1.Studies on the intratumoral microvascular density (IMD) and the VEGF expression in malignant mesothelioma

Objective To study the prognosis significance of intratumoral microvascular density(IMD)and the expression of vascular endothelial growth factor(VEGF)in malignant mesothelioma.Methods The data of 79 cases were reviewed of malignant mesothelioma confirmed by histology,immunohistochemistry and telectron microscope.CD34 and the expression of VEGF were detected by Immunohistochemical method.Results The age of the patients with malignant mesothelioma ranged from 11 to 79 years.The peak incidence of malignant mesothelioma was at the range of 40-50 years.The sex ratio of male to female was 1.78∶1.The survival period of the 79 cases was between 5 and 160 months with the medial survival of 23 months.According to histological structure,these cases were divided into epithelioid type(51 cases),sarcomatous type(20 cases)and mixed type(8 cases).For the VEGF positive expression,epithelioid type was 26 cases(50.98%),sarcomatous type was 13 cases(65.00)and mixed type was 5 cases(62.50%).Based on the mean IMD,the patients with malignant mesothelioma were divided into two groups.The survival time of the patients in the higher IMD group was significantly shorter than those in the lower IMD group.The analysis,on the relation of VEGF expression to IMD,as well as survival time and histological subtype,showed that VEGF expression was positively correlated to IMD(P=0.02,r=0.330 2).A significant difference existed between the survival of VEGF positive expression and negative expression(P0.05).Conclusion The results suggest that the prognosis of malignant mesothelioma probably related with IMD and VEGF expression.The higher index of IMD and the higher expression of VEGF might imply a worse prognosis.

Objective To investigate the mRNA expression of COX-2 and MMP-2 on transitional cell carcinoma of bladder(TCCB) and their relationship with the invasion and metastasis of the bladder neoplasm. Methods Surgical bladder specimens were obtained from 54 TCCB patients and 5 benign prostate hyperplasia (BPH) patients to make paraffin slices, and 8 specimens which against cancers. 3.Effects of excessive aluminum on osteoclasts and expression of mRNA related with bone metabolism in C57BL/6 mice

Objective To investigate the effects of excessive aluminum on osteoclastic formation and function in C57BL/6 mice.Methods Six-week-old male C57BL/6 mice (n =20) were randomized by weight and divided into control group and treatment group.The mice were assigned to distilled water and 270 mg/L aluminum ion,respectively.After the 15-week treatment period,the mice were euthanized by ether asphyxiation.Concentration of serum aluminum ion was determined by inductively coupled plasma (ICP).TRAP+ cells in vivo were observed using histomorphometry and osteoclasts microstructure using electron microscope (TEM).Osteoclasts were induced by macrophage colony-stimulating factor (M-CSF) and receptor activator of nuclear factor κB ligand (RANKL) with bone marrow macrophages separated from femur and tibia.The expression of mRNA related with bone resorption was detected,including c-FBJ osteosarcoma oncogene (c-Fos),nuclear factor of activated T-cells cl (NFATc1),c-nonreceptor tyrosine kinase (c-Src),dentrocyte expressed seven transmembrane protein (DC-STAMP),d2 isoform of vacuolar (H1) ATPase v0 domain (ATP6v0d2) and matrix metalloproteinase 9 (MMP-9).Results The data showed that the treatment group [(12.04 ± 0.21)mg/L] had significantly increased serum aluminum ion compared with the control group [(11.00 ± 0.04)mg/L,F =10.286,P < 0.05].TRAP+ cells examination confirmed osteoclasts in treatment group [(31.39 ± 9.80) number] were significantly higher than those in the control group [(5.46 ± 4.15) number,F =9.344,P < 0.05].A large proportion of osteoclasts in treatment group lacked ruffled borders and showed vacuolation degeneration.The expression of mRNA in treatment group was lower than that of the control group.The expression of mRNA in control group and treatment group was NFATcl (3.25 ± 0.93 vs.0.29 ± 0.18,F =11.602,P < 0.05),c-Fos (0.86 ± 0.16 vs.0.16 ± 0.02,F =9.405,P < 0.05),c-Src (8.82 ± 1.51 vs.2.29 ± 0.36,F =9.128,P < 0.05),DC-STAMP (3.70 ± 0.70 vs.1.36 ± 0.57,F =10.298,P < 0.05),ATP6v0d2 (15.60 ± 4.81 vs.1.39 ± 0.95,F =8.828,P < 0.05),and MMP-9 (18.64 ± 7.62 vs.2.10 ± 0.92,F =9.356,P < 0.05),respectively.Conclusions Aluminum can increase the number of osteoclasts under epiphyseal plate,but inhibits osteoclasts differentiation.This phenomena may be related with decreased expression of c-Src,DC-STAMP,c-Fos,NFATcl,ATP6v0d2 and MMP-9 mRNA which regulate the function of osteoclasts.

