1.Effect of propolis flavone on expression of matrix metalloproteinase-9 in human umbilical vein endothelial cell

BACKGROUND: It has been proved that matrix metalloproteinase-9 (MMP-9) has a correlation with stability of atherosclerotic plaque, and propolis flavone plays a role in anti-atherosclerosis. OBJECTIVE: To investigate the effect of propolis flavone on expression of MMP-9 in human umbilical vein endothelial cell (HUVEC). DESIGN, TIME AND SETTING: The comparative observation was performed at the Institute of Life Science, Taishan Medical University from January to August 2008. MATERIALS: The propolis flavone was extracted by the Institute of Life Science, Taishan Medical University. The neonatal umbilical cord with 1 hour was supplied by Tai’an Central Hospital, and the informed consent was obtained from the puerperant. METHODS: HUVEC was cultured and identified in vitro, followed by treated with propolis flavoneat with different concentrations for 24 hours. The blank was used as the control group. MAIN OUTCOME MEASURES: The effect of propolis flavone on expression of MMP-9 in HUVEC was detected by cytoimmunochemical staining and ELISA. RESULTS: Propolis flavonecan with all concentrations can restrain the expression of MMP-9 after co-culture with the cells for 24 hours, which showed in a dose-dependent manner. CONCLUSION: The propolis flavone can obvious depress the expression of matrix metalloproteinase-9 in HUVEC.
2.The characteristics of tidal breath lung function and growth level among infants with recurrent wheezing

Objectives To study the relationship of recurrent wheezing and obesity among infants. Methods In 118 recurrent wheezing infants, the growth level are assessed based on the WHO standard. Meanwhile, all the infants are divided into two subgroups (normal-weight, overweight/obese), and the tidal breath lung function (tidal breath flow volume loop) are analyzed. Results Among the infants of 1-3 years old, for the growth level, the recurrent wheezing infants have higher index of weight, but almost the same level of height, compared with the WHO standard. For the tidal breath lung function, tidal volume (VT/kg) is significantly impaired in overweight/obese subgroup in comparison to the normal-weight subgroup. But the ratio of time to peak tidal expiratory flow (tPF) to total expiratory time (tE) and ratio of volume to peak expiratory flow (VPF) to total expiratory volume (VE), which reflect the airway obstructions to a certain extent, have no statistical differences among the two subgroups. Conclusions Rather than the height index, the weight index is significantly higher in the recurrent wheezing infants than the level of WHO standard. Compared with normal-weight patients, the tidal volume of overweight/obese recurrent wheezing in overweight/obese group is lower, but no significant differences are found for other parameters of airway obstructions.
3.Activation of liver X receptor upregulates fatty acid synthase expression in diabetic kidney

Objective To investigate the effect and mechanism of liver X receptor ( LXR ) agonist on expression of fatty acid synthase( FAS) in diabetic kidney. Methods In the part of in vivo study, immunostaining was used to detect the FAS protein expression in kidney. 16-week-old male db/db mice on C57BL/6 background were administered via gavage a LXR synthetic agonist, TO901317, at a dose of 3 mg · kg-1 · d-1 or vehicle ( 0. 5%Carboxymethyl Cellulose Sodium, CMC-Na) alone for 7 d;Quantitative RT-PCR and Western blot were used to detect mRNA and protein levels of FAS and SREBP-1. In the part of in vitro study, MCT cell(a mouse murine proximal tubule cell line)was treated with 10μmol/L TO901317 for 24 h or transfected with active SREBP-1c expression vector (SREBP-1cN). HEK293 cells(a human renal tubule cell line)were transfected with mFAS-(1. 7 kb)-luc, LXR expression vector or SREBP-1cN for 12 h. Quantitative RT-PCR and luciferase reproter assay were utilized to examine FAS mRNA level and FAS promoter activity. Results FAS was abundantly expressed in renal cortex, with low expresson in renal glomeruli. The mRNA and protein expressious of FAS in kidney of db/db mice were lowered compared with db/m mice. TO90137 treatment increased FAS mRNA expression by 1. 3-fold. TO901317 increased expression of SREBP-1 in kidneys of db/m and db/db mice by 5. 1-fold and 17-fold, respectively. TO901317 and overexpression of SREBP-1c increased expression of FAS in MCT cells by 1. 5-fold and 1. 8-fold. Transcription activity of FAS were induced by TO901317, LXR, and SREBP-1cN overexpressions in HEK293 cells. Conclusions Both direct(LXRE)and indirect(SREBP-1c)mechanisms may contribute to the up-regulation of FAS expression by LXR in renal proximal tubule cells.
4.Effects of nutrition status on activities of daily living among nursing home residents: the mediating role of physical function

Objective To explore the relationship between nutrition status and activity of daily livingof elders in nursing homes and the mediating effect of physical activity. Methods From Apr to Sep in 2016,320 elders in nursing homes were selected and were surveyed using the standardized tools. The mediating effects were analyzed with PROCESS. Results The scores of elders′ nutrition status, physical function and activities of daily living were (11.88±2.27), (3.61±3.51), (79.13±26.00) points, respectively. And there were 66.9% elders suffering from activities of living life impaired. The nutrition status were significantly positive correlated with physical function and activities of daily living of elders, and the physical function were significantly positive correlated with activities of daily living of elders. The physical function partly mediated the association between nutrition status and activities of daily living, and the effect was reached to 39.1%. Conclusions The elders′activities of daily life was poor in nursing home in China. The physical function partly mediated the association between nutrition status and activities of daily living. The related departments could combine the long time and short time plans targeting at improving of nutrition status and physical function respectively, to enhance the outcomes of elders in nursing homes.
5.Construction of the care needs assessment index for institutional elderly

