1.Study on Accelerating Rat Fracture Healing by Injection of Soluble Multipeptides Abstracted from Bone Tissue

Objective: To study the effects of soluble multi-peptide agent,marked “Gu Kang Tai Ling”,derived from bone tissue on rat bone fracture healing.Methods 80 right tibia defects of rats were sawed.The “Gu Kang Tai Ling”were intramuscularly injected to the rats.The quality of the defect healing was investigated continuously and respectively by the defect bone histomorphometry,biomechanics,X-ray film,and bone mineral density(BMD).Results: Under the treatment of the agent,at early stage trabecular bone volume (TBV)ratio increased 24.7%,and osteoblast surface(OBS)26.7%,at late stage mean trabecular healing surface rate(MTHSR)was increased 38.8%,tibia anti-fracture strength 0.4527~1.4350N,and BMD 7.65% compared with the control group.Conclusion: “Gu Kang Tai Ling”can accelerate rat fracture healing and improve the quality of healing.
2.Evaluation on effectiveness of individual lingual orthodontics and labial straight wire orthodontics by X-ray cephalometrics

Objective To compare the X-ray cephalometric results of teeth and skeleton relationship between individual lingual orthodontic and labial straight wire orthodontics,and to provide reliable theoretical foundation for selecting orthodontic system in clinic. Methods The general conditions of the patients, classification of malocclusion,severity of denture crowding and orthodontic design were used as the selection criteria. Thirty familiar adult patients with malocclusion in clinic were divided into two groups randomly:individual lingual orthodontics group (A group)and labial straight wire orthodontics group (B group),15 cases in each group.X-ray cephalometrics were taken using Steiner and Tweed cephalometrics analysis,the results of analysis were compared between pre-treatment and post-treatment in two groups respectively, and the course of treatment in two groups was compared,too.The difference of treatment effect and course of treatment between two different orthodontic systems was evaluated.Results The relationship of first molar and canine of the patients in two groups was neutral relationship,overbite and overjet of anterior teeth was shallow,the teeth arrangement was orderly and no space was left after treatment.There was no statistically significant difference (P>0.05)between two groups in each measured values detected by Steiner Analysis after orthodontic treatment;there was no statistically significant difference (P>0.05)between two groups in each measured value detected by Tweed Analysis after orthodontic treatment;and there was no statistical significance (P>0.05 )in the D-values of Steiner Analysis and Tweed Analysis between two groups before and after treatment.The duration of treatment of the patients in two groups had no significant difference (P>0.05).Conclusion Both individual lingual orthodontics and labial straight wire orthodontics could obtain well teeth arrangement and occluding relationship and achieve satisfying clinical orthodontic outcome.
3.The clinical study on ankle joint fracture treated by open reduction and internal fixation

Objective To explore the surgical treatment of ankle fracture.Methods 326 cases with ankle joint fracture in our hospital treated by surgical treatment were followed up.There were 182 males and 43 females,aged from 7 ~ 75 years.The patients with medial malleolus fracture were cut through postero-medial process,and the lagged screw,absorbed screw,or tension band wire were used.Patients with bimalleolus and trimalleolus fracture were cut through medial and lateral two incision.When the fracture line of the fibula is high,the triangular needle,kirschner wire,plate and the screw,remember alloy were used,when the fracture line of the fibula was high,1 ~ 2 cortical or cancellous bone lagged screw were used.The patients with diastasis of inferior tibiofibular syndesmosis were cut through lateral process,1 lagged screw was used through 3 layer cortical bone.After the operation,the plaster or brace was used according to the stability of the fracture,then systemic functional exercises.Results All the patients were followed up from 4 ~ 120 months,according to the ankle-hind foot score of American Orthopedic Foot and Ankle Society Score(AOFAS),their clinical treatment results were assessed.In 225 cases,excellent and good rate was 86.7%.Connclusion Proper approach,anatomy reduction,articular surface recovery to smooth,stable internal fixation,necessary external fixation and systemic rehabilitation were the key aspects to the surgical treatment.Good result of most of the patients with fracture of the ankle joint can be obtained.
4.Experimental and clinical study on transplanted time of periosteal mesenchymal stem cell

