1.Cloning of 119-264 Amino Acid Fragment of Ro52 Antigen and Analysis of Its Antigenicity

Objective To investigate the role of Ro52 antigen fragment from amino acid (AA) 119 to 264 that contains leucine zipper motif in forming the epitopes of Ro52 autoantigen. Methods cDNA encoding Ro52 antigen fragment from aa 119 to 264 was amplified by PCR from human heart cDNA first strand. The obtained cDNA fragment was inserted into vector pMTY4 and transformed into E. coli pop2136 for fusion protein expression. The recombinant fusion protein was then purified, and the antigenicity was determined by immunoblotting. Results A 28 000 Dalton fusion protein was obtained. It was highly expressed in host E. coli pop2136. 61.90% of the anti-full-length Ro52 positive sera reacted with the fusion protein in immunoblotting. Conclusion The results suggest that the antigen fragment from the 119 AA to 264 AA that contains the leucine zipper motif represents an important epitope region in Ro52 autoantigen.
2.Struture Analysis and Selective Fragment Cloning of 52 kD Ro/SSA Antigen

Objective To detect the possible epitopes of 52 000 dalton Ro/SSA autoantigen and selectively clone the antigen fragment containing the leucine zipper motif. Methods The structure of 52 000 dalton Ro/SSA autoantigen was analyzed by protein structure analysis system. The cDNA encoding the antigen fragments from amino acid (aa) 119 to 264 was amplified from cDNA first strands of human heart by polymerase chain reaction (PCR). The PCR product was inserted into the vector pMTY4, and transfected E. coli pop 2136 for fusion protein production. Results Computer analysis showed that the 52 000 dalton Ro/SSA antigen had alpha helixes in its structure, and had good antigenicity, moderate flexibility and poor surface probability. The PCR product was 440bp in length. The recombinant fusion protein was 28 000 dalton. DNA sequencing proved the accuracy of the research. Conclusion The 52 000 dalton Ro/SSA autoantigen has good antigenicity. The cloning of the antigen fragment will help to elucidate the possible role of leucine zipper motif in forming the epitope of the antigen.
3.The correlation analysis between hepatitis B induced cirrhosis glucose metabolism disorders and hepatitis B immune related indexes and the regional epidemic characteristics

Objective To investigate the correlation between hepatitis B induced cirrhosis glucose metaboliam disorders and hepatitis B immune related indexes,to analyze the epidemic characteristics in the local region.Methods 128 cases with hepatitis B induced cirrhosis were divided into the NGT group,IGR group and DM group,according to 75g oral glucose tolerance test(OGTT).Results The difference of waist size,and body mass index data in the three groups had statistically significant difference( F =22.71,24.62,all P < 0.05 ).As the aggravating of glu cose metabolism disorder,the positive rate of HBsAg,HBeAg and former S1 antigen of NGT group,IGR group and DM group were gradually raised,and the differences were statistically significant ( F =17.35,21.47,19.37,all P <0.05).Conclusion There was positive correlation between virus infection in patients with hepatitis B induced cirrhosis and glucose metabolism disorder.There were intimate connection between the epidemiological factors such as age,waist size,weight,and body mass index and glucose metabolism disorder in patients with hepatitis B induced cirrhosis.
4.Correlation factors of liver injury in infectious mononucleosis

Objective To investigate the correlation factors of liver injury in infectious mononucleosis(IM).Methods One hundred and seventy-five IM patients were divided into two groups:IM with liver injury(patient group,85 cases)and IM without liver injury(control group,90 cases).Single factor and multiple factor analysis were completed by unconditional Logistic regression.Results Single factor analysis results showed that there were significance differences between the two groups in age(≥6 years old),male,course of disease,severe disease(involving ≥ 2 organs),heating time,the peak temperature,white blood cell(WBC)and C reactive protein(CRP),CD4+,CD8+,CD4+/CD8+,Epstein-Barr virus(EBV)-capsid antigen(CA)-IgM antibody positive and increased erythrocyte sedimentation rate(ESR)(P<0.05).Age(≥ 6 years old),male,course of disease,severe disease(involving ≥2 organs),heating time,the peak temperature,WBC,CRP,CD8+,EBV-CA-IgM antibody positive and increased ESR showed positive correlation with IM combined with liver injury,and CD4+,CD4+/CD8+ showed negatively correlated with IM combined with liver injury.Multiple factor analysis results showed that severe disease(involving ≥2 organs),CD8+ high levels were independent risk factors for IM combined with liver injury,and CD4+ high levels and CD4+/CD8+ high were protection factors for IM combined with liver injury(P<0.05).Conclusions Severe disease(involving ≥2 organs),CD8+ high levels are independent risk factors for IM combined with liver injury,and CD4+ high levels and CD4+/CD8+ high are protection factors for IM combined with liver injury.Therefore,immune modulators should be used to intervene the above factors when patients are treated with antiviral drugs.
5.The related factors analysis of hepatitis B liver cirrhosis patients with secondary glucose metabolism disorders

