6.Serum bisphenol-A concentrations from uterine leiomyoma patients.

OBJECTIVE: To investigate how many leiomyoma patients are exposed to bisphenol-A (BPA) and whether the serum concentration of BPA is related to leiomyoma growth. METHODS: Total 131 patients were recruited for measuring BPA. Initially, leiomyoma patients were divided into three groups, mild (n=38), moderate (n=33), and severe (n=30) according to the size of the leiomyomas. The control (n=30) group was defined as having no leiomyomas. The identification and diameter measurements of leiomyomas was performed by transvaginal ultrasonography. Serum BPA concentrations were measured by enzyme linked immunosorbent assay. RESULTS: BPA was detected in 83.9% out of 131 samples totally, and 83.1% out of 101 leiomyoma patients. In detail, the detection rates of serum BPA were 86.7% in control group, 71.1% in mild group, 84.9% in moderate group, and 96.7% in severe group. The mean BPA concentrations in the control group was 0.557+/-0.086 ng/mL and those in the leiomyoma groups were 0.273+/-0.052 ng/mL (mild), 0.336+/-0.063 ng/mL (moderate), and 0.636+/-0.075 ng/mL (severe) (P=0.0003). Values are mean+/-standard error. Conclusions: The detection rate of serum BPA in control and leiomyoma groups were 86.7% and 83.1% respectively. However, there was no statistical significance of serum BPA concentrations between control and leiomyoma groups. To verify the effect of BPA on the leiomyoma growth, close and sequential monitoring for the person who have exposure risk is recommended.

BACKGROUND: The aim of our study was to optimize and establish erythropoietin (EPO) enzyme linked immunosorbent assay (ELISA) system. METHODS: We prepared several monoclonal and polyclonal antibodies specific to human-EPO. The best combinations of antibodies for coating and detecting antibodies were selected for the establishment of ELISA. We tested several methods such as a competitive EIA and a sandwich ELISA. RESULTS: The best sandwich ELISA was optimized compared to competitive EIA when purified polyclonal antibody (PoAb) was used as a coating antibody and biotinylated PoAb as a detecting antibody. This sandwich ELISA easily detected EPO when PoAb pairs were used compared to the ELISA using monoclonal antibody and PoAb. There were no significant differences between the effects of various blocking solutions on the performance of sandwich ELISA using biotinylated antibody. The ELISA system using PBST containing 3% BSA as a blocking solution can sensitively detect EPO (10 mU/mL) in a broad range of EPO concentrations (10-2,000 mU/mL) and there were cross-reactions with other cytokines). CONCLUSIONS: EPO can be easily determined by using biotinylated PoAb as a detecting antibody and another PoAb as a coating antibody.

8.Preliminary study on protein A in some species of Staphylococcus aureus isolated in Vietnam

the ability to biosynthesis of protein A from some Staphylococcus aureus strains isolated from the patients of Bach Mai Hospital was studied in the years 2001-2002. The results showed that there were 3 strains (25, 28 and 55) have the ability to synthesize protein A in the highest levels among 8 strains investigated. Using ELISA technique with protein A served as an antigen from Staphylococcus strain 25 to analyse IgG from sera of 32 healthy subjects has determined that the average IgG level from human serum is 14.37mg/ml. This result was supported by the same technique using commercial foreign bio-kit. It is proven that protein A preparation can be used as an antigen in bio-kit capturing specifically IgG from human serum in clinical diagnosis and in immuno-pathological research using ELISA technique

9.Serological study of Hantavirus in worker of Hai Phong port at 2003

In the year 2003, for Hantaan virus, a serological surveillance ELISA method was performed on 155 serum samples collected from the workers of Haiphong port.26/155 samples were positive or in suspect with rodent sera. The samples submitted to 2nd examen by indirect fluoresceine immuno-assay showed that 3/26 samples were positive 3 positive samples were undertaken further studies by WB technique with specific SEO antigenes ,which confirmed anti-hantaan virus (IgG) and nuclecapsid protein (NP) of51 kD weigt. Results showed that Hantaan virus was found among Haiphong port workers
