Objective To evaluate early changes of femoral artery elasticity in type 2 diabetes mellitus (T2DM) combined with microalbuminuria (MA) patients using ultrasound radio-frequency technology, and to study the clinical significance. Methods 51 T2DM without MA patients were enrolled in Group A,and 53 T2DM with MA patients were enrolled in Group B.In order to observe the early elastic changes in the subjects,two-dimensional ultrasound was used to eliminate patients whose intima-media thickness (IMT) was >1 mm.The femoral arteries were imaged by two-dimensional ultrasonography, and then the IMT,femoral artery inner diameter (D),distensibility coefficient (DC),compliance coefficient (CC),elasticity coefficient (α and β),and pulse wave velocity (PWV) were real-time tested using quality intima-media thickness (QIMT) and quality arterial stiffness (QAS) analyzing systems.Besides, the differences and correlation analyses of these parameters between the two groups were conducted. In addition, some other parameters of sujects in both group A and B were also determined and analysed, including total cholesterol (TC),triglyceride (TG),low-density lipoprotein cholesterol (LDL-C),high-density lipoprotein cholesterol (HDL-C),creatinine (CREA),glycosylated hemoglobin (HbA1c),fasting blood glucose (FPG),2h postprandial plasma glucose (2hPG) and body mass index (BMI).Results There were no significant differences in the elastic parameters of femoral arteries between the male and female subjects (P>0.05),and between left and right sides (P>0.05) within a same group.Compared with group A,group B had lower CC and DC (P<0.05) and higher α,β and PWV (P<0.05).Pearsons correlation analysis indicateal that,CC was negatively correlated with α,β and PWV (P<0.05),and DC was also negatively correlated with α,β and PWV (P<0.05).There was no significant difference in the inner diameters of femoral arteries between the two groups (P>0.05).The levels of TG,HbA1c,FPG and 2hPG in group B were significantly higher than those in group A (P<0.05).There were no significant differences in TC,HDL-C,LDL-C,CREA and BMI between the two groups (P>0.05).Conclusions The decrease of arterial elastic function is earlier than morphological changes, that is,it may have occurred before MA in T2DM patients. The application of ultrasound radio-frequency analyzing technology has certain significance in the clinical early diagnosis and treatment.

OBJECTIVE:
To study the appropriate time to take out T tube, long-term effect and adverse consequence of children with chronic otitis media subjected to adenoidectomy and tonsillectomy plus tympanostomy.
METHOD:
One hundred and eighteen children with 226 ears operated were analyzed and summarized.
RESULT:
T tubes taken out at 1, 3, 6 and 12 month were 6, 102, 88 and 23 ears, respectively, and T-tubes were not taken out in 7 ears.
CONCLUSION
At present, the treatment is curable and reliable with fewer complications. The appropriate time to take out the T-tube should be 3 months after the operation.

Objective:To investigate the role of complement C3a receptor in the diabetic nephropathy pathogenesis of db/db mice, and to provide a new target for prevention and treatment of diabetic nephropathy. Methods:Twelve 8-week-old male mice with type 2 diabetes mellitus (db/db mice) and 6 wild-type (db/m) mice were reared in the special pathogen free environment. The mice were grouped into db/m group, db/db group and C3a receptor antagonist group, with 6 mice in each group. db/db model mice were intraperitoneally injected with C3a receptor antagonist (SB290157, 10 mg/kg) once every two days for 8 weeks in C3a receptor antagonist group. Blood and urine samples were collected, and body weight of mice, fasting blood glucose, serum creatinine, blood urea nitrogen, urinary microalbumin/urinary creatinine and urinary N-acetyl-β- D-glucosaminidase (NAG) were detected. Renal tissues were collected, and HE, PAS and Masson stainings were used to observe the pathological changes. Immunohistochemistry, immunofluorescence and Western blotting were used to detect the protein expression levels of C3 and C3a receptor. Western blotting was used to analyze the protein expression levels of kidney injury molecule-1 (Kim-1), α-smooth muscle actin (α-SMA), zonula occluden-1 (ZO-1), vimentin and E-cadherin in renal tissues. Immunofluorescence was used to analyze the protein expression levels and distribution of α-SMA, ZO-1 and Kim-1, and immunohistochemistry was used to analyze the protein expression levels of interleukin-1 (IL-1) and tumor necrosis factor-α (TNF-α). TUNEL assay was used to detect apoptotic cells in renal tissues. Results:Compared with db/m group, body weight, fasting blood glucose, urinary microalbumin/urinary creatinine and urinary NAG in db/db group were significantly higher, while these indicators in C3a receptor antagonist group were slightly lower than those in db/db group (all P<0.01). There were no significant differences in serum creatinine and blood urea nitrogen among the three groups (all P>0.01). Compared with db/m group, db/db group had glomerular hypertrophy, necrosis and exfoliation of renal tubular epithelial cells, and dilation of renal tubules, and C3 and C3a receptor protein expression levels were higher (both P<0.01). Compared with db/db group, C3a receptor antagonist group had less glomerular lesions, mild necrosis of renal tubular epithelial cells and less tubular dilation. Compared with db/m group, the protein expression levels of Kim-1, IL-1 and TNF-α in kidney tissues of db/db group were significantly higher, while Kim-1, IL-1 and TNF-α in C3a receptor antagonist group were significantly lower than those in db/db group (all P<0.01). Compared with db/m group, the protein expression levels of α-SMA and vimentin of renal tubular epithelial cells in db/db group were significantly higher, while the protein expression levels of ZO-1 and E-cadherin were significantly lower (all P<0.01). Compared with db/db group, the protein expression levels of α-SMA and vimentin of renal tubular epithelial cells in C3a receptor antagonist group were significantly lower, and the protein expression levels of ZO-1 and E-cadherin were significantly higher (all P<0.01). Compared with db/m group, the number of apoptotic cells of kidney tissues in db/db group was increased, while the number of apoptotic cells in C3a receptor antagonist group was reduced compared with db/db group. Conclusions:The expression levels of C3 and C3a receptor of kidney tissues in db/db mice are significantly increased. Antagonistic C3a receptor can reduce the body weight, blood glucose, urinary microalbumin/urinary creatinine and urinary NAG, alleviate renal pathological injury, inhibit renal tissue inflammation, apoptosis and renal tubule epithelial-mesenchymal transition in db/db mice.

Clustered regularly interspaced short palindromic repeats (CRISPR)/CRISPR-associated proteins (Cas) system is a hotspot of gene editing and gene expression research, in which CRISPR/Cas13 system provides a new direction for RNA interference and editing. In this study, we designed and synthesized the corresponding gRNAs of CRISPR/Cas13a and CRISPR/Cas13b systems in non-homologous end joining (NHEJ) pathway, such as Ku70 and Lig4, and then detected the expression of ku70 and lig4 in HEK293T cells. The CRISPR/Cas13a system could efficiently knockdown the mRNA expression of ku70 and lig4 more than 50%, and CRISPR/Cas13b system also suppressed ku70 and lig4 about 92% and 76%, respectively. Also, CRISPR/Cas13a, b systems could down-regulate Ku70 and Lig4 proteins level to 68% and 53%, respectively. The study demonstrates that the CRISPR/Cas13 system could effectively knockdown the expression of RNA and protein in HEK293T cells, providing a new strategy for gene function and regulation research.
