1.Further enhancing patient satisfaction after laser corneal refractive surgery

Corneal refractive surgery has been the major solution for the correction of ametropia.With the standardization of preoperative examination,intraoperative procedure and perioperative drugs,corneal refractive surgery has become much safer.Meanwhile,patients can get not only good visual acuity,but also favorable visual quality.However,the patient satisfaction has not been improved dramatically with the enhance of effectiveness and safety after surgery.Increase of the satisfaction degree to corneal refractive surgery is related to multiple factors such as experienced and highly skilled operation,operative safety,lessening operative complication,good postoperative visual and life quality of patients.Comprehensively analyzing existing problems in corneal refractive surgery in China and further obtain satisfaction of patient will be of an important significance for the healthy development of laser corneal refractive surgery.
2.Urea Combine with Glucocorticoid Treatment of Infancy Hemangioma Associated with Thrombopenia Syndrome

Objective To explore the treatment of hemangioma associated with thrombopenia(Kasabach-Merritte syndrome,KMS) in infant.Methods The clinical manifestation and the therapy of 14 cases patients with KMS in hospital from 2003 to 2006 were collected ang analyzed,the 400 g/L urea(manufactured by ourself)combined with methylprednisolone local injection were used and followed 0.5 to 1.0 year.Results Two in 14 cases were emergency exairesis,7 cases were recurred for several times,12 cases were cured,1 case was improved,1 case was loss the connection.Conclusions It is an ideal therapy method to use the urea combined with glucocorticoid to treat infancy KMS,little trauma,definite effective,low side effect,high cure rate.
3.Locating the displacement of the steel wire implantation with the stereotactic mammography

Objective To analyze the manifestation,reason,the processing method of the steel wire implantation with the sereotactic mammography to improve the accuracy of the preoperative positioning.Methods Seventy-nine cases which got the stereotactic steel wire implantation.In 96 lesions, 13 had steel wire displacement.Among them,5 cases got steel wire displacement during the sereotactic process,5 cases got steel wire displacement after the stereotactic process,2 cases got steel wire displacement during the operation,one case did not show the calcification on the postoperative radiography.Results The steel wire displacement occurred in 5 cases during the stereotactic process came from the patients and doctors respectively and the repositioning was needed.The steel wire displacement after the stereoscopic positioning was attributed to the overdose injection of local anesthesia,which led to the mismatch between the depth of Z axis of the mammary gland and the actual depth the computer given,the incorrect method for needle placement,and,neglecting whether the steel wire have got the lesion anchored when pulling out the needle set of steel wire hood,besides,these three kinds of instances above were all exaggerated by the accordion effect.For the displacement within 2 cm,the lesion can be excised toward the pathological change direction according to the position that steel wire prompted and re-place the second steel wire,putting the J-shaped steel wire into the needle hood and taking it out of the body.After repositioning,2 cases had the steel wire prolapse during operation,which resulted from the over-lifting of the steel wire.After placing the steel wire, the radiologist should give an accurate description on the depth and direction to the surgeon and the notch should be taken for incision from the steel wire head end which is proximate to skin.The postoperative specimen from one case had no calcification,which might be related to the condition that the calcification was located in the gland body,which got destruction from the surgical electrical electrotome.The excisionscope should be extended and the short term reexamination is recommended to make sure the complete excision of the calcification.Conclusion It is the gold standard method that implanting the steel wire with the stereotactic mammography to guide the surgical dissecting technique to diagnose non-palpable breast lesion(NPBL).Thorough understanding of the displacement manifestation of implanting steel wire with stereotactic technique and the treatment methods will be helpful in the surgical dissecting guidance.
4. Effect of Yishenbaozhen Recipe on serum erythropoietin level in renal anemia rats

Objective: To explore the intervention effect of Yishenbaozhen Recipe (YSBZR) on the renal anemia in rats with chronic renal failure and the related mechanism. Methods: Totally 43 rat models of chronic renal failure were produced by two-step 5/6 nephrectomy. The study was randomly divided into 5 groups: normal control group (n=9), model group (n=8), YSBZR high-dose group (n=9, 46. 5 g/[kg • d]), YSBZR low-dose group (n=9, 15. 6 g/[kg • d]) and Niaoduqing group (n=8, 2.5 g/[kg • d]). YSBZR was given intragastrically on a daily bases for 6 weeks, and then the serum erythropoietin (EPO), serum creatinine (SCr), blood urea nitrogen (BUN), hemoglobin (Hb), red blood cell count (RBC), and mean corpuscular volume (MCV) were examined. Results: Compared with the model group and Niaoduqing group, YSBZR high-dose group and low-dose group had significantly higher Hb, RBC and EPO levels (P<0.05), and had similar MCV levels. YSBZR high-dose and low-dose groups had significantly decreased SCr and BUN levels compared with the modll group (P<0.01), and had similar levels when compared with Niaoduqing group. Conclusion: YSBZR has interventional effect on renal anemia, which might be related to decreasing nitrogen retention, reducing the toxin-induced inhibition of hematopoietic system, and increasing EPO level.
5.Relationships between changes of kernel nutritive components and seed vigor during development stages of F1 seeds of sh2 sweet corn.

