1.Effect of levodopa on melanogenesis in and antifungal drug susceptibility of Penicillium marneffei

Objective To investigate the effect of levodopa on melanogenesis in Penicillium mameffei (PM),and to determine if melanization affects the antifungal drug susceptibility of PM.Methods A clinical isolate of PM,GXMU121011,was inoculated onto Sabouraud's dextrose agar (SDA) containing different concentrations (0.1-10 mmol/L) of levodopa at an inoculum density of 1.0 × 106 cfu/ml,or onto SDA containing 1 mmol/L levodopa at three inoculum densities (1.0 × 105,1.0 × 106,1.0 × 107 cfu/ml),and cultured at 37 ℃ for 7 days.Subsequently,melanization of PM colonies was observed.The paper-disk method was used for antifungal susceptibility testing,and the diameter of inhibition zones of itraconazole,fluconazole and amphotericin B against 8 clinical strains of PM was determined on SDA with or without levodopa.Results The melanization of PM colonies increased when the concentration of levodopa increased from 0.1 to 1.0 mmol/L,peaked when that reached 1.0 and 3.0 mmol/L,but mildly decreased when that continuously increased beyond 3.0 mmol/L,and a slight shrinkage was observed in PM colonies when that was 10.0 mmol/L.The color of colonies deepened along with the increase in inoculum density of PM.The average diameters of inhibition zones of itraconazole,fluconazole and amphotericin B against PM were all significantly lower on SDA with levodopa than on SDA without levodopa (all P < 0.05).Conclusions Levodopa concentration and inoculum density both affect melanogenesis in PM.Melanization may decrease the susceptibility of PM in yeast phase to itraconazole,fluconazole and amphotericin B in vitro.
2.Effect evaluation of meta-mood ability training on improvement of job satisfaction degree of nurses

Objective To explore the effect of meta-mood ability training for improving job satisfaction of nurses.Methods A total of 120 nurses were randomized into the observation group and the control group (60 cases in each group).The observation group received meta-mood ability training,while the control group was given routine training.The meta-mood ability and job satisfaction degree were investigated.Results After the intervention,job satisfaction degree and the meta-mood ability level of the observation group nurses were improved than that of the control group.Conclusions The meta-mood ability training is effective in improvement of job satisfaction degree of nurses,as well as increase clinical service quality of nurses.
3.Forensic Analysis of 33 Cases of Fatal Pulmonary Thromboembolism

Objective To explore the related risk facts of pulm onary throm boem bolism (PTE ) and analyze the relation betw een PTE and the traum a or m edical behavior by investigating the cases of PTE . Methods Thirty-three cases w ere selected from Institute of Forensic Science (IFS) from 2000 to 2014. Results In 33 cases, 16 decedents w ere m ale, 17 decedents w ere fem ale;different degrees of dyspnea, chest tight-ness and syncope sym ptom s w ere the clinical m anifestation of the deceased; the throm bus w as mainly distributed in the left and right pulm onary arteries. The main source of em bolism w as the deep vein of low er lim b and the left probability w as higher. Traum a, lim ited position, operation and cardiovascular disease show ed high-risk factors of PTE; D-D im er test, hem olytic test and com puter tom ography pul-m onary angiography w ere the diagnostic tools for PTE . In som e cases, traum a and m edical m alpractice could be involved in the cause of death. Conclusion N on-typical clinical sym ptom s present in the m ost cases caused by PTE , and these cases alw ays show m any high-risk factors. The relation betw een PTE and injury or m edical behavior should be considered carefully in the forensic pathological practice.
4.Ordinal regression analysis on influence factors of health literacy in middle-aged and elderly people in Hunan province

Objective To explore the influence factors of health literacy in middle-aged and elderly people in Hunan province.Methods With multi-stage randomized cluster sampling, people aged 45 years and over were selected from both urban and rural areas of six geographical regions of Hunan province, using the China residents health literacy questionnaire developed by Chinese Health Education Center, by face to face survey to get data. The influence factors were analyzed retrospectively with univariate and ordinal regression analysis.Results The 1158 persons were investigated and the valid questionnaires were 1154, the effective recovery rate was 99.7%. Univariate analysis showed that there were significant differences in heath literacy among different degree of education, age, gender, marital status, income, region and occupation (all P<0.05). The results of ordinal regression analysis Showed that the major influence factors were degree of education, income,age and occupation.Conclusions The people at low levels of education or income should be concerned about health education and health promotion strategies.
5.Construction and identification of recombinant lentivirus carrying human beta defensins-3 and connective tissue growth factor gene

