A variety of biochemical and physical examination indices can be used to predict adverse outcome of neurodevelopment caused by congenital heart disease and they contribute to early intervention and improvement of neurodevelopmental disorders. In recent years, new progresses in the laboratory tests, neurological physiccal examination and perioperative management of the neurodevelopmental abnormalities caused by congenital heart disease have been made. The predictive factors of abnormal neurodevelopment caused by congenital heart disease were reviewed in this article.
6.Protective mechanism of salubrinal against apoptosis induced by lipopolysaccharide in H9C2 cardiomyocytes
Chinese Pediatric Emergency Medicine 2012;19(5):507-509
Objective To investigate the mechanism of apoptosis induced by endoplasmic reticulum stress during sepsis. Methods Septic model of H9C2 cells was established by lipopolysaccharide (LPS).CHOP mRNA,mitochondrial membrane potential (MMP) and apoptosis were assessed by real-time PCR,flow cytometry respectively at LPS administration 24 h with or without salubrinal (inhibitor of endoplasmic reticulum stress). Results The expression of CHOP mRNA and apoptosis were upregulated, but MMP was decreased after LPS exposure 24 h compared with control group[2.40 ±0.30 vs 1.00 ±0,(16.58 ±2.71) % vs (3.85 ± 1.07) %,1.07 ± 0.33 vs 2.62 ± 0.51,P < 0.05).Compared with LPS group, salburinal pretreatment decreased LPS-induced upregulation of CHOP mRNA (1.60 ± 0.23 vs 2.40 ± 0.30,P < 0.05),meanwhile,salburianl alleviated MMP(1.85 ± 0.31 vs 1.07 ± 0.33,P < 0.05) and LPS-induced apoptosis [(6.05 ± 1.48) % vs (16.58 ± 2.71) %,P < 0.05].Conclusion Salubrinal plays a cytoprotective role against apoptosis induced by LPS through inhibition of CHOP mRNA,which leads to mitochondrial impairment.Endoplasmic reticulum stress may induce apoptosis via mitochondrial pathway in the septic model of H9C2 cardiomyocytes.
7.Relationship between liver X receptor (LXR) expression and hepatic glucose metabolism in the insulin resistant obese rats
Chinese Journal of Endocrinology and Metabolism 2011;27(8):677-679
Twenty-seven male SD rats were assigned to 3 groups: control, obese, and obese with diabetes groups. Serum corticosterone was determined and euglycemic-hyperinsulinemic clamp was carried out. Expressions of liver X receptor ( LXR), 11 β-hydroxysteroid dehydrogenase 1 ( 11 β-HSD1 ), and glucocorticoid receptor (GR) were determined by real-time PCR. Both groups of obese rats were insulin-resistant, with up-regulated LXR mRNA and down-regulated 11 β-HSD1 mRNA( both P<0.05 ).
8.Effects of limb ischemia-reperfusion injury on blood coagulation
The Journal of Clinical Anesthesiology 2010;26(3):224-226
Objective To investigate the effects of limb ischemia-reperfusion injury(I-RI) on blood coagulation. Methods Eighteen patients scheduled for unilateral knee arthroscopy surgery under epidural anesthesia were treated with an inflatable tourniquet to ptoduce ischemia for (42 ± 6) min.Venous blood was taken before tourniquet inflation,at 30 min during ischemia,3 min and 30 min during reperfusion for measuring blood coagulation by thrombelastography(TEG).Results TEG showed the decreases in reaction time(R value) and coagulation time(K value),and the increases in alpha angle(α) and maximal amplitude(MA),which were all within the normal limits and had no significant differences among four time points of testing. Conclusion I-RI of the limbs has no significant effects on blood coagulation.
9.Creep characterizes of rabbit tibia under torsion load following exercise
Chinese Journal of Tissue Engineering Research 2009;13(46):9101-9104
BACKGROUND:The creep performance of bone tissues during exercise is achieved through the microstrain, and the holosteous torsion can completely retiect the biomechanicel charactedstics of bone tissues.OBJECTIVE:To determine the creep characteristics of rabbit tibia under the torsion load following exercise.DESIGN,TIME AND SETTING:A parallel controlled animal observational experiment was performed at the Laboratory of Biomechanics in Orthopedic Hospital of Sichuan Province between May 2005 and July 2007.MATERIALS:A total of 32 Japanese white rabbits, irrespective of genders, aged 6 months, were divided into exercise group(n=24)and control group(n=8).METHODS:Exercise group animals were placed in cage to receive electrical stimulus.0.1-0.2seconds once every 20 seconds.Following the stimulation, the animals ran and jumped for several paces along with the cage and made a tum,15 minutes once, three times per day, totally 45 minutes,at the interval of 15 minutes. The exercise was carried out five days per week for 4successive weeks. The control group was fed as exercise group.but received no exercise.MAIN OUTCOME MEASURES:After 4 week exercise.the tibias were griped by the torsion clamping apparatus, the creep characterizes of anterior, posterior, medial and lateral sides of tibia under 0.52 N·m torsion load within 500 seconds were determined by strain gauge technique.RESULTS:Under torsion load, the creeps were present obviously in the rabbit tibia, reached the creep curve inflection point at 2seconds,and then gradually appeared. After 4 weeks, the creeps of tibias at anterior and posterior side in the exercise group were less than control group;in the control group, the anterior side of tibia was remarkably greater than the medial side. After exercise.the creep was the smallest in anterior side, which was less than the lateral side(P<0.05).CONCLUSlON:The creep characteristics of tibia are anisotropic, the deformation at the anterior and posterior sides is greater than that at the medial and lateral sides.Exercisecan alter the creep charactedstice of rabbit tibia and reduces the creep at the anterior and posterior sides.
10.The expression of LXR and its impacts on hepatic glucose metabolism in different glucose metabolic statuses in rats
Chinese Journal of Endocrinology and Metabolism 2011;27(2):148-151
Objective To investigate the mechanism of liver X receptor(LXR)signal pathway in regulating glucose metabolism by observing the variety of LXR expression and its impacts on regulating the mRNA expression of PEPCK and GCK, the key enzyme of hepatic glucose metabolism in various glucose metabolic status in rats. Methods SD rats were chosen and divided into four groups:the CON group, the induced DM group, the OB group, and the induced OB+DM group. For each group of rats, body weight, blood glucose, serum triglycerides, and cholesterol were measured. Then the rats were sacrificed and the livers were collected and studied. Real-time PGR was used to measure the expressions of LXR mRNA, PEPCK mRNA, and GCK mRNA in the livers. Finally, the Western Blot assay was used to measure the liver LXR protein expression. Results The expression of LXR mRNA was significantly higher in DM,OB, and OB+DM groups than in CON group(P<0.05).The Western blot results showed that the levels of protein were in accordance with the mRNA expression. Comparing to the CON and the OB groups, the PEPCK mRNA expression of the OB+DM and the DM groups was significantly higher, while the GCK mRNA expression of these two groups was significantly lower(P<0.05). Comparing to the CON group, the PEPCK mRNA expression of the OB group was significantly lower, while the GCK mRNA expression of OB group was significantly higher(P<0.05).Conclusions During non-diabetic phase, LXR could act as a protective receptor for glucose metabolism and keep glucose homeostasis by regulating the key enzymes of the hepatic glucose metabolism. While in the diabetic phase, the protective receptor LXR failed to reverse the change of the related enzymes caused by insulin deficiency, and finally the plasma glucose level was raised.