Objective To construct the care needs assessment index for institutional elderly. Methods A total of 30 experts were consulted by the Delphi method. The indexes were selected and identified according to the inquiry results. Results The response rates of three expert consultation rounds were 93.33%(28/30), 85.71%(24/28), and 79.17%(19/24) respectively. The authority coefficients of the three rounds were all above 0.80. The assessment index includes 4 first level indicators, 13 secondary level indicators, and 48 third level indicators. Weighting results indicated that in terms of the elderly in institutional care, the most important domain of care needs was physical function, followed by ability of activity. At the same time, psychological function and social function in the elderly cannot be ignored. Conclusions The study obtained key elements that should be included in a comprehensive care needs assessment index of the institutional elderly, which laid a solid foundation for further investigation on the formation of specific assessment tools for care needs of the elderly in institutional care.
6. Nursing care of one case of incontinent dermatitis complicated with sacrococcygeal pressure ulcer

A 67-year-old female patient with incontinent dermatitis complicated with sacrococcygeal pressure ulcer was admitted to our unit in November 2017. The wound was treated with a new dressing based on the concept of wet healing, management of exudation, prevention of infection to promote wound healing. Meanwhile, the fecal incontinence was properly treated with colostomy bag combined with disposable negative pressure drainage device to avoid fecal contamination and aggravation of pressure ulcer. Incontinent dermatitis was treated with wound protective powder and wound protective film. After 14 days of treatment, the wound of pressure ulcer was reduced and the incontinent dermatitis was cured.

9.Effect of down-regulation of lncRNA LINC00263 targeting miR-4458 on regulating radiosensitivity of breast cancer SK-BR-3 cells

Objective:To evaluate the effect of down-regulating lncRNA LINC00263 targeting miR-4458 on the proliferation, migration, invasion and radiosensitivity of breast cancer SK-BR-3 cells.Methods:The expression differences of LINC00263 in breast cancer tissues, adjacent tissues, normal breast epithelial cells and breast cancer cells were determined by qRT-PCR. Transfection of LINC00263 shRNA in breast cancer SK-BR-3 cells down-regulated the expression of LINC00263, and the cloning experiment was used to detect the radiosensitivity. Breast cancer SK-BR-3 cells were treated with 6 Gy irradiation. CCK-8 assay was employed to detect cell proliferation. Flow cytometry was adopted to detect cell apoptosis. Transwell chamber test was performed to detect cell migration and invasion. Western blot was used to detect the expression levels of C-Caspase-3 and C-Caspase-9, MMP-2 and MMP-9 proteins. Bioinformatics software predicted that LINC00263 and miR-4458 had complementary binding sites, and the luciferase reporter system was utilized determine the targeting relationship between LINC00263 and miR-4458. LINC00263 shRNA and miR-4458 inhibitor were co-transfected into breast cancer SK-BR-3 cells, and 6 Gy irradiation was given to detect the changes in cell proliferation, apoptosis, invasion and migration.Results:The expression level of LINC00263 in breast cancer tissues was higher than that in adjacent tissues. The expression level of LINC00263 in breast cancer cells was higher compared with that in normal breast epithelial cells. The radiosensitivity of breast cancer SK-BR-3 cells was increased after transfection of LINC00263 shRNA. Transfection of LINC00263 shRNA and radiation exerted a synergistic effect, jointly inhibited breast cancer cell proliferation, migration and invasion, promoted cell apoptosis, up-regulated the expression levels of C-Caspase-3 and C-Caspase-9 proteins in cells, and down-regulated those of MMP-2 and MMP-9 proteins. Down-regulation of LINC00263 targetedly up-regulated miR-4458 expression. miR-4458 inhibitor reversed the inhibitory effect of LINC00263 shRNA combined with radiation on the proliferation, migration, invasion and apoptosis promotion of breast cancer SK-BR-3 cells.Conclusion:Down-regulating lncRNA LINC00263 targeting miR-4458 inhibits the proliferation, migration and invasion of breast cancer SK-BR-3 cells, and improves cell radiosensitivity.
10.Post-marketing immunogenicity and safety of domestic 23-valent pneumococcal polysaccharide vaccine: a multicenter study

Objective:To evaluate the post-marketing safety and immunogenicity of a 23-valent pneumococcal polysaccharide vaccine (PPV23).Methods:From September 2020 to June 2021, a clinical trial of single-dose PPV23 was conducted in people ≥3 years old in Centers for Disease Control and Prevention of Guizhou, Hunan and Fujian provinces. Blood samples were collects from the subjects before and 30 d after vaccination. ELISA was used to quantitatively detect IgG antibodies against capsular polysaccharides of 23 Streptococcus pneumoniae serotypes in serum samples. The adverse events (AEs) were monitored within 7 d after vaccination. Results:A total of 409 subjects were enrolled and included in safety analysis. Except for one with antibody level inversion, the other 408 participants were included in immunogenicity analysis. The levels of antibodies against the 23 Streptococcus pneumoniae serotypes were all increased after vaccination by an average of 4.24 folds. The two-fold growth rates of the antibodies ranged from 51.72% to 96.81% with a total two-fold growth rate of 78.59%. The overall rate of AEs was 27.14% (111/409). Local AEs were mainly pain, induration, redness and swollen. No serious adverse events related to vaccination occurred. Conclusions:This study preliminarily demonstrated the good immunogenicity and safety of PPV23 vaccine.
