Ptosis is one of the most common diseases in oculoplastics. Surgical treatment is the only way to correcting ptosis. Most of doctors paid special attention to surgical technique, rather than to some related issues of operation. Timing is crucial for ptosis surgery. Both cosmetic and function need to be considered for a success surgery, the evaluation of extraocular muscle disorder is impor- tant for surgical result and postoperative complications.
2. Diagnosis value of DWI, T2 WI and dynamic enhanced MR imaging for early liver abscess

Objective: The imaging diagnosis of early liver abscess is difficult. This study is to evaluate the diagnostic value of DWI, T2WI and dynamic enhanced MR imaging for early liver abscess. Methods: DWI, T2WI and dynamic enhanced imaging scans were performed in 17 patients with 17 early liver abscesses. The signal intensities and ADC values of necrotic area, non-necrotic area and normal liver were measured and compared. Results: The ADC value of the necrotic areas of liver abscesses ([0.88±0.09] mm2/s) was significantly lower than those of normal liver ([1.31±0.12] mm2/s,P<0.05) and non-necrotic area of liver abscesses ([1.43±0.18] mm2/ s,P<0.05); there was also significant difference between the ADC values of the normal liver and non-neocrotic area (P<0.05). The necrotic areas had a hyperintensity characteristic on DWI, T2WI and a hypointensity characteristic on T1 WI; no enhancement was found on dynamic enhanced imaging. The non-necrotic areas had moderate hyperintensity on DWI, T2 WI and hypointensity on T1 WI; there was obvious enhancement on dynamic enhanced imaging. Conclusion: The early liver abscess is characterized on magnetic resonance imaging. The DWI scan is helpful for diagnosis of early liver abscess.
3. Value of magnetic resonance cholangiopancreatography for diagnosis of pancreas divisum

Objective: To investigate the value of magnetic resonance cholangiopancreatography(MRCP) in the diagnosis of pancreas divisum by comparing with endoscopic retrograde cholangiopancreatography (ERCP). Methods: The MRCP and ERCP images of 8 patients with pancreas divisum were retrospectively analyzed. The diagnostic accuracy and findings by MRCP were compared with those by ERCP. Results: MRCP had a diagnostic accuracy of 87.5%(7/8) based on the result of ERCP. ERCP displayed the dominant dorsal pancreatic ducts in all 8 cases and ventral pancreatic ducts in 6 cases; MRCP also displayed the dominant dorsal pancreatic ducts in all 8 cases, but the ventral pancreatic ducts only in 3 cases. Conclusion: As a non-invasive technique, MRCP has important clinical value in the diagnosis of pancreas divisum.
4.Effects of vascular endothelial growth factor receptor-1 on the invasion and metastasis of hepatocellular carcinoma

Objective To investigate the effects of vascular endothelial growth factor receptor-1(VEGFR-1) in the invasion and metastasis of hepatocellular carcinoma(HCC)by detecting the expression of VEGFR-1 in HCC tissues.Methods Paraffin-embedded tissue which containing HCC tissues and adjacent tissues were prepared from patients(n=82)with HCC,then were analyzed by immunohistochemical technique for the expression of VEGFR-1,epithelial marker E-cadherin and mesenchymal marker Vimentin.The relationship between the expression of VEGFR-1 and pathological parameters,and the correlation between VEGFR-1 and E-cadherin,VEGFR-1 and Vimentin were analyzed.Results The expression rates of VEGFR-1 in HCC tissues and adjacent tissues were 89%(73/82)and 0,respectively.The difference of VEGFR-1 expression in portal vein tumor thrombus,tumor capsule,stage,size and differentiation grade of tumor had statistical significance(x2=22.192,15.934,16.751,20.154,6.487,P<0.05).The expression of VEGFR-1 had effect on the 1-,2-year recurrence rate and 2-year survival rate (x2=0.983,0.958,0.847,P<0.05),but not on 1-year survival rate(x2=0.359,P>0.05).The expression of VEGFR-1 was negatively correlated with that of E-cadherin(r=0.765,P<0.01)but positively with that of Vimentin (r=1.469,P>0.05).Conclusions VEGFR-1 may play an important role in invasion and metastasis of HCC.Epithelial-mesenehymal transition that induced by VEGFR-1 may contribute to the invasion and metastasis of HCC.
5.Construction of siRNA expression vector targeting IGF-1R and its ability to induce cell apoptosls in hmnan lung cancer cells

