1.Construction and identification of the recombinant eukaryotic expression plasmid pHsp70-hsv-tk

Objective: To construct the recombinant eukaryotic expression plasmid pHsp70-hsv-tk(pHT)for further target gene therapy research.Methods: The oligomers of primers were synthesized based on the reported sequences of hsv-tk and with the pHSV-106 plasmid as the template.Then the hsv-tk gene fragment was amplified and inserted into the pD3SX vector.E.coli DH5? was transformed with the recombinant plasmids and the bacterial colonies containing the recombinant plasmids were identified by the PCR cloning method,and the recombinant plasmids were extracted and purified.Results: The recombinant eukaryotic expression plasmid pHT was obtained.The target gene obtained by PCR amplification had the same molecular size as predicted.The recombined pD3SX plasmid contained correct recombinant hsv-tk sequences.Conclusion: The plasmid pHT was successfully constructed,which can be used for further target gene therapy research.

3.The effect of integron on the drug-resistant of Acinetobacter baumannii

Objective To investigate the drug-resistant status of Acinetobacter baumannii in clinical strains isolated from our hospital,and get to know the distribution of integrons carried by Acinetobacter baumannii,analyze the correlation between integron and drug-resistant of Acinetobacter baumannii..Methods Collected 85 Acinetobacter baumannii isolated in our lab,the drug-sensitivity tests were completed by the method of KB,the integrase genes were amplified by polymerase chain reaction(PCR),and identified the type of integron..Analyzed the correlation between the drug-resistant and integron in Acinetobacter baumannii..The open reading frame of integron were amplified by PCR,the polymorphism of integron was identified,and sequenced the PCR products.Results Except for Imipenem and Cefoperazone/ sulbactam(drug resistant rate less than 10%),the drug resistant rate of other 15 antibacterial was no more than 30% among the 85 Acinetobacter baumannii isolated in our lab..66.1 %(57/85) stains carried the integrons I,we didn't find out the integron Ⅱ and Ⅲ.The drug-resistant rates of Acinetobacter baumannii with integron were higher than that of Acinetobacter baumannii without integron.The length of amplified products in variable region of open reading frame of integron varied from 03 to 25kb,.The sequencing results confirmed the integron included multidrug resistant gene code.Conclusion In our hospital,the drug-resistant rate of Acinetobacter baumannii was high,and the majority were multidrug resistant strains.The drug-resistant rate of Acinetobacter baumannii was enhanced with integron,The multidrug resistant of Acinetobacter baumannii was associated with integrons.
4.Askin Tumors in Children

Objective To analyze the clinicopathological features and enhance the knowledge of Askin tumor in children. Methods The clinicopathological features of 4 cases of children with Askin tumor were described. Results Clinically, the tumor was often seen in elder child patients with painful thoracopulmonary masses.Histopathologically, the nest-like or lobular structures composed of small round-to-oval cells were surrounded by sparse fibrovascular stroma. Among tumor cells there were no or few resticular fibers,however in the surrounding of tumor masses the fibers were found. PAS staining in the tumor in a part of cases was positive. The positive rates of CD99 and Syn detected by immunohistochemistry were high. Neurosecretory granules can be observed by electron microscopy. Two patients received both chemotherapy and radiotherapy three patients died of metastatic tumor of lung and brain and chemotherapy complication in 24,25 and 5 months, respectively. Conclusions Askin tumor is a highly malignant neurosecretory tumor in thoracopulmonary region, which often be misdiagnosed as neuroblastoma, lymphoma, embryonal rhabdomyo sarcoma, tuberculosis and lung abscess, and requires a synthetical treatment plan consisting of pre-and postoperative chemotherapy, radiotherapy and surgical resection. The treatment methods of Askin tumor needs to be further studied.
5.Analysis of Autologous Peripheral Blood Stem Cell Transplantation on Malignant Tumors in Children

20?10 9/L) were 12 (8-20), 17(10-28) and 21 days (7-78), respectively. The average follow-up period was 7 3 years(ranged from 1 month to 14 years). The overall survival (OS) and death rates after transplantation were 70%(18/26) and 30%(8/26) respectively. Transplant-related mortality was 7 7%(2/26). Conclusion The combination of chemotherapy and G-CSF is an effective and low toxicity protocol for the mobilization of peripheral blood stem cells. APBSCT can be safely performed in children with malignant tumors.
6.Evaluation and Countermeasures on OTC Cough and Cold Medicines for Children in New Zealand

OBJECTIVE:To introduce positive response to the security and effectiveness issues on OTC cough and cold medicines for children in New Zealand in order to provide reference for drug risk-benefit evaluation and putting forward reasonable management measures in China. METHODS:The concept of OTC cough and cold medicines for children were introduced firstly. Then the evaluation and countermeasures of OTC cough and cold medicines for children in New Zealand were also introduced. RESULTS & CONCLUSIONS:It has significance for the evaluation of drugs for sale that are review and evaluation,study and countermea-sures and dynamic tracking for OTC cough and cold medicines for children in New Zealand and the realistic attitude of New Zealand Drug and Medical Instruments Security Bureau.

