1.Expression of p120-catenin in different bladder cancer cells and effect of p120-catenin to the prolifer-ation, invasion and adhesion of human bladder carcinoma cell line BIU-87 in vitro

Objective To investigate the expression of p 120-catenin in human bladder cell line BIU-87 and T24.And to explore the effect of down-regulation of p120-catenin on proliferation, invasion and adhesion of BIU-87 cell line. Methods The study was from Aug .2010 to May.2011.Real-time RT-PCR and Western blot were used to examine the expression of p 120-catenin in human bladder cell line BIU-87 and T24.There were three study groups:non-transfected group , the group transfected with p 120-catenin siRNA and the group transfected with control siRNA .p120-catenin siRNA was used to decrease the expression of p120-catenin.MTT assay and colony formation assay were used to study the BIU-87 cell growth and cell pro-liferation.The invasion of BIU-87 cells was measured by Transwell chamber assay .Cell adhesion artificial re-constituted basement membrane assay was used to examine the change of BIU-87 cell adhesion capacity . Results In both BIU-87 and T24 cells there were the expressions of p 120-catenin.But the expression in BIU-87 cells (0.11±0.39) was more than that in T24 cells (0.48±0.17).The decreased of p120-catenin ex-pression could enhance the proliferation (182.7±13.4%) and the invasiveness (217.5±15.9) and decrease the adhesion capacity (57.3±6.4%) of BIU-87 cells. Conclusions There is higher expression level of p120-catenin in lower-grade malignant bladder cancer cells .The down-regulation of p120-catenin promotes the bladder cancer proliferation and invasiveness of bladder carcinoma cells , and inhibited the bladder canc-er cell adhesion .
2.Effect of the down regulation of p120-catenin on the proliferation and invasion of human bladder carcinoma cell line T24 in Vitro

Objective To investigate the regulating function of p120-catenin (p120ctn) on proliferation and invasion of human bladder cancer cell T24.Methods Special siRNA was used to silence the expression of p120ctn in T24 cells.MTT assay was used to examine the T24 cell growth rate.The invasion of T24 cells and the control cells were measured by transwell chamber assay.Results The silencing of p120ctn could improve the proliferation and the invasiveness of T24 tumor cells.Transwell chamber assay showed that the in vitro invasion function of T24 cells was significant increase after p120ctn-siRNA.Conclusion p120ctn inhibited the bladder cancer proliferation and invasiveness of bladder carcinoma cells.
3.Management of superficial bladder tumor by greenlight photoselective vapontion per urethra

Objective To explore the management,curative effect and safe of superficial bladder tumor(BT)by greenlight photoselective vapontion per urethra(GPVPU). Methods 32 patients of BT were treated by GPVPU,and operation time,postoperative complications,relapse rate and so on were observed. Results All cases were completed successfully. No complication happened in operations. Urinary canal was detained 2 ~4 days and bladder washout(BW) did not need after operation. Follow-up average time was 16 months. Irrigation chems of bladder and cystoscopy were applied routinely. Conclusion GPVPU was safe and effective in treatment of superficial bladder tumor.
4.Management of benign prostatic hyperplasia by photoselective vaporization of prostate

Objective To explore the management of benign prostatic hyperplasia(BPH)by photoselective vaporization of prostate(PVP)and curative effect in the near future. Methods Clinical data of 60 BPH patients underwent PVP were retrospctively aralyzed.The changes of IPSS and QOL and the max urine flow rate and residual urine volume(RUV)perioperatively were compared through observations of the operation time and complication. Results The therapeutic efficacy was favourable.No transurethral resection of prostate(TURP)syndrome and uroclepsia happens.IPSS(t=2.471,P <0.05)and QOL(t=2.392,P <0.05)and the max urine flow rate(t=2.287,P <0.05)and residual urine volume(t=2.513,P<0.05)obviously improved(P <0.05). Conclusion PVP had many advantages such as operated simply and no serious complications.It was a good operation method to treat BPH.
5.Genetic incorporation of unnatural amino acids into proteins and its translational application in biomedicine

Proteins in the human body are usually made of 20 natural amino acids.Through different amino acid combinations and isomerization, proteins of diverse functions are built.An emerging genetic code expansion technology can introduce unnatural amino acids into specific sites of target protein, endowing the protein with new biological characteristics including covalently binding with proximal proteins, carrying fluorescence, and mimicking specific protein post-translational modifications.In this paper, based on the structure and function of unnatural amino acids, the applications of different types of unnatural amino acids in regulating protein''s stability, studying protein''s conformation, expression level, and localization, and uncovering heretofore unknown protein-protein interactions were reviewed.Besides, genetic code expansion of unnatural amino acids is anticipated to find broad utilities in biomedicine by bringing new ideas and methods to the design and optimization of biologics.
6.Effect of interferon on HEL cell apoptosis and JAK2 V617F mutation gene expression

