1.Antigen-incorporated connective tissue mast cells promote specific T cell activa-tion

To study the effects of connective tissue mast cells ( CTMCs ) on T cell activation in an antigen-dependent manner.Methods: Connective tissue mast cells were differentiated from mouse bone marrow cells.Alexa Fluor 488-conjugated ovalbumin (OVA)/IgG immune complexes were generated and incubated with CTMCs ,and analyzed by flow cytometry to detect the antigen uptake by CTMCs via Fcγreceptors ( FcγRs ).The relevant effects on antigen-specific T cell activation and proliferation were further determined by CD 69 and Ki67 expression.Results: A magnificent increase in OVA uptake was seen in CTMCs treated with OVA/IgG immunocomplex after 24-hour incubation ,compared with that treated with fluorescent OVA alone.OVA-incorporated mast cells induced OVA-specific CD4+T-cell activation and proliferation.Conclusion:It was suggested that the binding of IgG receptor FcγRs could elicit CTMCs reaction and ,as a result,to mediate crosstalk between professional APCs and T cells.
2.Preparation and Quality Control of Compound Doxepin Hydrochloride Cream

OBJECTIVE:To study the preparation and quality control of compound doxepin hydrochloride cream.METHO DS:The compound cream was prepared with doxepin hydrochloride and triamcinolone acetonide,and the contents were determinated by UV-spectrophotometry and HPLC respectively.RESULTS:The calibration curves were linear in the range of the concentrations of the experiment respectively,r=0.9999.CONCLUSION:The method is simple,accurate and suitable for the quality control of this preparation.

Object A method was developed for the determination of fatty acid in Fructus Bruceae oil by precolumn derivation HPLC Methods Fatty acids were derivatized with p bromophenacylbromide as derivative and 18 crown 6 as catalyst The method used C 8 Kromasil C 8 (250 mm?4 mm, 5 ?m) column and isocratic acetonitrile water eluent and the internal standard was heptadecanoic acid The detection wavelenghth was 254 nm Column temperature was fixed at room temperature, and the flow rate was 1 3 mL/min Results The standard curves of linoleic, palmitic, oleic, and steraric acid are linear within the range of 0 022—0 330, 0 014—0 213, 0 028—0 416, and 0 012—0 177 ?g, respectively The four fatty acid recoveries are 99 2%, 97 2%, 101 8%, 97 8%, and the RSD are 1 2%, 1 5%, 0 4%, 2 3%, individuslly Conclusion Five fatty acids are separated within 30 minutes during a single run The present method is reliable and relatively simple for the determination of fatty acid in Fruetus Bruceae oil
4.Stereological study on syncytial cell of human placenta and determinations of total bile acid in cord blood of intrahepatic cholestasis of pregnancy

Objective To investigate the relation of changes of fetal bile acid and morphologic ultrastructure of human placental syncytial cells in intrahepatic cholestasis of pregnancy (ICP). Methods(Total bile acid of cord venous blood was measured by cycle enzyme method between intrahepatic cholestasis of pregnancy and control groups after birth. The ultrastructure of human placental syncytial cell was analyzed by morphologic study in two groups. Results (1) The total bile acid of cord venous blood in ICP group was significantly higher than in control group[(8.6?3.2) ?mol/L vs (4.6?1.5)?mol/L ](P0.05). The average volume, surface area density and volume density of mitochondrion in ICP group [respectively (0.0200?0.0020) ?m~3,(0.600?0.010) ?m~2/?m~3,(0.0800?0.0090)?m~3/?m~3] were significantly enlarged than in control group [respectively (0.0100?0.0050)?m~3, (0.500?0.030) ?m~2/?m~3,(0.0500?0.0020) ?m~3/?m~3](P0.05). Conclusion In ICP, high levels of bile acids may impair cellular organelle, resulting in abnormality of physiological function of syncytial cells, and affecting the synthesis and transportation functions of placenta.
5.Effects of phalcoemulsification on unilateral high myopia with cataract

