1.Preliminary Study of Transcatheter Arterial Chemoembolization for MalignantLimb Bone Tumor Before Limb-sparing Operation

Objective To evaluate the values of transcatheter arterial chemoembolization(TACE)for malignant limbs bone tumors before limb-sparing operation and the influence of TACE on operation.Methods TACE was performed in 10 patients with malignant limb bone tumor before limb-sparing operation.The complication of TACE,morphology and histology of neoplasm,incisional healing after TACE were observed.Results (1)The embolization syndrome (swelling,pain)appeared obviously in all patients at early stage and disppeared or alleviated greatly after 2 weeks of TACE.The severe complication-local skin necrosis appeared in 1 case;there was a subcrustal healing after 3 weeks.(2)In 6~28 days after embolization,all neoplasms underwent coagulation to variable extents,and the tumor cells was more than 60% necrosis in 8 cases.(3)All incisions had a good healing by first intention.Conclusion TACE is an effective and safe accessory therapeutic preecdure for malignant limbs bone tumors before limb-sparing operation.
2.A study on the changing tendency of IgG to coronavirus associated severe acute respiratory syndrome

Objective:To explore the changing tendency of IgG to coronavirus associated severe acute respiratory syndrome(SARS).Methods:Between 1 and 153 days after the onset of symptoms, the antibody IgG to SARS associated coronavirus were detected in 534 patients with SRAS by ELISA. 216 randomly selected people without SARS contact history and SARS symptoms were contrasted.Results:The positive rates of IgG between SARS patients and control group respectivel were 58 1%,0 5%.There is a statistical significance(P=0 000).Among SARS patients, the positive rates of IgG in 1~10 days,11~30 days,31~60 days,61~90 days,91~120 days,above 120 days and after the onset of symptoms respectively were 18 8%(3/16 cases),63 8%(37/58 cases),55 6%(30/54 cases),51 4%(71/138 cases),62 1%(159/256 cases),83 3%(10/12 cases).Conclusion:There are low positive rates of IgG in SARS patients in the first 10 days after onset of symptoms, and abruptly ascending positive rates of IgG in 11~30 days.The positive rates of IgG remains the high levels during 4th and 5th months.Further observation is essential to elucidating the duration of IgG.
3.Establishment of cross priming amplification for influenza A virus (H1N1) and its clinical application

In this study ,we established Cross Priming Amplification (CPA ) technology for detection of influenza A virus (H1N1) approach ,and evaluated the method through clinical specimens .A set of specific primers were designed for CPA ac‐cording to the conservative gene sequences ,designed and realized in the same temperature reverse transcription of RNA and DNA amplification . The amplification products can be totally enclosed nucleic acid detection device for testing . Fourteen healthy pharyngeal swab specimens ,seven other respiratory viruses ,and six arboviruses strains were used as the controls .We used a method that application of gradient dilution to the H 1N1 virus strain as the control to test the sensitivity of the CPA .We also used 102 clinical pharyngeal swab specimens of H1N1 patients for detection object to evaluate the feasibility of CPA clinical detection .Results showed that the CPA reaction did not appear cross reaction on health cases samples and other viruses .The sensitivity of the CPA was approximately 10 copies/uL in the established method that exactly titer H1N1 virus strain gradient dilution test .As to the positive results among the clinical pharyngeal swab samples collected from patients at different stages after onset ,the CPA had the highest positive detection rate during the first three days after onset (100% ) .While the detection rate from day 4 to day 6 after onset was 79 .31% .After 7 days ,the detection rate was 9 .09% .The established CPA assay was a highly sensitive ,specific and reproducible approach for rapid detection of H1N1 virus ,which is conducive to the early diagno‐sis of influenza A virus (H1N1) for basic medical units .
4.Envelope gene evolution analysis on type 1, 2, 3 dengue virus in Guangzhou in 2010

ObjectiveTo analyze the Envelope (E) gene of type 1,2,3 dengue virus isolated fromGuangzhouin2010, andtoinvestigatetheinfectionsourceandvirusgenotypes.MethodsEighty-five serum samples were collected from 85 patients in acute phase of dengue fever.Dengue virus was cultured and isolated by C6/36 cells.The whole length of E gene was amplified by reverse transcriptase-polymerase chain reaction (RT-PCR) and then sequenced.The phylogenetic tree was drawn by neighbor-joining method.The bioinformatics analysis was performed by combining the phylogenetic information and the epidemiologic data.ResultsSix strains of type 1 dengue virus,two strains of type 2 dengue virus and six strains of type 3 dengue virus were isolated from 85 samples.The E gene sequence of these strains was obtained by sequencing.The phylogenetic analysis showed that type 1 and 3 dengue virus belonged to two genotypes (Asian and South Pacific ocean,India subcontinent and Southeast Asia/South Pacific ocean,respectively),and type 2 dengue virus belonged to one genotype (Malaysia/India subcontinent).ConclusionIt's presumed that all strains of type 2 dengue virus are imported,four strains of type 1 dengue virus are imported and four strains of type 3 dengue virus arc imported,the remaining two stains of type 1 and two stains of type 3 dengue virus need mosquito intermediary research further to prove their origins.
5.Epidemiological and genetic characteristics of H5 subtype avian influenza virus in Guangzhou, 2014-2019

