Background and purpose:HIF-1? (Hypoxia-inducible factor-1?) is an important transcription factor under hypoxia condition. It plays the role of dominating the expressions of correlative genes. It also promotes tumor deterioration, tumor metastasis and tumor invasion. The molecular role of HIF-1? has been intensively studied in cancer basic research. This article is to investigate the expression of HIF-1? under hypoxic and reoxygenation induced by CoCl_ 2 and the impact of hypoxia-inducible transcription factor-1? on human ovarian carcinoma cell line HO-8910PM. Methods:Semi quantitative reverse transcription-polymerase chain reaction (RT-PCR) is performed to detect the expression of HIF-1? mRNA in human ovarian carcinoma cell HO-8910PM exposed during the phase of hypoxia and reoxygenation. The relations of the quantity-efficiency and the time-efficiency were analyzed. The effects of HIF-1? gene on the proliferation, invasion and adhesion of HO-8910PM cell were estimated by either MTT, Boyden cell or cell adhesion tests.Results:There was endogenous expression of HIF-1? in human ovarian carcinoma cell line HO-8910PM. 3.Culture of Donor Cells in Extracranial Pituitary Implantation

Objective To culture human pituitary adenoma cells to provide some useful experimental foundations to practice pituitary adenoma cells as donor cells in the extracranial implantation. Methods Human pituitary adenoma cells obtained by operation were cultured in vitro, and their growth state was dynamically observed. Immunohistochemical staining and RIA were used to evaluate their functional status. Results The cultured pituitary adenoma cells still had secretory function in vitro. 6 cases of GH adenoma cells secreted a large amount of GH, 5 cases of PRL adenoma cells secreted a large amount of PRL, and 3 cases of pituitary adenoma cells without function could also secrete a small quantity of FSH, LH and PRL. Conclusion 14 cases of cultured human pituitary adenoma cells had secretory function in vitro, which provides the useful experimental data to perform extracranial implantation with pituitary adenoma cells as donor cells.
4.Relationship between range of operative and postoperative ovary function in 55 endometriosis

ObjectiveTo study the relationship between the range of operations and the incidence of climacteric syndrom,the time of episode, degree and estrogen level in endometriosis after operations. MethodsThesexual hormone level was measured and climacteric syndrome was followed up after operation in 55 endometriosis. ResultsThe incidence of climacteric syndrom in oophorectomy was more frequent and earlier than partial oophorectomy. Meanwhile, the syndrome was much severe and the level of E2 was lower in the former group than those in the later one. ConclusionSo we should retain the ovary tissue and blood supply as much as possible. 6 months postoperation is suggested to be suitable to begin hormone replacement therapy. The dosage should be adviced to remain the blood hormone level just like one ovary does.
5.The dynamic changes in the numbers and functions of CD8~+T cells and NK cells in advanced ovarian cancer patients undergoing first line chemotherapy

Objective: The purpose of this study is to investigate the dynamic changes in the numbers and functions of CD8~+T cells and NK cells in patients with advanced ovarian cancer undergoing first line chemotherapy,so as to identify whether there was a "window period" of anti-tumor immune suppression reverse after chemotherapy.Methods: Peripheral blood samples from each ovarian cancer patient were obtained before (S_0) and at day 5-7 (S_1),day 12-14 (S_2) and day 25-28 (S_3) after Chemotherapy in 13 patients.The numbers and proportions of CD3~+,CD4~+,CD8~+ and nature killer (NK) cells were analyzed by flow cytometry technique.The percentages of specific IFN-γ-secreting CD8~+ cells were also calculated after that peripheral lymphocytes had been stimulated with self tumor lysates.Cytotoxicity of NK cells against K562 cells was detected by LDH releasing assay.Results: The numbers of CD3~+,CD4~+,CD8~+T cells and NK cells reduced to the lowest on S1.Compared to those of the control group,and the percentages of IFN-γ-secreting CD8~+T cells were remarkably higher on S_1,S_2 and S_3 when CD8~+T cells were stimulated with autologous tumor antigen,and the percentage of CD8~+IFN-γ~+ cell reached the highest on S2.No significant differences of NK cell cytotoxicity against K562 cells were found on S_1,S_2 and S_3 compared to S0.Conclusion: Paclitaxel and carboplatin induce lymphopenia,which triggers the temporary immune reconstitution.During immune reconstitution the enhanced priming of CD8~+T cell response by autologons tumor antigen is found while the function of NK cells does not change significantly.It probably turns out that the" window period"during immune reconstitution offers a best opportunity for cancer immunotherapy.
6.Artery stiffness is an independent factor for left ventricular remodeling and cardiac function in coronary heart disease patients

