Objective To research the effect of molecular chimeric mice pre-T cells on proliferation ability of allogeneic mouse T cells. Methods The MHC-Ⅰ gene (H-2Kband H-2Db gene) were extracted and amplified by RT-PCR,the identified pre-T cells were transfected by the constructed eukaryotic expression vector of C57BL/6mouse MHC-I (pIRES-H-2Db and pIRES-H-2Kb),non-transfected group and sham pIRES-transfected control group were set.The molecular chimeric cells were transfused back to BALB/c mouse. After 7 days,T lymphocyte cells of each group were extracted, the ability of molecular chimeric cells inducing spleen T lymphocyte response to allogeneic T cells was observed through mixed lymphocyte culture (MLC).Results Sequencing of the plasmid we have constructed showed that insertion sequence contained C57BL/6 mice H-2Kb and H-2Db series which could be retrieved from GenBank. The result of flow cytometry analysis indicated that H-2Kb and H-2Db protein had an increased expression in pre-T cells, the difference with other two groups was statistically significant (P<0.05).The result of MLC demonstrated that the stimulation index (SI) of T lymphocyte in co-injection transfected pIRES-H-2Kb and pIRES-H-2Db pre-T cells group (0.764±0.074) were significantly decreased compared with non-transfected group(0.983±0.081)and sham pIRES-transfected group(0.994±0.142) (P<0.05).Conclusions The molecular chimeric pre-T cells infusion can reduce spleen T lymphocyte response to allogeneic T cells and it may induce immune tolerance in vivo.

Background Diabetic cataract is one of the major ocular complications in diabetes mellitus, including cortical cataract, nuclear cataract, subcapsular cataract and mixed cataract, and different cataractogenesis may be associated with lens epithelial cells (LECs).Subcapsular cataract is one of diabetic cataracts.Studying the biological behavior of LECs in subcapsular cataract is crucial for prevention and treatment. Objective This study was to investigate the effects of Src-family tyrosine kinases (SFKs) on apoptosis and epithelial-mesenchymal transition (EMT) of human LECs cultured by high glucose. Methods Human LECs (HLE-B3) were cultured for 24 hours with DMEM containing 5.5 mmol/L glucose (normal control group),DMEM containing 35.5 mmol/L glucose (high glucose group) and DMEM containing 35.5 mmol/L glucose + 10 μmol/L PP1,a specific inhibitor of SFKs (PP1 group). In 3,6,12 and 24 hours after culture, the apoptosis of human LECs was detected by flow cytometry assay;morphological change of human LECs was observed under the inverted microscope, and the expressions of the markers of EMT,E-cadherin and a-smooth muscle actin (α-SMA),in the cells were detected by immnofluorescence staining. In addition, the alternations of p-Src418 (active c-Src),bcl-xl,survivin,caspase-3,E-cadherin and α-SMA proteins were assayed by Western blot analysis. Results An elevated expression level of p-Src418 was found in LECs in the high glucose group and peaked 6 hours after cultured. The expressions of p-Src418(grey levels) were 0.125 ±0.036 in the high glucose group, and which was significantly higher than 0.042±0.011 in the normal control group and 0.035 ± 0.018 in the PP1 group, respectively (both at P<0.01).No remarkable differences were seen in the apoptotic rates between the high glucose group and normal control group in 6,12 and 24 hours after culture (all at P>0.05).The apoptotic rates of human LECs were(6.433±2.084)%,(10.333±2.610)% and (8.033±2.967)% in the PP1 group, which were higher than (3.233 ± 1.320) %,(3.533 ± 1.159) %,(5.733 ±0.230) % in the high glucose group and (3.133±1.170)%,(2.833±0.751)%,(3.333±1.201)% in the normal control group (all at P<0.05),however,there were significant differences in the apoptosis between the high glucose group and the normal control group (all at P>0.05).In 6 hours and 12 hours after cell culture, the expression levels of bcl-xl and survivin (grey values) in human LECs were significantly declined, but the expression of caspase-3 was increased in the PP1 group compared with the the high glucose group and the normal control group (all at P<0.05).The LECs showed slender in shape 24 hours after culture in the high glucose group, but the cell shape was close to the normal in the PP1 group.Western blot and immunofluorescence assay revealed that the expression of E-cadherin in human LECs was significantly reduced and that of α-SMA was significantly increased 6 hours after culture in the high glucose group compared with the PP1 group and the normal control group (all at P<0.05).Conclusions High glucose activates c-Src kinase of LECs in high glucose environment and therefore induces EMT and inhibits apoptosis.However,PP1 impedes the biological process of EMT and apoptosis of LECs to maintain the epithelial characteristics even under the stress of high glucose.