Objective:To analyze the characteristics of spread and genetic evolution of H5 subtype avian influenza virus in Guangzhou from 2014 to 2019.Methods:H5 subtype virus was detected by fluorescence quantitative RT-PCR from the environmental samples in Guangzhou poultry markets. The genes of HA and NA of 48 isolates randomly selected were sequenced, including 46 isolates from environmental samples and 2 isolates from cases. The characteristics of molecular variation and genetic evolution were analyzed by using bioinformatics software.Results:A total of 1 094 strains of H5 subtype avian influenza virus were isolated from 52 284 samples (2.09%). All the strains belonged to Clade 2.3.4.4.C. NA gene belonged to H6N6 of Eurasian lineage. The cleavage sites of all the strains showed the characteristics of highly pathogenicity. Receptor binding sites were avian-derived receptors. However, mutations of S123P, S133A and T156A occurred, which implied that these strains could tend to bind to human receptors. There was an additional glycosylation site at 140 in strains isolated after 2017. The variation of antigen loci mainly occurred in B and E regions.Conclusions:H5 subtype avian influenza virus spread in Guangzhou from 2014 to 2019 with annual increased proportion of positive rate, and the sequencing results indicated that it belonged to Clade 2.3.4.4.C of H5N6 highly pathogenic virus, and genetic evolution and mutation continued, especially the common mutations which could enhance the binding capacity to human receptors. It is necessary to strengthen the surveillance.
6.High grade B cell lymphoma with concurrent MYC rearrangement and 11q aberrations:a clinicopathological analysis

Purpose To investigate the clinicopathological features,molecular genetics and prognosis of high grade B cell lymphoma with concurrent MYC rearrangement and 11q aberra-tions(HGBCL-MYC-11q).MethodsThree cases of HGBCL-MYC-11q were reviewed and analyzed using hematoxylin-eosin staining,immunohistochemistry,EBER in situ hybridization and fluorescence in situ hybridization.Clinical data were collected with follow-up.Results All three patients were male,age was 10,61,and 74 years,respectively.All patients had Ann Arbor stage Ⅳ disease.All three cases were biopsies occurring in the nasopharynx,upper pharynx and ileocecus,respectively.Three cases were morphologically similar to diffuse infiltrative growth of tumor cells,moderate or moderately large cells,round to slightly irregular nuclei and easily visible mitotic figures.Focal necrosis was noted in one case.One case exhibited the distinct"starry sky"pattern.All cases expressed CD20,BCL6 and MUM1 and high Ki67 index,two cases expressed CD10 and two cases ex-pressed BCL2.CD3,CD30 and TDT were all negative.EBER in situ hybridization was all negative.FISH analyses using C-MYC break-apart probes were all positive and all cases had 11q aberrations.One case only had the 11q23.3 amplification;and one case only had the 11q24.3 loss.After a follow-up for 1-18 months,one patient died and two patients survived with disease.ConclusionHGBCL-MYC-11q is rare,morphologically similar to BL/HGBCL,with MYC rearrangement and 11q abnormali-ties.We should enhance awareness of the disease and improve more accurate diagnosis and differential diagnosis of the disease.
7.Association between physical activity and exercise self-efficacy for adolescents with visual impairment

ObjectiveTo investigate the association between physical activity (PA), sedentary level and exercise self-efficacy for adolescents with visual impairment, and impacts of gender, age, body mass index (BMI), degree of visual impairment and exercise self-efficacy on PA. MethodsFrom October to December, 2022, 79 students aged 13 to 18 years with visual impairment from Nanjing School for the Blind were measured PA and sedentary level with accelerometer. The average percentage of daily sedentary time (ST), light physical activity (LPA) and moderate-to-vigorous physical activity (MVPA) were recorded. The students fininshed the Exercise Self-Efficacy Questionnaire with the help of teachers, and their height and body mass was measured. ResultsThe average percentage of daily LPA, MVPA and ST was (15.45±5.99)%, (4.55±2.15)% and (80.00±6.99)%, respectively. Compared with the girls, the boys spent more time in LPA (F = 5.330, P < 0.05) and less in ST (F = 5.760, P < 0.05). Compared with students aged 16 to 18 years, those aged 13 to 15 years spent more time in MVPA (F = 11.292, P < 0.001) and less in ST (F = 8.279, P < 0.01). The score of exercise self-efficacy correlated with ST, LPA, MVPA and age (|r| > 0.254, P < 0.05). The result of multiple linear regression showed that the exercise self-efficacy (β = 0.224) and age (β = -0.495) were the independently factors related with MVPA, which explained 34.7% of MVPA variation. ConclusionAdolescents with visual impairment aged 13 to 18 years spend more time in sedentary, and their PA level is very low. Improvement of exercise self-efficacy may help to improve PA level of them.
