1.Histological Classification and Prognosis of Neoplastic Osteoid Tissue of Osteosarcoma

A histological classification based on comprehensive morphological and stereometric observation of osteosarcoma was found that osteosarcoma might be divided into three types. Type Ⅰ (mature osteoid type) , Type Ⅱ (intermediate differentiated osteoid type ) and Type Ⅲ (immature osteoid type) . Fifty patients of the seventy were followed up for years, the longest up to twenty years. The five-year survival rate for 12 patients in Type Ⅰ is 92%; for 18 patients in Type Ⅱ and 20 patients in Type Ⅲ, none of them survived more than fiye years after operation. Statistically significant difference is found between individuals in the Type Ⅰ and Type Ⅱ, in Type Ⅰ and Type Ⅲ. The results indicate that the present pathological classification may be correlated with the prognosis. The prognosis of such Type was not correlated with site and size of the tumor, the dominating cellular component, pleomorphism, mito-tic figures and the number of giant cells.
2.Comparison of the clinical effect of laparoscopic partial hepatectomy and open surgery in treatment of hepatic hemangioma

Objective To investigate the efficacy of laparoscopic partial hepatectomy and open surgery in the treatment of hepatic hemangioma.Methods The clinical data of 45 cases of laparoscopic partial hepatectomy and 15cases of open surgery were retrospectively analyzed.The incision length,operative time,blood loss,start eating timeafter operation,postoperative drainage volume,postoperative hospital stay and incidence rate of complications werecompared between the two groups.Results The incision length,blood loss,start eating time after operation,postoperative drainage volume,postoperative hospital stay of laparoscopic partial hepatectomy group were [(4.5 ± 1.1) cm,(100.3 ± 33.7) ml,(1.5 ± 0.4) d,(112.2 ± 32.7) ml,(6.7 ± 1.5) d],which were better than those in the opensurgery group[(25.1 ±5.4)cm,(201.5 ±23.8)ml,(3.5 ±0.6)d,(230.3 ±43.7)ml,(11.3 ± 3.7)d],the differences were statistically significant between the two groups (t =1.977,2.023,1.989,2.015,2.025,P < 0.05).The operation time of the open surgery group was (159.4 ± 20.5) min),that in laparoscopic partial hepatectomy group was (139.0 ± 21.7) min,the difference was not statistically significant (t =1.453,P > 0.05).The incidence rate of complications in laparoscopic partial hepatectomy group was significantly lower than the open surgery group(x2 =3.013,P < 0.05).Conclusion Compared with open surgery,laparoscopic partial hepatectomy has the advantages of less blood loss,faster recovery,and fewer complications in the treatment of hepatic hemangioma.
3.Small cell carcinoma of the endometrium:case report and literature review

Objective:To explore etiology of small cell carcinoma of endometrium and clinical,histopathologic and features of the disease.Methods:We reported two case of this rare tumor and reviewed the literature.Results:Both patients presented with vaginal bleeding.Both of them underwent total abdominal hysterectomy and bilateral salpingo-oophorectomy.Two cases were stage Ⅲc according to postoperative pathology,Pathological examination revealed small cell carcinoma of endometrium Immunohistochemical evidence of neuroendocrine differentiation was demonstrated by using markers as neuron-specific enolase,synaptophysin.Case 1 died of her disease 5months after surgery,case 2 underwent chemotherapy combined with radiotherapy and was alive with no evidence of disease for 2 months.Conclusion:Primary small cell carcinoma of the endometrium is extremely rare and aggressive disease,The outcome is very poor,Surgery combined with chemotherapy and radiotherapy may contribute to an improved prognosis.
4.Research progress on human epidermal growth factor receptor-2 in gastric cancer

Amplification and overexpression of human epidermal growth factor receptor-2 (HER-2) gene are bound up with the process and the prognosis of gastric cancer.Meanwhile,recent researches have shown the great significance of HER-2 co-amplification with other genes in gastric cancer.Therefore,detection of HER-2 gene gradually becomes the basis of gastric cancer diagnosis,and it is one of the important targets in the treatment of gastric cancer.This article will review the gene amplification and co-amplification of HER-2 in the progress,prognosis,diagnosis and treatment of gastric cancer.
5.Innovation on informational management in the research-oriented hospitals

6.Study on the Growth for Selectivity to Negative Ion of Moderately Halophilic Actinomycetes

