3.Triple-Endobutton technique for the treatment of Tossy type III acromioclavicular joint dislocation.

OBJECTIVETo evaluate the clinical outcomes and complications of Triple-Endobutton plates in treating Tossy type III acromioclavicular joint dislocation.
METHODSFrom January 2011 to January 2013,45 patients with Tossy type III acromioclavicular joint dislocation were treated with Triple-Endobutton plates. There were 35 males and 10 females with an average age of 30.5 (ranged from 19 to 60) years old. At the final follow-up, VAS, DASH, Constant-Murley criterion were used to evaluate shoulder function.
RESULTSAll patients were followed up from 15 to 36 months. No neurovascular injury, wound infection and stress fractures were found,but 3 patients had a re-dislocation. At the final follow-up,the mean VAS score was decreased from (5.7±1.6) preoperatively to postoperative (0.2±0.1); DASH score was significantly decreased from (19.6±4.3) preoperatively to (0.3±0.1) postoperatively; Constant-Murley score was improved from (34.4±4.3) before operation to (94.8± 3.5) after operation.
CONCLUSIONClinical outcomes of treating Tossy type III acromioclavicular joint dislocation with Triple-Endobutton plates is satisfactory. However, re-dislocation is still the most common complication. Careful perioperative management is an important factor in preventing re-dislocation.

5.Application of preoperative CT scanning in three- dimensional positioning of puncture route for percutaneous vertebroplasty

Objective To discuss the feasibility of using three-dimensional simulation positioning of puncture route that is sketched according to the transversal and sagittal CT images to design percutaneous vertebroplasty (PVP) plan. Methods A total of 58 patients with 63 diseased vertebral bodies were enrolled in this study. Before PVP, the simulation puncture route was delineated according to the transversal and sagittal CT images, and the simulation puncture parameters were determined by using the software of PACS, in this way the three-dimensional simulation puncturing route was established , and the skin needle entry point as well as the puncture angle were calculated. Unilateral transpedicular approach was used when the simulation puncture route could meet the requirements of unilateral approach. Otherwise , bilateral transpedicular approach would be employed. Three months after PVP visual analogue score (VAS) was used to evaluate the degree of pain relief, and the result was compared with that determined before PVP. SPSS 13.0 software was used for statistical analysis. The estimating data were statistically analyzed with paired t-test and the results were expressed in the form of (x ± s). Results The measurement results showed that significant differences in the puncture angles and skin needle entry points existed between different vertebral bodies. Guided by the preoperatively determined simulation puncture route , PVP was successfully accomplished in all patients. The success rate of single puncturing was 100% (63 vertebral bodies in total). Among 63 vertebral bodies, unilateral transpedicular approach was adopted in 55 and the operation time was (28.5 ± 5.5) minutes, while bilateral transpedicular approach was carried out in 8 and the operation time was (37.5 ± 5.5) minutes. After PVP, complete pain relief was achieved in all patients. No serious complications occurred during the follow-up period. Conclusion Three-dimensional simulation positioning of puncture route that is sketched according to preoperative transversal and sagittal CT images is very useful in precisely determining the puncturing route before PVP, which is very helpful to increase the success rate of single puncture and to improve the procedural safety.
6.miR-193 regulates proliferation of rat marrow mesenchymal stem cells by cell cycle-related proteins

AIM:To investigate the effects of microRNA-193 (miR-193) on the proliferation and apoptosis of rat bone marrow mesenchymal stem cells (MSCs).METHODS:Cultured rat MSCs were transfected with pre-miR-193 or anti-miR-193 to regulate the expression of miR-193.The proliferation of the MSCs after transfection was evaluated by MTS assay, colorimetric BrdU cell proliferation assay and Ki-67 immunostaining.Cell apoptosis was analyzed by flow cytometry with Annexin V/PI staining.The effect of miR-193 on the expression of cell cycle-related proteins was evaluated by qRT-PCR.RESULTS:Transfection of pre-miR-193 or anti-miR-193 regulated the expression of miR-193 in MSCs effectively . Over-expression of miR-193 significantly promoted the proliferation of MSCs ( P<0.05 ) , and inhibition of miR-193 re-duced the proliferation of MSCs (P<0.05).miR-193 had no significant effect on the apoptosis of MSCs (P>0.05).The result of qRT-PCR indicated miR-193 promoted the expression of cyclin-dependent kinase 2 ( CDK2 ) significantly ( P<0.01).CONCLUSION:miR-193 promotes the proliferation of MSCs possibly through the CDK 2 pathway.
7.Simultaneous quantitative determination of four active components in Salvia miltiorrhiza tablets by HPLC

