2. Matrine induced apoptosis of murine H22 hepatocarcinoma cells

Objective: To investigate the apoptosis-inducing effect of matrine on murine hepatocarcinoma cell line H22 in vivo and in vitro and explore the underlying mechanisms. Methods: The H22 cell apoptosis induced by matrine at the early stage was detected with Annexin V-FITC/PI double staining assay, The expressions of Bcl-2 and Bax proteins in H22 cells as well as in the BALB/c H22 xenograft tumor tissues were detected using immunohistochemical method. Transmission electron microscopy (TEM) was used to observe the ultramicro-structure alterations of H22 xenograft tumor cells in BALB/c mice. The effect of matrine on the kinetics of tumor growth after subcutaneous injection of H22 cells in BALB/c mice was observed and the tumor inhibition rate was calculated. Results: Annexin V staining detected early apoptosis of H22 cells after treatment with matrine 1.0 mg/mL and 1.5 mg/mL for 48 h. The apoptotic rates were 11.71% and 17.86%, respectively, both of which higher than that of control groups (P <0.05). The tumor inhibition rate was above 60% after matrine treatment. Immunohistochemistry analysis revealed that matrine increased Bax protein expression and reduced Bcl-2 protein expression in both H22 cells in vitro and xenograft tumor tissues in vivo. TEM demonstrated the existence of apoptotic cells and apoptotic bodies in H22 xenograft tumor tissues after matrine treatment. Conclusion: Matrine significantly suppresses tumor growth and induced apoptosis both in vitro and in vivo. The apoptosis-inducing effects is related with up-regulation of Bax protein and down-regulation of Bcl-2 protein.

4.Tolerability and safety of enteral nutritionin in critical ill patients with hemodynamic instability

In patients with a functional gut,enteral nutrition is the preferred route of nutrition support.The favorable effects of enteral nutrition include prevention of mucosal atrophy,maintaining of the integrity of gut flora,and improvement of immunocompetence.The guidlines recommend EN be withheld in patients requiring significant hemodynamic support,because of highdose catecholamine agents which could reduce EN tolerance.Splanchnic perfusion is reduced in sepsis shock,yet vasoactive agents have demonstrated both improved and diminished perfusion.Inadequate perfusion increases the risk of experiencing rare but serious adverse events.This study summarizes the tolerability and safety of enteral nutritionin in critically iH patients with hemodynamic instability and provides theoretical basis for the early administration of EN.
5.A Clinical Study on the Serum Triglyceride and Total Cholesterol of Patients with Vascular Dementia and Alzheimer's Disease

Objective To explore the change and clinical significance of serum triglycerides and total cholesterol of patients with Vascular dementia (VD) and Alzheimer's disease (AD). Methods 54 cases of VD and 78 cases of AD were examined. 50 cases of healthy adult were enrolled as the control group. Results The serum triglyceride [(1.98?0.98) mmol/L, (12.13?1.0) mmol/L] in VD patients and total cholesterol [(6.52?1.42 mmol/L, (6.36?1.15) mmol/L] in AD patients were significantly higher than those of the control group [(1.32?0.88) mmol/L,(5.45?0.93) mmol/L](P0.05). Conclusion The serum triglyceride and total cholesterol of patients with VD and AD were much higher than those of normal healthy persons. It is conducive to lower the triglyceride and total cholesterol for the prevention and treatment of these two diseases.
6.Analysis on the Impact of Resource Allocation on Basic Public Health Service Provided by Township Health Centers

Objective: To describe the status of the basic public health service in township health centers, analyze the resource allocation which influences the supply of basic public health service. Methods: The data is based on monitoring project surveys collected by the health statistical information center, Spss13.0 and Excellare applied to make descriptive statistic analysis. Results:Overall, basic public health service has been carried out well in each area, but there are differences among the application rates of different basic public health service items; basic public health service can be effected by the level of basic public health input and the number of public health workers.
7.Quality of Life in Elderly with Cardiovascular Diseases in A Community in Beijing

ObjectiveTo investigate the quality of life (QOL) of old patients with cardiovascular diseases in community. Methods100 old patients with cardiovascular diseases in Xiluoyuan, Beijing were investigated with WHOQOL-BREF. ResultsThe QOL-BREF scores in the dimensions of physical health, psychology, social relationships, and environment were (53.21±11.50), (53.27±15.99), (54.29±14.41) and (57.65±15.30) respectively. There was not significant difference between self-rating scores and scores in four dimensions of QOL-BREF (P>0.05). There was not significant difference of QOL-BREF scores in four dimensions between male and female, nor the ones whether complicated with diabetes (P>0.05). The incidence of satisfaction was 31% for QOL, 23% for health, and 62% for the healthy system as self-report. ConclusionThe QOL of the patients is related to diseases.
9.Correlation between CD34+ cell and immature granulocyte in donor's peripheral blood by rhG-CSF

Objective To investigate the correlation between immature granulocyte and CD34+ cells, mononuclear cells (MNC) in donor's peripheral blood by recombinant human granulocyte colony-stimulating factor (rhG-CSF). Methods The stem cell were mobilized by rhG-CSF 7.25-10 μg·kg-1·d-1 from 122 allo-PBSCT donors. Before and after mobilization, to test CD34+ cells of peripheral blood stem cell graft and number of MNC, immature granulocyte, CD34+ cell per patient' weight were calculated. Results White blood cell count and immature granulocyte gradually increased, and reached the peak on the frith day. There was a good relationship between increased immature granulocytes and increased CD34+ cells. The patients all achieved completed donor engraftment and achieved hematopoietic recovery. The chromosome, blood type and HLA type were transformed to be donor's type. Ph1 changed to be negative in CML patients. Conclusion rhG-CSF (7.25~10 μg·kg-1·d-1) had a good effect to mobilize PBSC. There was a good relationship between in-creased immature granulocytes and increased CD34+ cells after mobilization by rhG-CSF. The number of immature granulocytes can reflect indirectly the count of stem/progenitor, so the MNC and immature granulo-cytes can become a threshold of dosage standard.
10.Discovery of potential nicotinic acid receptor agonists from Chinese herbal medicines based on molecular simulation.

Nicotinic acid could increase high density lipoprotein and reduce serum total cholesterol, low density lipoprotein cholesterol and triglycerides in human bodies, thus is frequently applied in treating low high-density lipoprotein cholesterol and hypertriglyceridemia in clinic. However, according to the findings, nicotinic acid could also cause adverse effects, such as skin flush, beside its curative effects. In this study, bioisosterism, fragment-based search and Lipinski's Rule of Five were used to preliminarily screen out potential TCM ingredients that may have similar pharmacological effects with nicotinic acid from Traditional Chinese medicine database (TCMD). Afterwards, homology modeling and flexible docking were used to further screen out potential nicotinic acid receptor agonists. As a result, eleven candidate compounds were derived from eight commonly used traditional Chinese medicines. Specifically, all of the candidate compounds' interaction with nicotinic acid receptor was similar to nicotinic acid, and their docking scores were all higher than that of nicotinic acid, but their druggability remained to be further studied. Some of the eight source traditional Chinese medicines were used to lower lipid according to literature studies, implying that they may show effect through above means. In summary, this study provides basis and reference for extracting new nicotinic acid receptor agonists from traditional Chinese medicines and improving the medication status of hyperlipidemia.
