1.Effects of Three Kinds of Mutagens on the Cell Survival Rate and the Chromosomal Aberration in V79 Cells

The cell survival rate and the chromosomal aberration induced by N-methyl-N-nitro-N-nitriosoguanidine (MNNG), mitomycin C (MMC) and ethyl methanesuiphonate (EMS) in V79 cells and its ?-ray-sensitive mutants (irs5 and mil) were observed in this study. The concentrations of mutagens to determine the survival rate were arranged at 2.5~ 30 ?mol/L in MNNG, 1~20?mol/L in MMC and 2~ 16 ?mol/L in EMS, respectively. The cell survival rates and the cell clones of irs5 were higher than that of the V79 cells and irs11; and of irs11 were significantly lower than that of V79 cells and irs5. The result of IC50 of mutagens to determine the chromosomal aberration was similar to the cell survival rate. The type and frequency of chromosomal aberration induced by MNNG, MMC and EMS in irs5 was same as in V79 cell; and there was a dose-response relationship between them. The spontaneous aberration frequencies in irsS and V79 cells were lower than 4%, however, in irs11 was higher than 10%. Data suggests that the cellular biological characterization in irs5 is steady, and further study to certain irsS whether it will be a new mutant for the mutagenicity or carcinogenicity assay or not.
2.Effects of ginsenoside Rg1 on free amino acids in brain of sports fatigue-mice

Objective: To study the effect and possible mechanism of Ginsenoside Rg1 in sports CNS fatigue and dose-response relationship.Methods: Kun Ming male mice were divided randomly into 6 groups: control group and five different doses groups(5,10,20,40,80 mg/kg).They were treated orally with Ginsenoside Rg1 for 2 weeks.The levels of amino acids of brain tissue were tested after swimming for 90 min and the exhaustive swimming time were recorded.Results: The levels of BCAA and the ratio of BCAA/AAA in groups 5,10 and 20 mg/kg were significantly higher than control group and the levels of AAA in groups 5,10,20 and 40 mg/kg were significantly decreased.The exhaustive swimming time in groups 5,10 and 20 mg/kg were much longer than in control group.Conclusion: Through adjusting the ratio of BCAA and AAA and decreasing the level of excitatory amino acids neurotransmitter,Ginsenoside Rg1 can elevate movement ability and resist against CNS fatigue.The roles of mid-low and low doses may be better.
3.Magnetic resonance imaging of nude mice model injected in USPIO-PEG-sLeX in vivo

Objective To explore the value of the molecular probe USPIO-PEG-sLeX on nasopharyngeal carcinoma xenograft in nude mice.Methods The USPIO nanoparticles was synthesized by physical deposition method,and which was modified by PEG to synthesize USPIO-PEG-sLeX .The nude mice of nasopharyngeal carcinoma xenograft were divided into experimental and control groups.USPIO-PEG-sLeX and USPIO-PEG were injected into nude mice of experimental and control groups by caudal vein,respectively.MR T2 mapping imaging was scanned before and after the injection,and analyzed the changes of T2 values between experimental and control groups. Results USPIO-PEG-sLeX had a good representation.The non-enhanced T2 values between control and experimental group had no statistical significance (P >0.05).However,T2 values of the mice in two groups before and after injections were statistically significant (P <0.05);and T2 values of experimental group were much lower than that of the control group after the injection,additionally,the difference of enhanced rate between the two groups was statistically significant (P <0.05).Conclusion USPIO-PEG-sLeX magnetic nanoparticles is potential to be a targeted contrast agent to ELAM-1 expression of nasopharyngeal carcinoma,and can be valuable in non-invasive dynamic monitoring the expression of ELAM-1.
4.Development and reliability and validity test of Pregnant Women Weight Self-Management Scale

Objective To develop Pregnant Women Weight Self-Management Scale and test its reliability and validity.Methods Based on the concepts and theories of self-management,the first draft of the scale was formed through literature analysis,semi-structured interviews,2 rounds of Delphi expert correspondence and pre-surveys.The scale was used to test the reliability and validity of the scale by facilitating the selection of 300 pregnant women who gave birth in April 18-23 2023 at a maternal and child healthcare hospital in Ningxia Hui Autonomous Region.Results The Pregnant Women Weight Self-Management Scale consisted of 8 dimensions and 38 entries with a cumulative variance contribution of 68.560%.The Cronbach's alpha coefficient for the total scale was 0.944,and each dimension ranged from 0.772 to 0.929.The split-half reliability for the total was 0.858,and each dimension ranged from 0.715 to 0.902.The content validity index at the level of the total scale was 0.921,and at the level of each entry was from 0.833 to 1.000.There was a positive correlation between the dimensions and with the total scale score(r=0.178~0.854,P<0.001).Conclusion The scale has good reliability and validity,and it can be used as a tool to evaluate the self-management ability of pregnant women.
5.Effect of early postoperative systemic inflammatory response syndrome (SIRS) on the short-term outcome of patients with acute Stanford type A aortic dissection

Objective To investigate the effect of early postoperative systemic inflammatory response syndrome (SIRS) on the short-term outcome of patients with acute Stanford type A aortic dissection (ATAAD). Methods The clinical data of 88 patients with ATAAD who were treated in our hospital from January 2018 to January 2020 were retrospectively analyzed. Patients were divided into a SIRS group (n=37) and a non-SIRS group (n=51) according to whether SIRS occurred within 24 hours after surgery. The perioperative data of the two groups were compared. Results There was no significant difference between the two groups in general clinical data, preoperative left ventricular ejection fraction, white blood cell (WBC) and body temperature (P>0.05). Compared with the non-SIRS group, the cardiopulmonary bypass time in the SIRS group was significantly longer (P<0.05), and the WBC and body temperature within 1 day after surgery in the SIRS group were higher (P<0.01). A significant difference was revealed in the mechanical ventilation time, ICU stay, total hospitalization time and hospitalization costs between two groups (P<0.01). Patients in the SIRS group had higher postoperative acute physiology and chronic health evaluationⅡscores, sequential organ failure assessment score as well as a greater risk of developing postoperative acute lung injury, acute kidney injury, continuous renal replacement therapy, delirium, liver dysfunction and morbidity (P<0.05). Conclusion Early postoperative SIRS significantly increases the incidence of major adverse complications and the mortality rate of patients with ATAAD.