Funding of research projects within the category of Endemiology (H2401) funded by National Natural Science Foundation of China (NSFC) from 2010-2019 was summarized in this paper. Granted projects were categorized and analyzed based on the funding type, support institute and disease classification. The characteristics and trends of granted projects supported by NSFC were analyzed and summarized in order to provide reference for researchers to apply for projects in the future.
5.An annual national monitoring report on drinking-water-borne endemic fluorosis in 2012

Objective To investigate the implementation effects of control measures and the dynamic prevalence of drinking-water-borne endemic fluorosis,in order to provide a scientific basis for developing control measures against the disease.Methods According to the Surveillance Scheme of Drinking-Water-Borne Endemic Fluorosis,137 counties were selected as monitoring counties in national 27 provinces (autonomous regions,municipalities) and Xin Jiang Production and Construction Corps.Three epidemic villages were selected as fixed monitoring villages in each county.The operating condition and the fluoride level of water improvement projects were investigated in the villages with water improvement projects.The fluoride level of drinking water was tested in the villages without water improvement projects.The fluoride content in drinking water was detected by the Standard Test Method for Drinking Water (GB/T 5750.5-2006).The preyalence of dental fluorosis of all children aged 8-12 were surveyed,and dental fluorosis was diagnosed by Clinical Diagnostic Criteria of Dental Fluorosis (WS/T 208-2011).Results ①427 villages were surveyed in 2012,among which the villages with water improvement projects accounted for 75.88% (324/427).② 299 water improvement projects were investigated.Normal operation rates of those were 89.30% (267/299).The qualified rate of fluoride content was 75.78% (219/289).Totally 103 villages without water improvement projects were monitored.Villages with fluoride level > 1.2-2.0 mg/L,> 2.0-4.0 mg/L and > 4.0 mg/L accounted for 48.54% (50/103),29.13% (30/103) and 9.71% (10/103),respectively.(③The national detection rate of dental fluorosis and community fluorosis index (CFI) in children aged 8-12 were 31.24% (8 677/27 773) and 0.66.The detection rate and CFI of the villages with normal operation projects and the qualified fluoride content were 24.04% (3 999/16 634) and 0.51.The detection rate and CFI of the villages with water improvement projects under abnormal operation or excessive fluoride were 40.77% (1 654/4 057) and 0.91.The rate and CFI of the villages without water improvement projects were 42.70% (3 024/7 082) and 0.86.Conclusions The rate of water improvement projects of the whole country is lower.The national normal operation rate of water defluoridation improvement projects is below 90%,and the qualified rate of fluoride content in drinking water is below 80%.The prevalence of dental fluorosis in children aged 8-12 is very mild.

In 2012,General Office of the State Council forwarded the'12th Five-Year Plan'on National Prevention and Control of Endemic Diseases,which was jointly developed by the Former Ministry of Health,Development and Reform Commission,and Ministry of Finance.During the 12th Five-Year Plan period,the national prevention and control of endemic diseases had made steady progress.This article mainly summarized the progress made in prevention and control of endemic diseases in China during the 12th Five-Year Plan period.The main problems encountered in prevention and control of endemic diseases in China were discussed.The prevention and key research tasks of the 13th Five-Year Plan on national prevention and control of endemic diseases were introduced.In the 13th Five-Year Plan,key issues in prevention and control of endemic diseases will be the existing main problems,to carry out targeted prevention and control,and to develop scientific and accurate prevention and control plan according to law.
7.Studies on the characteristics and cytotoxicity of gentamicin/chitosan microspheres for local injection

