1.Treatment of 21 Cases of Post-herpetic Neuralgia by Warm Acupuncture

Jiaji (Ex-B 2), Waiguan (TE 5), Taichong (LR 3), Zulinqi (GB 41) and Ashi points were mainly selected. Adjunct acupoints were added according to the affected positions. Neiting (ST 44) was added when the affected position was mainly on the abdomen, Kunlun (BL 60) was added on the back and Xingjian (LR 2) was added on the hypochondriac region. Twenty- one patients with post-herpetic neuralgia were treated. The result showed that pain and skin injury of 16 cases disappeared, skin injury of 4 cases disappeared with obviously alleviated pain and skin injury of 1 case disappeared with slightly alleviated pain.
2.Effects of Mannitol Combined with Dexamethasone on Related Indexes of Patients with Infectious Brain Edema

OBJECTIVE:To investigate the effects of mannitol combined with dexamethasone on related indexes of patients with infectious brain edema.METHODS:A total of 120 patients with infectious cerebral edema were divided into control group (60 cases) and observation group (60 cases) according to therapy plan.Control group was given Furosemide injection 20 mg+20% Mannitol injection 1 g/(kg,time)+0.9% Sodium chloride injection 10 mL intravenously,every 6 h,3 days later adjusting drug dose according to the disease condition of patients.Observation group was given Dexamethasone injection 10-20 mg+20% Mannitol injection 1 g/(kg·time)+ 0.9% Sodium chloride injection 10 mL intravenously,every 6 h,3 days later adjusting drug dose according to the disease condition of patients.A treatment course lasted for 7 d,and both groups were treated for 2 courses of treatment.The levels of NO,IL-1 and TNF-α,mortality and the occurrence of sequelae before and after treatment as well as the occurrence of ADR were observed in 2 groups.RESULTS:Before treatment,there was no statistical significance in the levels of NO,IL-1 or TNF-α between 2 groups (P>0.05).After treatment,the levels of NO,IL-1 and TNF-α in 2 groups were significantly lower than before treatment,and the observation group was significantly lower than the control group,with statistical significance (P<0.05).The incidence of mortality,sequelae and ADR in observation group were significantly lower than control group,with statistical significance (P<0.05).CONCLUSIONS:Mannitol combined with dexamethasone can reduce inflammatory factor level,the incidence of sequelae and mortality,without increasing the incidence of ADR.
3.Determination of Benzoic Acid and Cinnamic Acid in Suhexiang Pills by HPLC with Dual Wavelength De-tection

Objective:To establish an HPLC method with dual wavelength detection for the determination of benzoic acid and cin-namic acid in Suhexiang pills. Methods:A Waters Sun Fire TM C18 column(150 mm × 4. 6 mm,5 μm) was adopted with 0. 1% ace-tic acid-methanol(60∶40) as the mobile phase at a flow rate of 1. 0 ml·min-1 . The detection wavelength was set at 228nm for benzoic acid(0-18min) and 285nm for cinnamic acid (18. 1-35min). The injection volume was 10μl, and the column temperature was room temperature. Results:The linear range of benzoic acid and cinnamic acid was 1. 0-50. 0 μg·ml-1(r=0. 999 7) and 0. 2-10. 0 μg· ml-1, respectively(r=0. 999 9). The average recovery was 96. 88%(RSD=1. 6%, n=9) and 99. 35%(RSD=1. 7%, n=9), re-spectively. Conclusion:The method is accurate, fast and simple in the determination of benzoic acid and cinnamic acid in Suhexiang pills.
4.Clinical analysis of pulmonary arterial hypertention in systemic lupus erythamatosus

