Objective To develop a sensitive, simple, and accurate method for the determination of shionone in rat plasma after ig administration of Asteris Radix petroleum ether extract (RAPE). Methods The separation was achieved by HPLC on a RP18 column (150 mm × 3.9 mm, 5μm) with a mobile phase composed of acetotitrile-0.05% phosphoric acid water (98: 2) at a flow rate of 1.0 mL/min. UV Detector was set at 200 nm and friedelin was chosen as an internal standard. Results The linear range of the standard curves was (0.3443-22.0) μg/mL with the correlation coefficient of 0.9968. The intra- and inter-day precisions were all below 10% and the relative error was -3.5%-1.1%.Conclusion The developed method can be successfully applied to the pharmacokinetic study. After ig administration of RAPE, T1/2(ka) is (33.09 ± 7.32) min and T1/2(ke) is (84.95 ± 22.34) min.

Aim To investigate the change in neuropeptide Y(NPY) Y1 receptor level in infarcted myocardium tissues of (MI) rats. Method MI was induced by ligating the left descending anterior coronary artery (LAD) in the heart of rats. The techniques of reverse transcription polymerase chain reaction (RT-PCR) were used to identify the exist of NPY Y1 receptor mRNA in myocardial tissues. The technique of semi-quantitative PCR wa s used to observe the change of NPY Y1 receptor mRNA level. Results NPY Y1 receptor mRNA distributed in the heart tissues of rat. Compared with sham operated rats ,the NPY Y1 receptor mRNA level both in infarction area and non-infarction area was increased significantly after MI fo r 1d and 3d. The NPY Y1 receptor mRNA level in the heart tissues of sham-op erated rats was also significantly increased compared with that in control rats . Conclusion These results suggest that MI may result in increase NPY Y1 receptor level in the heart tissues of rats. Stre ss stimulation such as surgery may also increase the NPY Y1 receptor level in the heart.

ObjectiveTo explore the expression of N-cadherin and β-catenin mRNA in human brainstem and supratentorial gliomas. MethodsN-cadherin and β-catenin mRNA expression in 18 cases of brainstem gliomas and 18 cases of supratentorial gliomas tissues were detected with PT-PCR. Resultsβ-catenin mRNA expression was more in human brainstem gliomas than in supratentorial gliomas (t=2.255,P<0.05), but was not significantly different of N-cadherin mRNA (P>0.05). The expression of N-cadherin mRNA in human brainstem gliomas of grades Ⅰ~Ⅱ were less than those in human gliomas of grades Ⅲ~Ⅳ (t=2.711,P<0.05), but was not of β-catenin mRNA (P>0.05). N-cadherin mRNA expression was positively correlated with the β-catenin mRNA expression in either brainstem gliomas or supratentorial gliomas (r=0.480,r=0.809 respectively, P<0.05). ConclusionThe over expressions of N-cadherin and β-catenin may play an important role in the invasion and malignant progress of human brainstem gliomas.

Objective:
To summarize the research hotspots and knowledge structure of pediatric teaching model by introducing the co-word clustering analysis into the research on the current status of pediatric teaching model and conducting a visualized analysis of related articles in the past decade, and to provide a reference for further development of pediatric teaching model.
Methods:
In July 2017, CNKI and Wanfang Data were searched for related articles published from January 2006 to June 2017, with "pediatrics" and "teaching model" as the key words. The articles which met the research criteria were screened out manually, and Word and Bicomb 2.0 were used to identify high-frequency key words. SPSS 20.0 was used to plot the diagram of the cluster analysis, and Ucinet 6.0 and Newdraw were used to generate the co-occurrence network map to show the relationship between high-frequency key words.
Results:
A total of 367 articles were included and 31 high-frequency key words were extracted, among which the most frequently used key word was "PBL teaching method". Studies in this field were roughly classified into four types, i.e., teaching method in pediatric practice/and clerkship, contents of cultivation and assessments in pediatric teaching, teaching methods for pediatric nursing, and objects of the reform of teaching methods. The key words of "PBL teaching method", "multimedia teaching", "case-based teaching", and "nursing" were located at the center of the co-occurrence network map.
Conclusion
Exploration and practice of various teaching methods have been the main research topics in pediatric teaching model in China over the past decade. PBL is a research hotspot in this field and is often combined with multimedia, case teaching, or other methods in pediatric teaching. In addition, the application of evidence-based medicine has attracted more and more attention.

OBJECTIVE: To compare the quality of original preparations of Thioctic acid injection and generic preparations from 2 domestic manufacturers, discuss the acute toxicity test of mice and to investigate the project of consistency evaluation methods. METHODS: According to the quality standard that stated in Chinese Pharmacopeia, physicochemical properties (characters, pH, osmotic pressure, etc., contents and related substances of samples of Thioctic acid injection as well as LD50 in acute toxicity test (n=10), and mortality of mice after administration of injection solution (n=30) were compared among 3 manufacturers. RESULTS: The physicochemical properties as and related substances of the original drug and 2 generic drugs were all in line with the quality standard; the contents of 3 samples ranged 95%-105%. The acute toxicity test results showed that the LD50 values of 2 generic drugs (LD50: 247.911 mg/kg, 95% confidence interval: 222.209-277.999 mg/kg;LD50: 215.291 mg/kg, 95% confidence interval: 196.637-235.053 mg/kg) were smaller than that of original drug (LD50: 266.534 mg/kg, 95% confidence interval: 250.597-283.418 mg/kg), but there was no statistical difference (P>0.05). The results of 3 repeated experiments showed that there was statistical significance in the number of animal death caused by the 2 generic drugs (26, 28) was more than that of the original drug (19) (all P<0.05), when injection solution was injected into mice in a single dose. After administration of the original drug, mice showed excitatory reactions such as movement and squeal, while 2 generic drugs showed inhibitory reactions. CONCLUSIONS: 2 generic drugs of Thioctic acid injection and the original drug all conform to the relevant regulations of Chinese Pharmacopoeia in terms of preparation quality standards, but the results of acute toxicity test are quite different, so it is difficult to prove the consistency between the 2 generic drugs and the original drug. Therefore, acute toxicity test is necessary for the consistency evaluation of injections.