The changes of kernel nutritive components and seed vigor in F1 seeds of sh2 sweet corn during seed development stage were investigated and the relationships between them were analyzed by time series regression (TSR) analysis. The results show that total soluble sugar and reducing sugar contents gradually declined, while starch and soluble protein contents increased throughout the seed development stages. Germination percentage, energy of germination, germination index and vigor index gradually increased along with seed development and reached the highest levels at 38 d after pollination (DAP). The TSR showed that, during 14 to 42 DAP, total soluble sugar content was independent of the vigor parameters determined in present experiment, while the reducing sugar content had a significant effect on seed vigor. TSR equations between seed reducing sugar and seed vigor were also developed. There were negative correlations between the seed reducing sugar content and the germination percentage, energy of germination, germination index and vigor index, respectively. It is suggested that the seed germination, energy of germination, germination index and vigor index could be predicted by the content of reducing sugar in sweet corn seeds during seed development stages.

6.Effect of nicotinamide mononucleotide on insulin secretion and gene expressions of PDX-1 and FoxO1 in RIN-n5f cells

Objective To investigate the effect of nicotinamide mononucleotide (NMN) on insulin secretion and gene expressions of pancreatic and duodenal homeobox 1 ( PDX-1 ) and forkhead box-containing protein O-1 ( FoxO1 ),which were important transcription factors for insulin secretion.Methods Insulin secretion level in RIN-m5f cells was detected by rat insulin ELISA detection kit.The mRNA expression levels of PDX-1 and FoxO1 in RIN-m5f cells were analyzed by real-time PCR.The protein expression of PDX-1 was measured by Western blot.Results Insulin secretion levels in RIN-m5f cells treated with repaglinide ( 10 nmol/L) plus NMN ( 100 μnol/L) was significantly higher than those in the blank control,the DMSO control group,and the NMN (50μmol/L) treated group (P <0.05 ).The mRNA expression levels of PDX-1 in RIN-m5f cells treated with NMN ( 10,50 and 100 μmol/L) for 36 h were significantly higher than those in the control group (P <0.05,P < 0.01,and P < 0.001,respectively).There was marked differences in the mRNA expression levels of PDX-1 among different concentrations of NMN (P <0.001 ),but no significant differences in the mRNA expression level of FoxO1 ( P > 0.05).No significant difference was found in the protein expression levels of PDX-1 in RIN-m5f cells treated by NMN (50,100,and 200 μmol/L) for 36 or 48 h compared with the control group (P > 0.05).Conclusion NMN can stimulate insulin secretion and upregulate the mRNA expression of PDX-1 in RIN-m5f cells.
7.Sanggenon C induces apoptosis of prostate cancer PC3 cells by activating caspase 3 and caspase 9 pathways.

OBJECTIVETo investigate the effects of Sanggenon C in inducing apoptosis of prostate cancer PC3 cell line and explore the underlying mechanism.
METHODSThe proliferation of PC3 cells treated for 24 h with 1, 5, 20, 50, and 100 µmol/L sanggenon C or treated with 20 µmol/L Sanggenon C for 0, 6, 12, 24 and 48 h was evaluated using MTT assay. Flow cytometry was performed for analysis of apoptosis of PC3 cells after exposure to sanggenon C with different treatment protocols, and the activity of caspase 3 was detected using spectrofluorometry. The inhibitory effect of sanggenon C on PC3 cells pretreated with DMSO, z-DEVD-fmk, z-LEHD-fmk or z-IETD-fmk for 1 h was detected by MTT assay.
RESULTSSanggenon C inhibited the proliferation of PC3 cells in a dose- and time-dependent manner (P<0.05 except for 1 µmol/L group) with a 24-h ICof 18.76 µmol/L. Sanggenon C at 20 µmol/L caused inhibition rates of PC3 cells of 10.57%, 27.09%, 51.88%, 80.73% and 87.99% after treatment for 6, 12, 24, 48, and 72 h, respectively (P<0.05), and resulted in apoptosis rates of 7.43%, 20.91% and 37.56% at 12 h, 24 h and 48 h, respectively. Sanggenon C significantly increased caspase-3 activity in the cells, and its effect on PC3 cell proliferation was partially reversed by caspase 3 and caspase 9 inhibitors.
CONCLUSIONSanggenon C can dose-dependently induce growth inhibition and apoptosis of PC3 cells possibly by activating caspase 9 and caspase 3 pathways.
8.Clinical application of anterolateral femoral skin flap pedicled with high site cutaneous branches of the transverse branch of lateral circumflex femoral artery

