1.Isolation and Identification of cytopathogenic Effect Strains of Epidemic Hemorrhagic Virus in Vero-E_6 Cell Culture

Five strains out of the 8 of epidermic hemorrhagic fever(EHF)virus isolated from the serum of the acute phase of EHF patients in Nanchong, Sichuan were found to have cytopathogenic effect(CPE)through 4th to 10th passages in Vero-E6 cell culture. The biological, physico-chemical and antigenic characteristics of HFN-04 and HFN-19, 2 out of the 5 CPE strains, were identified. The results were as follows;( 1 ) The CPE development and the dynamic proliferation of the virus areconsistent. ( 2 ) The main biological and physico-chemical characteristics of the 2 CPEstrains arc the same as those of EHF virus.( 3 ) When paired samples of EHF patients' serum were tested with the slide antigens of E6 cells infected with these CPE strains, all the convalescent serum demonstrated an increase by 4 fold or more of the IFA. titer.( 4 ) The CPE of the virus can be neutralized by EHF patient's serum or EHF and KHF antisera, but not by normal rabbit serum or the multivalent serum of Reovirus type 1-3.( 5 ) The E6 cells infected by CPE strains of virus can almost fully absorbthe specific antibodies in EHF patients or in EHF and KHF antisera.( 6 ) The suspension of the mouse brain infected by CPE strains can formpositive agglutination reaction with the sentitized blood cells by EHFvirus strain A9 McAb 25-1.On the basis of our observation, it can be concluded that at least certain strains of EHF virus can produce CPE in Vero-E6 cell culture.
2.Character of Hospital Infection and Multidrug Resistance of Citrobacter

OBJECTIVE To investigate the flora distribution,enzyme producing and drug resistance of Citrobacter in our hospital,and analyzed multi drug resistance(MDR) character in order to guide the clinical medication.METHODS Totally 147 clinical isolates of Citrobacter were detected out ESBLs and AmpC ?-lactamases by three-dimensional test,MBL by the double-disk synergy test.at the same time,drug resistance to fifteen antibiotics was also detected by K-B method.RESULTS Hospital infection caused by Citrobacter.most commonly by C.freundii and then C.amalonaticus.Respiratory tract and urinary tract were prone to be infected than other sites(P0.05).The rate of ESBLs,AmpC,ESBLs+AmpC and MBL which produced by Citrobacter were 36.05% 10.20%,7.48% and 2.72%.The Citrobacter were sensitive to imipenem and meropenem,and their resistance rates to imipenem and meropenem were 4.76% and 3.40%,respectively.The resistance rate to cefoperazone/tazobactam was 23.81%.Otherwise,the resistance rate to 12 kinds of other antibiotics were all higher than 40.0%.MDR strains in the ICU ward were 81.08%,while in other wards were 53.64%,with significant difference(P
3.AN INDIRECT PEROXIDASE ANTIBODY TEST (IPA) FOR DETECTION OF ANTIBODIES TO HEMORRHAGIC FEVER WITH RENAL SYNDROMES (HFRS)

Hemorrhagic fever with renal syndrome (HFRS) occurs endemically in certain localities in our country.At present, the diagnostic method usually used is indirect flurorescent antibody (IFA) technique, but due to the requirement of costly equipments and trained technicians, it is not feasible in grass-root clinics. We used an indirect peroxidase antibody method (IPA) to detect HFRS specific IgM. and their results were compared with that of IFA. It was found that the IPA titre was 2.6 times higher than IFA in sera of early cases, and 20 times higher than IFA titers in convalescent sera. Of the 49 sera tested. 40 gave a positive result by IPA. 35 positive by IFA. There was discrepancy between IPA and IFA in 1 serum. Only ordinary light-microscope is necessary to perform IPA test, and the test is sensitive, specific, and easy to perform. The procedure is simpler and can be used in grass-root medical units
4.Detection of Japanese B Encephalitis Specific Antibody in pig's serum with Single Radical Hemolysis

This paper is to report our study of using single radical hemolysis(SRH) technique to detect the specific antibody for Japanese B encephalitis (JBE) virus in 101 samples of pigs' serum at Chongqing area. It was found that SRH was more sensitive and more specific in the detection of the JBE virus antibody in the pig's serum than CF or HI. SRH is simple in its technique and easy to perform. In addition, it is very sensitive and specific and it can be reproduced easily. It is suggested that SRH be used in clinical diagnosis and in seroepidermic survey of JBE virsus infection.
5.SOMATOSTATIN IN SYNAPTIC STRUCTURES OF SUBSTANTIA GELATINOSA OF THE SPINAL CORD

The distribution of somatostatin (SRIF) in substantia gelatinosa of the rat spinal cord was studied by means of immuno-electron microscopy. The ultrastructural features showed that nerve terminals containing SRIF take part in forming presynap- tie elements of axe-somatic, axe-dendritic and axo-axonic synapses. The immune- reactive products locatl at the external membrane of mitochondria, around the small clear synaptie vesicles and in the large granular vesicles. Most of synaptic vesicles are round or ovoid in shape. Only a few of them are flattened. Based on the ultrastructural characteristics mentioned above and related experimental results the authors believe that SRIF in substantia gelatinosa of the rat spinal cord is probably involved as a neurotransmitter instead of neuromodulator.
6.THE ULTRASTRUCTURAL CHARACTERISTICS AND THEIR FUNCTIONAL SIGNIFICANCES OF THE ARCUATE NUCLEUS AND MEDIAN EMINENCE

The ultrastructural characteristics of the arcuate and median eminence in the rat have been studied by means of folmaldehyde-osmic acid fixation method. The observations showed that there are two kinds of neurons (dark and light) in the arcuate nucleus which might be responsible for producing both dopamine and releasing hormones. The tanycytes of the ependyma of the third ventricle run longitudinally through various zones of the median eminence and reach the perivascular space of the portal capillaries. The neurosecretory substance-containing nerve terminals may travel between ependymal cells or end around the basal membrane of the capillaries of the median eminence, even keep close to the endothelial fenestrae of "open-type capillary". The axo-somatie and axo-dendritic synapses are formed at the soma and dendrites of neurons of the arcuate nucleus. Both agranular type and granular type axo-axonic synapses are encountered in the fibrous zone of the median eminence. There are also synaptic connections between the basic processes and the large granular vesicles containing nerve terminals in the palisade zone of the median eminence. The ultrastructural characteristics mentioned above suggest that (1) the releasing (or inhibiting) hormones of the hypothalamus might be released into the portal capillaries from nerve terminals directly or into the cerebro-spinal fluid of the third ventricle and then uptaken by the tanycytes, and transported to the portal capillaries by their processes. (2) each step of synthesis, storage, transport and release of the releasing (or inhibiting) hormones could be regulated by nervous mechanism

8.Explorations on improving the quality of medical genetics teaching for international students

The teaching methods were explored to improve the quality of medical genetic teaching for foreign students according to the common problems during the teaching process. The negative effects of communication barriers in medical genetic teaching could be reduced by interactive teaching or problem-based learning in groups,in which the ability to resolve problems by themselves could be improved. In order to improve the teaching systematicness and teaching quality,the teaching contents in class should be from simple to deep,covering genetic laws,pathogenesis,diagnosis and control measure of genetic diseases. From the perspective of practical application and combining with the construction of self-de-signed teaching textbook and cases, the quality of medical genetic teaching ultimately could be further improved.