Objective To study the correlation of artery stiffness with left ventricular remodeling and cardiac function in coronary heart disease (CHD) patients .Methods Four hundred and eighty patients were divided into CHD group (n=373) and non-CHD group(n=107)according to their coronary angiography .Patients in CHD group were further divided into normal left ventricular re-modeling group (n=139) and abnormal left ventricular remodeling group (n=243) .Their brachi-al-ankle pulse wave velocity (BaPWV ) was measured .The patients underwent echocardiography and 42 patients in CHD group received Doppler tissue imaging .Results The BaPWV was signifi-cantly higher in CHD group than in non-CHD group (P<0 .01) and in abnormal left ventricular remodeling group than in normal left ventricular remodeling group (1689 ± 305 cm/s vs 1508 ± 280 cm/s ,P< 0 .05) .The BaPWV was positively related with the IVST ,PWT ,RWT ,LVMI ,E/Ea and negatively related with the LVEF ,Ea ,Ea/Aa in CHD group (P<0 .05 ,P<0 .01) .Partial cor-relation analysis showed that the BaPWV was significantly related with the IVST ,PWT ,RWT , LVMI ,LVEF ,Ea ,Ea/Aa and E/Ea when the effect of hypertension ,etc ,was excluded (P<0.01) .Multivariate regression analysis demonstrated that the BaPWV was an independent factor for LVMI and LVEF .Conclusion BaPWV is an independent factor for left ventricular remodeling and cardiac function in CHD patients .

Objective To investigate the changes of Tc1/Tc2 profiles and the cytotoxic function of CD8~+ cells in ovarian cancer patients undergoing paclitaxel and carboplatin chemotherapy, so as to identify whether there is a "window period" of anti-tumor immune suppression reverse after chemotherapy. Methods Blood samples from each ovarian cancer patient were obtained before (S_0) and at day 5-7 (S_1), day 12-14 (S_2) and day 25-28 (S_3) after chemotherapy in 13 patients. Thirteen age-matched healthy female volunteers were enrolled as a control group. Flow cytometry technique was employed to analyze the proportion of Tc1/Tc2. The numbers of specific IFN-γ, secreting CD8~+ cells were also calculated after peripheral lymphocytes had been stimulated with self tumor lysates. Results The proportion of Tel in CD8~+ cells increased re-markably on S2 while the proportion of Tc2 in CD8~+ cells remained no significant changes after chemothera-py. The ratio of Tc1 to Tc2 cells reached the highest on S_2. IFN-γ/secreting CD8~+ T cells also increased re-markably on S_2, especially when CD8~+ T cells were stimulated with autologous tumor antigen. Conclusion Paclitaxel and carboplatin induce the changes of Tc1/Tc2 profile and augment anti-tumor immune response by immune reconstitution. It probably turns out that the "window period" during immune reconstitution offers a best opportunity for cancer immunotherapy.

9. Steroidal saponins from the seeds of Trigonella foenum-graecum L.: Research advances

Seeds of Trigonella foenum-graecum L., as a raditional Chinese medicine, are rich n steroidal saponins. The systematic research on steroidal saponins of Trigonella foenum-graecum, including the study on furostanol saponins and spirostanol saponins, the rapid characterization and dentification of steroidal saponins, and he analytical and preparative separation of C-25 R/S-spirostanol saponin diastereomers using supercritical fluid chromatography, was performed. The results of research are helpful to the development and utilization of Trigonella foenum-graecum seeds, and the methods could provide reference for the chemical research on other traditional Chinese medicines.