Objective To study the clinic manifest-rations, mechanism, diagnosis and treatment of pulmonary arterial hypertention (PAH) in systemic lupus erythamatosus (SLE). Methods The clinic symptoms, laboratory tests. Doppler tests of patients who were diagnosed with PAH and SLE during last five years were analyze retrospectively. Results Anti-RNP antibody's prevalence rate was high. The presence of pulmonary arterial perssure (PAP) was not related to SLE disease active index (SLEDAI). Those who had PAP higher than 45 mm Hg were prone to have Raynaud's phenomenon PAH had no association to auto-antibodies and other systemic lesions. There were no differences in clinic features, auto-antibodies and other examinations between those with kidney injure and without kidney injure. Conclusion PAH may develop insidiously in SLE, so early diagnosis and prompt treatment of PAH can improve the prognosis.
5.Influence of metformin and paclitaxel in proliferation and apoptosis of ovarian cancer SKOV3 cells in vitro

Objective:To study the influence of metformin and paclitaxel in the proliferation and apoptosis of human ovarian cancer SKOV3 cells in vitro, and to clarify the synergistic effect of metformin and paclitaxel. Methods:The ovarian cancer SKOV3 cells were divided into blank control group, different concentrations (0.01, 0.50, 1.00, 5.00, 10.00mmol·L-1) of metformin groups and combined treatment groups(metformin combined with paclitaxel with different concentrations).The inhibitory rates of proliferation of SKOV3 cells after treated with different concentrations of metformin were detected by MTT method.The apoptotic rates of SKOV3 cells after treated with different concentrations of metformin were measured by flow cytometry.The inhibitory rates of proliferation of SKOV3 cells after treated with metformin and paclitaxel were determined by MTT method.Results:The MTT results showed that the inhibitory rates of proliferation of SKOV3 cells in different concentrations of metformin groups were increased in concentration-and timedependent manner;there were significant differences compared with blank control group (P<0.05).The flow cytometry results showed that the apoptotic rates of SKOV3 cells in different concentrations of metformin groups were increased;compared with control group, with the increasing of concentrations of metformin, the apoptotic rates of SKOV3 cells in experimental groups 48 h after treatment were increased significantly (P<0.05);the percentages of SKOV3 cells in G0/G1 phase were decreased with the increasing of metformin concentrations(P<0.05) and the percentages of SKOV3 cells in G2/M phase were increased(P<0.05).The MTT results showed that the inhibitory rate of proliferation of SKOV3 cells in 1.00 mmol·L-1 metformin+paclitaxel group was higher than that in 0.50 mmol·L-1 metformin+paclitaxel group was higher than that in 0.50 mmol·L-1 metformin+paclitaxel group(P<0.05),and the inhibitory rates of proliferation of SKOV3 cells in combined treatment groups were higher than those in paclitaxel alone groups(P<0.05).Conclusion:Metformin inhibits the proliferation of ovarian cancer SKOV3 cells through the induction of apoptosis.Metformin can enhance the cell proliferation inhibition of paclitaxel on ovarian cancer SKOV3 cells.The combination of metformin and paclitaxel has a synergistic reaction on SKOV3 cells.
6.Expression of CD163 in patients of non-small cell lung and effects of cancer cell migration and proliferation

Objective:To investigate the expression of CD 163 in the plasma and tumor tissue of non-small cell lung cancer (NSCLC)patients,and the effects of cancer cell migration and proliferation .Methods:35 cases of NSCLC patients in our hospital as the study group,and 35 healthy volunteers as control group ,we used Real-time RCR to detect the expression of CD163 in the plasma and tumor tissue of NSCLC patients.Using small interfering RNA (siRNA) to inhibit the expression of CD163 in MH-2,and co-culture with A549.The level of cancer cell migration was observed by transwell and the level of cell proliferation was observed by CCK -8.Results:The expression of CD163 in the plasma and tumor tissue of NSCLC patients was significantly higher than the control group , the difference was statistically significant (P<0.05);after the expression of CD163 was suppressed in MH-2,the levels of cancer cell migration and proliferation were significantly decreased , the difference was statistically significant ( P<0.05 ) .Conclusion: The expression of CD163 in the plasma and tumor tissue of NSCLC patients is increased ,and may elevate the levels of cancer cell migration and proliferation.
7.Valproic acid exerts differential effects on cytokine synthesis in human peripheral lymphocytes

