Objective To elucidate the effects of endothelin-1(ET-1) on expression of extra-regulated kinase 1(ERK 1), cell cycle and changes of Ca 2+ in activated hepatic stellate cells (HSCs). Methods Effects of different concentra tions of ET-1 on ERK 1 expression were determined by Western blotting. Effects of ET- 1 on cell cycle were observed by flow cytometry(FCM) analysis. Effects of ET-1 on changes of calcium concentration and cell area were observed by laser scanning confocal microscopy (LSCM). Objective To evaluate the efficacy and safety of brachytherapy by percutaneous paracentesis implantation of iodine-125 seeds for elderly non-small cell lung cancer.Methods 18 patients with peripheral lung cancer were undergone brachytherapy by percutaneous paracentesis implantation of iodine-125 seeds.The effect was evaluated by periodically chest CT.The complications were analyzed.Results 18 patients were observed one,two and six months after operation.CR,PR,SD were seen in 27.8%(3/18),66.7%(14/18),5.6%(1/18)respectively of the patients.Pneumothorax,hemoptysis were seen in 16.67%(3/18),95.37%(17/18),no pleural cavity bleeding.Conclusion Brachytherapy by percutaneous paracentesis implantation of iodine-125 seeds for lung cancer was effecitve.The complications were easy to treat.The methods of implantation of iodine-125 seeds could fully play the advantages of CT,and reduce the damage to the body when raising the tumorous control rate.
3.Effect of externally applied Honghua Liquid and catus on phlebitis induced by amiodaone

Objective To observe the effect of externally-applied Honghua liquid and catus on phlebitis induced by amiodaone.Methods One hundred and two patients with phlebitis induced by injection of amiodaone for the treatment of arrhythmia were randomly divided into control group(n=50)and treatment group(n=52)from January 2012 to December 2013. Patients in the control group were independently treated only by 50 % Honghua liquid, while the patients in the observation group were treated by hot dressing with Honghua liquid followed by wet dressing of catus. The curative effects in both groups were observed and compared.Result The total effective rate in the treatment group was significantly higher than that in the control group(χ2=6.275, P<0.05).Conclusion Hot dressing with Honghua liquid combined with wet dressing with catus is a simple and effective method to treat the phlebitis induced by injection of amiodaone.
4.Dynamic monitoring of leukocytes in patients undergoing renal transplantation

BACKGROUND: Changes of leukocytes are related with some postoperative complications after renal transplantation. OBJECTIVE: To observe dynamically the level of white blood cell (WBC) and its morphological changes using automatic blood cell analyzer to evaluate its relationship with infection, immunosuppressive intoxation and acute rejection. DESIGN, TIME AND SETTING: Observation and controlled trial was performed at Second Xiangya Hospital, Central South University from June 2004 to October 2005. PARTICIPANTS: Sixty-eight patients who received renal transplantation were selected. According to clinical symptoms and laboratory biochemical indexes, X-ray, B ultrasound, and renal puncture biopsy, the patients were divided into stable group (n=36), infection group (n=9), intoxation group (n=6) and acute rejection group (n=13). METHODS: The blood routine was tested with Sysmex XE-2100 automatic blood analyzer following renal transplantation and WBC manual differentiation was done to observe peripheral blood morphology, their liver function. In addition, renal and liver functions were detected by automatic biochemical system 7600A. MAIN OUTCOME MEASURES: WBC count and its morphological changes in peripheral blood and liver and renal function after renal transplantation. 5.Detection and antimicrobial resistance of hypermutable Pseudomonas aeruginosa strains

Objective To investigate the isolation and antimicrobial resistance of hypermutable Pseudomonas aeruginosa strains.Methods The cultured bacteria were identified by API 20NE system.The susceptibilities of bacteria were detected by disk diffusion method and the results were confirmed with the rules of NCCLS/CLSI.The hypermutable strains were identified byhigh concentration of rifampin-resistant growth assay.Results Among analyzed 100 strains,37 strains(37.0%) were hypermutable.The resistant rates of hypermutable strains to imipenem,meropenem,cefoperazone/sulbactam,piperacillin/tazobactam,ceftazidime,cephfime,aztreonam,amikacin and ciprofloxacin were significantly higher than those of non-hypermutable strains.The multidrug resistance rates of hypermutable strains were also higher than those of non-hypermutable strains.Conclusion The hypermutable strains of Pseudomonas aeruginosa sourced from chronic respiratory tract infections dominantly.The hypermutable strains were hyper-resistant and multidrug resistant to many antibiotics,so strengthened monitoring should be encouraged to control the prevalence of hypermutable Pseudomonas aeruginosa strains.
6.Calcium/Calmodulin-dependent Protein Kinase-II δ Involved in Doxorubicin Induced Cardio-toxicity in Experimental Rats

