To establish a good training model,we should take into consideration its operability and practicability, as well as the positivity of the teachers.The key to making the training succeful is to ascertain the direction and contents, to set up an instructors' team, and to make a specific training plan.
2.Protective effect of Danshen Tablet on vascular endothelial function

AIM:To investigate the protective effects of Danshen Tablet on the brachial artery flow-mediated dilatation(FMD)in the subjects accompanied with risk factors of coronary heart disease(CHD). METHODS: Fifty-five subjects were randomly assigned to two groups,therapy group(32 cases)were administrated Danshen Tablet(three tablets once,three times a day),the control group(23 cases) were received placebo and then observed their brachial artery FMD before and after three months' therapy.Meanwhile 19 healthy cases were compared with the subjects. 4. Expression of GUI in gastric carcinoma tissue and its effect on biological characteristics of gastric carcinoma cells

Objective: To study the expression of glioma-associated oncogene 1 (Glil) in gastric carcinoma tissue, and to explore the effects of Glil on the proliferation and migration abilities of gastric carcinoma cells. Methods: A total of 95 cases of human gastric carcinoma tissue and paracancerous tissue were collected. Immunohistochemistry was used to detect the expression levels of Glil protein in gastric carcinoma tissue and paracancerous tissue; Real-time PCR method was used to detect the expression levels of Glil mRNA in gastric carcinoma tissue and paracancerous tissue. The human gastric cancer cell lines MKN28, BGC823, SGC7901 and immortalized gastric epithelial cells GES-1 were cultured, GES-1 as a reference, and the expression levels of Glil mRNA in cell lines were detected by RT-qPCR. The Glil-siRNA and con-siRNA were transfected into the gastric carcinoma cell line BGC823, and control group, con-siRNA and Gill-siRNA group were set up. The expression levels of Glil mRNA in the cells in various groups were detected by RT-qPCR; CCK8 was used to detect the proliferation of cells; Transwell migration assay was used to detect the cell migration ability. Western blotting was used to detect the expression levels of the P27, matrix metalloproteinase-2 (MMP-2), and MMP-9 proteins in the cells in various groups. Results: The positive expression rates of Glil mRNA and protein in gastric carcinoma tissue were significantly higher than those in paracancerous tissue (t=27. 606, P<0. 01; χ2 =54. 782, P<0. 01). There were significantly differences in the expression levels of Glil mRNA between GES-1, MKN28, SGC7901, and BGC823 cell lines (F=86. 341, P<0. 01). The expression level of GUI mRNA in Glil-siRNA group was significantly lower than those in control and con-siRNA groups (F=48. 322, P<0. 01). The proliferation rate of gastric carcinoma cells in Glil-siRNA group was significantly lower than those in control and con-siRNA groups (F=54. 428, P<0. 01). The results of Transwell migration assay showed that the number of migration cells in Glil-siRNA group was significantly lower than those in control and con-siRNA groups (F=257. 788, P<0. 01). The Western blotting results showed that the expression levels of P27, MMP-2, and MMP-9 proteins in GlilsiRNA group had no significant differences compared with control group (P>0. 05). Compared with con-siRNA group, the expression level of P27 in the cells in Glil-siRNA group was significantly increased (t= - 3. 776, P= 0.020), and the expression levels of MMP-2 and MMP-9 proteins were significantly dereased (P=3. 497, P = 0. 025; t=5. 487, P=0. 005). Conclusion: The expression level of Glil in gastric carcinoma tissue is higher than that in paracancerous tissue, and the inhibition of the expression of Glil gene could inhibit the cell proliferation and migration.
5.Protective effect of human umbilical cord-derived mesenchymal stem cells against severe acute pancreatitis in rats

