1.Withdrawal symptoms of ethanol are ameliorated by thymopentin in mice

Objective:To explore the effect of thymopentin (TP5) on the choice of ethanol and ameliorating withdrawal symptoms (anxiety) of ethanol in mice. Methods: Mice were administered ethanol (v/v) in schedular fashion: 5% (1 week), 10% (1 week) and 15% (4 weeks), followed with the free choice between ethanol and water. Ethanol/(ethanol+water)?100% E/(E+W)?100% was measured as an index of ethanol selection. Light-dark box test and elevated plus maze test were chosen for the assessment of anxiety pre-drug and post-drug. After TP5 0.2 mg/(kg?d), 0.4 mg/(kg?d), ip or saline(vehicle control), ip for 14 days,the procedure was repeated. Result: (1) E/(E+W)?100%: the post-drug values of TP5 (0.2 mg/kg, 0.4 mg/kg) were lower significantly than the pre-drug values. (2) Light-dark box test: the post-drug values of number of entries and time spent in the light chamber of TP5 (0.2 mg/kg, 0.4 mg/kg) were more than the pre-drug values themselves and the post-drug value of saline. (3) Elevated plus maze test: the post-drug values of time spent on open arms of TP5 (0.2 mg/kg, 0.4 mg/kg) were more than the pre-drug values themselves and the post-drug value of saline,and the post-drug values of time spent on close arms of TP5 were less than the pre-drug values. Conclusion: TP5 could decrease the uptake of ethanol and ameliorate anxious behavior associated with ethanol withdrawal in mice.
2.5-HT_3 receptor of hippocampus mediate immunomodulation in rat

Objective:The study is designed to explore the neuroimmunomodulation mediated by 5 HT 3 receptor of hippocampus in rats.Methods:ConA or lipopolysaccharide stimulated splenocytes proliferative responses are measured by TdR incorporation.Results:1 phenylbiguanide(PBG) 1 mg/kg,ip,10 ?g,icv and microinjection in the ventral hippocampus could enhance the splenocyte proliferative responses stimulated by ConA and LPS in mice and rats respectively.But the inhibitory effect on ConA stimulated response was only obtained by bilateral microinfusion of PBG in the dorsal hippocampus,while had no effect on the LPS induced proliferative responses.All of effects on mitogen stimulated splenocyte proliferation could be antiagonized by coadministration of a selective 5 HT 3 antiagonist tropisetron.Conclusion:It suggested that the 5 HT 3 receptors in the hippocampus might mediate neuro immuno modulation in which ventral and dorsal hippocampus have different effects.
3.Effects of ST-1 against focal cerebral ischemia-reperfusion injury in rats

AIM: To demonstrate the protective effects of ST-1 against focal cerebral ischemia-reperfusion injury in rats.METHODS: The model of focal ischemia-reperfusion was induced by middle cerebral artery occlusion 2 h followed by reperfusion 22 h in rats.Neurological deficit scores were evaluated.Infarct size,malondialdehyde(MDA) content and superoxide dismutase (SOD) activity were measured,pathological changes of neurons in hippocampal CA1 region were observed.RESULTS: Compared with vehicle group ST1 at dose of 10 and 20(mg?kg~(-1)) could reduce the infarct size,MDA content,enhance SOD activity and relieve neurons injury in hippocampal CA1 region in dose-dependent way;ST-1 at dose of 20(mg?kg~(-1)) could also improve neurological function.CONCLUSION: ST-1 can protect brain tissue from focal ischemia-reperfusion injury.
4.A meta-analysis of clinical effects of Bryan cervical disc replacement versus anterior cervical discectomy and fusion

BACKGROUND:Bryan cervical disc replacement and anterior cervical discectomy and fusion have a dispute in the treatment of cervical spondylosis.
OBJECTIVE:To evaluate the clinical effects of Bryan cervical disc replacement and anterior cervical discectomy and fusion by meta-analysis, thereby providing clinical evidence for treatment strategy of cervical spondylosis.
METHODS:The authors searched Medline, PubMed, EMBASE, OVID, CBM, CNKI and also searched manual y seven relevant Chinese orthopedic journals for articles pertinent to clinical research of Bryan cervical disc replacement and anterior cervical discectomy and fusion. Extracted data included the range of motion of the cervical spine, visual analog scale score, neck disability index, the Japanese Orthopaedic Association score. Meta-analysis and forest plots were conducted with RevMan4.2.2 Software.
RESULTS AND CONCLUSION:There are eight articles in the meta-analysis, including 883 patients (430 patients receiving Bryan cervical disc replacement, and 453 patients receiving anterior cervical discectomy and fusion). Meta-analysis did detect statistical y significant differences in the range of motion of the cervical spine at 3 months and 24 months postoperatively between the two groups, but did not detect statistical y significant differences in visual analog scale score, neck disability index, the Japanese Orthopaedic Association score between the two groups. These findings indicate that Bryan cervical disc replacement is superior to anterior cervical discectomy and fusion in the range of motion of the cervical spine. However, the current literature offers no evidence to support superiority of the Bryan cervical disc replacement over the anterior cervical discectomy and fusion.
5.Effects of hyperbaric oxygen on subgingival obligately,facultatively anaerobes and black pigmented bacteroides in periodontics pocket

