1.Research advances on ventilator-associated pneumonia

Ventilator-associated pneumonia (VAP) is the most common hospital acquired infection,which increases the burden of patients significantly.Early diagnosis and effective prevention are important for VAP management.The guidelines for diaguosis,prevention and treatment of ventilator-associated pneumonia was issued by Chinese Society of Critical Care Medicine Chinese Medical Association in 2013.This article reviews the new progress of the diagnosis,treatment and prevention of VAP after the guideline was published.
2.Protective effect of large doses of vitamin C on acute lung injury

Objective To investigate the protective and therapeutic effect of large doses of vitamin C on the acute lung injury in the model of rabbits induced by oleic acid. Methods A total of 18 rabbits were divided into four group randomly: control group(CG); oleic acid group(OG); preventive group(PG) and treatment group(TG). Oxygen pressure in arterial blood (PaO_(2)), superoxide dismutase (SOD) and malondialdehyde(MDA) in blood were measured in 0, 30, 90 and 150min, respectively. All the animals were killed in 150min later. Total protein in broncho-alveolar lavage fluid (BALF) and wet dry weight ratio of the right lung were measured, and the left lung sample was treated by formaldehyde for pathological test. 3.Smoking Behavior and Self-Esteem of Students of Private Schools

Objective:To study the relationship between smoking behavior and self-esteem of adolescents.Methods:1307 students of the seventh and eighth grades of 8 private schools completed questionnaires about smoking behavior and self-esteem. Results: More cigarettes smoking, less score of self-esteem. Those had quit smoking had higher scores in self-esteem than those can not quit. Students who chose“will smoke in the future”had lower self-esteem than the students chose“won't smoke in the future”.Conclusion:Smoking behavior is a marker of low self-esteem.

Colon capsule endoscopy( CCE)is a new technique for the diagnosis of colonic lesions,it has many advantages such as high safety,good tolerance,etc. . Bowel preparation is an important procedure in CCE. The quality of bowel preparation is closely related with the accuracy of CCE. This article reviewed the progress in research on bowel preparation for CCE.

6.Cloning of human ScFv against digoxin from a semi-synthetic phage antibody library

Objective:To clone human anti digoxin ScFv from a human semi synthetic phage antibody library.Methods:①Digoxin BSA conjugate(Dig BSA) was preparaed by a modified periodate oxidation method.②A semi synthetic phage antibody library was panned against immobilized Dig BSA.Collected clones were analysed by ELISA,inhibition ELISA and DNA Sequencing.Results:①During the four rounds panning against Dig BSA,the enrichment of the eluted phage particles was observed;②Analysis of the eluted clones identified one clone that could bind Dig as well as other digitalis.③Sequencing analysis showed that the variable genes of the positive clone belonged to VH5 and V?1 subgroup respectively.Conclusion:Human Dig specific ScFv that could bind Dig and other digitalis has been cloned from a semi synthetic phage antibody library which may provide a potential reagent for the diagnosis and therapy of Dig toxication.
7.Effect of sodium butyrate combined with TRAIL on biological behaviors of lung cancer stem cells

