Tianjin Medical Journal 2016;44(8):1014-1017
Objective To explore the effects of aerobic exercise on 6-minute walk distance and serum level of omentin-1 in maintenance hemodialysis (MHD) patients. Methods A total of 89 MHD patients (MHD group) in the 5th Central Hospital of Tianjin were included in the study from December 2013 to November 2014. Patients were randomly divided into two groups:routine treatment group (n=43) and aerobic exercise treatment group (aerobic exercise group, n=46). Fifty healthy subjects were selected as normal control group. Both groups received the same hemodialysis and regular treatments, and the aerobic exercise group received 6-month interdialytic aerobic exercise, and the routine group did not take any exercise intervention. The 6-minute walk distances (6MWDs),body mass, height and blood pressure were recorded, and serum levels of omentin-1, C-reactive protein (CRP), blood routine test and biochemistry test were determined in two groups. The relationship between 6 MWDs, age, CRP, left ventricular ejection fraction (LVEF) and omentin-1 were analyzed. Results The serum levels of omentin-1 and CRP weresignificantly higher, LVEF was significantly lower, in MHD group than those of healthy control group (P<0.01). After 6 months of aerobic exercise, 6MWDs, LVEF, hemoglobin and serum level of omentin-1 were significantly increased than those before treatment in aerobic exercise group. The 6MWDs, LVEF and hemoglobin were significantly increased after treatment than routine group. The levels of CRP, fasting glucose and blood pressure were significant decreased after treatment. The levels of CRP, fasting glucose and systolic blood pressure were decreased in treatment group than those of routine group. Before treatment, 6MWDs was negatively related with age and CRP, and positively related with LVEF and serum level of omentin-1 in MHD patients (r=-0.418,-0.229, 0.252 and 0.234, P<0.05). Conclusion Aerobic exercise can significantly increase values of 6MWDs, LVEF, hemoglobin and serum level of omentin-1, and reduce values of CRP, fasting blood glucose and systolic blood pressure in MHD patients. Aerobic exercise can improve physical performance, heart function and micro inflammatory state, thereby reduce cardiovascular events and mortality risk in MHD patients.

Surgical resection (usually lobectomy) is considered the treatment option for individuals with stage Ⅰ and Ⅱ non-small cell lung cancer. The surgical treatment of stage Ⅰ and Ⅱ non-small cell lung cancer (NSCLC) continues to evolve in the areas of intraoperative lymph node staging (specifically the issue of lymph node dissection vs sampling), the role of sublobular resections instead of lobectomy for treatment of smaller tumors (especially peripheral carcinoma ≤2 cm in diameter), and the use of video-assisted techniques to perform anatomic lobectomy. Video-assisted thoracic surgery (VATS) lobectomy provides a minimally invasive approach for the management of early-stage lung cancer. Questions about the safety of VATS lobectomy and its adequacy as a cancer operation compared with open thoraeotomy have hindered its universal acceptance among thoracic surgeons. Evidence suggests that VATS lobectomy can be safely performed and is an adequate cancer operation for early-stage NSCLC. Recently, robots have been introduced into surgical procedures in an attempt to facilitate surgical performance. However, adequately powered well-balanced studies comparing VATS with open thoracotomy for lobectomy are lacking in the literature.

Objective To know the actual level of hair lead levels in children of Xi'an. Methods Hair samples were collected from 203 children aged 7-13 years of an urban primary school and a suburban one during April-June, 2001. The hair lead levels were measured with flame atomic absorption spectrophotometer. Results The geometric mean of hair lead levels of children in urban area was 4.69 ?g/g, while in suburban area it was 1.67 ?g/g. There was a significant difference between them (t=7.78, P=0.001). The younger group had a higher hair lead levels in urban area and suburban area. With age, hair lead levels showed a significant down trend. There was no significant difference had been seen between boys and girls living in the city, but the hair lead levels of rural boys(median 2.92 ?g/g) were higher than rural girls (median 1.52 ?g/g), P=0.02. Rate of high hair lead (13.2%)of urban children, based on the standard (≥10.0 ?g/g), was higher than rural children (1.2%) (?2=9.18, P=0.002). Conclusion The hair lead levels of the children living in urban districts are higher than that in the suburb area of Xi'an city. The rate of high hair lead of urban children was higher than that of the suburban children. The hair lead levels of children aged 7-13 years in Xi'an have showed a down trend with age. There is no significant difference between boys and girls in urban area, but the hair lead levels in suburban boys is higher than that of the suburban girls.