AIM: To observe the clinical effect of phacoemulsification and posterior chamber intraocular lens ( IOL) implantation in the treatment of unilateral high myopia combined with age-related cataract.
METHODS: A retrospective clinical study was carried out in a single ophthalmology center. All patients diagnosed of unilateral high myopia with age- related cataract were performed phacoemulsification and posterior chamber IOL implantation. The axial length of operation eye was ≥27mm, and the axial length of unoperated fellow eye was≤25mm. Visual acuity and complications were followed up for 1, 7d, 1, 3mo postoperative.
RESULTS:Thirty-three patients with average age 68. 8±9. 26 (41-87) y were enrolled in this study, 7 cases were men, 26 cases were women. The mean axial length of operation eyes was 29. 44±1. 80(27. 1-32. 71) mm and the mean axial length of unoperated fellow eyes was 23. 39±0.92 ( 21. 72 - 24. 90 ) mm. The average axial length difference of binocular eyes was 6. 06±1. 97(2. 78-9. 92) mm. The best corrected visual acuity ( BCVA ) increased from 0. 02±0. 02(light perception~0. 12) preoperatively to 0. 20±0. 20 ( 0. 02 - 0. 8 ) at 3mo postoperatively. There was statistically significant difference (t= -5. 278, P<0. 05). Among the 10 patients whose axial length difference of binocular eyes was 2. 01 - 5. 00mm, 60% of their postoperative BCVA were >0. 3. Among the 13 patients with binocular axial length difference of 5. 01-7. 00mm, 46.2% of BCVA were >0. 3,46.2% achieved 0. 05-0. 25. Another 10 patients of binocular axial length difference between 7. 01-9. 99mm, 90% of BCVA were <0. 3, of which 40% were<0. 05. No complications occurred during operative and postoperative .
CONCLUSION: Phacoemulsification and posterior chamber IOL implantation can effectively improve the postoperative visual acuity and it is safe for unilateral high myopic patients combined with age-related cataract. The recovery of visual acuity is related with binocular axial length difference.
6.Enhancing the research on the mechanism of the occurrence and development of retinal neovascularization to prevent and treat retinal neovascularization diseases

Retinal neovascularization is a complicated pathophysiological process as a result of imbalance between angiogenic and anti-angiogenic factors. Correct understanding of the signaling pathways,exploring the critical factors involved in retinal angiogenesis, looking for new strategies by reconstructing the new vessels are helpful for knowing the mechanism of the occurrence and development of reitnal neovascularization, which would be good for preventing and treating retinal neovascularization diseases.
7.Study on the clinical effect of the massage method of micro-regulating with vertical cross pressing lying on one side in treating cervicogenic headache.

OBJECTIVETo evaluate the clinical effects of the massage method of micro-regulating with vertical cross pressing lying on one side in treating cervicogenic headache (CEH).
METHODSTotal 136 patients with CEH were collected in the study from August 2012 to April 2014. They were divided randomly into two groups according to random digits table. Sixty-nine patients accepted the treatment of micro-regulating with vertical cross pressing lying on one side (pressing micro-regulating group), including 29 males and 40 females with an average age of (50.55 ± 11.38) years old; 67 patients received the treatment of traditional massage (traditional massage group), including 28 males and 39 females with an average age of (51.20 ± 11.90) years old. Clinical effect was observed according to the standard of curative effect of State Administration of Traditional Chinese Medicine; the function of cervical vertebra and all body status were evaluated according to NDI score. VAS score, frequency and time of headache were recorded and compared before and after treatment.
RESULTSNo adverse reactions were found after treatment, all patients were followed up from 1 to 6 months with an average of 3.1 months. In pressing micro-regulating group, 25 cases got fully recover, 26 excellence, 14 effectiveness and 4 inefficiency; and in traditional massage group, the results were 12,21,22, 12;clinical effect of pressing micro-regulating group was better than that of traditional massage group (P<0.01). NDI score in pressing micro-regulating group decreased from preoperative 13.48 ± 4.83 to postoperative 6.23 ± 3.76; in traditional massage group also decreased from preoperative 13.82 ± 5.78 to postoperative 8.25 ± 4.75; the improvement of the pressing micro-regulating group was obviously better than that of traditional massage group (P < 0.01). VAS score in pressing micro-regulating group decreased from preoperative 4.75 ± 0.97 to postoperative 1.88 ± 1.78; and in traditional massage group decreased from pre-operative 4.78 ± 0.98 to postoperative 2.84 ± 1.94; pressing micro-regulating group was more notable than that of traditional massage group (P < 0.01). The frequency per week,the pain time in pressing micro-regulating group decreased from preoperative (5.38 ± 1.96) times and (6.87 ± 3.67) hours to postoperative (1.71 ± 2.04) times and (0.97 ± 1.74) hours,respectively,in traditional massage group the above parameters decreased from preoperative (5.22 ± 1.81) times and (6.90 ± 3.79) hours to postoperative (2.81 ± 2.42) times and (1.83 ± 2.21) hours;pressing micr-regulating group was more notable than that of traditional massage group (P < 0.01 or P < 0.05).
CONCLUSIONUsing the tuina method of micro-regulating with vertical cross pressing lying on one side to treat CEH can improve function of cervical vertebra and all body status, lessen the intensity, frequency, duration time of pain, and had advantage of higher security, simple operation, and evident effect.