Objective: To investigate the effects of hyperbaric oxygen(HB0_2) on black pigmented bacteroides groups(BPB), obligately and facultatively anaerobes of periodontitis. Methods: 60 periodontitis cases were divided into 2 groups. The HB0_2 group(gingival scaling was done at the first visit)was exposed in 0.25 MPa pure oxygen 90 minutes each day, and treated 10 days continuously. The control group received gingival scaling at the first visit. The clinical indices of each group were measured. The microorganisms in periodontal pocket were incubated and analyzed. The number of subgingival anaerobes as well as the number of BPB,obligately and facultatively anaerobes were measured and counted by routine anaerobic culture. Results: Highly significant differences in Gingival Indices (GI), Probing Depth (PD), Attachment Loss (AL), and also significant differences in Plaque Index (PLI),and teeth mobility were seen between the HB0_2 group and the control group. There were significant difference of PLI, GI,PD,AL, teeth mobility in HB0_2 group pre-therapy and post-therapy. There were significant difference of subgingival anaerobes amounts, obligately and facultatively anaerobes sorts between pre-therapy and post-therapy. The positive rate of BPB of group HB0_2 post-therapy were reduced more than that of pre-therapy(P<0.01). Conclusion: HB0_2 has beneficial therapeutic effects on chronic and aggressive periodontitis. The mechanism of the HB0_2 therapy might inhibit the growth of subgingival obligately and facultatively anaerobes and BPB of human periodontitis, especially the obligately anaerobes and BPB in periodontal pockets.
6.Construction and function of the recombinant vector expressing human glutamic acid decarboxylase 65

Objective To construct the recombinant rAAV2-hGAD65 vector and detect its function both in vitro and in vivo. Methods The cDNA of human glutamic acid decarboxylase 2 (hGAD65) gene, which was one of gamma-aminobutyric acid (GABA) synthetase, cloned by the method of RT-PCR, was subcloned into the adeno-associated virus (AAV) vector and formed the recombinant vector of AAV-hGAD65 (rAAV2-hGAD65). The recombinant vector was packaged by the AAV Helper-Free System and its titer was detected. The primary fibroblast, cultured from the rat lung, was transfected by the rAAV2-hGAD65. The expression of the hGAD65 in fibroblast was detected by immunohistochemical method and the level of GABA in the media was assayed by high performance liquid chromatograph (HPLC). In vivo, the hGAD65 was detected by immunohistochemical method in STN and the concentration of the GABA in the reticular part of substantia nigra (SNr) was assayed by HPLC after the rAAV2-hGAD65 was injected into the subthalamic nucleus (STN) by the stereotaxic method. Results The sequence of hGAD65 cDNA was in accordance with that in the Genebank. The amino acid sequence of hGAD65 had no mutation and the titer of rAAV2-hGAD65 was reached 4.5 ×10~(11) per milliliter. The efficiency of infection to the rat primary firoblasts was 80%, and the concentration of GABA in the media of fibroblasts was (45.66±6.07)nmol/L per liter. In vivo, hGAD65 could be detected in STN, and the concentrateion of the GABA in the SNr was increased from (5.66±1.07)nmol/g to (12.66±2.59)nmol/g.Conclusion The cDNA of hGAD65 was cloned by RT-PCR and the recombinant vector of rAAV2-hGAD65 was constructed. The AAV can infect the primary fibroblast in vitro and the hGAD65 can catalyse the glutamic acic to GABA. In vivo, the concentration of GABA in the SNr was heighten after the rAAV2-hGAD65 was injected into the STN.
7.Correlation of telomerase reverse transcriptional gene expression with astrocyte activation