Objective Investigating transplanted time of periosteal mesenchymal stem cell (PMSC) Methods Rat PMSCs were allografted into rat tibia defects made in different time of granulation Then the quantity of formatted bone,i.e.,trabecular volumm rate (TVR),in the defects were measured in histology and bone histomorphometry by 2,3,4 and 8 weeks respectively after transplantation The long bone draft fracture defects of patients were allografted with PMSC of youth children after 2 week of fracture Results TVR was higher and the defects healed more fast in those rats transplanted PMSCs within 2 weeks after the defect were made However there was insignificant difference between the groups of PMSC transplantation was carried out after two weeks of the defect The fractured defects were healed in 15 cases (57 7%) and were not healed in 11 cases (42 3) after 9 months of transplantation Conclusion That rat bone defects were transplanted PMSC within 2 weeks showed expected bone formed effect and insignificant effect on accelerating defect healing by PMSC transplantation in rats or patients after 2 weeks.
5.Experimental study of rat bone marrow stromal cell proliferation and differentiation in vitro

Objective:To study the conditions of in vitro proliferation and differentiation of rat bone marrow stromal cells(MSC) for its application in transplantation.Methods:MSC were obtained by flushing the shaft with DMEM using a sterile syringe,dissociated by density gradient centrifuge,and cultured in the medium containing DMEM and 10% fatal bovine serum(FBS).Then we added neural growth factors into the medium and observed their influence on MSC proliferation,differentiation and survival by phase contrast microscopy.Immunocytochemistry was used to identify cell types.Results:MSC cultured in medium containing DMEM/ 10% FBS/ neural growth factors proliferated quickly.The neuron-like cells appeared at 7-9 d with the expression of neurone specific enolase(NSE) at(52?9.2)% and glial fibrillary acidic protein(GFAP) at(15?7)%,and could survive more than 7 days;In contrast,cells induced by ?-mercaptoethanol(BME) died gradually after 24 hours.Conclusion:MSC can not only proliferate and differentiate into neuron-like cells effectively in the medium containing neural growth factors but also survive longer.
6.Application of Tracheal Intubation Guided by Fiberoptic Bronchoscopy in Children Snoring Disease Operation

Objective To explore the value of tracheal intubation guided by fiberoptic bronchoscopy in children snoring disease operation.Methods A total of 50 patients underwent tracheal intubation guided by fiberoptic bronchoscope or laryngoscope from August 2015 to June 2016.There were 25 cases of fiberoptic bronchoscopy assigned into group A, 25 cases of laryngoscopy into group B.The intubation time, success rate and adverse reactions of the two groups were compared and analyzed.Results All the intubation in group A succeed at the first time.The intubation time was (20.1±4.3) s and the position was right.There was no need for replacement during the operation.Three cases in group B succeed at the second time because the tonsil was too big to expose the pharynx.The intubation got out of the pharynx in 3 cases in group B and were adjusted to the right position.The intubation time was (50.7±6.9) s and the success rate at the first time was 76% (19/25).Groups A behaved better than group B in aspects of the success rate of signal intubation and intubation time (Fisher''s Exact Test, P=0.022;t=-18.725, P=0.000).There were no adverse reactions in group A, such as pharyngorrhagia, odontoseisis or loss of tooth, laryngospasm and so on.There were 3 cases of hemorrhage from throat after intubation, 3 cases of loss of tooth in different degrees and 1 case of laryngospasm at second intubation time in group B.There was no significant difference in adverse reactions between the two groups (P>0.05).ConclusionFiberoptic bronchoscopy can be used for children snoring disease with good safety as well as rare adverse reactions.
7.Effect of NF-?B Decoys on development and maturation of dendritic cells and in itiation of Tcells immune response in vitro

