1.Study of P53 in Cutaneous Squamous Epithelial Cell Tumour

Gene mutation is known to play an important role in carcinogenesis.There are two types of p53 gene,wild-type and mutant-type.In order to observe the relationship between p53 gene mutation and cutaneous squarnous epithelial ceil tumours,immunohistochemical study was carried out for p53 ex- pression in the epidermis from 29 patients with cutaneous squamous epithelial cell tumouts using a new immunohistochemical technique labelled streptavidin biotin system.The results showed that p53 positive staining was only confined to areas of atypical proliferation of epithelia in 3/6 cases of actinic keratosis. 6/10 cases of Bowen's disease and 8/13 cases of squamous cell carcinoma showed that p53 positive staining was seen throughout the tumour.It is suggested that p53 gene mutation might play an impor- tant role in cutaneous squamous epithlial cell tumour formation.
2.Effects of Yiweining recipe on expressions of metalloproteinase-2 and cyclooxygenase-2 mRNAs in ectopic endometrium of rats with endometriosis.

OBJECTIVE: To explore the effects of Yiweining Recipe (YWNR), a compound Chinese herbal medicine, on expressions of metalloproteinase-2 (MMP-2) and cyclooxygenase-2 (COX-2) mRNAs in rats with endometriosis (EM). METHODS: Operational self-transplantation was applied in establishing the rat models. Detection of MMP-2 and COX-2 mRNAs was conducted with hybridization in situ. RESULTS: There were significant differences in the expressions of MMP-2 and COX-2 mRNAs between the untreated group and the high-dose YWNR-treated group. YWNR could reduce the expressions of MMP-2 and COX-2 mRNAs. CONCLUSION: YWNR can treat EM through reducing the positive expressions of MMP-2 and COX-2 mRNAs.
3.OX40 stimulation down-regulates the expression of Foxp3 in CD4+ CD25+ regulatory T cells

Objective To evaluate the regulatory effect of OX40 co-stimulatory signal on the expression of Foxp3 in inductive regulatory T cells (iTreg) in vitro.Method CD4+ CD25+ naive T cells were isolated from C57BL/6 mouse lymphocyte suspension by MASC CD4+ CD25+ regulatory T cell isolation kit.Inductive Tregs were generated by stimulation of naive T cells in the presence of transforming growth factor beta (TGFβ1),anti-CD3,anti-CD28 and IL-2.The regulatory effect on iTregs was shown by use of OX40 stimulation monoclonal antibody (OX86) or control antibody.Using flow cytometric analysis (FACS),we examined the antibody-based identification of Tregs surface markers CD4 and CD25,along with the intracellular activation marker FoxP3.Results The ratio of CD4+ CD25+ nTregs isolated from mouse lymphatic node was (5.0 ± 0.4)% vs.(71.8 ± 13.4)% of TGFβ1-driven iTregs.The ratio of CD4+ CD25+ Tregs was (80.0 ± 1.6) % in OX40 stimulation McAb group vs.(86.0 ± 1.4)% in control antibody group.Furthermore,the expression of Foxp3 was (59.2 ± 0.7) % in OX40 stimulation McAb group vs.(70.0 ± 0.8) % in control antibody group (P<0.05).Conclusion TGFβ1-dependent protocol may induce the conversion of naive CD4+ T cells into CD25+ Foxp3+ iTregs.OX40 stimulation can down-regulate the expression of Foxp3 in CD4+ CD25 + iTreg significantly.Thus OX40 molecular may become an attractive target in Tregs-induced transplant tolerance.Further study should be performed to increase the suppressive activity of iTregs through blockade of OX40 signal.
4.Interactions between proteins and cation exchange adsorbents analyzed by NMR and hydrogen/deuterium exchange technique.

In silico acquirement of the accurate residue details of protein on chromatographic media is a bottleneck in protein chromatography separation and purification. Here we developed a novel approach by coupling with H/D exchange and nuclear magnetic resonance to observe hen egg white lysozyme (HEWL) unfolding behavior adsorbed on cation exchange media (SP Sepharose FF). Analysis of 1D 1H-NMR shows that protein unfolding accelerated H/D exchange rate, leading to more loss of signal of amide hydrogen owing to exposure of residues and the more unfolding of protein. Analysis of two-dimensional hydrogen-hydrogen total correlation spectroscopy shows that lysozyme lost more signals and experienced great unfolding during its adsorption on media surface. However, for several distinct fragments, the protection degrees varied, the adsorbed lysozyme lost more signal intensity and was less protected at disorder structures (coil, bend, and turn), but was comparatively more protected against exchange at secondary structure domains (α-helix, β-sheet). Finally, the binding site was determined by electrostatic calculations using computer simulation methods in conjunction with hydrogen deuterium labeled protein and NMR. This study would help deeply understand the microscopic mechanism of protein chromatography and guide the purposely design of chromatographic process and media. Moreover, it also provide an effective tool to study the protein and biomaterials interaction in other applications.

