1.Relationship between cerebral artery stenosis and obstructive sleep apnea

Objective To investigate the relationship of obstructive sleep apnea and atherosclerotic cerebral artery stenosis.Methods One hundred and twenty patients with acute cerebral infarction were enrolled in the study.Patients underwent polysomnography monitoring;according to the results, the sleep apnea/hypopnea index ( AHI) was ≥5/h in 60 patients ( OSA group) and AHI was <5/h in another 60 patients ( non-OSA group) .The degree, quantity and distribution of cerebral artery stenosis were compared between OSA and non-OSA group.Results The results showed that there were no significant differences in gender, age, NIHSS score and rates of diabetes, coronary heart disease, hyperlipidemia and smoking between two groups (P>0.05).The rates of increased BMI [(26.07 ±2.39)vs.(23.59 ±2.73) kg/m2] and hypertension [70%(42/60)vs.43%(26/60)] in OSA group were higher than those in non-OSA group (t=5.304,χ2 =7.603, all P<0.05).There were 47 (78.3%) patients in OSA group and 27 (45.0%) in non-OSA group presenting cerebral artery stenosis (χ2 =16.535, P<0.05), and both OSA group and non-OSA group displayed high proportion of intracranial artery stenosis ( 68.0% vs.81.5%, P>0.05 ) . The severity of cerebral artery stenosis in OSA group was higher than that in non-OSA group (χ2 =17.801, P<0.05).There were no significant differences in location distribution of stenosed cerebral arteries between two groups (P>0.05).Conclusion The study indicates that there is a close relationship between OSA and stenosis of atherosclerotic cerebral arteries.

3.Comparison of on -line hemodiafiltration and high -flux hemodialysis for correcting calcium and phosphate metabolic disorder in uremia patients

Objective To compare the effect on correcting calcium and phosphate metabolic disorder and the impact on quality of life in uremia patients between on -line hemodiafiltration and high -flux hemodialysis.Methods 90 patients with uremia,digital watches were randomly divided into QL -HDF group and HFHD group,Ca2 +,P3 +, serum parathyroid hormone(PTH)level were measured before and after dialysis blood.The survival quality of patients were evaluated by the KDQOL -SFTM1.3 scale.The incidence of adverse reactions were collected.The similarities and differences of two groups were compared.Results The comparison of Ca2 + levels in the two groups before and after dialysis showed no significant difference(P >0.05);Of the QL -HDF group after treatment,P3 + was (1.82 ± 0.19)mmol/L,PTH was (401.6 ±16.7)pg/L,which were significantly lower than those of QL -HDF group[P3 +(2.14 ±0.22)mmol/L,PTH(425.0 ±17.2)pg/L](t =24.256,21.059,all P <0.05)in addition to the burden of kidney disease;life(BKD),the other ten areas scores of the QL -HDF group were higher than those of the HFHD group;The symptoms and discomfort(SPL)and the effect of nephropathy in life(EKD),cognitive function(CF), quality(QSI),social function(SEXF)and sleep(SLEEP),patient satisfaction(PS)had significant differences in areas such as score(all P <0.05),working condition(WS)and social support(SOS),dialysis staff support(DSF) and other domain scores had no significant difference (all P >0.05 );6 cases of adverse reactions was in the QL -HDF group (13.33%),14 cases of adverse reactions was in the HFHD group(31.11%),there was significant difference between the two groups(χ2 =5.361,P <0.05).Conclusion The two methods can improve the metabolic disorders of calcium and phosphorus in patients with uremia,and QL -HDF showed stronger ability to remove the ability,and the quality of life is higher,it is worth promoting in clinical practice.
4.Screening the down-regulated genes in HepG2 cells transfected by IFN-?

