1.Advancement and the value of targeted gene related to lung cancer

The occurrence and development of lung cancer are closely related to gene expression and mutations.Recent studies of lung cancer related genes show that the expression of COPS3 gene may play a key role in the lung cancer cell proliferation.Echinoderm microtubule associated protein like 4-anaplastic lymphoma kinase fusion gene and epidermal growth factor receptor mutations coexist.GA733 gene is closely related to lung cancer survival.Fibroblast growth factor receptor 1,high mobility protein family,members of the scavenger receptor class 5 and epidermal growth factor receptor provide new ideas for targeted therapy of lung cancer.For the therapy of lung cancer,multiple gene therapy and gene therapy combined with conventional treatment is required.
2.CLONING OF HUMAN ANGIOPOIETIN-1 GENE AND CONSTRUCTION OF ITS EUKARYOTIC AND PROKARYOTIC EXPRESSION VECTORS

Angiopoietin is a novel angiogenesis related molecule. To investigate its roles in tumor angiogenesis and metastasis,angiopoietin 1 gene was obtained from human placental tissue by reverse transcription polymerase chain reaction (RT PCR). The sense, antisense eukaryotic expression vector pcDNA3.1/hangiopoietin1(+), antisense eakaryotic expression vector pcDNA3.1/hangiopoietin1(-), and prokaryotic expression vector pRSETC hangiopoietin1 were successfully constructed by directional cloning the target gene into pcDNA3.1 V5 His C and pRSET C vectors, respectively. This lays good foundation for further study on the roles of angiopoietin 1 in angiogenesis in tumors of the digestive tract.
3.Clinical Observation of Yangyin Tongluo Pills Combined with Acupuncture for Treatment of Cerebral Infarction Sequela

Objective To observe the clinical effect of Yangyin Tongluo Pills combined with acupuncture for the treatment of cerebral infarction sequela. Methods One hundred patients with cerebral infarction sequela were evenly randomized into treatment group and control group. The control group was given acupuncture therapy combined with oral use of clopidogrel, and the treatment group was given acupuncture therapy combined with oral use of Yangyin Tongluo Pills (mainly composed of Fructus Lycii, Flos Chrysanthemi, Radix Rehmanniae, Cortex Moutan, Radix Salviae Miltiorrhizae, Flos Carthami, Semen Persicae, Pheretima, Rhizoma Gastrodiae, Ramulus Uncariae cum Uncis, Spica Prunellae, Radix Glehniae, Radix Ophiopogonis, Fructus Schisandrae Chinensis, Radix Pseudostellariae) . One month constituted a treatment course, and the treatment covered 2 courses. After treatment, the therapeutic effect of the two groups was evaluated, and the changes of National Institute of Health Stroke Scale ( NIHSS) scores and Barthel Index Scale for activities of daily living ( ADL) , and hemorrheology indexes were observed before and after treatment. Results (1) After treatment for 2 courses, NIHSS scores were decreased, ADL scores were increased in the two groups (P<0.01 compared with those before treatment) , and the improvement of the treatment group was superior to that of the control group ( P<0.01) . ( 2) After treatment, hemorrheology indexes were much improved in the treatment group ( P<0 . 05 or P<0 . 01 ) , but remained unchanged in the control group ( P>0.05) . The treatment group had better effect on improving hemorrheology indexes than the control group (P<0.05) . (3) The total effective rate of the treatment group was 90.00% and that of the control group was 64.00%, the overall therapeutic effect of the treatment group being superior to that of the control group (P<0.05) . Conclusion Yangyin Tongluo Pills combined with acupuncture are effective for the treatment of cerebral infarction sequela, and the effect is superior to that of acupuncture combined with clopidogrel.
4.Comparison of HIV-1 DNA in different parts of early antiretroviral treatment patients

ObjectiveTo study HIV-1 DNA levels in different parts of HIV patients during the early stage of antiretroviral therapy.MethodsThe peripheral blood,gut associated lymphoid tissues and lymph nodes samples were collected before and 12 weeks after treatment in regular follow-up HIV-1/AIDS patients in Beijing Youan Hospital ( n =11 ).The average age was 39 years old ( 25 to 55 ).Mononuclear Cells were isolated by density gradient centrifugation and then used DNA extraction kit to extract DNA.Realtime quantitative polymerase chain reaction was used to examine HIV-1 DNA copy-number.Non-parametric test was used to analyse the differences of HIV-1 DNA copy numbers among groups.Results Before treatment,HIV-1 DNA copy-number in both gut associated lymphoid tissues ( 10 714 ± 2043 ) copies/106 cells and lymph nodes (9145 ± 1202) copies/106 cells were higher than that in the peripheral blood (66 ± 8) copies/106 cells ( U =0.00,P <0.05 ),There was no significant difference between lymph nodes and gut associated lymphoid tissues (U =46.00,P >0.05).After 12 weeks of treatment,HIV-1 DNA copy-number in both gut associated lymphoid tissues (1701 ± 790) copies/106 cells and lymph node (11 591 ± 1781 ) copies/106 cells were higher than the peripheral blood ( 18 ± 3 ) copies/106 cells ( Z =- 2.934,P < 0.05 ).There was a significant reduction of DNA copy-number in gut associated lymphoid tissues and peripheral blood after treatment (Z =- 2.934,P < 0.05 ).Conclusion Gut associated lymphoid tissues and lymph nodes may be important latent reservoirs for HIV-1 DNA.
5.Utilization reseach of medical insurance drugs in the hospitals of Suqian district