To improve bond selectivity of recombinant β-glucuronidase in Escherichia coli (PGUS-E), based on the PGUS-E structure guidance, three key points R329, T369 and N467 were identified to be responsible for the bond selectivity of PGUS-E, and further saturation mutagenesis was conducted. Two positive mutants R329K and T369V were obtained by a combined selection technique of thin-layer chromatography and high performance liquid chromatography. Compared to PGUS-E, the bond selectivity of mutants R329K and T369V increased by 26.9% and 34.3%, respectively; whereas the biochemical properties such as pH and temperature profile were unchanged. Nevertheless, the activity was decreased compared to PGUS-E. These results further confirmed that sites R329 and T369 played important roles for the bond selectivity and activity. In summary, this study significantly increased the bond selectivity of PGUS-E by structure guided saturation mutagenesis, providing experimental support for elucidating the relationship between the structure and function of PGUS-E.

Chinese Journal of Microsurgery 2017;40(4):337-341
Objective To explore the related factors with skin flap necrosis,we concluded the cases of patients with skin defects after free flap plantation. Methods From 2001 to 2016,188 cases about 20 influencing factors were analyzed (The characteristics of patients:age, sex, smoke, diabetes, high blood pressure;Preoperative factors:injured sections, injured causes, preoperative wound infection, preoperative wound osteomyelitis, the time from injury to operation;Intraoperative factors:operator, operation time, anesthesia time, intraoperative rehydration fluids, the way of vascular anastomosis, the number of venous anastomosis, the area of flap;Postoperative factors:flap hematoma, flap infection, vascular crisis) and multivariate logistic regression analysis was used to analyze the relationship between these risk factors and flap necrosis. Results All 188 cases were treated with free anterolateral thigh flap to repair soft tissue defect and it was revealed that the 174 cases were successful (92.55%) and 23 cases were occured vascular crisis (12.23%),8 cases were arterial crisis,11 cases were vein crisis,4 cases were ateriovenous crisis. After the treatment, the rescue was successful in 5 cases (38.46%).After the analysis we made the conclusion that the number of venous anastomoses, flap hematoma and vascular crisis were related with the skin flap necrosis. Conclusion The number of venous anastomose (≥2) will increase blood return to make the flap easier to survive.Intraoperative stanching and drainage tube placement work will reduce the skin flap hematoma as a result of reducing the skin flap necrosis.Artery and venous crisis handled in time, can enhance the survival rate of flap.

Objective To study the characteristics of EEG after tramsient ischemic attack and to offer reference for screening procedure of aircrew and astronaut selection. Method The dominant frequency uncertainty of alpha band EEG in 12 pilots(males; age 30±5) with transient ischemic episodes in middle cerebral artery(MCA) territories and in 20 normal healthy pilots was analyzed with frequency-fluctuation analysis. Result The dominant probability of the main frequency coinciding with sites affected by transient ischaemic attack(TIA) in patient pilots was higher than that in healthy pilots (P<0.01),and the dominant probability ratio logarithmic index I≥0 in all patient pilots with normal EEG, but I<0 in all healthy pilots. It was also found that not only I≥0, but the second component shifted to lower frequency(8 Hz) in patients with slight focal EEG alterations,i.e. slowing of frequency. The relative entropy values (percentage) were decreased significantly in pilots with TIA as compared with healthy pilots (P<0.05). Conclusion The dominant frequency uncertainty analysis of alpha band showed clear superiority of computerized evaluation over routine visual assessment for the diagnosis of minor cerebral ischemia. It offers not only a possibility of studying pathophysiological functional parameter, but also the reference for screening procedure in aircrew and astronaut selection.