Objective To screen the down-regulated genes in HepG2 cells transfected by pcDNA3.1(-)-IFN-? to further investigate the biological mechanism of IFN-?. Methods pcDNA3.1(-)-IFN-? was transfected into HepG2 cells as treatment group and the pcDNA3.1(-) transfected cells as the control to perform suppression subtractive hybridization (SSH). The subtractive library for down-regulated genes was obtained when pcDNA3.1(-) transfected HepG2 as tester and pcDNA3.1(-)-IFN-? transfected cells as driver. A housekeeping gene, G3PDH, was used to estimate the subtractive efficiency. Genes with lower expressions in IFN-? transfected HepG2 cells were obtained from the library after being randomly sequenced and analyzed. Among the obtained genes, regulation of IFN-? on heat shock 90kD (Hsp90) mRNA was further investigated by RT-PCR. Results G3PDH was subtracted efficiently indicating that the subtractive library was constructed successfully. 50 positive clones were randomly isolated for PCR identification. Results showed that most of the plasmids in the clones contained 200-1 000bp inserts. The down-regulated genes obtained were as follows: eukaryotic translation elongation factor 1 beta, ferritin, RAD23 homolog B, signal sequence receptor 2 (SSR2), tissue factor pathway inhibitor, heat shock 90kD protein 1, and adenosylmethionine decarboxylase 1. RT-PCR showed that hsp90 mRNA had a reduced expression in pcDNA3.1 (-)-IFN-? transfected HepG2 cells. Conclusion The down-regulated cDNA subtractive library in HepG2 cells after pcDNA3.1 (-)-IFN-? transfection was constructed successfully. IFN-? could down-regulate the hsp90 mRNA expression.
5.Immunohistochemistry study on the over expression of ERBB3 in gastric cancer

Objective To investigate the expression of ERBB3 protein,which encoded by ERBB3 gene,in gastric cancer(intestinal type and diffuse type)as well as the normal tissue,and to elucidate the possible role of ERBB3 gene in the progress of gastric cancer.Methods 65 samples of gastric cancer tissue(of which 43 samples were intestinal type and 22 were diffuse type,classified according to the Lauren patho-classification)and 65 samples of normal tissue(control group)were investigated immunohistochemically.The expression of ERBB3 in both control group and gastric cancer group was determined.Comparisons were made according to different pathological types and TNM clinical stages within and between groups.Results 7 of 65 samples(10.8%)in gastric cancer group were found to over-express the ERBB3 protein,while only 1 of 65 samples(1.6%)in control group was found to over-express the ERBB3 protein.Statistical differences(P
6.Clinical assessment of the effect of intravenous PCA and epidural PCA in postoperative analgesia

To compare the clinical effect and side-effect of intravenous PCA(PCIA) with epidural PCA (PCEA). Method: One hundred and eighty postoperative patients, who were randomly divided into three groups: group PCIA, group PCEA and control, were observed for 3 days after operation. Result: Overall patients in two PCA groups were satisfied with the postoperative analgesia. The incidence of urinary retention in group PCIA were significantly lower than that in group PCEA (P0.05). 10% in group PCIA were in medium sedation whereas no eases in group PCEA were in sedation. At 6th hour after operation,serum cortisol level of control patients was much higher than that of PCA patients. Conclusion=Both PCIA and PCEA have excellent analgesia and reduce stress response after operation. PClA has lower incidence of urinary retention and is performed easily,but inhibits gastrointestinal motility much more and has higher sedative incidence compared with PCEA.
7.Investigation on Weight Variations and Microbial Contaminations of5Tablets Broken off with Hands

OBJECTIVE:To improve medication safety and efficacy.METHODS:5different tablets broken off with hands were investigated with respect to weight variations and microbial limits by reference to Chinese Pharmacopeia.RESULTS:The5kinds of tablets broken off with hands were not in conformity with Chinese Pharmacopeia with respect to weight variation and which involved different degrees of microbial contaminations.CONCLUSION:It is recommended that pharmaceutical enter?prises should add more dosage forms of standard drugs that suitable for the use of children,women,the aged and particular patients.If the separately packaged drugs must be broken off with hands,aseptic method should be applied in which the drugs were broken off by the pharmacists on the sterile air bench before being separately packed with sterilized plastic pouches(which were sterilized with anprolene),and which should be administered to the patients for taking during the same day.
8.Dynamic detection of RNA of SARS-associated coronavirus in blood and excreta of SARS patients