8.Interventional effect of fluoxetine on the quality of life in depressive patients with breast cancer

BACKGROUND: There are many studies on the quality of life (QOL) of patients with breast cancer at home and abroad at present. But the effect of antidepressants on the QOL of depressive patients with breast cancer is not clear yet.OBJECTIVE: To investigate the effect of fluoxetine on the QOL of depressive patients with breast cancer.DESIGN: A self-controlled and observational comparative study of taking the patient as the subjects.SETTING: The research institute of medical psychology and psychiatric specialty in two universities and a psychiatric department in a city hospital.PARTICIPANTS: Sixty-three female post-operative patients with breast cancer hospitalized in the First Affiliated Hospital and Second Affiliated Hospital of China Medical University and Dalian Tumor Hospital, with an average age of (50 ± 9) years.INTERVENTIONS: Sity-three depressive patients with breast cancer were treated with fluoxetine for 8 weeks. Treatment emergent symptom scale (TESS) and relative laboratory examinations were used to evaluate the safety of fluoxetine to depression and anxiety of depressive patients with breast cancer. QOL of WHO scale was used to assess the QOL of the patients before and after administration.MAIN OUTCOME MEASURS:①Score of every index of QOL of WHO;②Side effects and adverse effects.RESULTS: The scores of physiology, psychology and independence in QOLafter 8 weeks' administration(16.30 ±2.17, 14.41 ± 1.85, 16. 90 ± 1.53)were significantly higher than those before grouping (13.61 ±2.46,12.98 ± 2.65, 13.65 ± 2.45) ( t = 6.52, P < 0.01; t = 2.41, P< 0.05; t= 8.93, P < 0.01 ). No obvious side effect was observed during administration.CONCLUSION: Fluoxetine can significantly improve the QOL of patients with breast cancer and there are fewer side effects during the treatment.
9.EFFECT OF NERVE REGENERATION FACTOR ON THE NEURITE GROWTH OF RAT DORSAL ROOT GANGLIONS

Objective To study the effects of Nerve Regeneration Factor on the neurite growth and Neurofilament-H expression of rat dorsal root ganglions in vitro. Methods DRG was cultured and treated with NRF,observation made by fluorescent immunocytochemistry and the mRNA and protein level of NF-H of DRG neurons was detected by real-time fluorescence quantitative RT-PCR and Western blot. Results NRF can promote the neurite growth of DRGs in vitro.The mRNA and protein levels of NF-H of DRG neurons can be up-regulated by NRF in a dose-dependent manner,with the best concentration of 2.0?(mg/L.) Conclusion\ NRF can promote the neurite growth of DRGs in vitro.NRF exerts a neurotrophic action on developmental sensory neurons.
10.Comparison of 3.0T MRI and SPECT-CT in the diagnosis of osteoporotic vertebral compression fractures

BACKGROUND:There have been many studies on the diagnosis of osteoporotic vertebral compression fractures. However, there are few studies on the diagnostic methods of fracture in different periods. OBJECTIVE:To comparatively analyze the application value of high-intensity magnetic resonance imaging (MRI) and single photon emission computed tomography combined with CT scan image fusion (SPECT-CT) in the diagnosis of osteoporotic vertebral compression fractures. METHODS:Clinical data of 35 patients with osteoporotic vertebral compression fractures were retrospectively analyzed, including 56 vertebrae. There were 14 males and 21 females, with an average age of 72.6 years. The course of disease was within 3 weeks in 22 patients, belonging to acute fractures. The course of the disease was between 4 and 12 weeks in 10 patients, belonging to the subacute fractures. The course of disease was 6 months in 3 patients, belonging to the fracture healing period. Al patients were tested with high-intensity MRI and SPECT-CT examination, and were treated with vertebroplasty or percutaneous kyphoplasty. The Fisher exact method was used to compare the two diagnostic methods in the diagnosis of vertebral fracture. RESULTS AND CONCLUSION:(1) In the 56 responsible vertebral bodies, 32 vertebral bodies belonged to the acute-stage fractures, and 24 vertebral bodies belonged to the non-acute-stage fractures. In al the fractures, MRI diagnosed 49 segments;SPECT-CT diagnosed 52 segments. The sensitivity of SPECT-CT testing was higher than MRI, and its specificity was less than MRI. There was a high consistency of diagnosis between fractures. (2) In the 32 acute vertebral fractures, the two tests diagnosed 29 segments. The sensitivity of SPECT-CT was higher than that of MRI, but its specificity was lower than that of the MRI. There was a high consistency between the diagnoses of fractures. (3) In the 24 non-acute vertebral fractures, MRI diagnosed 20 segments;SPECT-CT diagnosed 23 segments. SPECT-CT was more sensitive than MRI;the specificity of SPECT-CT was lower than MRI. There was a high consistency between the two diagnoses of fractures. (4) There was a high consistency in the MRI and SPECT-CT examination to determine the fracture of different periods of responsibility of the vertebral body, but the sensitivity of SPECT-CT is higher than MRI examination;SPECT-CT is an effective inspection method in the diagnosis of vertebral fractures.
