1.The relationship between urodynamic findings and detrusor changes of diabetic cystopathy

150 ml in 22 patients (31%).Ten patients (14%) had detru-sor instability.Six patients were incapable to void during the test. The volume of first bladder sensation was(203.25?107.53)ml (range,125 -630 ml) in the diabetes patients.The bladder capacity was (428.09?227.89)ml (range,220 -1350 ml).The maximum flow rate was (10.70?3.27) ml/min.The residualurine volume was (100.57?108.08) ml.In early stage group the volume of first bladder sensation was(151.67?24.07) ml;while in progressive stage group it was (268.16?13.90)ml,and bladder capacitywas (592.97?252.51)ml.The maximum flow rate was (8.61?2.04) ml/min. PQmax was (33.16?19.81)cm H2O (1 cm H2O=0.098 kPa).The residual urine volume was (169.03?137.25) ml. Theseparameters were all abnormal. The changes in excitability and autorhthmicity of bladder detrusor in experimental non-insulin dependent diabetes mellitus (NIDDM) rats were observed. Sixty-nine NIDDM rats as NIDDM group and 69 normal rats as control group were enrolled into this experimental study. At 6th, 10th, 14th, 18th, 22nd and 26th week after the rats were injected last time, the changes in the excitability and autorhthmicity of detrusor strips in vitro were observed. The results showed that the threshold of the tension which made the detrusor strips contract was significantly higher in NIDDM group (0.716 +/- 0.325 g) than in control group (0.323 +/- 0.177 g) (F = 59.63, P < 0.001). At different stages, the threshold of the tension resulting the contract of the detrusor strips in NIDDM group was also higher than in control group. At 18th week after STZ injection, the frequency of spontaneous contract of the detrusor strips in NIDDM was significantly higher than in control group (P < 0.05), whereas at 22nd week, that in NIDDM group was significantly lower than in control group (P < 0.05). It was concluded that the decreased excitability of the bladder detrusor was the earliest and most obvious changes in bladder function in diabetes rats and the autorhthmicity had also changed at the early stage of diabetic bladder.

4.Impacts of platelet-derived growth factor-D on prostate cancer cells migration

Objective To investigate the effect of platelet-derived growth factor-D (PDGF-D) on the prostate cancer cells migration and its possible mechanism. Methods The expressions of PDGF-D in LNCaP and PC-3 cells were detected with western blot.PDGF-D siRNA was synthesized according to mRNA sequence of PDGF-D gene and was transfected into PC-3 cell.The cells were treated with PDGF-D and PDGF-D siRNA,the cell migration was examined by Boyden chamber migration assay.The expression changes of VEGF and MMP-9 mRNA were detected by RT-PCR. Results The results of western blot indicated that the PDGF-D protein expression level was lower in LNCaP cells (29.47 ± 1.68) than that in PC-3 cells (63.43 ±2.10),(P < 0.05).PDGF-D siRNA could down-regulate the PDGF-D protein expression in the transfected group (35.19 ± 1.51).The exogenous PDGF-D could promote migration of LNCaP and PC-3cells,and up-regulate the expression of VEGF,MMP-9 mRNA in PC-3 cells (P < 0.05,compared with control group).PDGF-D siRNA inhibited PC-3 cells' migration and decreased the level of VEGF,MMP-9mRNA expression (0.72 ± 0.09 vs 0.43 ± 0.18,0.65 ±0.07 vs 0.22 ± 0.08) (P < 0.05). Conclusion PDGF-D is involved in the promotion of prostate cancer invasion and angiogenesis.
5.Computational fluid dynamics study on the effects of morphology of prostatic urethra in mid-voiding after minimally invasive surgery with BPH

It is important to investigate the mechanical effects of morphology of prostatic urethra (PU) for benign prostatic hyperplasia (BPH). PU and bladder neck transverse diameter ratio (RPU-1), which related to vortex, and their influence on urine flow were observed by CFD. The results showed that vortexes appeared and expanded with increasing RPU on both sides of PU when RPU-1>0.79, and velocity of external urethral orifice decreased gradually. CFD is an available method for urodynamics research. The quantitative simulation of the relationship between RPU-1 and vortex was proposed for the first time, providing a new idea for advancing theory of PU lumen repair in BPH.
6.Effectiveness and safety of sodium glycididazole for nasopharyngeal carcinoma radiotherapy:a Meta-analysis

Objective To assess the efficacy and safety of radiotherapy combined with sodium glyci-didazole (CMNa)in treating nasopharyngeal carcinoma by conducting a Meta-analysis.Methods Randomized clinical controlled studies (RCTs)of radiotherapy plus CMNa versus radiotherapy alone in the treatment of nasopharyngeal carcinoma were retrieved from China Biology Medicine disc,China National Knowledge Infrastruc-ture,digital journal of Wanfang Data,China Science and Technology Journal Database,PubMed and Cochrane Library.According to the inclusion and exclusion criteria,data extraction and quality assessment were done by two researchers independently,and clinically important measures were selected.Outcomes were pooled with Rev-Man 5.2.Results The rate of complete remission (CR)in the radiotherapy combined CMNa group was better than that in the radiotherapy alone group in 1 .41 multiple to primary lesion,RR =1 .41 ,95%CI:1 .30-1 .52, Z =8.20,P =0.00.To lymphonodi cervicales metastasis,the rate of CR in the radiotherapy combined CMNa group was better than that in the radiotherapy alone group in 1 .45 multiple,RR =1 .45,95%CI:1 .30-1 .62, Z =6.50,P =0.00.There were no significant differences in the incidence rates of adverse reactions of skin (RR =0.97,95%CI:0.87-1 .08,Z =0.57,P =0.57 ),mucosa (RR =1 .03,95%CI:0.93-1 .15,Z =0.65, P =0.52 )and decrease in white blood cells (RR =0.84,95% CI:0.56-1 .25,Z =0.87,P =0.38). Conclusion In the treatments of nasopharyngeal carcinoma,the rates of CR of primary lesion and lymphonodi cervicales metastasis in radiotherapy combined with CMNa group are superior to the radiotherapy alone group. However,the occurrence rates of adverse reactions in the two groups have no obvious differences.