ObjectiveTo investigate the effect of NF-?B on the differentiation and maturation of mu rine bone marrow-derived dendritic cells (DC) in vitro.MethodsThe activity of NF-?B in dendritic cells was blocked by oligodeoxyribonucleot ide Decoys (ODN Decoys) containing two special binding sites for NF-?B. Morpho logical changes of DS were observed. The expression of assistant molecules on th e cell surface was detected by using flow cytometer, and the stimulating activit y of allogeneic T lymphocyte proliferative response was determined in culture mu rine DC. The effect of NF-?B on the maturation and immunobiological activity o f murine DC was studied.ResultsNF-?B ODN Decoys were efficiently incorporated by DC, markedly suppressed the maturation of DC, the expression of assistant costimulatory molecules (CD80, CD8 6 and CD40) on the surface of DC and the secretion of IL-12, blocked the develo pment of DC and this blocked function was not reversed by lipopolysaccharide (LP S). In mixed lymphocyte reactions, DC treated with NF-?B ODN Decoys could indu ce allogeneic T cells hyphoresponsiveness, and this was associated with the inhi bition of Th1-type cytokines (IL-2 and IFN-?) production.ConclusionsNF-? B is a key gene to the development and maturation of DC. Specific interference w ith NF-?B in DC using ODN Decoys approaches could offer a novel strategy for i nducing and generating tolerogeneic immature DC and provide a promising means to induce transplant immune tolerance.
8.Study of the effect of murine dendritic cells modified with IFN-? inducible protein-10 gene on prostatic carcinoma cells in vitro

Objective To investigate the effect of murine dentritic cells vaccine modified with IFN-? inducible protein-10 gene on CTL induction. Methods DC was propagated from bone marrow (BM) of mice and pulsed with mouse prostate cancer cell line RM-1's whole lysate (Tuly-DC).IP-10 DNA fragments were inserted into pcDNA3.1(+) vector to construct recombinant plasmid IP-10/pcDNA3.1.Tuly-DC was transfected with IP-10/pcDNA3.1 by DOTAP liposome.Reversal transcript-polymerase chain reaction (RT-PCR) was used to evaluate gene transfer efficiency and chemotaxis assay was used to estimate the tansfected DC's chemotactic activity on T cells.Antitumor activity of the DC vaccine was studied in vitro by using Mixed leukocyte reaction (MLR) and cytotoxic assay (MTT assay). Objective To investigate the effect of recipient dendritic cells (DC) loaded with donor-derived apoptotic cells on inducing murine cardiac allograft tolerance. Methods Apoptosis of donor-derived splenocytes (SC) was induced by ultraviolet B irradiation(UVB). UVB-irradiated allogenic SC were co-cultured with recipient bone marrow-derived DC that maturation was inhibited by NF-?B ODN Decoy, so that could acquire tolerogenic-immature DC loaded with donor-derived apoptotic cells (Decoy Apo-SC DC). A heterotopic vascularized heart transplantation was performed from BALB/c to C57BL/6 mice, and recipients were given one injection of recipient immature (Decoy Apo-SC DC) or mature (Apo-SC DC) DC engulfed donor-derived apoptotic cells (2?10 6 cells) through the portal vein at 7 days before the heart transplantation in the absence of immunosuppression. The cardiac survival and the expression of intragraft cytokines (IL-2, IL-10 and IFN-?) were evaluated.Results DC had potent phagocytosis of allogenic apoptotic SC (Apo-SC). NF-?B ODN Decoy inhibited engulfment of apoptotic cells-induced maturation of DC and then induced recipient tolerogenic DC. Recipient tolerogenic DC loaded with donor-derived apoptotic cells were able to cross-tolerate recipient T cells, which revealed by alloantigen-specific T-cell hyporesponsiveness in primary and secondary mixed leukocyte reaction. Objective To study the diagnosis and trea tm ent of multiple endocrine neoplasia type 2b. Methods On e case of MEN2b was reported.The clinical features,the diagnosis and treatment o f MEN2b were reviewed and presented. Results The diagno sis was established on B-ultrasonography and CT scanning,and the patient was cu red by operation. Conclusions MEN2b has been rare.Endoc rine check up,imaging procedures,family investigation and DNA analysis are very important to early diagnosis.Surgical removal of the tumors is effective.