Objective To construct the drug delivery system of gentamicin/chitosan microspheres for local injection, and evaluate its physicochemical properties and cell cytotoxicity.Methods Gentamicinwas used as model drug, chitosan as carrier, lecithinand hydroxypropyl-β-cyclodextrin as accessories, and the microspheres of gentamicin/chitosan/lecithin/hydroxypropyl-β-cyclodextrinwas prepared by spray drying method.The physicochemical properties and cell cytotoxicity of themicrospheres were investigated by UV spectrophotometry , scanning electron microscopy, X-ray diffraction, dynamic membrane dialysis and MTT assay.Results Five kinds of chitosan microspheres ( A, B, C, D and E) with different drug/carrier ratios were successfully prepared by spray drying method.The yield, drug loading and entrapment efficiency of the drug-loaded microspheres were 34.38%~46.94%, 10.20% ~18.67%, 61.20% ~74.72%, respectively.SEM results showed that compared with microspheres A, B and C, microspheres D and E own the spherical shape with wrinkled surface and uniform particle size, particle size between 0.5 ~3 μm, no adhesion.X-ray diffraction analysis showed that the drug was encapsulated in the microspheres.The results of in vitro release indicated that microspheres D had a good sustained release effect in the four drug-loaded microspheres.The results of cytotoxicity test showed that when the concentration of gentamicin reached 400 μg/mL, the relative growth rate of microspheres D was still higher than 80% with the cytotoxicity grade was one, ie, no cytotoxicity.Conclusion The microspheres D with good physicochemical properties, sustained-release effect and biocompatibility, is expected to be a good carrier of prostate local drug delivery.
8.Value of preoperative neutrophil-to-lymphocyte ratio as a prognostic predictor after radical resection of bladder cancer for muscle-invasive bladder cancer

Objective To investigate the value of preoperative neutrophil?to?lymphocyte ratio(NLR) as a prognostic predictor after radical resection of bladder cancer for muscle?bladder cancer. Methods Two hundred and fifty?two cases consecutive muscle?invasive bladder cancer patients treated with total excision radi?cal bladder at the Rology Surgery Department of Affiliated Hospital of Weifang Medical College from January 2006 to March 2010 were retrospectively analyzed,and all patients were diagnosed bladder cancer by pathological examination. Based on the cut?off value of 2. 7,all the patients were divided into two groups:a low NLR(<2. 7,n=142) group and a high NLR(≥2. 7,n=110) group. None accepted neoadjuvant therapy. Results The 5?year overall survival rate was 75. 2% in patients of the low NLR group and 53% in the high NLR group,the difference was significant(P=0. 005). Univariate analysis showed that NLR(P=0. 002),tumor size(P=0. 035),T stage (P=0. 0022),tumor grade(P=0. 04),hydronep?hrosis or no(P=0. 031),Lymph node metastasis or no(P=0. 003) were associated with survival. Cox multivariate analysis revealed that NLR was independent risk factors of prognosis(HR:2. 586;95%CI:1. 256-2. 851;P=0. 039). Conclusion Preoperative NLR≥2. 7 may be a convenient biomarker to predict patients with a poor prognosis after radical cystectomy for muscle?invasive blad?der cancer.
9.Human electrocardiogram changes caused by exposure to arsenic through drinking water

Objective Through studying the relationship between different concentrations of arsenic in drinking water and ECG changes,to investigate the effects of arsenic on human cardiovascular system.Methods During 2008-2013,people that over 20 years old and over 10 years of drinking arsenic water were investigated in 15 villages of Shanxi Province and Inner Mongolia according to the historical data of endemic arsenic poisoning through drinking water,who were divided into control group (< 0.01 mg/L),low arsenic group (0.01-< 0.05 mg/L),medium arsenic group (0.05-< 0.10 mg/L) and high arsenic group (≥0.10 mg/L) according to the concentration of water arsenic.Arsenic concentrations in drinking water samples were detected by the method of hydride generation atomic fluorescence spectrometry.ECG-site inspections were finished to record 12-lead ECG.The effect of different concentrations of arsenic in drinking water on ECG was studied.Results ECG of 1341 people were surveyed,and abnormal rate was 11.56% (155/1 341),including abnormal rate of control group,low arsenic group,medium arsenic group and high arsenic group which was 5.7% (9/158),12.85% (59/459),12.02% (28/233),and 12.02% (59/491),respectively.The abnormal rate of control group was lower than that of low arsenic group,medium arsenic group and high arsenic group (x2 =6.141,4.391,5.090,all P < 0.05).ECG changes were characterized by cardiac arrhythmias and ST-T changes.A variety of arrhythmias abnormal rate of control group [0(0/158)] was lower than that of low arsenic group,medium arsenic group and high arsenic group [4.58% (21/459),3.86% (9/233),3.46% (17/491); x2 =7.483,6.247,5.618,all P < 0.05].In addition,there were significant differences among the four groups in the rates of right ventricular enlargement and myocardial ischemia (x2 =9.525,9.848,all P < 0.05).Conclusions ECG changes of the residents in the areas of water-borne-endemic arsenic poisoning are characterized by cardiac arrhythmias and ST-T changes.Water arsenic concentration exceeding 0.01 mg/L could significantly increase the ECG abnormal rate,indicating that arsenic in drinking water has a certain effect on heart tissue.
10.Effects of sub-chronic arsenic exposure on arsenic exposure markers and brain total NOS activity in rats