The pathogenesis of diabetic cystopathy(DCP)is complex and early diagnosis is hindered by a lack of specific and sensitive criteria and more research is needed to establish guidelines for its clinical diagnosis and treatment.Exosomes are membrane vesicles carrying various biological information such as proteins and nucleic acids, which are critical for communication between different tissues and organs.This review discusses the potential role of exosome miRNA in the pathogenesis of DCP and its application in therapy, aiming to provide new insight into the diagnosis and treatment of DCP.

ObjectiveTo understand the possible role of some proteins expressed by human synovial fibroblasts(SFs)in the pathogenesis of rheumatoid arthritis.MethodsThe expression difference of synovial fibroblast proteins between rheumatoid arthritis(RA)patients and healthy controls was analyzed by 2-DE.The differential expression spots were identified by matrix-assisted laser desorption ionization-time-of-flight mass spectrometry(MALDI-TOF-MS),followed by bioinformatics analysis,some of which were validated by Western blot.ResultsUsing 12% SDS-PAGE following pH 4-7 IPG strips in IEF,averagely 837 and 852 protein spots were detected in RA patients and normal subjects,respectively.Gel image analysis revealed that there were 49 differential protein spots.By peptide mass fingerprinting strategy,we identified 40 protein spots derived from gels of SFs in RA patients among differential spots and 23 valid proteins were obtained.Western blot analysis showed that expressions of Enolase ?,Annexin I,Cathepsin D,SOD2,Peroxiredoxin 2 were significantly higher in SFs from RA patients than those from normal subjects,which was consistent with proteome analysis.ConclusionThe differential proteins might be involved in inflammation of synovitis in RA.

10.Identification and study of regulatory axis that miRNA-224-5p targets NEDD4L gene in clear cell renal cell carcinoma

Objective:To screen key genes of renal clear cell carcinoma based on bioinformatics methods, identify possible microRNA (miRNA)-mRNA action axis, and explore the expression of related genes in clear cell renal cell carcinoma tissues and cells.Methods:Gene expression profiles of GSE40435 and GSE71302 datasets were obtained from the Gene Expression Omnibus (GEO) database. TCGA-KIRC datasets were obtained from The Cancer Genome Atlas (TCGA) database. R software was used to identify the differentially expressed mRNA and miRNA, and the functional enrichment analysis was performed. STRING database and Cytoscape software were used to perform the protein interaction analysis. The prognosis-related differentially expressed miRNA was evaluated by the Oncomir database. The potential targeted genes regulated by miRNA were determined by using TargetScan and miRDB targeted gene prediction tools. The tissue samples and clinicopathological features of 34 patients with clear cell renal cell carcinoma in the First Hospital of Shanxi Medical University from June to December 2021 were collected, and normal renal cell line 293T and clear cell renal cell carcinoma cell line 786O were selected. The real-time fluorescence quantitative polymerase chain reaction (qRT-PCR), was used to detect the relative expression of genes; Western blotting and immunohistochemical staining were used to detect the expression levels of the targeted proteins. The dual luciferase reporter gene assay was carried out to verify the targeting relationship between genes.Results:A total of 1 351 differentially expressed mRNA and 50 differentially expressed miRNA were screened and identified. The result of functional enrichment analysis suggested that the fatty acid metabolism pathway and xenobiotic metabolism pathway were suppressed in clear cell renal cell carcinoma, while the apoptosis and immune response pathways were activated. Protein interaction analysis suggested that the signal transduction and protein ubiquitination pathways might play a key role in clear cell renal cell carcinoma. The screening results showed that miRNA-224-5p (miR-224-5p) was most closely associated with clear cell renal cell carcinoma progression and was highly expressed in tumor tissues, and its prognosis-related target gene was NEDD4L. The relative expression of NEDD4L mRNA in clear cell renal cell carcinoma tissues and paraneoplastic tissues were 0.138±0.103 and 1.000±0.026 ( t = 46.23, P < 0.05), and the relative expression of miR-224-5p was 1.000±0.043 and 0.129±0.108 ( t = 45.28, P < 0.05). The differences of NEDD4L mRNA and miR-224-5p expressions in different grades and stages of clear cell renal cell carcinoma tissues were statistically significant (all P < 0.05). The expression of NEDD4L protein was decreased in clear cell renal cell carcinoma. The relative expression of NEDD4L gene in 293T and 786O cells were 1.000±0.125 and 0.210±0.044 ( t = 17.52, P < 0.05); the relative expressions of miR-224-5p gene were 0.209±0.049 and 1.000±0.234 ( t = 10.61, P < 0.05). The relative expressions of NEDD4L mRNA in miRNA mimic group and negative control group were 0.236±0.062 and 1.000±0.024, and the difference was statistically significant ( t = 43.56, P < 0.05). NEDD4L protein expression was reduced in the miRNA mimic group. Dual luciferase reporter gene assay suggested that NEDD4L was a direct target gene of miR-224-5p. Conclusions:In clear cell renal cell carcinoma, miR-224-5p targets and regulates NEDD4L expression, and this mechanism may be related to carcinogenesis and progression of clear cell renal cell carcinoma.