Objective To study the protective effects of human umbilical cord-derived mesenchymal stem cells (ucMSCs)against severe acute pancreatitis (SAP) in rats.Methods A total of 135 Sprague-Dawley male rats were randomly divided into Sham group,SAP group and SAP+ucMSCs group (45 each).SAP+ucMSCs group:Severe acute pancreatitis was induced by injecting 5% sodium taurocholate (0.1ml/100g) into the common bilio-pancreatic duct and then CM-DiI-labeled ucMSCs at 1 × 107cells/kg were injected via the tail vein.All the rats were sacrificed 12,24 and 72 hours after SAP.The 72h death rate was counted.Pathological changes in the pancrease were detected by HE staining and pathological score was graded,ucMSCs colonization was observed by fluorescence microscopy.The serum levels of amylase,lipase,TNF-α,IL-1β,IL-4 and IL-10 were determined by ELISA.Results ucMSCs colonize the injured area of pancreatic tissue,the 72h death rate was reduced,and the serum amylase and lipase were also reduced significantly.Moreover,ucMSCs significantly reduced the pathological score of the pancrea and the level of proinflammatory cytokines (TNF-α and IL-1β),but the levels of anti-inflammatory cytokines were increased (IL-4 and IL-10).Conclusion Transplantation of ucMSCs can reduce the severity of pancreatic injury and inflammation in SAP rats.
6.Expression and significance of c-Jun N-terminal protein kinase 1/2 protein in chronic hibernated myocardium of domestic pigs

BACKGROUND: Acute myocardial ischemia can activate the c-Jun N-terminal protein kinase(JNK) and, in turn, the ischemia damage can be aggravated by JNK. While in chronic hibernating myocardium, whether chronic myocardial ischemia can activate JNK1/2 or not and what is the role of JNK1/2 in developing the chronic hibernating myocardium, is not clear.OBJECTIVE :To identify protein expression of JNK1/2 and the p-JNK1/2 changes in chronic hibernating myocardium.DESIGN: Randomly controlled experimental research.SETTING: This study was performed in Cardiovascular Disease Institute, the Affiliated Hospital of Xuzhou Medical College.MATERIALS: Animal models were prepared in the Catheter Department of the Affiliated Hospital of Xuzhou Medical College, protein expression of JNK1/2 and the phosphorylation(p-JNK1/2) changes were measured in the Department of Biochemistry of Xuzhou Medical College. Fourteen small Chinese domestic pigs were randomly divided into two groups, experiment group( n = 8) and control group( n = 6) . Right coronary artery was selected as target vessel in experiment group, and home-made constrictor was delivered through right femoral artery to induce chronic hibernating myocardium and myocardial infarction.MAIN OUTCOME MEASURES: Normal myocardial tissue in control group, normal myocardial tissue and chronic hibernating myocardium sample in experiment group were checked with light microscope and electron microscope,protein expression of JNK1/2 and the phosphorylation changes in myocardium tissue of three groups were analyzed by immunoblotting (Western blotting).RESULTS: p-JNK1/2 was higher in chronic hibernating myocardium of experiment group than that of normal myocardium in experiment and control group. Immunological activity of p-JNK1/2 in control group, normal myocardium, and chronic hibernating myocardium in experiment group was 1,1.42 ± 0.52, 2.6 ± 0.59, respectively.CONCLUSION: JNK1/2 can be activated in chronic hibernating myocardium tissue and also participated in its occurrence and development.
7.Effects of celecoxib on cardiac myocyte apoptosis after myocardial infarction

AIM:To investigate the effects of celecoxib,a selective cyclooxygenase-2 inhibitor,on antioxidative capability and apoptosis of cardiac myocytes after myocardial infarction.METHODS:24 New Zealand rabbits were divided into three groups randomly(8 in each group):sham-operated group(sham group),myocardial infarction group(MI group),celecoxib group(Cele group,10 mg kg-1?d-1,qd,with the drugs gastric gavage for six weeks).The NO concentration,total antioxidative capability(T-AOC),the activity of constitutive nitric oxide synthase(cNOS)and inducible NOS(iNOS)in cardiac tissue homogenate,adjacent to the infracted area,were detected.The pathological changes were observed by light microscope and electron microscopy.The expressions of Bcl-2 and Bax protein in myocytes were observed using immunohistochemistry,and the degree of apoptosis were examined by TUNEL.RESULTS:Cardiac tissue in MI group presented interstitial edema,fibroplastic proliferation,inflammatory cellular infiltration,and vacuolar degeneration in cardiac myocytes.The results of electron microscopy showed that myocytes presented more changes caused by ischemic injury:widened interspace of myofibril,disordered myofibrillae,focal lysis of myofilament,ectasia of sarcoplasmic reticulum.In Cele group,the pathological changes were light,the NO-_2/NO-_3 concentration,the activity of iNOS were lower(P
8.Expressions of Krüppel-like factor 12 in cancer tissue and serum of patients with colorectal cancer and their significances

