BACKGROUND: The aim of our study was to optimize and establish erythropoietin (EPO) enzyme linked immunosorbent assay (ELISA) system. METHODS: We prepared several monoclonal and polyclonal antibodies specific to human-EPO. The best combinations of antibodies for coating and detecting antibodies were selected for the establishment of ELISA. We tested several methods such as a competitive EIA and a sandwich ELISA. RESULTS: The best sandwich ELISA was optimized compared to competitive EIA when purified polyclonal antibody (PoAb) was used as a coating antibody and biotinylated PoAb as a detecting antibody. This sandwich ELISA easily detected EPO when PoAb pairs were used compared to the ELISA using monoclonal antibody and PoAb. There were no significant differences between the effects of various blocking solutions on the performance of sandwich ELISA using biotinylated antibody. The ELISA system using PBST containing 3% BSA as a blocking solution can sensitively detect EPO (10 mU/mL) in a broad range of EPO concentrations (10-2,000 mU/mL) and there were cross-reactions with other cytokines). CONCLUSIONS: EPO can be easily determined by using biotinylated PoAb as a detecting antibody and another PoAb as a coating antibody.

9.Study on immune response of antibodies IgA1 and IgG fron sera of hepatoma and acute leukaemia patients by lectin-ELISA and lectin-protein A

Healthy human serum IgA, purified lectin-sepharose 4B column affinity chromatography was assessed by sodium dodecyl sulfate polyacrylamide electrophresis. The lectin from wild Jackfruit reacted specifically to IgA1 from human serum. This lectin used as antigen in the technique of ELISA-Lectin to determine the concentration of IgA1 from healthy and pathological human sera. Using protein A from Staphylococcus aureus isolated from Vietnamese infectious patients to capture IgG average concentration of IgA1 from healthy human serum as 14.37 mg/ml. No change of IgG concentration from patients sera (hepatoma and leukemia) was obtained in comparison with healthy subjects

10.Quick diagnosis of japanese encephalitis by MAC-ELISA technique

On 138 serum samples taken in the first 7 days of acute syndrome of brain, by MAC-ELISA technique 95 samples were determined as positive, 68.84% was positive. 43 samples taken in the first 7 days of the disease there was no detected antivirus IgM antibody but this could not concluded as negative, but may be a late antibody response or may be another cause of acute brain syndrome. For assuring the accuracy of the diagnosis, the samples taken in the first 7 days of the disease must be taken again after 7 day to verify the results.
