1.Clinical analysis of serum interleukin-8, interleukin-4, interleukin-9 levels on the children with bronchus asthma

Objective To observe the effects of serum intefleukin-8,-4,-9 levels on the children with bronchus asthma.Methods 186 cases of children with bronchus asthma in our hospital during May 2010 to December 2010 were randomly selected,and given routine clinical therapy for 6 months,observing the serum interleukin-8,-4,-9 levels before and after treatment.Results The serum interleukin-8,-4,-9 levels of the children with bronchus asthma was higher than normal,but after treatment,the levels were totally decreased,there had a significant difference in the indexes.Conclusion The serum interleukin-8,-4,-9 levels was a good kind of reference index of disease progressed and effect determination in the children with bronchus asthma,it can more accurate evaluated the occurrence,development,metastasis and curative effect of asthma.
2.Intelligent prediction of HER2 status based on breast histopathology

Objective:To study the association between histopathological features and HER2 overexpression/amplification in breast cancers using deep learning algorithms.Methods:A total of 345 HE-stained slides of breast cancer from 2012 to 2018 were collected at the China-Japan Friendship Hospital, Beijing, China. All samples had accurate diagnosis results of HER2 which were classified into one of the 4 HER2 expression levels (0, 1+, 2+, 3+). After digitalization, 204 slides were used for weakly supervised model training, and 141 used for model testing. In the training process, the regions of interest were extracted through cancer detected model and then input to the weakly supervised classification model to tune the model parameters. In the testing phase, we compared performance of the single- and double-threshold strategies to assess the role of the double-threshold strategy in clinical practice.Results:Under the single-threshold strategy, the deep learning model had a sensitivity of 81.6% and a specificity of 42.1%, with the AUC of 0.67 [95% confidence intervals (0.560,0.778)]. Using the double-threshold strategy, the model achieved a sensitivity of 96.3% and a specificity of 89.5%.Conclusions:Using HE-stained histopathological slides alone, the deep learning technology could predict the HER2 status using breast cancer slides, with a satisfactory accuracy. Based on the double-threshold strategy, a large number of samples could be screened with high sensitivity and specificity.
3.Myxiod pleomorphic liposarcoma: a clinicopathological and molecular genetic analysis of six cases.

Objective: To investigate the clinicopathologic and molecular genetic characteristics of myxoid pleomorphic liposarcoma (MPLPS). Methods: Six cases of MPLPS diagnosed and consulted in Fujian Provincial Hospital from 2015 to 2021 were collected for histomorphological observation, immunohistochemistry, and fluorescence in situ hybridization (FISH) detection of DDIT3 (CHOP) gene translocation and MDM2/CDK4 gene amplification. Results: There were four males and two females, aged 26-74 years (mean 53.8 years). The tumor size was 3.8-16.0 cm (mean 11.8 cm). All six cases had similar histopathologic features, showing overlapping histologic morphology of myxoid liposarcoma and pleomorphic liposarcoma. Four cases (4/6) were positive for S-100 protein, and the Ki-67 index was 50%-95%. All cases (6/6) were negative for DDIT3 (CHOP) translocation and MDM2/CDK4 amplification by FISH. TP53 (p.R248w) germline mutation was found in one case. Conclusions: MPLPS is a rare subtype of liposarcoma, characterized by overlapping morphology of myxoid liposarcoma and pleomorphic liposarcoma. Genetically, a few of them have TP53 gene germline mutations, but they lack of DDIT3 (CHOP) translocation or MDM2/CDK4 amplification.