Objective To investigate the related factors analysis of hepatitis B liver cirrhosis with secondary glucose metabolism disorders.Methods 184 patients with hepatitis B liver cirrhosis were selected as the research object and were given 75g oral glucose tolerance test( OGTI),then they were divided into three groups[ normal glucose tolerance(NGT) group,impaired glucose tolerance and impaired fasting glucose regulation impairment(IGR) group,diabetic( DM ) group ] according the results.The general information,laboratory examination and viral load of three groups were compared.Results ( 1 ) The difference of age,waist size,weight,body mass index data and hepatitis B virus load in IGR group and DM group compared with NGT group were statistically significant difference ( all P <0.05 ),IGR compared with DM group,all index comparative differences were not significant (all P > 0.05 ).(2)As the aggravating of glucose metabolism disorder,the positive rate of HBsAg,HBeAg and former S1 antigen of NGT group,IGR group and DM group were gradually raised ( all P < 0.05 ).Conclusion The body mass index and hepatitis B virus load are relatively independent risk factors of secondary glucose metabolism disorders in patients with hepatitis B liver cirrhosis,and which are positive correlation with it.However,gender,age,blood lipids are not obvious correlation with secondary glucose metabolism disorders.

Objective To observe the therapeutic effect of spleen-invigorating and superficies-consolidating therapy combined with life style intervention for improving constitution of recurrent respiratory tract infection(RRTI) children.Methods One hundred and twenty RRTI children were classified into excessive yang with deficiency yin syndrome(N = 60) and excessive yin with deficiency yang syndrome(N = 60).Then the excessive yang with deficiency yin syndrome were equally randomized into herbal medicine group Ⅰ and western medicine group Ⅰ,and excessive yin with deficiency yang syndrome were equally randomized into herbal medicine group Ⅱ and western medicine group Ⅱ.Herbal medicine groups Ⅰ and Ⅱ were given Fuganning Oral Liquid and life style intervention,and western medicine groups Ⅰand Ⅱ were given thymic peptide capsules and life style intervention.Eight weeks constituted one treatment course.The score of symptoms before and after treatment,and the therapeutic effect were compared after 2 treatment courses.Results(1) The symptoms score was improved obviously in herbal medicine groups and western medicine groups(P 0.05).The difference of therapeutic effect was significant between both of herbal medicine groups(P
7.EFFECTS OF ZINC DEFICIENCY ON BONE HISTOMORPHOMETRIC PARAMETERS OF RATS

Objective:To study the effects of zinc deficiency on bone histomorphometric parameters of femoral distal diaphysis in rats. Methods:Thirty Wistar rats were randomly allocated into three groups:the zinc-deficient group(ZD), the control group(Cont),and the pair-fed group(ZP). After the rats had been fed for eight weeks, the histomorphometric and dynamic parameters of the rats were analysed using bone histomorphometric method. Results:The number, volume and connectivity of trabecular bone, and the mean trabecular plate density of ZD rats were significantly decreased, but their mean trabecular plate space was significantly increased. In addition, the data showed that ZD animals had significantly decreased trabecular osteoid surface, reduced velocity of bone formation as compared with Cont and ZP animals. The results showed that in ZD rats the mineral deposit rate was significantly slow, while the mineralization lag and osteoid maturation period were obviously prolonged. Conclusion: Zinc deficiency reduces velocity of bone formation and prolongs bone mineralization and destroys bone structure.
8.Biodistribution and estimation of radiation-absorbed doses in humans for 13N-ammonia PET/CT

