Objective:To investigate the effect of human Leptin on the proliferation of human circulating T lymphocytes in vitro and the influence of Leptin on the action of PHA on the proliferation of human circulating T lymphocytes. Methods:PBMC were isolated from hepartinized venous blood of normal donors by density-gradient sedimentation over Ficoll-Hypaque and cultured in the 1640 complete medium.T lymphocytes were then obtained by depleting the monocyte and B cell populations.Cells were then cultured in the 1640 complete medium for 56 h in the presence of PHA.The proliferative effect was assessed by means of incorporation of thymidine.TdR was added and cultured for 16 hours.Cells were then processed in the harvester to measure the incorporation of [()~3H] thymidine.Radioactivity was determined by liquid scintillation counter. Results:The results showed that Leptin alone did not affect the proliferation of T lymphocytes, but it could enhance the effect of PHA on the proliferation of T lymphocytes at the concentrations of PHA from 2-8 ?g/ml.A dose-response studies of T lymphocyte proliferation showed that the maximal effect of Leptin were observed at 10 nmol/L.Leptin did not affect the proliferation of T lymphocytes when the concentrations of PHA were over 8 ?g/ml.Conclusion:The studies demonstrated that Leptin alone did not affect the proliferation of lymphocytes, but it could enhance the action of PHA on cell growth in a dose-dependent manner.

Objective To explore the relationship between apoptosis promoting effector molecules TFAR19,Apaf-1 and lubar intervertebral disc protrusion. Methods 99 of operation patients with lubar intervertebral disc protrusion in the First Affiliated Hospital of Guangxi Medical University Department of Spine and Osteoputhy were recruited.Among them, single segment of lubar intervertebral disc protrusion were 70 (Group A),more than one segments of lubar intervertebral disc protrusion were 29 (Group B).In addition,40 unrelated healthy people from physical examination center were enrolled as controls (Group C).Enzyme-linked immunosorbent assay (ELISA) was used to examine serum TFAR19,Apaf-1 levels in lubar intervertebral disc protrusion patients. Results The level of TFAR19 in Group A,Group B and Group C respectively were 1.85±0.14,2.33±0.25 and 1.30±0.09 ng/ml.The TFAR19 activity in each group was statistically significant difference (F=7.979,P<0.01).Compared with Group C,the TFAR19 activity in Group B (q=5.59,P<0.01),Group A (q=3.60,P=0.012) was statistically significant difference. The TFAR19 activity in lubar intervertebral disc protrusion patients subgroups (Group A,Group B) was statistically significant difference (q =2.93,P =0.012).The level of Apaf-1 in Group A,Group B and Group C respectively were 159.22±11.87,203.20±20.21 and 107.52±11.58 pg/ml.The Apaf-1 activity in each group was statistically significant difference (F=8.828,P<0.01).Compared with Group C,the Apaf-1 activity in Group B (q=5.86,P<0.01),Group A (q=3.89,P=0.007) was statistically significant difference. The Apaf-1 activity in lubar intervertebral disc protrusion patients subgroups (Group A,Group B) was statistically significant difference (q =2.97,P=0.037).Male/female ratio between each groups was not statistically significant difference (x2=0.229,P =0.892).Age between each groups was not statistically significant difference (F=0.091,P =0.91).Conclusion The augment of TFAR19 and Apaf-1 promotes apoptosis of lubar intervertebral disc protrusion. It is connected with quantity of protrusive segments. The more segments of protrusion, the higher TFAR19 and Apaf-1 level of examination will be.

With the continuous progress of research methodology in the real world and the growing maturity of artificial intelligence technology, a method for conducting “quantitative” research to guide clinical practice based on traditional Chinese medicine (TCM) diagnosis and treatment data was gradually developed. However, there is still a need for further improvements in the overall design of studies and the transformation of findings into clinical practice. Based on this, we put forward a comprehensive overall design concept and application approach for real-world study and artificial intelligence research based on clinical diagnosis and treatment data of TCM. This approach consists of five steps: Constructing a research-based database with a large sample size and high data quality; Mining and classification of core prescriptions; Conducting cohort studies to evaluate the effectiveness of core prescriptions; Utilizing case-control studies to clarify the dominant population; Establishing predictive models to achieve precision medicine. Additionally, it is imperative for researchers to establish a standardized system for collecting TCM variables and processing data, optimize the determination and measurement methods of confounding factors, further improve and promote methodologies, and strengthen the training of interdisciplinary talents. By following this research method, we anticipate that the clinical translation of research findings will be facilitated, leading to advancements in TCM precision medicine. Real-world study and artificial intelligence research share similar research foundations, and clinical applications complement each other. In the future, the two will merge together.
