased PCT levels which can be reduced by anti-inflammatory treatment indicate the likelihood of bacterial infection and sepsis.The increase of PCT induced by viral and mycoplasma infections is not obvious,but bacterial infection can not be completely ruled out.
10.Antibody array analysis of atherosclerosis cytokine levels in rats

Objective To investigate the changes of the cytokines on the aortas of atherosclerosis (AS) rats. Methods Zymosan-liquid medical paraffin suspension (contain Zymosan 20 mg/kg) was injected intra- peritoneally once a week for 10 times and given intraperitoneal injections of ovalbumin, 2.5 mg/kg, once a week for 7 times after initial subcutaneous sensitization. All animals were fed cholesterol-rich diet to induce AS. Electron microscopy was used to observe the foam cells, and RayBio Rat Cytokine Antibody Array 1.1 was employed to detect the cytokines in the aorta wall. Results Electron microscopy showed all models had monocytes migration from lumen to subendothelium in the aorta and formed to foam cell for phagocytosis lipid, indicating typical characteristic of AS in the rats. Most pro-inflammatory cytokines were increased except Fractalkine and IL-6, but anti-inflammatory cytokines were all slightly decreased. Conclusion Inflammation and immune can induce the formation of foam cells. The balance of pro-inflammatory cytokines and anti-inflammatory cytokines is broken is the important reasons of atherogenesis。
