1.Advancement and the value of targeted gene related to lung cancer
Journal of International Oncology 2013;40(7):512-515
The occurrence and development of lung cancer are closely related to gene expression and mutations.Recent studies of lung cancer related genes show that the expression of COPS3 gene may play a key role in the lung cancer cell proliferation.Echinoderm microtubule associated protein like 4-anaplastic lymphoma kinase fusion gene and epidermal growth factor receptor mutations coexist.GA733 gene is closely related to lung cancer survival.Fibroblast growth factor receptor 1,high mobility protein family, members of the scavenger receptor class 5 and epidermal growth factor receptor provide new ideas for targeted therapy of lung cancer. For the therapy of lung cancer, multiple gene therapy and gene therapy combined with conventional treatment is required.

Angiopoietin is a novel angiogenesis related molecule. To investigate its roles in tumor angiogenesis and metastasis, angiopoietin 1 gene was obtained from human placental tissue by reverse transcription polymerase chain reaction (RT PCR). The sense, antisense eukaryotic expression vector pcDNA3.1/hangiopoietin1(+), antisense eakaryotic expression vector pcDNA3.1/hangiopoietin1(-), and prokaryotic expression vector pRSETC hangiopoietin1 were successfully constructed by directional cloning the target gene into pcDNA3.1 V5 His C and pRSET C vectors, respectively. This lays good foundation for further study on the roles of angiopoietin 1 in angiogenesis in tumors of the digestive tract.

Background Tear meniscus gained by OCT device is essential to the dry eye diagnosis.Currently,RTVue Fourier-domain optical coherence tomography (FD-OCT) is commercially available to image the tear menisci. However, there were few studies to report the repeatability and reproducibility of lower and upper tear meniscus parameters measured by RTVue FD-OCT.Objective The purpose of this study was to assess the repeatability and reproducibility of lower and upper tear meniscus parameters measured by RTVue FD-OCT in normal and dry eyes. Methods In this prospective study,20 dry eyes of 20 patients were finally recruited, and 20 normal eyes of 20 age-and gender-matched healthy subjects were included during the same period.Successive three-time scans were performed at a 3-minute interval with RTVue FD-OCT in optional eye by one well-trained examiner, and the other operator performed the same measurement 10-15 minutes later.Six parameters of tear meniscus were yielded by custom software, including upper meniscus curvature (UMC),upper meniscus height(UMH),upper meniscus area (UMA),lower meniscus curvature (LMC),lower meniscus height (LMH) and lower meniscus area (LMA).The between-image repeatability was assessed using mean coefficient of repeatability (MCOR),while between-operator reproducibility were evaluated using coefficient of repeatability (COR) and interclass correlation of coefficients (ICC).Results As for the between-image repeatability, the results of MCOR for UMH,UMA,LMH and LMA were 5.11%,3.58%,4.98% and 4.82% respectively in the normal eyes and 5.60%,5.80%,5.03% and 5.74% respectively in the dry eyes.As for the between-operator reproducibility, the results of COR and ICC for UMA were 5.38% and O.975,the COR and ICC for LMA were 5.28% and 0.961 respectively in the normal eyes, and the results of COR and ICC for LMA were 9.69% and 0.964 in the dry eyes. Conclusions The UMA,LMH and LMA tear meniscus parameters obtained by RTvue FD-OCT show good between-image repeatability, and LMA also presents good between-operator reproducibility.Measurement of tear meniscus parameters with RTVue FD-OCT measuring is a potential diagnostic index for the diagnosis of dry eye.

Journal of Biomedical Engineering 2014;31(1):210-213
The use of actigraphy, which can be used to estimate sleep-wake patterns from activity levels, has become common in sleep research. Actigraphy is a simple, cost-effective and non-invasive method for healthcare providers and researchers to assess patients sleep quality and screen for potential sleep disorders in recent years. But, there is no wide recognition and application of actigraphy in China up till now. This review summarized the application of actigraphy in evaluation of sleep and diagnosis of sleep disorders.

Objective To analyze the role of cysteinyl aspartate specific proteinase-1 (caspase-1) in a mouse model of D-galactosamine (D-GalN) and lipopolysaccharide (LPS) induced acute liver failure (ALF) and to study the possible mechanism. Methods C57BL/ 6 mice were randomly divided into four groups including control group, Z-WEHD-FMK (caspase-1 inhibitor) treatment group, ALF model group and Z-WEHD-FMK-treated ALF group. The mouse model of ALF was established by intraperitoneally injec-ting the mice with D-GalN (450 mg/ kg) and LPS (10 μg/ kg). The damages in liver tissues were evaluated based on the histopathological examination and the levels of alanine transaminase (ALT) and aspartate trans-aminase (AST) in serum samples. Western blot assay was performed to analyze the expression of caspase-1 and the phosphorylation of glycogen synthase kinase 3β (GSK-3β). The qRT-PCR was used to measure the expression of inflammatory cytokines at transcriptional level. Results The expression of caspase-1 at both mRNA and protein levels were gradually increased during the development of ALF. Compared with the mice with ALF, those in the Z-WEHD-FMK-treated ALF group showed less severe liver damages on histopatholog-ical examination and decreased levels of ALT and AST in serum samples [ALT: (479. 2±39. 5) U/ L vs (998. 5±60. 4 ) U/ L, P<0. 05; AST: ( 478. 5±28. 6) U/ L vs ( 1 180. 7±91. 4) U/ L, P<0. 05]. The expression of TNF-α, IL-1β, IL-18 and IL-33 at transcriptional level were significantly suppressed in mice with ALF upon the Z-WEHD-FMK intervention. Results of the Western blot assay indicated that Z-WEHD-FMK suppressed the activities of GSK-3β by enhancing its phosphorylation. Conclusion This study demon-strated that caspase-1 could promote the activation of GSK-3β resulting in the development of inflammation responses and liver damages during the development of ALF in mice.
