1.Whether nano-hydroxyapatite particles can influence apoptosis of mononuclear macrophage in rat abdominal cavity?

BACKGROUND: Because of their size effect, nanometer particles (NPs) can combine molecular within cells, which can result in cell necrosis or apoptosis. But there are no systematic mechanisms of apoptosis induced by NPs about biological safety of NPs.OBJECTIVE: To investigate the effect of nano-hydroxyapatite particles on mononuclear macrophage in rat abdominal cavity at celluar and molecular level.DESIGN, TIME AND SETTING: A materials-cytology observation was performed at Shanghai Biomaterials Research & Testing Center from January 2001 to December 2008.MATERIALS: SD rats of clean grade were provided by SINO-BRITISH SIPPR/BK LAB. ANIMAL Co., Ltd.; NPs were provided by Shanghai Institute of Ceramics.METHODS: Peritoneal fluid was extracted under a sterility environment to in vitro separate and culture mononuclear macrophage using adherence method. The concentration of cell was adjusted at 2×10~9/L. At 300 W/40 kHz ultrasound, cell suspension containing 20,100 and 200 mg/L nano-hydroxyapatite particles was prepared to induce mononuclear macrophage for 24 hours, respectively. A normal control group was established.MAIN OUTCOME MEASURES: Ultrastructural phenotype was detected using transmission electron microscope; apoptotic rate was measured using AnnexinV-EGFP/PI staining; variation of apoptosis-related P53 gene expression was detected using Western Blot.RESULTS: Pseudopodia of mononuclear macrophage in the normal control group were intact, nuclear membrane was normal, and nucleoplasm was uniformed. After inducing by 20,100, 200 mg/L nano-hydroxyapatite particles for 24 hours, apoptotic morphological characteristics were typical in mononuclear macrophage. Compared with normal control group, apoptotic rate was significantly increased following the induction of 20 and 100 mg/L nano-hydroxyapatite (P< 0.01), and the increasing in the 100 mg/L nano-hydroxyapatite particle group was greater than 20 mg/L nano-hydroxyapatite particle group (P < 0.05). P53 protein was not observed in mononuclear macrophage in the normal control group. Following induction of varying concentrations of nano-hydroxyapatite particles, P53 protein expression was increased in the mononuclear macrophage, and the expression was positively related to concentration of nano-hydroxyapatite particles.CONCLUSION: Nano-hydroxyapatite particles can induce p53 expression through phosphorylation, which promotes downstream genes and finally results in cell apoptosis.
2.The clinical effects of tumor necrosis factor-α antagonist combined with glucocorticoid in patients with ulcera tive colitis

Objective To explore the effect of tumor necrosis factor-α antagonist (Remicade) on the patients with active moderate to severe ulcerative colitis (UC) who failed to response to mesalazine or sulfasalazine Sah'cyloyl treatment for six months period.Methods The patients were collected in our hospital,who diagnosed by colonoscopy as moderate to severe ulcerative colitis and failed to response to mesalazine or sulfasalazine Salicyloyl treatment for six months period;These patients were randomly divided into control group(22 cases) and treatment group(28 cases) ;glucocorticoid was used alone in control group while it was combined with Remicade in treatment group.After treatment for six months,they were suffered from colonoscopy and clinical evaluation.Results There were significant differences between control and treatment group of the recover rate concerning the clinic symptoms such as abdominal pain,diarrhea,and mucous bloody stool.There were 7 cases complete remission,13 cases effective outcome and 9 cases ineffective,with complete remission rate of 31.67% .efficiency rate 59.09% in control group.There are 17 cases complete remission,25 cases effective outcome and 3 cases ineffective,with complete remission rate of 60.71% ,efficiency rate 89.28% in treatment group.There was significant difference (P<0.05) between two groups of complete remission rate and efficiency rate.There were 3 cases recurring in control group,with the recurrence rate of 13.67% ,while there were 2 cases recurring in treatment group with the recurrence rate of 7.14%.There was significant dif-ferene( P<0.05) between two groups with regards to recurrence rate;Besides,there were no significant adverse effect.Conclusion The combination with Remicade and glucocorticoids in the treatment of refractory severe ulcerative colitis was more effective and can also reduce the relapse rate.
3.Research status of hyaluronan composite materials

BACKGROUND:Hyaluronan with special linear macromolecular structure and rheologic property has good biocompatibility and becomes a signal matter of the cel surface, which possesses a promising application potential in tissue-engineering field. OBJECTIVE:To review the research progress in medical hyaluronan composites. METHODS:Related literatures concerning preparation, safety evaluation and clinical trials of hyaluronan composites were extensively reviewed and comprehensively analyzed in the three folowing aspects: cartilage scaffold, cornea scaffold and submucous bulking agent. RESULTS AND CONCLUSION:The hyaluronan composite has a potential use in cartilage scaffold and cornea scaffold as its suitable porosity and pore size, biomimetic three-dimensional microstructure, excelent physical and chemical property and biological property. The hyaluronan composite as a bulking agent has been applied clinicaly, and results from some clinical trials have shown that the composite used as bulking agent has the problems about low efficacy and complications, which need to do further improvement. More researches are needed in product design, evaluation of performance and safety for expending and optimizing the clinical applications of the hyaluronan composite.