AIM: Valproic acid (VPA) is a histone deacetylase inhibitor and is believed to have anti-tumor activity. The present study aims to investigate the effect of VPA on the, apoptosis and cytokine synthesis of human peripheral lymphocytes. METHODS: The activation and cytokine synthesis in lymphocytes in whole blood stimulated with phorbol dibutyrate (PDB) and ionomycin were evaluated with flow cytometry after fluorescent staining. The mitochondrial membrane potential was examined using 3, 3-dihexyloxacarbocyanine iodide [DiOC6(3)]staining. RESULTS: VPA at low doses (1 and 5 mmol/L) promoted CD69 expression in activated lymphocytes, whereas it turned to inhibit the expression of CD69 at a high dose (25 mmol/L). Meanwhile, VPA at low doses increased the mitochondrial membrane potential, while a high dose of VPA decreased it in activated lymphocytes. Furthermore, interleukin-2 (IL-2) synthesis was enhanced by low doses of VPA but inhibited by a high dose. However, interferon-γ (IFN-γ) and tumor necrosis factor-α (TNF-α) synthesis were dose-dependently enhanced by VPA as compared with those of PDB plus ionomycin-treated cells. CONCLUSION: VPA exerts biphasic effect on the further activation and apoptosis of human peripheral lymphocytes stimulated with mitogens and exhibits differential activity on the synthesis of several important cytokines in human lymphocytes.
8.Exploring the mechanism of moxibustion in myocardial protection of rats with long-term fatigue exercise based on the classical pyroptosis pathway

Objective: To explore the possible mechanism of moxibustion in myocardial protection of rats undergoing long-term fatigue exercise based on observing the classical pyroptosis pathway mediated by nuclear factor kappa-B (NF-κB)/nucleotide-binding oligomerization domain-like receptor protein 3 (NLRP3)/cysteinyl aspartate specific proteinase 1 (Caspase-1).Methods: A total of 50 specific-pathogen-free male Sprague-Dawley rats were bought. Ten unqualified rats were excluded, and the remaining 40 rats were divided into a normal group, a normal + Shenque (CV8) group, a model group, a model + non-meridian non-point group, and a model + Shenque (CV8) group according to the random number table method, with 8 rats in each group. Except for rats in the normal group and the normal + Shenque (CV8) group, rats in the other three groups were trained with an incline running table exercise protocol to create a long-term fatigue exercise model, 1 h/time, once a day for 5 d with 2 d off, for a total of 8 weeks. Rats in the normal group received no modeling or intervention. Rats in the normal + Shenque (CV8) group were not modeled but received mild moxibustion at Shenque (CV8); those in the model group were modeled only without intervention; those in the model + non-meridian non-point group received moxibustion at non-meridian and non-point spots after the modeling; those in the model + Shenque (CV8) group received moxibustion at Shenque (CV8) after modeling. The above moxibustion interventions were performed for 15 min/time once daily, for 5 d with 2 d off per week and a total of 8 weeks. Blood was collected from the femoral artery 4 h after the last exercise, and the serum interleukin (IL)-1β and IL-18 levels were measured. The NF-κB, NLRP3, apoptosis-associated speck-like protein containing a CARD (ASC), Caspase-1, and gasdermin D (GSDMD) expression levels were detected by Western blotting. Myocardial morphology and pyroptosis were observed by hematoxylin-eosin (HE) staining and electron microscopy. Results: The HE staining results showed that the myocardial cells in the model group and the model + non-meridian non-point group were disorganized with blurred transverse lines, widened interstitial spaces, interstitial edema, and inflammatory cell infiltration. The structure of myocardial cells in the model + Shenque (CV8) group was clearly visible, with slightly widened interstitial spaces and occasional infiltration of inflammatory cells in the interstitium. Compared with the normal group, the serum IL-1β and IL-18 levels were increased, and myocardial NF-κB, NLRP3, ASC, Caspase-1, and GSDMD expression levels were elevated in the model group and the model + non-meridian non-point group (P<0.01). Compared with the model group, the above indicators did not change significantly in the model + non-meridian non-point group, while all the above indicators were decreased in the model + Shenque (CV8) group (P<0.01). Compared with the model + non-meridian non-point group, all the above biochemical indicators were decreased in the model + Shenque (CV8) group (P<0.01). Transmission electron microscopy showed that the mitochondria number was increased in the model group and the model + non-meridian non-point group, some of the mitochondrial lumen was irregularly enlarged, the cell membrane structure was unclear, and chromatin was aggregated. The mitochondria number was increased, the swelling was reduced, and the nuclear membrane structure was more intact in the model + Shenque (CV8) group. Conclusion: Moxibustion at Shenque (CV8) regulates the NF-κB/NLRP3/Caspase-1 pathway and reduces the pyroptosisin the myocardium of rats with long-term fatigue exercise, thus reducing the myocardial injury caused by long-term fatigue exercise.
9.Cucurbitacin E induces autophagy in HeLa cells by inhibiting mTORC1 activity