BACKGROUND:Sodium butyrate, a histone deacetylase inhibitor, can inhibit cell proliferation, and induce apoptosis and differentiation of various cancer cells. However, the role of sodium butyrate combined with tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) on lung cancer stem cells remains unclear. OBJECTIVE:To explore the effect of sodium butyrate combined with TRAIL on biological behaviors of lung cancer stem cells. METHODS:Magnetic bead separation was used to separate lung cancer stem cells (CD133+) from human lung adenocarcinoma A549 cells. After the lung cancer stem cells were treated with simple DMEM/F12, DMEM/F12 containing sodium butyrate (5 mmol/L), TRAIL (50 μg/L) or sodium butyrate combined with TRAIL, the cell proliferation within 96 hours of culture was determined by MTT assay; the apoptosis within 24 hours of culture was measured by flow cytometry; the cell migration within 48 hours of culture was detected by cell scratch test; the expression levels of pluripotent transcription factors (Oct4, Sox2 and Nanog) within 48 hours of culture were detected using western blot analysis. RESULTS AND CONCLUSION:The CD133+ lung cancer stem cells were successfully enriched from human lung adenocarcinoma A549 cells. MTT assay showed that sodium butyrate and TRAIL significantly inhibited the proliferation of lung cancer stem cells (P< 0.05), and the combination effect was even stronger (P < 0.05). Results from flow cytometry analysis and scratch test showed that sodium butyrate or TRAIL induced apoptosis and inhibited cell migration of lung cancer stem cells (P < 0.05), and the combination of sodium butyrate and TRAIL showed a stronger effect (P < 0.05). In addition, the expression levels of Oct4, Sox2 and Nanog were significantly down-regulated by sodium butyrate (P < 0.05), TRAIL or sodium butyrate combined with TRAIL, and the combination effect was stronger (P < 0.05). In conclusion, sodium butyrate and TRAIL have synergistic effects on lung cancer stem cells, indicating a new way for treatment of lung cancer.
8.Neural stem cell transplantation and cerebral infarction

BACKGROUND:Neural stem cells transplanted into the infarct region can promote the recovery of damaged nerve cells after cerebral infarction.
OBJECTIVE:To analyze the relevant influential factors of neural stem celltransplantation in the treatment of cerebral infarction.
METHODS:In this paper, we analyzed the experimental studies concerning neural stem celltransplantation for cerebral infarction published in recent years based on database search. There were main two aspects for discussing the progress in neural stem cells and cerebral infarction, the Chinese clinical trial registration and basic experimental studies.
RESULTS AND CONCLUSION:After cerebral infarction, neural stem cellproliferation and differentiation is closely related to the brain microenvironment. Large amount of nerve cellloss can be found in the infarct region. Cytokines can play a role in the neural stem celltransplantation to repair neurological injury after cerebral infarction, and also can induce neural stem cellproliferation, differentiation and migration, including epidermal growth factor, brain-derived neurotrophic factor, insulin-like growth factor 1, nerve growth factor, and basic fibroblast growth factor. Acupuncture and traditional Chinese medicine can promote the proliferation, migration and differentiation neural stem cells in the subependymal zone after cerebral infarction. Neural stem celltransplantation in the treatment of cerebral infarction has yielded progress, but there are stil many issues that need to be resolved in the future.
9.Clinical Application and Assessment of Chinese Version of Checklist Individual Strength Questionnaire in Stroke Patients

Objective To introduce Checklist Individual Strength(CIS)into China,and evaluate its reliability and validity.Methods CIS was translated into Chinese language and the reverse translation was done by several experts.Validity and reliability were implemented in 214 cases with cerebral infarction.Results 4 components were extracted in factor analysis,and the total cumulative contribution was 78.984%.By correlation analysis,the twenty items were divided into 4 domains:subjective feeling of fatigue,concentration,motivation and physical activity,it was same as in the English version.Cronbach α of four domains were 0.9256,0.9072,0.7598,and 0.9157,respectively.Conclusion Psychometric properties(reliabilities and validities)of CIS Chinese version is met with satisfaction and seems to be adaptable to Chinese cerebral infarction patients.
10.Evaluation of Chinese Version of 3 Fatigue Questionnaires for Stroke Patients

Objective To analyze the instruments for post stroke fatigue, Fatigue Severity Scale (FSS),Checklist Individual Strength (CIS) and Fatigue Impact Scale (FIS). Methods The FSS,CIS and FIS were applied in 214 cases with cerebral infarction. They were combined together at the (sub)scale level, and evaluated with Mokken Scale Analysis for Polytomous Items, for validity and reliability. Results and Conclusion Three scales were and The combined scale proved to be acceptable (H>0.5). 3 components were extracted in factor analysis, and the their total cumulative contribution was 77.504%, with reliability (α) of 0.8097,0.7094 and 0.8019 respectively. FSS put emphasis on the fatigue experiences of stroke patients, CIS on the influence of fatigue on, and FIS on the social function.