Objective To analyze the current situation and developing trend of utilizaion of medical insurance drugs.Methods Data were collected from Suqian Hospital.Drug Utilization Analysis System and DDD were taken as the basic unit of measurement.Results The consumption of cost,DDDs and variety of type A medical insurance drugs were rising year by year,but the proportion in total was in declining trend.When analyzed according to DDDc,the type A,non medical insurance drugs and the type B ranked the first,the second and the third place during the 2009-2010,the DDDc of type A was lower than the non medical insurancedrugs and the type B.For example,the DDDc of type A was 3.32 yuan/day,the DDDc of the non medical insurance drugs and the type B was 15.04 and 15.97 yuan/day respectively.Conclusion Analyzed in respect to DDDs,the protortion of type A was in high level and the type B was in low.But analyzed in respect to the comsumption cost,the protortion of type B was in high level and the type A was in low.

8.The role of actigraphy in monitoring sleep and sleep disorders.

The use of actigraphy, which can be used to estimate sleep-wake patterns from activity levels, has become common in sleep research. Actigraphy is a simple, cost-effective and non-invasive method for healthcare providers and researchers to assess patients sleep quality and screen for potential sleep disorders in recent years. But, there is no wide recognition and application of actigraphy in China up till now. This review summarized the application of actigraphy in evaluation of sleep and diagnosis of sleep disorders.

This article was aimed to review and make a commentary on modern medicine and traditional Chinese medicine (TCM) treatment for intracerebral hemorrhage. This article introduced applications and evaluations on medi-cations to reduce intracranial pressure, blood pressure control medications, ultra-early hemostatic medications, hy-pothermia and neuroprotective agents from modern medicine treatment on acute intracerebral hemorrhage. The article also introduced the current large-scale TCM clinical trials for acute intracerebral hemorrhage treatment and some systematic reviews on medications in order to provide theoretical evidences for the clinical treatment of acute intrac-erebral hemorrhage.
10.Activated peroxisome proliferator-activated receptor αinhibiting the lipopolysaccharide-induced macrophage-mediated inflammatory responses by promoting the autophagy

Objective To investigate the effects of peroxisome proliferator-activated receptor α( PPARα) on macrophage-mediated inflammatory responses with the interference of lipopolysaccharide and the possible mechanism.Methods The bone marrow stem cells were isolated from the femora of mice.The granulocyte-macrophage colony stimulating factor ( GM-CSF) was used to stimulate the in vitro differentiation from bone marrow stem cells into primary macrophages.An in vitro model with cultured cells expressing in-flammatory cytokines was established by treating the primary macrophages with lipopolysaccharide ( LPS) .A specific chemical agonist, Wy-14643, was used to activate PPARα. Autophagy inhibitors including 3-methyladenine (3-MA) and small interfering RNA against Atg7 ( Atg7 siRNA) were used to inhibit the autophagy.Western blot assay was performed to detect the expression of autophagy-related proteins ( Atg5, Atg7, Beclin-1 and LC3).The transcriptional levels of TNF-α, IL-1β, IL-6, Atg5, Atg7 and Beclin-1 were analyzed by qRT-PCR.Results Compared with the macrophages treated with LPS alone, those pretreated with various concentrations of Wy-14643 (10 μmol/L, 25 μmol/L and 50 μmol/L) showed inhibited ex-pression of proinflammatory cytokines ( TNF-α,IL-1βand IL-6) and enhanced expression of autophagy-relat-ed proteins (Atg5, Atg7 and Beclin-1) at mRNA level in a dose-dependent manner.The expression of auto-phagy-related proteins (Atg5, Atg7, Beclin-1 and LC3) by macrophages was promoted with the pretreatment of Wy-14643 as indicated by Western blot assay.The transcriptional levels of TNF-α, IL-1βand IL-6 were increased in Wy-14643 pretreated-macrophages after stimulation with 3-MA or Atg7 siRNA .Conclusion PPARαsuppressed the macrophage-mediated inflammatory responses by promoting autophagy, suggesting that the PPARα-autophagy pathway might be one of the signaling pathways regulating LPS induced-inflamma-tory responses.