Small ubiquitin-like modifiers(SUMO) are ubiquitin-related proteins that can be covalently conjugated to cellular target proteins.Frequently,this modification has a key role in regulating the activities of those targets and,hence,their cellular functions.Researches show that some oncoproteins,tumor suppressors,DNA repair proteins and cyclins are target proteins of SUMO modification and the altered expression of some enzymes related to SUMO conjugation occurs in some neoplasm.It is indicated that sumoylation is important in tumor origination,development and metastasis.
6.Handling and prevention of medical disputes caused by suspected AIDS infection via blood transfusion

Medical disputes caused by suspected AIDS infection via blood transfusion are characterized by high sensitivity, involvement of many persons, great difficulty in carrying out investigation and obtaining evidence, and demand for a large compensation. The key to handling this kind of medical dispute lies in clarifying whether there is a causal relationship between the blood transfusion and the AIDS infection, whether there have been illegal activities in the process of blood collection and transfusion, and whether there is a causal relationship between such activities and the AIDS infection. In addition, the partys right of privacy should be protected. Measures of prevention include: handling without gloves activities of illegal blood supply and collection and the arrangement for other people selling their blood; strictly assessing and consolidating the blood centers; blood centers and hospitals jointly preventing the spread of AIDS via blood transfusion; taking proper care of the raw data of blood supply and collection; and speeding up the establishment of an insurance system for blood transfusion and the implementation of the States plans for AIDS prevention and treatment.
7.Pathogens and risk factors of healthcare-associated infection in children with acute leukemia

Objective To explore the main sites,pathogen distribution and risk factors of healthcare-associated in-fections (HAI)in children with acute leukemia,and provide scientific evidence for prevention and treatment. Methods Data of 828 children with acute leukemia admitted to a hospital in 2011 -2012 were collected,infection site,pathogen distribution and risk factors of infection were analyzed.Results Of 828 patients,184 cases and 196 times of HAI occurred with the incidence of 22.22% and 23.67%,respectively.HAI occurred mostly in respiratory tract (52.56%).A total of 96 pathogenic strains were isolated,among which gram-negative and gram-positive bac-teria accounted for 58.33%(n=56)and 29.17%(n=28)respectively.Univariate analysis revealed that risk factors for HAI were leukemia remission induction chemotherapy,hospital stay ≥ 30 days,peripheral leukocyte count (WBC)≤3×109/L,granulocyte count ≤0.5 ×109/L,and acute nonlymphocytic leukemia.Multivariate analysis revealed that hospital stay ≥30 days was independent risk factors for HAI.Conclusion Children with acute leuke-mia have a high incidence of HAI,infection mainly occurs in respiratory system,and gram-negative bacteria are ma-jor pathogens.The incidence of HAI is correlated with remission induction chemotherapy,long length of hospital stay,low WBC,low number of neutrophils,and acute myeloid leukemia.
8. Role of STAT3 antisense nucleic acid in promoting radiosensitivity of A549 cells

Objective To investigate the influence of self-designed STAT3 antisense oligodeoxynucleotides (STAT3 ASODN) on irradiation sensitivity of lung adenocarcinoma. Methods Lung adenocarcinoma cells were exposed to different doses of irradiation (4, 8, 12, 16, and 20 Gy 60Co γ) after transfected with STAT3 ASODN. The survival rates of cells in each group were evaluated by CCK-8 assay. Early apoptosis of cells was observed by FACS. Local irradiation models were established with tumof-bearing mice. STAT2 ASODN was intratumorally injected for two weeks at a dose of 15 mg/kg, and 2 h alter injection the animais were locally irradiated with 60Co γ(totally 20 Gy, 2 Gy each time, 5 times each week for 2 weeks, with a irradiation rate of 1 Gy/min). The following parameters were observed, including the tumor size, delay of tumor growth, relative growth rate, growth inhibition rate, q value, and tumor mass. Results In vitro experiment showed that compared with irradiation alone, combination of irradiation with STAT3 ASODN signiflcantly decreased the survival rate and increased the early apoptosis rate of A549 celis. In vivo experiment found that combination of STAT3 ASODN with local irradiation greatly inhibited the growth rate of lung adenocarcinoma xenografts (q= 1.27). Conclusion STAT3 ASODN not only possess chemotherapeutic effect, but also can improve the sensitivity of tumor cells, making it a promising agent for future research and clinical application.
9.The influence of obstetric complications and pregnancy outcome because of prenatal anxiety and depression