Objective To observe the effect of Interferon-α2b on HEL cells (human erythroleukemia cell line) growth, apoptosis and JAK2 V617F mutation gene expression. Methods HEL cells were placed in RPMI1640 containing 10% FBS and incubated in a cell incubator. Cells in the logarithmic growth phasem were collected, adjusting the cell density to 1 × 105/mL for experimental research. The interferon concentration in five groups were 0, 5 × 105, 10 × 105, 50 × 105, 100 × 105 U/L, with different incubation time (0, 24, 72, 120 h), respectively. The cell growth status in different groups was observed in the inverted optical microscope; MTT was used to detect the inhibition of interferon on HEL cell proliferation. Cell apoptosis was detected by flow cytometry. Fluorescence quantitative PCR was used to detect the mutation gene of JAK2 V617F expression. Results Inhibition rates of Interferon on the HEL cell proliferation in 5 × 105 U/L, 10 × 105 U/L, 50 × 105 U/L, 100 × 105 U/L groups were 18.57%, 25.10%, 42.10%, 57.00%, respectively. JAK2 V617F/GAPDH by fluorescence quantitative was 1.556, 1.213, 0.870 respectively under the concentration of interferon 100 × 105 U/L for 24, 72, 120 h. Conclusions Interferon-α2b can inhibit HEL cells proliferation and induce HEL cells apoptosis. Increasing concentration of interferon increases HEL cell apoptosis rate. Interferon can inhibit JAK2 V617F expression of HEL cells in a dose-dependent manner.
7.Study on the correlation between mental health status and social support and coping style in patients with occupational diseases

Objective To explore the related factors on the mental health and the correlation between mental health status and social support and coping style in patients with occupational diseases in order to provide the scientific basis for making appropriate countermeasures to system intervention .Methods Eighty-seven patients with occupation diseases in our hospital were surveyed by the symptom checklist 90 ( SCL-90 ) , social support rating scale (SSRS), coping style questionnaire (CSQ).Results The mental health status of patients with occupational diseases was poor , and the scores of all factors on SCL-90 in patients were higher than the national norm, and the difference was statistically significant (P<0.01).The scores of all factors on social support in patients were significantly lower than the national norm , and the difference was statistically significant (P<0.01).The scores of total SSRS, the factors on objective support and support utilization had negative correlation with the scores of total SCL-90 and the part factors in patients (P<0.05).The scores of the part factors on SCL-90 had positive or negative correlation with the scores of the part factors on CSQ (P<0.05), but had not relation with the mature or immature coping style in patients .The scores of problem solving and mature coping style had positive correlation with the scores of subjective support and the total SSRS in patients ( P<0.05).Conclusions The patients with occupation diseases should be given more social support and care in clinical practice , and we should help them adjust mental status and improve the coping style and maintenance of physical and mental health .
8.Causal relationship between thyroid dysfunction and sepsis: a bidirectional two-sample Mendelian randomization

Objective:To explore the causal relationship between thyroid dysfunction and sepsis based on the bidirectional two-sample Mendelian randomization (MR) method.Methods:The genome-wide association study (GWAS) dataset were selected to screen single nucleotide polymorphisms (SNP) associated with thyroid dysfunction as instrumental variable (IV) for genetic variation, using hypothyroidism and hyperthyroidism as exposure factor and sepsis as outcome factor. Potential causal relationship between thyroid dysfunction and sepsis was analyzed using a bidirectional two-sample MR method primary analysis method of inverse-variance weighted (IVW). Potential pleiotropic analysis of SNP was performed using the MR Egger regression intercept test. Sensitivity analysis was performed using the "leave one out" test. Reverse MR method was used to prove the causal relationship.Results:The GWAS data were screened based on the three main assumptions of MR, resulting in 101 SNP strongly associated with hypothyroidism and 10 SNP strongly associated with hyperthyroidism entering the MR analysis. The results of the MR using the IVW method showed that the risk of sepsis in individuals with hypothyroidism was 2.293 times higher than those without hypothyroidism [odds ratio ( OR) = 2.293, 95% confidence interval (95% CI) was 1.199-4.382, P = 0.012]. There was no significant difference in the risk of sepsis between hyperthyroid and non-hyperthyroid populations ( OR = 1.049, 95% CI was 0.999-1.100, P = 0.560). MR Egger regression intercept test showed that the included SNP did not have pleiotropy, and the MR-PRESSO test did not find outliers. Sensitivity analysis suggested that the results of MR were stable. The results of the reverse MR analysis showed that the reverse causal relationship between hyperthyroidism and sepsis was not proved ( OR = 0.996, 95% CI was 0.988-1.004, P = 0.338), which further confirmed the robust MR analysis result. Conclusion:The results of the bidirectional two-sample MR analysis show that hypothyroidism can increase the risk of sepsis onset, while there is no causal relationship between hyperthyroidism and sepsis.
9.Dermoscopic features of onychomycosis and nail psoriasis

