1.Trace Metal Contents in Human Hair of Korean.

Analysis for 6 metals (Cd, Zn, Cu, Ni, Pb, Mn) by Atomic absorption spectrophotometer were made on hair samples of healthy 135 in urban area and 130 in rural area who have not dealt with the above metals in their daily working life. Marked variations were found. Samples of urban area contained more cadmium, copper, lead and manganese than those of rural area. No sexual difference in mean value could be observed and the amount of metals in hair did not increase with age. And no age dependency was found. There was a statistically significant correlation between the concentrations of cadmium and lead in both urban and rural people; Pb=1.50 Cd + 6.69 (r = 0.213, p<0.05) in urban area. Pb=1.44 Cd + 3.67 (r = 0.327, p<0.001) in rural area.

3.The development of predicting the lung cancer resistance to gemcitabine by the expression levels of RRM1

Gemcitabine is important pyrimidine antimetabolites drug. Ribonucleotide reductase is oneof its targets whose overexpression has been observed in several gemcitabine- resistant cell lines. Meanwhile,the NSCLC patients with high RRM1 expression levels have a relatively poor survival when treated with gem-citabine. To this day, the relationship between RRM1 expression levels in peripheral blood mononuclear cellsand gemcitabine is still uncertain and it deserves further research.

5.Prevention of chondrocyte apoptosis following acute osteochondral injury

Objective To explore the characteristics of chondrocyte apoptosis following acute osteochondral injury and the effect of intra-articular sodium hyaluronate injection in prevention of chondrocyte apoptosig. Methods Acute osteochondral injuries were created by drilling holes in knees at both sides of 28 rabbits according to Costouros' method. The experimental and control sides were randomized in each animal. Another four rabbits served as a normal control which received dissection of knee joint capsule without osteochondral injury. The experimental knee was injected with 1 % sodium hyaluronate at the end of the operation and then every week postoperatively. TUNEL and fluorescence-activated cell sorter (FACS) were applied to study the injured femoral condyles obtained at different intervals following the drilling injury. Results From day 4 after injury, the injured cartilage specimens displayed a statistically significant increase in overall level of apoptosis. Chondrocyte apoptosis was significantly reduced in the experiment knees compared with the injured ones. Conclusions Chondrocyte apoptosis may contribute to the subsequent development of post-traumatic arthritis. Intra-articular sodium hyaluronate injection just after acute osteochondral injury can suppress chondrocyte apoptosis, and thus help prevent the knee from post-traumatic arthritis.
6.Biomonitoring Ethylene Oxide Sterilization Process and Reason of Sterilization Failure

OBJECTIVE Through biomonitoring the effect of ethylene oxide sterilization process,to discuss the precision usage and management of the technology in operating room,and instruct the sterile job in operating room in the future.METHODS The process of ethylene oxide sterilization was monitored by biological indicators to realize the whole controlling of sterilization effects.RESULTS In the whole 210 sterile articles,202 were qualified,and 8 were disqualified.CONCLUSIONS With correct procedures of sterilization and assurance the qualification of the biomonitoring indicators,the optimal quality of sterilization would be achieved.
7.Apoptosis of cell line HL-60 induced by matrine

Objective To investigate the apoptosis of cell line HL-60 induced by matrine and its possible mechanism. Methods CCK-8 assay was used to observe the proliferation inhibitation of HL-60 cells treated with matrine. FCM was applied to detect apoptotic cells and mitoehondrial transmembrane potential.Spectrophotometry was used to detect the activity of Caspase-9. Results Matrine (0.25-2.0 mg/ml) could significantly inhibit proliferation of HL-60 cells (F=67.83, P <0.05). Ater 48 hours, the apoptosis rate increased obviously in dose dependence (t = 4.685, 6.300. 9.641, 6.786, P <0.05). Within 48 hours, the mitochondrial transmembrance potential of HL-60 cells treated with matrine (1.0 mg/ml) gradually decreased (F = 54.83, P <0.05), and the activation of Caspase-9 gradually decreased in time dependence (F = 72.31,P <0.05). Conclusion Matrine can induce apoptosis of HL-60 cells by reducing mitochondrial transmembrance potential and activating Caspase-9.
9. Detection of expression of RRM1 mRNA by real-time fluorescent quantitative PCR in non-small cell lung cancer tissues and peripheral blood

Objective: We tired to develop a method of detecting the expression level of RRM1 mRNA in tumor tissues and peripheral blood by real-time fluorescent quantitative PCR and compare the detection methods and results between the two different samples. We aimed to provide the basis for predicting clinical outcome from the expression of RRMI gene. Methods: The plasmid standard of RRM1 gene was constructed. Real-time quantitative analysis was performed using ABI7000 PCR kit. The PCR condition was optimized and the standard curve was established to detect the clinical samples. Results: The RRMI gene expression was detected in lung cancer tissues and normal lung tissues from 18 patients and in peripheral blood samples from 17 patients. The relative expression of RRM1 normalized by β-actin was 4.46 × 10-3 in lung cancer tissues, 3.43 × 10-3 in normal tissues, 2.54 × 10-3 in peripheral blood. The linear correlation between Ct value and the logarithm of original concentration was favorable. Conclusion: The expression of RRM1 mRNA in non-small cell lung cancer tissues and peripheral blood was successfully detected by using SYBR Green I fluorescence real-time quantitative PCR assay. There is no significant difference between the two sources of samples. The detection method has relatively higher sensitivity and specificity and can be used in clinical analysis.