Objective To evaluate the biodistribution and radiation-absorbed doses of main organs in healthy people with 13 N-ammonia.Methods Five healthy volunteers underwent whole-body PET and CT scans after injection of 666-814 MBq of 13 N-ammonia.The serial dynamic emission images of each healthy volunteer were acquired.ROI were drawn manually based on the transverse CT images and transferred to the corresponding PET slices.Radiation-absorbed doses were calculated using the medical internal radiation dosimetry (MIRD) method.Results The highest concentrations of 13 N-ammonia were found in the heart,liver and kidneys,followed by pancreas,brain,spleen and stomach.The organ of highest absorbed dose was heart with (7.14±3.63) × 10-3 mGy/MBq.The whole-body absorbed dose was (2.11±0.44) × 10 3 mGy/MBq.The whole-body effective dose was (6.58± 1.23) × 10-3 mSv/MBq.Conclusion As one of the most important myocardial perfusion tracers,the whole-body 13 N-NH3 · H2 O PET appears to be safe for humans.
9.Tetrandrine induces retinoblastoma cells apoptosis via ROS induction and PI3K/Akt inhibition

Background and purpose:Tetrandrine is a natural compound whose role in retinoblastoma remains unclear. This study investigated the effects of tetrandrine (Tet) on human retinoblastoma cells.Methods:CCK-8 assays were performed to analyze the effects of Tet on viability of retinoblastoma cells. The apoptosis rate was determined by Annexin V/PI assays. After staining with 2′,7′-dichlorolfuorescin diacetate (DCFH-DA), cellular reactive oxygen species (ROS) was measured by lfow cytometry. Akt and p-Akt were detected by Western blot.Results:Tet inhibited cell viability of retinoblastoma cells. After treatment with Tet (4, 8, 10 and 20μmol/L) for 24h, cell viability inhibition rates of WERI-Rb-I were 5.7%, 25.0%, 55.1% and 84.9%, whereas inhibition rates of Y79 cells were 2.4%,2.9%, 23.8% and 54.2% (P<0.01). In cells treated with 10μmol/L of Tet for 12, 24 and 48 h, cell viability inhibition rates of WERI-Rb-I were 6.0%, 45.5% and 74.7%, whereas inhibition rates of Y79 cells were 2.9%, 19.4% and 43.3% (P<0.01). Tet induced retinoblastoma cell apoptosis. After treatment with Tet (10 μmol/L) for 24 and 48 h, apoptosis rates of WERI-Rb-I were (23.70±1.75)% and (34.83±3.15)%, respectively, whereas apoptosis rates of Y79 cells were (9.62±2.69)% and (14.97±1.50)%, respectively (P<0.01). Apoptosis inhibitor Z-VAD-FMK attenuated Tet-induced cell death (P<0.05). ROS levels were indeed increased in cells treated with Tet (10 μmol/L) for 6 and 12 h (P<0.01), while N-Acetyl-L-cysteine (NAC) decreased Tet-induced ROS (P<0.01). After ROS was inhibited by NAC, apoptosis rate was decreased compared with the control (P<0.01). Further study indicated that Tet inhibited PI3K/Akt pathway in retinoblastoma cells.Conclusion:Tet induces cell apoptosis via increasing ROS synthesis and inhibiting PI3K/Akt pathway.
10.EFFECT OF HOMOCYSTEINE ON POTENTIATING ?ETA-AMYLOID NEUROTOXICITY

Objective To examine the possibility that homocysteine(Hcy) may augment ?-amyloid(A?) neurotoxicity.Method Cultured hippocampal neurons were treatd with Hcy and/or A?42 and it's apoptosis,calcium influx,DNA damage and oxidative injury were examined.Results Combined treatment with 250?mol/L Hcy and 10?mol/L A?42 increased the apoptosis rate of hippocampal neurons significantly more than either agent alone,and even the sum of each agent treated alone.The combined treatment significantly exceeded the cytosolic calcium accumulation and MDA obtained with either alone or the sum of each agent.A significant increase in DNA damage also occurred in neurons exposed to A?42 in the presence of Hcy,but the magnitude of the increase was not greater than that seen with either treatment alone.Conclusion Hcy could potentiate A? neurotoxicity by neuron exitotoxicity,oxidative stress and inducing apoptosis.