Objective To serially examine virus carrying condition of clinical specimens collected from SARS patients, in order to evaluate the dynamic changes of virus in the bady and its clinical significance. Methods A total of 494 samples, including plasma, urine, feces, sputum and throat swab, obtained from 84 cases of clinically diagnesed SARS patients at different time-points were examined for the study. Nested RT-PCR was employed for the detection of SARS-CoV using 2 pairs of primers targeting P and N region of the viral genome. The amplified viral genomic fragments were confirmed by DNA sequence analysis. Results Specific bands for SARS-CoV were amplified from various specimens of some SARS patients. Highest positive rate was found in sputum compared with the others. However, simultaneous examination of more than two specimens increased detectable rate of the virus. Moreover, dynamic examination revealed that the highest positive rate occurred in the 2nd week after the onset of the disease and next to the highest during the 3rd to 4th week. However, dynamic detectable rates for different specimens were not identical. Conclusion Nested RT-PCR is a practicable method for the detection of SARS-CoV and helpful for the early diagnosis or confirmation of the disease. Dynamic examinations for SARS-CoV by combined employment of the N and P primers and by involving multiple specimens may significantly increase the positive detectable rate, therefore would be helpful in diagnosis and eratuation of therapeutic effect of SARS.
9.Dynamic analysis of anti-Cov antibody and SARS coronavirus in the plasma of 25 SARS patients

Objective To observe dynamic responsive regularity of specific IgM and IgG antibodies in SARS patients. Methods 145 specimens of plasma from 25 cases of clinically diagnosed SARS patients were examined in the study. ELISA was employed to detect IgM and IgG antibodies against SARS coronaviral antigens. Nested RT-PCR was used to qualitatively determine SARS coronavirus (SARS-CoV). Results 49.0% (71/145) and 54.5% (79/145) of the samples were positive for IgM and IgG antibodies, respectively. Both antibodies were found to be detected in 84.0% of the patients. The antibodies were found to be detectable from the 2nd to 4th week after the onset of disease in most patients, and there was a fendeney of sising in positive rate until the 5th week after the onset. Thereafter, the detectable rate of IgM antibody began to decline, while that of IgG antibody remained to rise. Positive rate for serum SARS-CoV was 15.8% (18/114) for all samples or 40.0% (10/25) of patients. Most virus-positive samples were those which were collected within 4 weeks after disease onset. Conclusions Anti-SARS-CoV IgM/IgG antibody and detection of virus in plasma could serve as practical diagnostic indicators for SARS. In most cases, when the serum was pasitive for antibody the serum virus positive rate would soon declined. However, concomitant existence of antibody and viral sequence in plasma was observed in a few patients.
10.Inhibitory effects of tea polyphenols on telomerase and its ability to induce ap optosis of BEL-7404 human hepatocellular carcinoma cell *

AIM To investigate the effects of Tea Po lyp henols on induction of apoptosis in human liver cancer cell etc. Telomerase acti vity of BEL-7404 cells was dramatically declined during the cell incuba tion with Tea Polyphenols. METHODS MTT colorimetric assay and trypan b lue exclusion method were used to examine the growth inhibition ofTea Polyphenols on BEL-7404 cells. Telomerase activity in cell was determined by PCR-ELISA method. Cell morphology and DNA gel electrophoresis were used to observe the apoptosis. RESULTS The solution of Tea Polyphenols showed dose-dependent and time-dependent inh ibitory effects on the proliferation on BEL-7404 cell line. After treatment wit h 0 2 and 0 1 g?L -1 tea polyphenols for 48 hours, cells displayed DNA ladder bands and typical morphological change of apoptosis included: ce ll shrinkage, membrane blebbing, chromatin condensation, nuclear fragmentation etc. Telomerase activit y of BEL-7404 cells was dramatically declined during the cell incubation with Tea Polyphenols. CONCLUSION Tea Polyphenols has growth inhibiti ng effect and may induce apoptosis in BEL-7404 in a certain dose range. The ant icancer mechanism of Tea Polyphenols might be related to the inhibition of telom erase activity.