Objective To investigate the relationship between telomerase reverse transcriptase (TERT) gene expression and astrocyte activation.Methods Twenty neonatal 3-day-old male SD rats were used for culture of the astrocytes.The astrocytes were divided into Group A (activated,non-transfected astrocytes),Group B (activated,transfected astrocytes),Group C (unactivated astrocytes) and Group D (activated,empty plasmid transfected astrocytes) according to the random number table,with 5 rats per group.The cell proliferation rate in each group was detected by cell counting kit-8 (CCK-8) ;TERT expression by immunocytochemical method; expressions of TERT and glial fibrillary acidic protein (GFAP) genes by RT-PCR assay.Results Astrocytic proliferation ability in Group B lowered significantly as compared with that in Groups A,D and C (F =43.418,P < 0.01).Expressions of TERT and GFAP mRNAs in Groups A and D were significantly higher than those in Group B and C,and no significant difference was found between Groups A and D.Besides,there was a linear correlation between mRNAs expressions of both genes in Groups A and D (r =0.701,0.704,P < 0.01),while no significant linear correlation was observed in Groups B and C (r =0.260,P > 0.05).Expressions of TERT and GFAP proteins in Groups A and D were markedly higher than those in Groups B and C and no significant difference was found between Groups A and D.Conclusion TERT genes are involved in the activation of astrocytes and exert effect on promoting the activation of astrocytes.
8.Arthroscopic debridement and meniscectomy in treatment of meniscus injury combined with osteoarthritis

Objective To investigate the effect of arthroscopic debridement and meniscectomy in treatment of moderate or severe meniscus injury combined with knee osteoarthritis in early or middle stage. Methods 156 cases diagnosed with moderate or severe meniscus injury combined with knee osteoarthritis in early or middle stage were collected from October 2011 to October 2014. Lysholm knee score and preoperative examinations such as anteroposterior, lateral, axial radiographs, the standing full leg length X-ray film and MRI scan of the knee were recommended to definitively understand the osteoarthritis staging and meniscus injury grading. All patients were treated with arthroscopic debridement and meniscectomy. After operation, physical rehabilitation exercises and regular clinical follow-up were carried out as planned. The Lysholm knee score data from preoperation and terminal follow-up was statistical analyzed. Results No patient experienced any perioperative and postoperative complications. Statistical analysis showed that the Lysholm knee score of postoperation was significantly higher than that of preoperation [(87.3 ± 7.9) vs (67.5 ± 4.9), P < 0.05). Conclusion Arthroscopic debridement and meniscectomy in treatment of moderate or severe meniscus injury combined with knee osteoarthritis in early or middle stage, gains beneficial effects for its minimal invasion and quick recovery.
9.Molecular genetic analysis of four cases with weak D variant of Rh blood type.

OBJECTIVETo explore the molecular basis of 4 cases with weak D variant of Rh blood type.
METHODSRoutine serological testing was applied to determine the D, C, c, E and e antigens of the Rh blood group. The D antigen was further detected with an indirect antiglobulin test. RHD zygosity was detected by sequence-specific primer PCR method. All exons and flanking intron regions of the RHD gene were sequenced.
RESULTSThe samples were determined as weak D phenotype by serological testing. DNA sequencing showed that the 4 cases were heterozygous for 17C>T mutation in exon 1, 29G>C mutation in exon 1, 1212C>A mutation in exon 9, and IVS4+5G>A mutation in intron 4 of the RHD gene, respectively. According to the rule of Rhesus Base Nomenclature, the 4 samples were respectively named as weak D type 31, weak D type 71, weak D type 72, and weak D type 82.
CONCLUSIONSerological and molecular testing for the weak D can facilitate in-depth understanding of its immunology and genetics, and provide guidance for clinical blood transfusion and prevention of hemolytic disease in newborns.

10.Infection characters of Actinobacillus actinomycetemcomitans and Porphyromonas gingivalis in immunodeficient guinea pigs.

OBJECTIVEThe aim of this study was to investigate effects of immunodeficiency on the periodontal infection characters of the specific pathogens of juvenile periodontitis.
METHODSA total of 36 immunodeficient guinea pigs produced by twice whole-body irradiation with 60Co were divided randomly into four groups, in which Actinobacillus actinomycetemcomitans, Porphyromonas gingivalis, and A. actinomycetemcomitans with P. gingivalis were inoculated into the gingival sulcus of two mandibular incisors respectively. The pigs in the control group did not receive any inoculation. At 2, 3 and 6 weeks after inoculation, three animals in each group were sacrificed successively. Clinical and histological examinations were used to examine the changes in the periodontal tissues. The other 36 normal guinea pigs were divided into four groups and treated in a similar way described above.
RESULTSSignificant periodontal damages were noted in immunodeficient pigs inoculated with A. actinomycetemcomitans, P. gingivalis or A. actinomycetemcomitans and P. gingivalis in 2 and 3 weeks after bacterial inoculation. The damages were more severe than in the normal groups. The immunodeficient groups demonstrated larger numbers of osteoclasts than the normal groups (P < 0.05).
CONCLUSIONThe loss of periodontal tissue in immunodeficient hosts is much serious than those with normal defence system, after they are infected with A. actinomycetemcomitans and P. gingivalis. Abnormal defence system in hosts may play an important role in onset and development of juvenile periodontitis.