Aim To study the mechanism of cucurb-itacin E ( CuE )-induced autophagy in HeLa cells. Methods Improved MTT assay was adopted to meas-ure the effect of CuE on cell proliferation. Western blot was used to determine the phosphorylation levels of downstream signaling proteins of mTORC1 and the ex-pression of autophagy associated proteins. ResultsCuE inhibited the proliferation of HeLa cells in a dose-dependent manner, and the 24-h IC50 of CuE was 4. 01μmol· L-1 . CuE significantly inhibited the phospho-rylation of p70 S6 K in a time-and dose-dependent man-ner as evidenced by decreased phosphorylation levels of
the mTORC1 substrate. Meanwhile, the expression of LC3-II, a marker for autophagosome formation, was elevated by CuE treatment, and was further increased in the presence of chloroquine. Furthermore, CuE re-duced the levels of p62/SQSTM1 . These results indi-cated that CuE induced autophagy in HeLa cells. The decreased levels of phosphorylated ULK1 S757 were posi-tively correlated with autophagy induction in HeLa cells. Conclusion CuE is likely to induce autophagy through inhibiting mTORC1 activity.
10.Effects of subcutaneous transfection of human beta-nerve growth factor gene on expression of substance P in dorsal root ganglion in a rat model of diabetic neuropathic pain

Objective To investigate the effects of subcutaneous transfection of human beta-nerve growth factor (Ad-hNGFβ) gene on the expression of substance P (SP) in the dorsal root ganglion in a rat model of diabetic neuropathic pain (DNP).Methods Male SD rats weighing 180-220 g were used in this study.Ten rats were randomly collected as normal control without DNP (group C).DNP model was made by intraperitoneal injection of streptozocin (STZ) 75 mg/kg.Seventy-five rats with DNP were randomly divided into 3 groups ( n =25 each):DNP group; Ad-hNGF group and Ad-LacZ group.Groups Ad-NGF and Ad-LacZ received subcutaneous transfection of 1.12 × 1010 PFU Ad-hNGFβ 10 μl and 1.12 × 1010 PFU Ad-LacZ 10 μl respectively after pain threshold was measured on 21d after STZ injection.The mechanical and thermal pain threshold was measured before STZ injection (baseline) and at 21,35 and 49 d after STZ injection.The expression of SP in the dorsal root ganglion was determined after the measurement of pain threshold on 49 d after STZ injection.Results Compared with group C,the mechanical and thermal pain threshold was significantly decreased at each time point after STZ injection in groups DNP,Ad-NGF and Ad-LacZ,and the expression of SP in the dorsal root ganglion was signilicantly downregulated in groups DNP and Ad-LacZ (P < 0.05).Compared with group DNP,the thermal pain threshold was significantly increased on 49 d afar STZ injection,and the expression of SP in the dorsal root ganglion was significantly up-regulated in group Ad-NGF ( P < 0.01 ),and no significant change was found in the mechanical and thermal pain threshold and the expression of SP in the dorsal root ganglion at each time point in group Ad-LacZ ( P > 0.05).Conclusion Subcutaneous transfection of Ad-hNGFβ can attenuate DNP to some extent through upregulation of the expression of SP in rat dorsal root ganglion.