Objective To report the clinical effect of free transplanting for soft tissue defects pedieled with high site cutaneous branches of the transverse branch of lateral cirumflex femoral artery.Methods Cu- taneous branches of the descending branch of lateral circumflex femoral artery were found small or abscent in 7 patients.The anterolateral femoral skin flap was pedicled with high site cutaneous branches of the transverse branch to repair the soft tissue defects of the arm,hand,leg and foot,rather than with the descending ones. The size of the flap ranged from 15 cm?6 cm to 28 cm?13 cm,with part muscle valve,iliotibal tract and lat- eral femoral cutaneous nerve.The fractures were performed with internal or external fixation.Results All of the anterolateral femoral skin flap survived well postoperatively in the 7 cases and had good appearance and sensation at one stage.The function of the repaired extremities recovered well.Conclusion The anterolat- eral femoral skin flap pedicled with high site cutaneous branches of the transuvrse branch of lateral circumflex femoral artery has many advantages of good blood supply and large size.The flap was secluding,and can be taken with some muscle and lateral femoral cutaneous nerve.When cutaneous branches of the descending branch of lateral circumflex femoral artery is small or abscent,the anterolateral femoral skin flap with high site cutaneous branches of the transverse branch of lateral circumflex femoral artery is an optimal alternative.
9.Role of NF-κB in factor VIIa-induced proliferation and migration of colon cancer cell line SW620 cells.

OBJECTIVETo explore the roles of NF-κB in factor VIIa-induced proliferation and migration of a colon cancer cell line (SW620) in vitro and its possible mechanism.
METHODSThe expression levels of NF-κB (p65), inhibitory protein of NF-κB (IκB-α), caspase-7, interleukin 8 (IL-8) and tissue factor (TF) in SW620 cells treated with factor VIIa, PDTC (an inhibitor of NF-κB) and other factors were measured by Western-blotting and real-time PCR. Proliferation and migration of the cells were analyzed by flow cytometry and Transwell assay, respectively.
RESULTSFactor VIIa down-regulated the IκB-α level in SW620 cells and increased the intranuclear level of NF-κB. Those effects of factor VIIa were blocked by anti-TF or anti-PAR2 antibodies. The effects of factor VIIa on proliferation and migration of SW620 cells, expression of IL-8, TF as well as caspase-7, were interfered by PDTC (the inhibitor of NF-κB).
CONCLUSIONSTF/VIIa complex activates NF-κB pathway via PAR2, thereby up-regulates IL-8 and down-regulates caspase-7 expression in SW620 cells, finally promotes proliferation and migration of colon cancer cells. In addition, TF/VIIa/PAR2/NF-κB pathway also upregulates TF expression, thus to create a positive feedback loop of TF/VIIa/PAR2/NF-κB/TF.

10.Studies on paclitaxel-loaded nanoparticles of amphiphilic block copolymer.

AIMTo investigate the paclitaxel-loaded nanoparticles of poly(ethylene glycol)-b-poly(D,L-lactic acid) amphiphilic diblock copolymer (PMT).
METHODSPMT was prepared by solid dispersion technique. The average size and size distribution were determined by dynamic light scattering (DLS). The morphology was characterized by transmission electron microscopy (TEM) and 1HNMR. The influences of the copolymer molecular weight and the paclitaxel-fed amount on PMT were studied. Therapeutic effect of PMT was studied on Kunming mice liver cancer H22.
RESULTSPMT showed nanometer size and spherical morphology with core and shell. The sizes of PMT increased with increasing the molecular weight of the hydrophobic segment in PEDLLA or increasing the drug-loaded amount. The tumour inhibiting effect of PMT was similar with that of Taxol.
CONCLUSIONIt will provide an experiment basis for the development of new kind of intravenous administration of paclitaxel.