Objective: To explore the role of calcium/calmodulin-dependent protein kinase-II δ (CaMK-II δ) in doxorubicin (DOX) induced cardio-toxicity in experimental rats.
Methods:①The rat’s cardiomyocytes were treated by DOX and the cell proliferation, protein expression and activity of CaMK-II δ were examined.②CaMK-II δ gene was knocked out by CRISPR method, the changes of DOX induced cell apoptosis and NF-κb activity and miR-146a expression were detected.
Results: DOX could inhibit cardiomyocyte proliferation, the protein expression level of CaMK-II δ was similar and the activity was increased. CRISPR method may effectively knock out CaMK-II δ gene. Compared with normal cells, the cells from CaMK-II δ knocked out rats had decreased sensitivity to DOX induction, suppressed NF-κb activation and miR-146a up-regulation.
Conclusion: CaMK-II δ participated in DOX induced cardio-toxicity in experimental rats and NF-κb and miR-146a were involved in this process.

8.Resistance and ?-Lactamases of Acinetobacter baumannii

OBJECTIVE To investigate the relationship of resistance and ?-lactamases of Acinetobacter baumannii.METHODS The bacteria cultured were identified with API 20NE system.The susceptibilities of the bacteria were detected by disk diffusion method.The ?-lactamases produced by these strains were characterized by three-dimensional test and 2-mercaptopropionic acid inhibition assays.RESULTS In 150 strains,the resistant rates of strains were 4.0% to cefoperazone/sulbactam.The resistant rates to imipenem,meropenem,piperacillin/tazobactam,ampicillin/sulbactam and levofloxacin were between 20.0% and 55.0%.The resistant rates to ceftazidime,cefepime,aztreonam and amikacin were above 60.0%.Three-dimensional test showed that 37 strains(24.7%) produced extended spectrum ?-lactamases(ESBLs),66 strains(44.0%)produced AmpC enzymes and 16 strains(10.7%)produced carbapenemases,in which 12 strains(8.0%) produced metallo-?-lactamases.CONCLUSIONS The resistant rates of A.baumannii to routine antibiotics are high except cefoperazone/sulbactam and carbapenem.It is the most important reason for multi-drug resistance that the strains of A.baumannii produce ESBLs,AmpC enzymes and metallo-?-lactamases.
9.Application of admission test in labor room

Objective To investigate the application of admission test as a medical management in the labor room.Methods Eight hundred and fifty singleton pregnant women underwent admission test in labor room at Women's Hospital,School of Medicine,Zhejiang University from Dec.2009 to Dec.2011 were enrolled into this study.They were divided into two groups:normal group (admission test was normal,n =658) and abnormal group (admission test was abnormal or undetermined,n =192).Deliveries and perinatal outcomes of them were observed.The difference between the two groups were compared with two independent sample t test and Chi-square test.Results There were no significant differences in the mean age at delivery of the women and mean birth weight of neonates between the two groups (all P>0.05).The cesarean section rate of normal group (34.2%,225/658) was lower than that (59.4%,114/192) of abnormal group (x2=3.93,P<0.05).The rates of prematurc rupture of membranes (72.4%,139/ 192),fetaldistress (42.2%,81/192),neonatal asphyxia (16.5%,27/192) and neonatal complications (31.8%,61/192) in abnormal group were significantly higher than those [31.0% (204/658),24.2% (159/ 658),0.2%(1/658) and 2.6% (17/658)] in normal group (x2 =105.78,40.84,52.54 and 151.92,P<0.01respectively).Conclusions Admission test is a good method to forecast placental function during labor and perinatal outcomes,which might play an important role in medical management in labor room.
10.A clinical observation of combined TUINA with Chinese herbs fumigating for bony arthritis of knee joints:A 73 cases reports

Objective To explore the clinical therapeutic effect of combined TUINA with Chinese herbs fumigating in patients with bony arthritis of knee joints.Methods A total of 73 patients with bony arthrithis of knee joints were divided into treatment group(37 cases)and control group(36 cases)randomly.The patients of treatment group were used the combined TUINA with Chinese herbs fumigating and patients in control group were used Chinese herbs fumigating alone.After three courses of treatment,therapeutic effect was compared the two groups patients.Results The results showed that the cure rate in treatment group was higher than control group markedly(P <0.01).The improvement rate between the two groups was not significently different.Conclusion The therapeutic effect that combined TUINA with Chinese herbs fumigating for patients with bony arthritis of knee joints is very effective treatment procedure.