Aim:To develop a method for rapid and sensitive simultaneous determination of the hydrophilic and lipophilic active components in Salvia miltiorrhiza tablets. Methods:RP-HPLC condition was established to separate danshensu,salvianolic acid B,cryptotanshinone and tanshinone ⅡA from each other on the Diamonsil C18(250 mm×4.6 mm ID,5 μm)column with the mobile phase consisted of methanol and 0.5% formic acid in gradient elution. Results:The four active components had good linearity. The average recoveries ranged between 96.63% to 100.3% with RSDs of less than 2%. Conclusion:The method appeared to be precise and accurate for the simultaneous determination of the four active components in Salvia miltiorrhiza Bunge tablets within 35 min.
8.Effects of Exposure to Arsenic through Drinking Water on Simple Visual Reaction Time and IQ of Combined Raven's Test in Children

Objective To explore the intellectual effects of arsenic in drinking water on children. Methods Intellectual effects were evaluated by computer-based reaction time and Combined Raven's Test. Children aged 9 to 11 years living in three villages of Inner Mongolia were investigated in April, 2006. The arsenic concentrations in the drinking water of the three villages were 0.16 mg/L, 0.09 mg/L and less than 0.05 mg/L (control group). Results The mean, fastest and slowest visual reaction time of children exposed to the high levels of arsenic through drinking water were longer than those of children in control. The differences of mean, fastest and slowest visual reaction time between girls exposed to 0.16 mg/L arsenic and control were significant; the differences of the mean and slowest visual reaction time between 10-year old children exposed to 0.16 mg/L arsenic and control were significant. IQ of girls exposed to 0.16 mg/L arsenic was significantly lower than that in control. Conclusion Intellectual function of children can be affected by arsenic in drinking water, and the intellectual effects of arsenic on girls are more obvious.
9. Real-time fluorescent quantitative PCR detection of key enzyme genes expression of alkaloid biosynthesis promoted by endophytic fungal elicitor in Sophora alopecuroides

Objective To establish a method of detecting the expression of Lysine decarboxylase (LDC) -a key enzyme for the synthesis of alkaloid in the host promoted by the endophytic fungal elicitor of Sophora alopecuroides by using real-time fluorescence quantitative PCR (qRT-PCR). Methods Target gene primers QLDC-F/QLDC-R and reference gene primers Lectin-F/Lectin-R were designed according to LDC and Lectin gene sequences of S. alopecuroids; Five-fold gradient dilution of cDNA was used as the standard sample for the construction of the standard curve of target gene and the reference gene. Reaction system and reaction conditions of qRT-PCR were optimized, and the sensitivity of semi-quantitative PCR and qRT-PCR were analyzed and compared. Under different eliciting time of endophytic fungal elicitors NDZKDF13 of S. alopecuroides, the content of oxymatrine in the host was determined by HPLC, the expression of LDC gene was detected by qRT-PCR, and the relationship between LDC gene expression and the accumulation of OMA was analyzed. Results The results of qRT-PCR were better when the cDNA content in the system was 200 ng/μL and the annealing temperature was 61 ℃. The standard curve of the target gene and the reference gene was constructed, in which the cycle threshold and template concentration showed a good linear relationship, the amplification efficiency was above 99%, and the sensitivity was 25 times that of semi-quantitative PCR. Under the induction effect of endophytic fungal elicitor NDZKDF13, expression of host LDC gene reached the peak on the 6th day, which was 25.58 times that of the control. The increase of OMA content lagged the change of the LDC gene expression and reached the highest amount on the 9th day after the induction. Conclusion The qRT-PCR technique was successfully applied to the functional gene research of S. alopecuroides. Through the optimization of various conditions, a platform for accurate and simple detection of functional gene expression in S. alopecuroides was established.
10.HPLC Determination of Magnolin in Xuetie Dingchuan Plaster

Objective To establish a HPLC method for the determination of magnolin in Xuetie Dingchuan Plaster.Methods The HPLC chromatographic condition was as follows: the analytical column was Lichrospher C8(4.6mm?250mm,5?m),the mobile phase consisted of A(acetonitrile)∶B(1.55%tetrahydrof uran-water) 34∶66,the flow rate was 1.0 mL?min-1,the detected wav elength was 278 nm,and the column temperature was 30 ℃.Results The calibr ation curve was linear(r=0.9999) in the range of 0.42~ 5.25 ?g for magnolin,the average recovery of the method was 100.50 %,with RSD 2.22 %(n=9).Conclusion The method is accurate,simple and stable,and can be used for the quality control of Xuetie Dingchuan Plaster.