Objective To investigate the influence of obstetric complications and pregnancy outcome because of prenatal anxiety and depression.Me~ods One hundred primipara were selected as the research objects,their emotions were evaluated by Self-Rating Anxiety Scale(SAS),Self-Rating depression Scale(SDS).The patients' scores of SAS ≥50points or SDS ≥ 53points were selected as the observation group,the patients'scores of SAS ≥ 50points and SDS≥53points were selected as the control group.The pregnant women childbirth way,preeclampsia,hyperemesis gravidarum,gestational diabetes,perinatal infection,hydramnios,oligohydramnios,premature rupture of membranes,postpartum hemorrhage,fetal distress,premature birth,low birth quality,neonatal asphyxia of two groups were compared.ResultsThe primipara cesarean delivery rate of the control group was lower than that of the observation group(4.00% vs 22.00%,x2 =7.161,P <0.05),the obstetric complications incidence of the observation group was higher than that of the control group (32.00% vs 2.00%,x2 =15.946,P < 0.05),the birth outcomes incidence of the observation group was lower than that of the control group (4.00% vs 20.00%,x2 =6.061,P < 0.05).Conclusion Prenatal anxiety and depression significantly increase pregnant women obstetric complications,increase the perinatal fetal intrauterine distress and the probability of premature birth,low birth quality and neonatal asphyxia.
10.Expression of nitric oxide synthase in the process of cerebral ischemia/reperfusion

BACKGROUND: Nitric oxide synthase(NOS) is the key factor for the synthesis of nitric oxide (NO) . Because NO combines with oxygen, hemoglobin and other substances in vivo easily and deactivates quickly, and it is not exactly determined, so determining the activity of NOS is the important link for further studying the pathogenesis of NO in cerebral ischemia/reperfusion (I/R)injury.OBJECTIVE: To study the effect of different types of NOS in the process of cerebral I/R injury.DESIGN: Randomized and controlled animal experiment.SETTING: Instituteof Cerebrovascular Disease, Affiliated Hospital of Medical College of Qingdao University.MATERIALS: This experiment was carried out in the Shandong Key Laboratory of Prevention and Treatment for Encephalopathy from May to December 2005. Twenty-eight adult healthy male Wistar rats, of clean grade, weighing from 220 to 260 g, were provided by the Experimental Animal Center of Shandong University. The involved rats were randomly divided into sham-operation group (n =4) and cerebral ischemia group (n =24). Six time points were set in cerebral ischemia group: ischemia 1 hour reperfusion 6 hours, 12 hours, 1 day, 3 days, 7 days and 14 days, 4 rats at each time point.METHODS: Rat models of middle cerebral artery occlusion/reperfusion were established by suture-occluded method through inserting a suture into the left internal-external carotid artery. The expressions of different types of NOS at different time points after cerebral I/R were detected by immunohistochemical technique.MAIN OUTCOME MEASURES: ①Toluidine blue-stained two groups of nerve cells; ② The expression and distribution of neuronal NOS (nNOS), endothelial NOS (eNOS) and inducible NOS(iNOS) at different time points.RESULTS: ①Karyopyknosis and cell debris appeared in the nerve cells of the injured region of cerebral ischemia group,and there were no significant differences of cells among different time points. ② Six hours after reperfusion, the expressions of nNOS, eNOS and iNOS were found in the neurons of brain tissue and increased with the elongation of time of reperfusion. The regions in which different types of NOS in neurons of brain tissue were expressed were cortical area and corpora striata. nNOS and iNOS were highly expressed within 12 hours to 7 days after reperfusion in the brain, and eNOS was highly expressed within a short time period, i.e. 6 hours to 3 days after reperfusion. eNOS expression increasing and decreasing occurred earlier than nNOS and iNOS. But the expressions of three kinds of NOS all reached peak on the first day after reperfusion. The changing tendencies of the expression of three kinds of NOS in the cortical area and corpora striata were the same basically.CONCLUSION: After cerebral I/R injury, the high expression of eNOS occurs early and lasts for a short time, while that of nNOS and iNOS occurs late and lasts for a long time.