Objective To observe the changes of the totle nitric oxide synthase (NOS) activity in brain tissue,the metabolism of arsenic speciations in urine and the totle contents in blood,brain after rats drinking water containing different doses of arsenic.Methods Forty SD rats were divided into 4 groups according to random number table,10 rats in each group:control group,5 mg/L NaAsO2 group,10 mg/L NaAsO2 group and 50 mg/L NaAsO2 group.The animals were allowed free access to water and food.Body mass was weighted once a week.Expose to arsenic was continued for three months,then the animals were put to death and their blood,urine and brain tissues were collected.Determination of four kinds of speciations of arsenic (3 valence inorganic arsenic,iAs3+;5 valence inorganic arsenic,iAs5+;monomethylated arsenic,MMA;dimethylated arsenic,DMA) in urine was carried out by high performance liquid chromatography-hydride atomic fluorescence spectrometry.Total arsenic concentration in blood and brain tissue was detected by Atomic Fluorescence Spectrometry.The activity of total NOS in blood and brain tissue was detected using the spectrophotometer method.Results ①Weight:at the 5th-12th week after arsenic exposure,compared with the weight of control group [(420.93 ± 21.13),(441.52 ± 28.85),(462.45 ± 30.57),(470.16 ± 31.17),(484.92 ± 32.93),(483.79 ± 29.63),(482.02 ± 29.14),(483.89 ± 29.31) g],weight of rats in 50 mg/L NaAsO2 group [(391.66 ± 32.88),(410.17 ± 33.47),(426.96 ± 33.49),(427.15 ± 32.20),(441.78 ± 33.69),(438.27 ± 33.05),(440.98 ± 33.33),(441.46 ± 32.45) g] was significantly lighter (all P < 0.05).② Urine arsenic:the medians of iAs3+ content (0.00,57.30,236.33,857.80 μg/L) were compared between control group,5,10 and 50 mg/L NaAsO2 groups,the differences were statistically significant (x2 =31.982,P < 0.01);the medians of iAs5+ content (0.00,0.00,80.75,162.90 μg/L) were compared between control group,5,10 and 50 mg/L NaAsO2 groups,the differences were statistically significant (x2 =24.206,P < 0.01);the medians of DMA content (12.83,1 711.13,l0 386.20,37 038.90 μg/L) were compared between control group,5,10 and 50 mg/L NaAsO2 groups,the differences were statistically significant (x2 =34.338,P < 0.01).③Blood arsenic:total arsenic content in serum of rats [(5.04 ± 1.57),(25.40 ± 7.33),(32.28 ± 7.75),(56.11 ± 19.87) mg/L] was compared between control group,5,10 and 50 mg/L NaAsO2 groups,the differences were statistically significant (F =27.78,P < 0.05).④Brain arsenic:total arsenic content in brain tissue of 5,10 and 50 mg/L NaAsO2 groups [(0.57 ± 0.20),(1.56 ± 0.52),(3.63 ± 0.48) μg/g] was respectively compared with that of control group [(0.11 ± 0.06) μg/g],the differences were statistically significant (all P < 0.05).⑤NOS activity:compared with control group [(27.69 ± 5.56) kU/L],total NOS activity [(33.63 ± 2.26),(34.19 ± 2.55) kU/L] in serum of rats in 10 mg/L NaAsO2 group and 50 mg/L NaAsO2 group increased significantly (all P < 0.05);compared with control group [(1.79 ± 0.79) U/(mg·prot)],total NOS activity [(2.63 ± 0.60)U/(mg ·prot)] in brain tissue of 50 mg/L NaAsO2 group increased significantly (P < 0.05).Conclusions A high dose of arsenic exposure can increase totle contents of arsenic in blood,brain and the activity of total NOS in rat brain tissue.