Objective To investigate dermoscopic features of onychomycosis and nail psoriasis.Methods From March 2017 to March 2018,128 patients with onychomycosis and 128 with nail psoriasis were enrolled from Department of Dermatology,Xiangyang Central Hospital.Dermoscopic images of the lesional nails were compared.Enumeration data were analyzed by chi-square test and Fisher's exact test.Results There were 7 kinds of dermoscopic patterns in onychomycosis,including jagged edge (69/128,53.91%),longitudinal striae (72/128,56.25%),cone-shaped keratosis (30/128,23.44%),nail plate thickening (21/128,16.41%),black dots (5/128,3.91%),onycholysis (26/128,20.31%),and nail discoloration (23/128,17.97%).The jagged edge and longitudinal striae occurred more frequently in distal/ lateral subungual onychomycosis (DLSO) than in the other 3 clinical subtypes of onychomycosis (all P < 0.001),and cone-shaped keratosis occurred more frequently in total dystrophic onychomycosis (TDO) than in the other 3 clinical subtypes of onychomycosis (x2 =42.020,P < 0.001).There were no significant differences in the prevalance of nail plate thickening,black dots,onycholysis and nail discoloration among the 4 clinical subtypes of onychomycosis (all P > 0.05).There were 7 kinds of dermoscopic patterns in nail psoriasis,including pitting corrosion (61/128,47.66%),oil drop sign (41/128,32.03%),splinter hemorrhage (41/128,32.03%),dilated hyponychial capillaries (23/128,17.97%),nail plate thickening (20/128,15.63%),onycholysis (20/128,15.63%),and nail discoloration (13/128,10.16%).The oil drop sign occurred more frequently in psoriasis vulgaris and psoriasis arthropathica than in the other 2 clinical subtypes of psoriasis (P =0.019).The dilated hyponychial capillaries occurred more frequently in pustular psoriasis than in the other 3 clinical subtypes of psoriasis (P =0.047).There were no significant differences in the prevalance of pitting corrosion,splinter haemorrhage,nail plate thickening and nail discoloration among the 4 clinical subtypes of psoriasis (all P > 0.05).Conclusions The dermoscopic features of onychomycosis include jagged edge,longitudinal striae,cone-shaped keratosis and black dots,and the dermoscopic features of nail psoriasis include pitting corrosion,oil drop sign,splinter haemorrhage and dilated hyponychial capillaries.Their common dermoscopic features include nail plate thickening,onycholysis and nail discoloration.
10.Oxidative stress injury and transcriptome analysis of human myocardial AC16 cells induced by zinc oxide nanoparticles

Objective:
To investigate the oxidative stress injury of nano zinc oxide nanoparticles(ZnO NPs) on human myocardial cells (AC16) ,and to analyze the mechanism of ZnO NPs from the transcriptome level.
Methods:
Dynamic light scattering (DLS) was used to characterize and detect ZnO NPs.After AC16 cells were exposed to ZnO NPs at different doses and at different times,the cell survival rate was determined by CCK-8 method.AC16 cells were divided into control group,ZnO NPs (50,100,200 μmol /L) ,after 6 h treatment,the mitochondrial membrane potential (MMP) and reactive oxygen species (ROS) were measured.AC16 cells were divided into control group,50 μmol /L ZnO NPs group and 200 μmol /L ZnO NPs group.After 6 h exposure,total RNA was extracted by TRIzol for transcriptome analysis ,and the differentially expressed genes were enriched by gene body ( GO) ,Kyoto Encyclopedia of Genes and Genomes (KEGG) .
Results :
The results of DLS showed that the hydrodynamic diameter was ( 192. 2 ± 1. 63 ) nm and the Zeta potential was ( -23. 26 ± 1. 05 ) mV. CCK-8 results showed that the survival rate of AC16 cells decreased with the increase of dose and time of exposure to ZnO NPs. Fluorescence quantification showed that with the increase of ZnO NPs exposure dose,MMP significantly decreased at 100 μmol /L ZnO NPs(P<0. 05) ,and ROS significantly increased at 50 μmol /L ZnO NPs(P<0. 05) .Using the multifunctional microplate reader,it was observed that MMP and ROS were statistically significant at 100 and 50 μmol /L ZnO NPs,respectively,showing a decrease in MMP and an increase in ROS.Transcriptome analysis showed that 1 071 genes were enriched in the 50 μmol /L ZnO NPs group compared with the control group,including 561 up-regulated genes and 510 down-regulated genes.Compared with the control group,7 164 genes were enriched in 200 μmol /L ZnO NPs group,including 4 098 up-regulated genes and 3 066 down-regulated genes.GO and KEGG analysis showed that the differential genes were mainly concentrated in ROS,antioxidant activity,mitochondrial cytochrome C release,apoptosis and other signaling pathways.
Conclusion
ZnO NPs can decrease the survival rate of AC16 cells and induce mitochondrial damage and oxidative stress,among which ROS-mediated oxi- dative stress and mitochondrial function changes are important toxic mechanisms of ZnO NPs induced AC16 cytotoxicity.