Cl~-was used as main negative ion long time for isolation and culture of moderately halophilic or halotolerant actinomycetes, but there are other negative ions such as NO_3~-、SO_4~(2-)、CO_3~(2-)and HCO_3~-in salt lake. Media with only Cl~-could not reflect their living natural environments, which was limited to study on moderately halophilic or halotolerant actinomycetes. In this study, the results show that some negative ion, such as NO_3~-and SO_4~(2-), have not much more selectivity to moderately halophilic or halotolerant actinomycetes, Cl~-can be substituted by NO_3~-and SO_4~(2-). CO_3~(2-)and HCO_3~-has little effect on the genus Nocardiopsis,but have much more on other genera such as Prauserella,Streptomonospora and Saccharomonospora, etc. These results will be some instructive to the isolation and cultivation of moderately halophilic or halotolerant actinomycetes.
7.The altered gene profile of human osteosarcoma cell after knock-down of double functional gene apurinic/apyrimidinic endonuclease

Objective To explore the molecular mechanism of inhibited malignant phenotype by knock-down of apurinic/apyrimidinic endonuclease(APE1) in human osteosarcoma cell HOS,as well as the possible interacting molecules among APE1.Methods Knock-down of APE1 by synthesized APE1 siRNA in HOS cell was identified first by Western blotting and AP endonuclease assay,then the gene profile was determined with GEArray~(TM) series Human CancerPathwayFinder gene array.Results The specific knock-down of APE1 was found in HOS cell transfected with APE1 siRNA,and its inhibited rate was 91.5%.Six biological pathways of the gene array were involved in HOS cell after knock-down of APE1,but the cell cycle pathway was less influenced.Forty-two out of 96 genes were altered(43.8%),in which only CDK4,FGFR2,KAI1 and NCAM were increased,38 others were decreased.Conclusion The knock-down of APE1 was detected to be involved in all the pathways including cell cycle and DNA repair,apoptosis,signal transduction,adhesion,angiogenesis,invasion and metastasis.These findings are significant to further elucidate APE1 role in tumor genesis and to provide experimental evidence for gene therapy targeting APE1 gene in the future.

9.DNA damage and repair gene N-methylpurine DNA glycosylase expression in human osteosarcoma

Objective To determine N-methylpurine DNA glycosylase (MPG) expression in human osteosarcoma and analyze its association with clinicopathology and prognosis, to construct adenoviral vector of MPG and study its ability to sensitize human osteosarcoma cell HOS to DNA damage agents. Methods The MPG expression was determined by immunohistochemical staining in 60 specimens and 3 cell lines (HOS, SAOS-2, U2OS) of human osteosarcoma. High expression group and low expression group were divided according to their staining intensity, and their association with clinicopathological data and prognosis was analyzed statistically with SAS software. The adenoviral infection and MPG expression, as well as enzyme activity were determined by flow cytometry, Western blotting, and HEX labeled oligonucleotide-based assay respectively. The cell survival/proliferation was measured using MTS, SRB, and thymidine incorporation assay. Apoptotic cells were assayed by flow cytometry after treated with phycoerythin (PE)-conjugated Annexin V and 7-amino-actinomycin (7-AAD). Results The high expression of MPG was found in 40 cases (67%), while low expression in three cell lines of osteosarcoma. MPG expression was significantly associated with WHO histological classification and prognosis by single factor regression analysis. The recombinant adenovirus of 10 MOI was found to infect more than 90% of HOS cells within 24 h by EGFP fluorescence, in which the MPG overexpression and MPG enzyme activity were also detected. The HOS cells with MPG overexpression were more sensitive to the DNA damage agents including MMS, MNNG, and TMZ, and the IC_(50) of three DNA damage agents decreased by 6.0, 4.5, and 2.5 fold respectively. Conclusion Transient MPG overexpression is a potential therapeutic approach for increasing HOS cellular sensitivity to DNA damage agent in chemotherapy.
10.Recombinant adenovirus containing PUMA gene regulated by human telomerase reverse transcriptase promoter transfected into ovarian cancer COC1 cells

Objective To over-express the P53 upregulated modulator of apoptosis(PUMA) in ovarian cancer COC1 cells for playing the role to restrain the cell growth .Methods Adenovirus vector (Ad)-human telomerase reverse transcriptase promoter (hTERT)-PUMA was transfected into ovarian cancer COC1 cells .Then the growth of COC1 cells was studied by CCK-8 and the apoptosis kit and the level of PUMA was analyzed .Results After transfection of Ad-hTERT-PUMA ,PUMA was over-expressed in COC1 cells ,which restrained the cells growth and promoted the cell apoptosis .Conclusion Ad-hTERT-PUMA is successfully transfected into ovarian cancer COC1 cells and PUMA is over-expressed to play the role for restraining the cells growth .