Objective:To detect the expressions of Krüppel-like factor 12 (KLF12)protein in the cancer tissue and serum of the patients with colorectal cancer,and to investigate their clinical values.Methods:A total of 120 colorectal cancer patients were selected as case group,and 100 normal cases were regarded as control group. The cancer tissue and adjacent mucosa tissue were collected.Immunohistochemical method was used to detect the expressions of KLF12,nm23,CyclinE1,matrix metalloproteinase-2 (MMP-2),and MMP-9 proteins,and the relationships among these proteins were evaluated.ELISA assay was applied to detect the serum KLF12 levels of the patients in case group before operation and 1 month after operation the levels of serum KLF12 of the subjects in control group.Results:The positive expression rates of KLF12,CyclinE1,MMP-2 and MMP-9 proteins in cancer tissue were higher than those in adjacent mucosa tissue (χ2 =66.155,52.795,64.515,52.632;P <0.001),while the positive expression rate of nm23 protein in cancer tissue was lower than that in adjacent mucosa tissue (χ2 =13.019,P <0.001).The Spearman analysis results showed that the expression of KLF12 protein was positively correlated with the expressions of MMP-2 and MMP-9 proteins (r=0.3963,P <0.001;r=0.3264,P <0.001);the expression of KLF12 was negatively correlated with the expression of nm23 protein (r = - 0.2273,P =0.013).The level of serum KLF12 of the patients in case group before operation was higher than that in control group.The cut-off value of ROC curve of serum KLF12 protein expression level was 3.795 μg·L-1 ,and the area under the ROC curve was 0.834.The sensitivity and specificity were 60.8% and 94.0%,respectively. After operation,the serum level of KLF12 protein in the patients in case group was decreased,which was still higher than that in control group (t=2.708,P =0.007).The expression level of KLF12 protein in cancer tissue was positively correlated with the level of serum KLF12 protein (r =0.4069,P <0.001).According to degree of lymph node metastasis,and the serum level of KLF12 protein in N3 group was the highest,and the lowest serum level of KLF12 protein was found in N0 group (F =21.731,P <0.001).Conclusion:KLF12 protein might be involved in the invasion and metastasis of colorectal cancer,and the detection of serum level of KLF12 protein could be valuable for the judgement of disease and prediction of lymph node metastasis of the patients.
9.Expression and clinical significance of Gli1 in peripheral blood and cancer tissues of gastric cancer patients

Objective To explore the relationship and clinical significance of Gli1 expression in peripheral blood and cancer tissues of gastric.Method 95 gastric cancer patients were recruited as surgery group,and 40 recurrent gastric cancer patients were recruited as recurrent group.50 normal subjects were recruited as control group.Paraffin specimens of gastric cancer tissues and gastric mucosa adjacent to cancer were collected to test expression of Gli1 protein.ELISA was applied to test Gli1 in peripheral blood.Results Levels of Gli1 in peripheral blood of preoperative surgery group were higher than in control group [(9.21 ± 1.37) ng/ml vs.(0.79 ±0.18) ng/ml,t =43.193,P <0.001].Post-up Gli1 decreased significantly to the level of control group (t =0.722,P =0.472).Levels of Gli1 in peripheral blood of recurrent group were higher than that in preoperativesurgery group (t =14.575,P < 0.001).Gli1 protein in cancer tissues was higher than that in adjacent gastric mucosa (t =33.471,P <0.001),and Gli1 protein in cancer tissues of surgery group was higher than that in cancer tissues of recurrent group (t =8.808,P <0.001).Conclusion In postoperative gastric cancer patients increased Gli1 level in peripheral blood and cancer tissues predicts recurrence.
10. Evaluation of the effectiveness of nasal feeding registration form based on energy intake optimization design for patients with oral cancer

Objective:
To evaluate the effect of a new kind of nasogastric feeding registration form based on energy intake in perioperative nutrition support for patients with oral cancer.
Methods:
According to the specification of nasal feeding and summarizing the ingredients in common foods, the form was optimized and applied. By the cluster sampling method, the knowledge about energy intake of 12 nurses were investigated before and after the use of the form. The accuracy of energy intake assessment of 5 patients by 12 nurses respectively and the time required for single registration was compared before and after the use. Meanwhile, the knowledge about nasal feeding of 18 patients before the use and 17 patients after the use was compared.
Results:
The mean score of knowledge about energy intake of nurses was 57.50±17.77 before the use, which was lower than that after the use (89.00 ±6.18)