Objective To investigate the effect of hypoxia on the expression of vascular endothelial growth factor (VEGF) in villous explants, and the effect of hypoxia-inducible factor 2? (HIF-2?) on the VEGF expression. Methods The villous were obtained from women who underwent suction and curettage for termination at 6-8 gestational weeks. Four groups were divided: normoxia group; hypoxia group; sense HIF-2? oligonucleotide group and anti-sense HIF-2? oligonucleotide group. All villous were cultured in vitro. In anti-sense and sense group, villous were incubated under hypoxia with HIF-2? oligonucleotide. The villous explants were collected after 48 h of cultivation. The protein level of HIF-2? was detected by Western blot and the mRNA of HIF-2? and VEGF were examined by real-time PCR. Results The expression of HIF-2? mRNA and VEGF mRNA as well as the protein level of HIF-2? were increased in hypoxic group comparing with the normoxia group. Compared with the sense group, the expression of HIF-2? protein and HIF-2?, VEGF mRNA showed significant decrease in anti-sense group. Conclusions Hypoxia may result in the increased expression of VEGF in villous explants during the early trimester, which might be mediated by HIF-2?.

Objective To unmask clues for the investigation of PIH pathogenesis by detecting the altered gene expression profile of placentas from PIH patients. Methods Clinical data and placentas were collected from 42 PIH patients and 22 normotensive pregnancies in seclective cesarean section to construct PIH database from Mar. 2001 to Sep. 2002 in Xi’an district. Suppression subtractive hybridization (SSH) was performed to set the subtractive cDNA library and differential screening was used to identify the positive clones;insertion of positive clones were sequenced by T7 primer method. RT-PCR and immunohistochemistry were employed to confirm the putative gene inhibin A expression. Results A database was set up that consists of pre- and postpartum clinical data and placenta samples of 42 preeclamptic pregnancies and 22 normotensive ones From which,150 features of placental tissue microarray were made. One hundred and three positive clones were isolated by SSH and differential screening. Sequencing and BLAST analysis showed that 90 insertion shared more than 95% homology with sequences in the GenBank/EMBL database. We identified 36 putative genes including pregnancy-specific glycoproteins gene (BC005924),serine protease inhibitor gene (BC012868),VEGFR-1 gene (AF063657),cytokeratine 7 gene(CK7) (AF509887),etc. Inhibin beta A gene was highly expressed in placentas in PIH patients. Conclusion The gene expression profile in PIH placenta was greatly changed;it might be necessary to investigate the role of VEGFR1,serine protease inhibitor, inhibin and CK7 in the pathogenesis of PIH.

Objective To study the effects of TSPG on apoptosis of K562 cells and the probable mechanism involved. Methods MTT was used to investigate the proliferation of K562 cells;Flow cytometry(FCM) was used to investigate the effects of TSPG on apoptosis of K562 cells; The expression of Survivin in K562 cells treated with differ-ent concentraction of TSPG were examined by RT-PCR. Results The growth of K562 cells was inhibited by TSPG in the concentration dependent manner (P < 0.05). FCM showed that the apoptosis rates of cells in 100 μg/L (23.78%) and 200μg/L TSPG group(33.98%) were higher than those in 101μg/L TSPG group(16.67%), with significant difference(P <0.01 or P < 0.05). The expression rates of Survivin were decreased by the treatment with the increasing concentrations of TSPG (P < 0.05). Conclusion TSPG can restrain the human leukemic cell growth and induce cell apoptosis, which may be related to the decreased expressions of Survivin.

Objective To evaluate the targeting and photodynamic activity of photosensitizer Ⅰ (PSⅠ)for laryngocarcinoma Hep-2 cells. Methods The photostability of PSⅠ was measured by bleaching assay. The cellular uptake of PSⅠ by Hep-2 cells were evaluated by fluorescence spectroscopy, the effects of PSⅠconcentration, irradiation dose and etc on the viability of Hep-2 cells were investigated by MTT assay. Results After irradiation of 50 min,the OD value of PS Ⅰ decreased 11%,which indicated that the stability of PSⅠ can meet the requirement of PDT.The cellular uptake of PS Ⅰ by Hep-2 cells increased with the concentration of PSⅠ and can be obviously inhibited by the presence of excessive free folic acid (P < 0.01).The results of MTT assays demonstrated that the viability of Hep-2 showed negative correlation with the PSⅠ concentration and irradiation dose. The viability of Hep-2 was only 34 % after PDT with 14 μ mol/L of the PS Ⅰ and 18 J/cm2 of irradiation. However, the viability of Hep-2 was still 100 % without irradiation, even though the concentration of PS Ⅰ was up to 1 10 μmol/L.The PDT carried out after 24 h of PSⅠ administration can efficiently inhibited the growth of Hep-2 with the survival rate of 32 %.Conclusion The PS Ⅰ posses satisfactory photostability and lower dark cytotoxicity, and displays significant targeting and photocytotoxicity for Hep-2 cells.