8.Analysis of Helicobacter pylori infection in different groups of population in Yangzhou

Objective To investigate the infection status and distribution characteristic of Helicobacter pylori (Hp)in different groups of population in Yangzhou.Methods 2 917 individuals from outpatients,hospitalized patients,and physical examination pop-ulation were recruited in the study.Serum samples from the subjects were rapidly detected using Helicobacter pylori urease antibody detection kits(Colloidal Gold method).The results were statistically analyzed.Results The totally Hp-detection rate of the 2 917 individuals was 32.9% (959/2 917).The Hp-detection rates of outpatients,hospitalized patients,and physical examination popula-tion were 33.1%,34.5% and 30.0% respectively.There were no significant difference in the positive rates between the male and female individuals(P >0.05).Overall,The Hp-positive rates increased with age.The minors(juveniles and children)had the lowest Hp-positive rate 16.0%(285/1 529);The Hp-positive rate of young adults was 34.0%(221/650),which was at the middle;elderly group had the highest Hp-positive rate 61.38%(453/758).59.8% of the patients diagnosed with gastrointestinal diseases were Hp positive.Among which,patients with peptic ulcer(74.4%)and gastric cancer(70.6%)have the highest detection rates.Some pa-tients with blood diseases,cardiovascular disease and some other diseases were also found with high Hp-detection rates.Conclusion The distribution of the Hp infection in population is positively correlated with age,but not with gender.Gastrointestinal diseases, especially peptic ulcers and gastric cancer,have high Hp infection rates,the relationships of Hp infections with some other diseases cannot be ignored and need to be further explored.
9.Analysis of chromosome 9 open reading frame 72 intermediate hexanucleotide repeat expansions in familial amyotrophic lateral sclerosis population from mainland China

Objective To identify the existence of chromosome 9 open reading frame 72 (C9ORF72) intermediate hexanucleotide (GGGGCC) repeat expansions as well as the mutation-related clinical phenotypes in familial amyotrophic lateral sclerosis (fALS) population from mainland China.Methods DNA samples and clinical data of 62 fALS cases were collected,then the pathological GGGGCC repeat expansions were detected.Haplotype analysis for 20 single nucleotide polymorphisms (SNPs) was performed finally.Results The C9ORF72 intermediate repeat expansions were found in one fALS patient (1.61%,1/62),and repeat number was 24.In addition,2 siblings of this fALS patient also had a 24 repeats allele.These 24 repeat alleles were associated with the pathogenic founder haplotype.Conclusions This study illustrates the existence of the C9ORF72 intermediate repeat expansions in fALS population from mainland China for the first time.An intermediate repeat length in C9ORF72 is related to fALS in mainland China.
10.A clinical and neurophysiologic study of upper motor neuron-dominant amyotrophic lateral sclerosis

Objective To investigate the clinical and electrophysiological characteristics of upper motor neuron-dominant amyotrophic lateral sclerosis (UMN-D ALS ).Methods The clinical and electrophysiological characteristics were analyzed retrospectively in 76 patients with UMN-D ALS and 19 patients with primary lateral sclerosis (PLS).Electrophysiological study included the examination of median nerve,ulnar nerve,tibial nerve,peroneal nerve and sural nerve except for the electromyogram of bulbar region,cervical region,thoracic region and lumbosacral region.Results The diagnosis in 8 PLS patients were changed to UMN-D ALS after detailed review.In UMN-D ALS patients,there were more females in age group of older than 40 (male:female =1∶1.37) and 32 patients (38.1%) had onset with bulbar dysfunction.UMN-D ALS patients showed electromyograph evidence of denervation at 30 months (median)after onset,and clinical lower motor neuron (LMN) signs 6 months later.Seventy-seven patients (91.6%)developed LMN sign by 4 years from symptom onset.ALS functional rating score-revised changed from 40 ±3 to 32 ±4 in 4 years in U MN-D ALS patients (t =1.83,P < 0.05 ).The amplitude of motor unit action potential (MUAP) on the first interosseus dorsalis was higher (( 1003.7 ± 25.2) μV vs (353.5 ±21.5) μV,t=2.34,P <0.05) and the duration of MUAP was longer ((19.8 ±2.3) ms vs (9.6 ±1.3) ms,t =1.85,P <0.05 ) in UMN-D ALS patients than in PLS patients.Conclusions UMN-D ALS occurs more in female cases,with more bulbar onset and with faster progression than PLS does.It also presents focal denervation in electromyograph.