5.Ultrasound monitoring of cervical canal length combined with measurement of fibronectin in cervical secretion for predicting preterm delivery

The length of cervical canal was monitored by perineal ultrasonography in 377 pregnant women with 22 to 36 weeks of gestation,at the same time the contents of fibronectin (fFN) in cervical secretions were measured.The rate of cervical canal length shortening from the time of first visit to the appearance of preterm signs was calculated.When the cervical length shortening rate was 16% ~ 60%,fFN (+) was correlated with higher preterm birth rate (P <0.01),while when the shorten rate > 60% there was no significant difference in preterm delivery rate between fFN (+) and fFN (-) groups (P > 0.05).The results indicate that the monitoring of cervical canal length combined with fiberonectin measurement in cervical secretion may better predict pretenn birth.
6.Controlled study of nimodipine in treatment of patients with diffuse axonal injury.

OBJECTIVE: To determine the efficacy and safety of early intervention with nimodipine treatment in diffuse axonal injury. METHODS: Based on the characteristic radiological signs and criteria for diffuse axonal injury (DAI), 89 patients with the diagnosis of DAI were enrolled in this randomized, double-blind, placebo-controlled trial. RESULTS: Nimodipine proved to be safe and well tolerated. With TCD sonography we found that there was a higher incidence of cerebral vasospasm in this series (38.2%). Overall, Nimodipine produced a better clinical result than placebo, but there was no statistically significant difference in favorable outcome at 3 months after injury (P=0.11) between the two groups. A trend toward a favorable effect was suggested by the analyses in two small subgroups, either in the patients suffering from clinical Grade III DAI (P=0.04), or in those with the TCD-evidence of cerebral vasospasm during clinical observation (P=0.049). CONCLUSIONS: We postulate that a clinically valuable benefit is possible with nimodipine treatment in DAI patients. However, the effects on outcome should be verified by further controlled study.
7.Association between bullying and family factors among middle school students in a city of central China

Objective:
To explore the relationship between bullying among middle school students and family factors in a city of central China, so as to provide support for the prevention and reduction of school bullying among middle school students.
Methods:
The stratified cluster random sampling method was used to investigate the bullying involvement and family factors of 2 996 middle school students from first grade in junior high school to third grade in high school in a city in central China. Chi-square test and Logistic-regression analysis were used to analyze the relationship between family factors and bullying participation of middle school students.
Results:
Among 2 996 students, 390 students(13.0%) were found of having bullying behavior, and 1 127 students(37.6%) were found of being bullied. Univariate analysis showed that there were statistically significant differences in family factors such as whether she or he is the only child, father-child relationship, mother-child relationship, marital status of parents, whether the mother work away from hometown, education level of father and mother(χ2=8.88, 56.49, 30.85, 30.91, 3.89, 10.36, 11.72;25.00, 69.33, 46.76, 57.09, 3.93, 23.19, 45.49, P<0.05). Logistic regression analysis showed that the only child and mother’s education degree was junior college and below were the risk factors for middle school students’ bullying involvement (OR=1.37,1.39). Parents’ harmonious marital status and father’s not working outside are the protective factors of middle school students’ bullying(OR=0.53, 0.83).The only child is the risk factor of bullying in middle school students (OR=1.42), and good father relationship is the protective factor of bullying in middle school students (OR=0.38).
Conclusion
Family factors have a certain impact on the involvement of middle school students in bullying in a city of central China, and corresponding preventive measures should be formulated from the perspective of family to focus on the intervention of high-risk groups.
8.Application of integrated nursing process in intravenous thrombolysis in patients with cerebral infarction in superacute period

Objective To study the impact of integrated nursing process in intravenous thrombolysis in patients with cerebral infarction in superacute period.Methods A total of 31 intravenous thrombolysis patients from January to December 2015 were as experimental group given integrated nursing process and 31 cases from May to December 2014 were as control group given conventional nursing,thrombolysis patients' behavioral life quality by NIHSS,dementia by MMSE and depression by CES-D before and after thrombolysis were compared.Results There was no significant different in experimental group and control group in NIHSS score,MMSE score and CES-D scores before thrombolysis;while there was statistically significance between the two groups in NIHSS scores after thrombolysis in 24 hours,in 15 days or in one month(P <0.05).There was no statistically significance between the two groups in MMSE scores after thrombolysis in 24 hours (P > 0.05),but was statistically significant in 15 days after thrombolysis and one month (P < 0.01).There was statistically significant difference between the two groups in CES-D scores in 24 hours,15 days and 1 month after thrombolysis (P < 0.05).Conclusion Quality of care for the patients with acute cerebral infarction thrombolysis associates with their behavioral life quality,dementia and depression.Integrated nursing process improves their behavioral life quality,and relieves dementia and depression of patients with acute cerebral infarction thrombolysis.

10.Practice of communication strategies of health scientific popularization of menopausal women

This paper aims to push health scientific popular knowledge and carry out targeted patient health education for certain patients, based on the demand of health education for specific diseases and population. Taking the female climacteric health scientific popularization as an example, the authors summarized the practice of popularizing scientific education in a tertiary specialized hospital based on the cognitive level and health education needs of outpatients and the general public. A multidisciplinary health scientific popularization team was set up to communicate popular science knowledge to patients and other menopausal women in need through multi-channel and multi-platform forms. Such means include online ones, offline ones, and cooperation with various social organizations. The implementation of health scientific popularization mode meets the needs of patients and the general public. The implementation of this mode of health popularization could improve their self-health management ability and health accomplishment. It provides a good reference for public hospitals to carry out health science popularization.
