Objective To evaluate the targeting and photodynamic activity of photosensitizer Ⅰ (PSⅠ)for laryngocarcinoma Hep-2 cells. Methods The photostability of PSⅠ was measured by bleaching assay. The cellular uptake of PSⅠ by Hep-2 cells were evaluated by fluorescence spectroscopy, the effects of PSⅠconcentration, irradiation dose and etc on the viability of Hep-2 cells were investigated by MTT assay. Results After irradiation of 50 min,the OD value of PS Ⅰ decreased 11%,which indicated that the stability of PSⅠ can meet the requirement of PDT.The cellular uptake of PS Ⅰ by Hep-2 cells increased with the concentration of PSⅠ and can be obviously inhibited by the presence of excessive free folic acid (P < 0.01).The results of MTT assays demonstrated that the viability of Hep-2 showed negative correlation with the PSⅠ concentration and irradiation dose. The viability of Hep-2 was only 34 % after PDT with 14 μ mol/L of the PS Ⅰ and 18 J/cm2 of irradiation. However, the viability of Hep-2 was still 100 % without irradiation, even though the concentration of PS Ⅰ was up to 1 10 μmol/L.The PDT carried out after 24 h of PSⅠ administration can efficiently inhibited the growth of Hep-2 with the survival rate of 32 %.Conclusion The PS Ⅰ posses satisfactory photostability and lower dark cytotoxicity, and displays significant targeting and photocytotoxicity for Hep-2 cells.

Hemagglutinin (HA) contains a head domain with a high degree of variability and a relatively conserved stem region. HA is the major viral antigen on the surface of the influenza virus. To define the biologic activities of chimeric HA bearing different head domains and stem regions or their potential use, a HA chimeric gene containing the head domain of the H7 subtype virus and stem region of the H3 subtype virus was modified and expressed using a baculovirus expression vector. Then, the secreted protein was purified and its biologic activities characterized. Approximately 1.4 mg/mL cH7/3 HA could be obtained, and its molecular weight was ≈ 70 kD. The trimer form of cH7/3 protein had hemagglutination activity and could be recognized by specific antibodies. The method described here can be used for further studies on the screening of HA stem-reactive antibodies or the development of vaccines with conserved epitopes.

Chinese Journal of Orthopaedic Trauma 2020;22(6):529-535
Objective:To compare the biomechanical performance between 5 internal fixation methods for fracture of the acetabular posterior column using finite element analysis. Methods:One healthy female volunteer (50 years old, 160 cm in height and 63 kg in weight with a body mass index of 24.6 kg/m 2) was recruited for this study to undergo CT scan. Her CT scan data were imported into software Mimics15.0 in the format of DICOM to generate a 3D model of the pelvis. After pre-processing and accessory ligament structures added, a 3D finite element model of the pelvis was established and verified by software ANSYS. Next, software ANSYS was used to generate a model of acetabular posterior column fracture, on which models of 5 internal fixation methods (internal iliac-ischial plating, conventional posterior column plating, anterograde tensile screwing, lesser sciatic notch screwing and ischial tuberosity screwing) were established. Then stress was loaded onto the 5 internal fixation models to compare stress displacements of the fracture line on the articular surface and sites of stress concentration between the 5 internal fixation models. Results:The average stress displacement of the fracture line at the standing position was 6.13 μm for conventional posterior column plating, <6.85 μm for ischial tuberosity screwing, <7.07 μm for lesser sciatic notch screwing, <7.08 μm for internal iliac-ischial plating, and <7.85 μm for anterograde tensile screwing; the average stress displacement of the fracture line at the sitting position was 7.77 μm for conventional posterior column plating, <9.65 μm for internal iliac-ischial plating, <9.69 μm for anterograde tensile screwing,<10.1 μm for lesser sciatic notch screwing, and<10.20 μm for ischial tuberosity screwing. In all the 5 internal fixation models, the stress was chiefly concentrated at fracture ends. Conclusion:Internal iliac-ischial plating and conventional posterior column plating provide better stability than the hollow screws (anterograde tensile screwing, ischial tuberosity screwing and lesser sciatic notch screwing) for fracture of the acetabular posterior column.

De novo variants (DNVs) are one of the most significant contributors to severe earlyonset genetic disorders such as autism spectrum disorder, intellectual disability, and other developmental and neuropsychiatric (DNP) disorders. Presently, a plethora of DNVs have been identified using next-generation sequencing, and many efforts have been made to understand their impact at the gene level. However, there has been little exploration of the effects at the isoform level. The brain contains a high level of alternative splicing and regulation, and exhibits a more divergent splicing program than other tissues. Therefore, it is crucial to explore variants at the transcriptional regulation level to better interpret the mechanisms underlying DNP disorders. To facilitate a better usage and improve the isoform-level interpretation of variants, we developed NeuroPsychiatric Mutation Knowledge Base (PsyMuKB). It contains a comprehensive, carefully curated list of DNVs with transcriptional and translational annotations to enable identification of isoformspecific mutations. PsyMuKB allows a flexible search of genes or variants and provides both table-based descriptions and associated visualizations, such as expression, transcript genomic structures, protein interactions, and the mutation sites mapped on the protein structures. It also provides an easy-to-use web interface, allowing users to rapidly visualize the locations and characteristics of mutations and the expression patterns of the impacted genes and isoforms. PsyMuKB thus constitutes a valuable resource for identifying tissue-specific DNVs for further functional studies of related disorders. PsyMuKB is freely accessible at http://psymukb.net.