Objective Insulin-like growth factor-1 receptor (IGF-1R),similar to insulin receptor,is one of the families of re- ceptor tyrosine kinases.,which has been found to be overexpressed in a variety of cancer.It is the main proliferation and survival sig- nal molecule in cancer cell and plays an important role in cancer growth and progress.Blocking signal transduction of IGF-1R by vari- ous strategies can suppress tumor growth and induce regression of established tumor.This study is to construct the siRNA expression vector targeting IGF-1R and to evaluate its ability to induce cell apoptosis in human lung cancer cells.Methods Two siRNA expres- sion vector,pENTR/U6-shRNA-1 and pENTR/U6-shRNA-2 targeting IGF-1R,were constructed using pENTR/U6 vector,and a vector targeting hieiferase gene,pENTR/U6-shRNA-Iuc,was constructed as control.After vectors were transfected into A549 for 48h, knockdown of IGF-1R mRNA and protein and Akt phosphorylation were accessed,and DNA ladder and flow cytometry were used for cell apoptosis.Results siRNA expression vectors targeting IGF-1R were successfully constructed,which was confirmed by PCR and DNA sequencing,pENTR/U6-shRNA-1 and pENTR/U6-shRNA-2 demonstrated the expression were (22.1?2.5) % and (80.1? 3.9) % in IGF-1R mRNA level,(15.2?3.1)% and (47.1?4.1)% in protein level,respectively,compared with pENTR/U6- shRNA-luc.Suppression of IGF-1R by pENTR/U6-shRNA-1 blunted Akt phosphorylation,increased cell apoptosis induced by 3% ethanol,and retained 77.5 % A549 cells in the G0/G1 phase.Conclusion siRNA expression vector targeting IGF-1R can effectively suppress the expression of IGF-1R expression in A549.This study suggests that DNA vector-based RNAi has the potential to be effec- tive and practical cancer gene therapy strategy.

Xenotransplantation is a feasibility way of solving the shortage of human organs for transplantation. Although it is urgently needed to satisfy the demand of people and sustain the function of human organs, there are multiple hurdles existed to clinical application, such as the immune rejection between human body and the xenografts, the infection of pathogens and a series of ethic, morality and social issues. A historical retrospect of xenotransplantation was given, and then probe into the strategies according to the main problems and the actualities. Finally, the prospect in the field of xenotransplantation was showed.

Viable but non-culturable state in bacterial cells as received uncultured microorganism has be- come the fundamental research issues in medical microbiology, epidemiology, general microbial ecology and sanitation quarantine since it was put forward in 1982. When in this state, bacterium are no longer able to grow and form colonies on conventional culture media, but maintain their patheogenicity, may become latent infection escaping detection to threaten enviroment and human security. With the development of modern molecular biology technique and metagenomics, it provide new research method and opportunity for uncul- tured microorganism in the environment. Recently, with the application of the metagenome technique, ge- netic fingerprinting technical etc, it becomes more and more popular. Simultaneously, along with this state become maturity in the laboratory. It provides a new research route for development and exploitation the uncultured microorganism.

10.Effect of RNA interference on the bionomics of Cathepsin L of hepatoma carcinoma cells

Objective To explore the effect of RNA interference on the bionomics of Cathepsin L of hepatoma carcinoma cells. Methods In this study, the experimental group was the Cathepsin L RNA interference group. Control groups were the normal blank hepatoma carcinoma cell group (the blank group) and the Cathepsin LRNA interference blank group (the fluorescence control group). Observing times were ld,3d and 6d after RNA interference. Transfection efficiency in each group was observed. Expression of Cathepsin L of hepatoma cells was detected by immunofluorescence, RT-PCR and WB. Cell vigor was detected by MTT assay. Changes in the cell cycle and apoptosis were observed by flow cytometry. Transwell cabin was used to detect the changes of cell invasive power.Results In the experimental group, Cathepsin L mRNA level and protein level significantly decreased, the proliferation index significantly decreased and apoptosis index significantly increased. The invasive power also decreased. Conclusion RNAi interference can inhibit Cathepsin L expression, cell proliferation and cell invasive power efficiently.
