1.The clinical application value of color doppler ultrasound in the fetal malformation screening

Objective Objective To study the clinical application value of color doppler ultrasound in the fetal malformation screening.Methods Pregnant women underwent prenatal screening were collected.According to the examination,the pregnant women who had the high risk factors of congenital malformation fetus were selected as the observation group,the normal pregnant women were selected as control group.They all underwent antenatal color doppler ultrasound examination,the prenatal diagnosis accuracy and the missed dignosis rate were confirmed through the induced labor or after delivery inspection.Results After prenatal color doppler ultrasonography,congenital malformation found in 53 cases,the diagnostic accuracy rate was 100%,the rate of missed diagnosis was 5.66%.Fetal congenital malformation found in 46 cases in the observation group,which was significantly higher than that in the control group.Conclusion The accuracy of prenatal color doppler ultrasound in the diagnosis of congenital malformation fetus was 100%,it is an effective method in prenatal screening of fetal congenital malformations.
2.The determination of p53 protein in maxillary sinus carcinoma and its prognosis value

Objective:To determine whether p53 protein overexpression correlated with early tumor recurrence in carcinoma of maxillary sinus.Method:Forty-five patients with T2NoMo squamous cell carcinoma of maxillary sinus were treated with operation and/or radiotherapy . The median follow up time was 3 years. The ratio of local control was 62.2% (28 patients) , as control group ; The ratio of local recurrence and/or metastasis was 37.8% (17 patients),as experiment group . Cancer specimens were obtained before treatment.p53 protein overexpression, which indicated a mutated p53 gene ,was analyzed by immunohistochemistry, using the mouse monoclonal antibody DO-7.Result:Approximately 88.2% of the carcinoma that recurred locally overexpressed p53 protein , compared with only 28.6% of those with local control (P<0.01) .No significant relation was noted between p53 protein overexpression and pathohistologic grade .Intensity of staining did not predict tumor recurrece.Conclusion:The study demonstrated that the p53 protein is an important prognostic marker on gene level for maxillary sinus carcinoma.
3.Effects of C-peptide on apoptosis induced by hyperglycemia and the relationship between the apoptosis and the expression of eNOS in human umbilical vein endothelial cell (HUVEC)

Objective To investigate the effects of C-peptide on apoptosis induced by high glucose and analyze the relationship between the apoptosis and expression of eNOS in HUVEC. Methods We determined the HUVEC apoptosis by Annexin-V, the expression of eNOS mRNA of HUVEC by RT-PCR, the apoptotic quantity of HUVEC by DNA fragment ELISA. Results High level of glucose could increase HUVEC apoptosis (P<0.01) and reduce the expression of eNOS. After adding physiological or high concentration of C-peptide, the apoptosis decreased and the expressions of eNOS increased markedly, with the relationship coefficient of -0.854 (P<0.01). Conclusion High glucose may induce apoptosis. Physiological or high concentration of C-peptide may reduce the apoptosis induced by hyperglycemia, the relationship between apoptosis and eNOS is negative.
4.The effect of low molecular weight heparin in the treatment of patients with intrahepatic cholestasis pregnancy ;and regulation of cytokines

Objective To observe the effect of low molecular weight heparin in the treatment of patients with intrahepatic cholestasis pregnancy(ICP)and regulation of cytokines.Methods 124 patients with ICP were randomly divided into observation group and control group.The control group was treated with SAMe +UDCA therapy,the con-trol group was added low molecular weight heparin.All patients were treated for 10 days.Alanine aminotransferase (ALT),aspartate aminotransferase (AST),total bile acid (TBA),tumor necrosis factor alpha (TNF -α)and IL-18 were observed before and after treatment.Results After treatment,ALT,AST and TBA levels were lower in the two groups(t=64.48,110.14,45.75,40.76,62.54,24.67,all P=0.000),which of the observation group were higher(t=62.42,49.42,23.03,all P=0.000).After treatment,the TNF-αand IL-18 levels were lower in the two groups(t=13.14,25.07,all P=0.000),the TNF-αlevel in the observation group was lower than the control group(t=4.91,P=0.000).After treatment,the incidence rates of neonatal meconium,neonatal asphyxia of the observation group were significantly lower than those of the control group(7.14%vs.20.69%,χ2 =4.33,P=0.037;8.93% vs.24.14%,χ2 =4.75,P=0.029).The incidence rate of fetal and maternal hemorrhage between the two groups had no statistically significant differences (1.79% vs.1.72%,χ2 =0.00,P=0.980;5.17% vs.3.57%,χ2 =0.17,P=0.680).Conclusion The effect of low molecular weight heparin in the treatment of patients with intrahepatic cholestasis of pregnancy is exact,it can reduce the biochemical markers and inflammatory cytokine expression and improve perinatal outcome,which deserves to promote in the clinical treatment application.