Objective To construct a recombinant lentivirus containing human beta defensins -3 ( hBD3 ) , connective tissue growth factor gene (CTGF) and enhanced green fluorescent protein (EGFP), and to detect its translation in rabbit bone marrow mesenchymal stem cells (BMSC).Methods The lentivirus containing hBD3, CTGF and EGFP genes was constructed in vitro.The titer of lentivirus was tested with end-paint dilution assay .Rabbit BMSCs were transfected with recombinant virus.The best value of multiplicity of infection (MOI) was tested.The expression condition, transfection efficacy and genetic stability of the target genes were evaluated by using fluorescence microscopy and flow cytometry . Western blotting was used to detect the expression of the target protein .Results Recombinant lentivirus vectors: Lenti-CTGF-hBD3-EGFP, Lenti-hBD3-EGFP, and Lenti-EGFP, were successfully obtained . The titer of the recombinant lentiviruses was 3.21 ×108, 5.80 ×108, and 1.16 ×109, respectively.The best MOI value to transfect BMSCs was 150. The transfection efficacy of these lentivirus vectors was high , reaching 79.72%as assessed by flow cytometry , and it could be stably inherited .Western blotting displayed that target protein expression was successful .Conclusion The construction of recombinant lentiviruses carrying hBD3 and CTGF genes is successful and can be effectively transfected into BMSCs .
6.Two kinds of goniosynechialysis for primary acute angle-closure glaucoma with cataract

AIM: To study the effect of two kinds of phacoemulsification combined with goniosynechialysis for primary angle-closure glaucoma with cataract.
METHODS: Phacoemulsification combined with goniosynechialysis was performed in 60 patients 62 eyes for primary angle-closure glaucoma with cataract. They were randomly divided into two groups, 31 eyes underwent phacoemulsification with goniosynechialysis combined with I/A catheterization were performed in Group A; in Group B, 31 eyes were performed phacoemulsification with goniosynechialysis combined with viscoelastics. Best corrected visual acuity, the intraocular pressure, anterior chamber depth, anterior chamber angle width and pupil size were measured before the operation, and 3mo after the operation.
RESULTS: Intraocular pressure of all cases were reduced obviously after the operation(P<0.05). Visual acuity of the 62 eyes were improved in different extent; the anterior chamber depth and anterior chamber angle width after 3mo of the operation were deeper and wider than those before the operation(P<0.05). There were no different in the intraocular pressure, best corrected visual acuity and anterior chamber depth between the two groups(P>0.05), but, Group A was superior to Group B in anterior chamber angle width(P<0.05). At 3mo after operation, there were 8 eyes in Group A with more than 1mm lessen pupil size, two eyes in Group B had the same.
CONCLUSION: Two kinds of goniosynechialysis combined with phacoemulsification in the treatment of primary acute angle-closure glaucoma with cataract are effective. Among them, the goniosynechialysis with I/A was more effective to increase the anterior chamber angle width.
7.Predictive efficacy of different intrauterine adhesion (IUA) classification systems on the prognosis of patients with IUA

Objective To explore the effects of different intrauterine adhesion (IUA) classification systems on predicting the IUA prognosis.Methods One hundred cases were selected as the subjects in present study from those diagnosed with IUA and underwent surgery in Zhujiang Hospital of Southern Medical University from Jan.2010 to Jan.2017,and were followed up for two years.According to the actual situation,all patients were scored by March,AFS,ESGE and Chinese classification for comparing the effects of different IUA classification systems on predicting the pregnancy rate,live birth rate and effective rate within 2 years after surgery.Results ESGE classification had a good effect on predicting the postoperative live birth rate and effective rate,and a certain predictive effect on pregnant rate,with the area under curve (AUC) of 0.722,0.754 and 0.635,respectively.March classification had a certain effect on predicting the postoperative live birth rate and effective rate with AUC of 0.635,0.754,respectively,but had a poor effect on predicting pregnant rate.AFS classification and China classification had poor effect on predicting the IUA prognosis.Conclusion ESGE classification system is better than the other systems including March,AFS and Chinese classification,on predicting the IUA prognosis,but further verification in large sample size is still required.
8.Phenotype and Genotype of Nosocomial Infection Strains of Klebsiella pneumoniae