A standardized near-drowning procedure was carried out on 16 adult normal sheep. Six milliliter sea water/kg body wt were introduced into the trachea by a tube. Ten minutes after near-drowning, eleven sheep were treated with HFV(HFVgroup), and the other 5 without HFV(NHFV group). Before near-drowning and at 5,15,30,40, 70,and 130 minutes afterward the measurement were made and it was found that: 1. Serum lipoperoxide(LPO) decreased for a short time, and then increased ten folds in both groups. 2. TXB_2 and 6-keto-PGF_(1?) increased in both groups. 3. pH decreased in NHFV groups(7.133—7.219). 4. PaO_2 decreased much mose in NHFV group. 5.PaCO_2 increased slightly at 5, 15 minutes after near-drowning; 6.K~+, Na~+, CI-increased slightly in both groups, and then returned to normal at 70 minutes after near-drowning. In HFV group; the changs of pulmonary ultrastructure after near-drowning were similar in both groups. The authors concluded that HFV showed some effects but is not satisfactory. We must find other treating methods to improve survival rate.

8.Role of DRP1 and OPA1 in hyperoxia induced lung injury of premature rats

Objective To investigate the role of DRP1 and OPA1 in the process of apoptotic in hyperoxia induced lung injury of premature rats .Methods 48 premature Wistar rats were divided into control group in which rats were exposed tO 210 ml .L 1 oxy-en ,and hyperoxia group in which Rats were exposed to 950 mL .L 1 oxyen .The lung tissues of 2 groups were gained on the first day ,the second day and the seventh day with which that all mice were killed by cutting neck .Section of lungs were stained with hea-toxylin eosin to observe the pathological changes ,the protein expression in the lung cells which were linked with the change of plas-tosome ,then the premature rat lung tissue apoptosis in each group were evaluated .Results Compored with the contnx group ,the expression level of OPA1 in the hyperoxia group began to decrease on the first day and apparently decreased to the buttom line on the seventh day .On the other hand ,DRP1 in hyperoxia group began to increase on the first day ,while reached the summit on the seventh day .A few of TUNEL positive cells began to increase with time dependence .A lot of TUNEL positive cells could be found in hyperoxia group ,and the apoptotic index reached the peak on the 7 d .There is a significantly positive correlation between the cell apoptosis and the ratio change of DRP1 and OPA1 in high oxygen group(r=0 .725 ,P<0 .01) .Conclusion The classical pathologic characters of lung injury were found out in hyperoxia group ,and it changed obviously with time .
9.Evaluation of pulmonary function and neurophysiological change in patient with chronic obstructive pulmonary disease

Objective To explore the association between the lung function and the related electroneurophysiological parameters in patients with chronic obstructive pulmonary disease (COPD). Methods Fifty COPD patients were studied. The lung function and the phrenic motor nerve conduction (PNC) were examined, respectively. Thirty cases of non respiratory disorders served as control. The lung function tests included maximal voluntary ventilation (MVV%), forced vital capacity (FVC), force expiratory volume (FEV 1) and RV/TLC% (residual volume, RV; total lung capacity, TLC). The phrenic nerve was stimulated electrically at the midpoint of the posterior border of the sternomastoid muscle, and the diaphragmatic compound muscle action potential (dCMAP) was recorded between the 7th and 8th intercostal space and xiphoid process. Results The relative values of MVV%, FVC 1/FVC 1% and RV/TLC% were 49.6?18.8, 62.5?16.4 and 54.1?8.0, respectively, which was significantly lower than that expected. There was no significant difference with regard to the PNC latency between the patients and the controls. The dCMAP amplitude ratio in the COPD patients was significantly lower than that of the controls, i.e., the dCMAP amplitude decreased in the COPD patients. Conclusion The decreased dCMAP might be associated with abnormal lung function. The PNC examination would provide valuable information for the evaluation of diaphragmatic dysfunction in those with abnormal lung function.