OBJECTIVE To investigate the prevalence,antibiotic resistance and genotype of the extended-spectrum ?-lactamases(ESBLs)-producing clinical isolates of Klebsiella pneumoniae.METHODS A total of 104 isolates of K.pneumoniae were examined for the ESBLs production and the susceptibilities of the bacteria to 15 antimicrobial agents.PCR was performed to detect the genes encoding the ESBLs belonging to SHV and TEM families as well as CTX-M-1 and CTX-M-9 groups.RESULTS The ESBLs-producers of K.pneumoniae were 54.0% in the total of 104 isolates.Almost all of the ESBLs-producing isolates were resistant to the antibiotics commonly used,and only remained susceptible to carbapenems and the combination of cefoperazone with sulbactam.The genes of SHV,CTX-M-1 and TEM groups were detected in the ESBLs-producing isolates by 64.3%,46.4%,and 32.1%,respectively,and 35.7% and 8.9% of ESBLs-producing K.pneumoniae strains carried two and three genes.CONCLUSIONS The clinical isolates of K.pneumoniae in Tianjin Nankai Hospital are shown a high rate of ESBLs-producing and antibiotic resistance.SHV and CTX-M-1 groups of ESBLs are the dominant genotypes in the isolates of ESBLs-producing K.pneumoniae.
9.Effects of estrogen on the fibrosis process of intrauterine adhesions and the expression of forkhead box F2

Objective To investigate the effect of estrogen on the fibrosis process of intrauterine adhesions and the expression of forkhead box F2 (FoxF2).Methods Primary human endometrial stromal cells (HESCs) were obtained by separation with 0.2% collagenase Ⅰ digestion-mesh filtration-differential adherence,and identified by immunocytochemistry.HESCs affected with 10ng/ml transforming growth factor β1 (TGF-β1) for 48 hours.HESCs in model group were affected with 0,10-6,10-8,10-10 and 10-12mol/L estrogen,the expressions of smooth muscle actin alpha (α-SMA),Collagen I (COL Ⅰ) and FoxF2 were detected by quantitative PCR (qPCR) and Western blotting.Results HESCs with high purity and good activity were obtained by using 0.2% collagenase Ⅰ digestion-mesh filtration-differential adherence separation method.Immunocytochemistry showed positive vimentin and negative cytokeratin 18 in HESCs.The results of qPCR and Western blotting showed that the mRNA and protein expression levels of α-SMA,COL Ⅰ and FoxF2 were higher in model group than in control group (P<0.05),the model was built successfully.qPCR revealed that the mRNA expression levels ofα-SMA,COL Ⅰ and FoxF2 were significantly lower in 10-6,10-8 and 10-10mol/L estrogen groups than in model group (P>0.05 in 10-10mol/L estrogen group,P<0.05 in other groups),while in 10-12mol/L estradiol group,the expression levels of FoxF2 mRNA significantly decreased (P<0.05),and of α-SMA and COL Ⅰ mRNA increased,but no significant difference were found (P>0.05).Compared with the model group,the protein expression levels of α-SMA,COL Ⅰ and FoxF2 in 10-6,10-8 and 10 10mol/L estrogen groups decreased,but no significant difference was found (P<0.05),while in 10-12mol/L estradiol group,the expression levels ofα-SMA protein increased (P>0.05),and of COL Ⅰ and FoxF2 proteins decreased (P<0.05).Conclusions The expression of FoxF2 in intrauterine adhesions is increased.Estrogen can reverse the fibrosis process of intrauterine adhesions in a certain range and inhibit the expression of FoxF2.
10.Genetic analysis of translocation of chromosome t(4;9) in a child with rare chromosome abnormality.

A 10-month-old boy suspected of genetic abnormality was admitted for fever and coughing. Routine G-banding chromosome analysis of the peripheral blood lymphocytes and hereditary pattern analysis on the basis of the karyotypes and disease history revealed that the karyotype of the boy's mother was 46, XX,t(4;9)(q31;p24), and that of the boy was XY,der(9)t(4;9)(q31;p24)mat. The mother was identified as a carrier of balanced translocation of the chromosome who gave the abnormal chromosome 9 to her son, and she had only a chance of 1:18 to have a normal offspring. This case reiterates the importance of antemarital examination and prenatal diagnosis for preventing chromosomal diseases.
