1.Application of thoracoscopic technology in elderly patients undergoing lobe resection

Objective To explore the efficacy and clinical value of video-assisted thoracoscopic surgery (VATS) in elderly patients undergoing lung resection surgery.Methods A total of 100 elderly patients with lung cancer in our hospital from January 2009 to March 2012 were randomly divided into VATS group and traditional operation group (n=50,each group).Patients in VATS group were given VATS with 3 minimally invasive cuts.Patients in traditional operation group were treated with traditional thoracotomy.There were 23 cases undergoing upper left lobectomy,24 cases undergoing left lower lobectomy,24 cases with right upper lobectomy,14 cases with right lobectomy,6 cases with right lower lobectomy,3 cases with right pneumonectomy and 6 cases with left pneumonectomy.Results The operative time was shorter in VATS group than in traditional operation group [(109± 15) min vs.(125±22) min,t=4.25,P<0.05].The intraoperative bleeding volume and total drainage volume were less in VATS group than in traditional operation group [(105.0±21.8) ml vs.(121.0±28.2) ml,(225.0±50.4) ml vs.(121.0±28.2) ml,respectively,t=3.17,4.99,both P<0.05].The hospital stay and surgery extubation days were less in VATS group than in traditional operation group [(3.1 ± 0.8) d vs.(3.8 ± 1.1) d,(5.8 ± 1.8) d vs.(8.7 ± 2.4) d,respectively,t =3.64,6.84,both P< 0.05].The incidence of pulmonary infection was lower in VATS group than in traditional operation group (10% vs.28%,x2 =5.26,P<0.05).Conclusions VATS is safe and effective for lobe resection in elderly lung cancer with a high cure rate and fewer complications,which can be widely applied in elderly patients with lung cancer.

OBJECTIVE:To establish an HPLC method for determination of ganciclovir in serum of dogs and to study pharmacokinetics and relative bioavailability of Ganciclovir dispersible tablets.METHODS:The determination was performed on Dikma DiamonsilTM C18 column.The mobile phase consisted of 0.015 mol?mL-1 monobasic potassium phosphate buffer solution (pH 2.50,containing 0.25% acetonitrile) with flow rate of 1.75 mL?min-1 at detection wavelength of 254 nm.The column temperature was set at 40 ℃.In a randomized cross-over study,a single dose of dispersible tablets and the capsule were given to 6 dogs.An HPLC method was used to determine plasma concentration of ganciclovir in dogs and the pharmacokinetic data were treated by DASver1.0 software.RESULTS:The linear range of ganciclovir were 0.050 25~100.5 ?g?mL-1;the methodological recovery was larger than 90% (RSD
3.Clinical effect analysis of hemocoagulase topical spraying during the neurosurgical operation

Objective To explore the clinical effect and influence of hemocoagulase on bleeding control and coagulation function in neu -rosurgery .Methods A total of 300 patients with surgical treatment in our hospital from June 2013 to June 2014 were randomly divided into control group (n=150) and observation group (n=150).The patients in control group were treated with physiological saline 10 mL topical spraying,while the patients in observation group were treated with physiological saline 10 mL plus hemocoagulase 4U topical spraying.The clinical effect and coagulation index were compared between the two groups .Results The bleeding volume of the observation group was sig-nificantly lower than that of the control group (P<0.01).No significant difference was found in the coagulation index of the two groups (P>0.05).Conclusion Hemocoagulase treatment during the operation can reduce the volume of bleeding and transfusion of blood ,and do not affect the coagulation function in the patients .
4.Radiofrequency ablation therapy for breast cancer:case report and reference review

Objective To evaluate the effect of ultrasound guided percutaneous radiofrequent ablation treatment on breast cancer .Methods Two patients,were pathologically confirmed as breast cancer ,underwent percutaneous ultrasound guided radiofrequency ablation of breast lesions ,followed with radical excision of breast within 3 months after the ablation .Postoperative pathological examination was conducted .Patients were followed up for over 12 months by now.Results The postoperative pathology of case No .1,in which the original breast le-sion was within 2 cm in diameter,found no tumor cells left but filled with inflammatory cells instead .Case No.2 with the longer diameter as 27 mm showed majority necrotic tissues but the tissue around left with some ductal carcinoma in-situ components .One-year′s follow up found no local recurrence or distant metastasis of tumor . Conclusion Radiofrequency ablation is a promising therapy with minimally invasion and better outlooking for early breast cancer within 2 cm in diameter .
5.Intracranial aneurysm isolation combined with extra-intracranial bypass in treatment of middle cerebral artery aneurysm

Objective To investigate the effect of intracranial aneurysm isolation combined with extra-intracranial bypass in treatment of middle cerebral artery aneurysm.Methods From January 2013 to March 2015,there were 32 patients with complex middle cerebral artery aneurysm in our hospital,who were treated by intracranial aneurysm isolation combined with extra-intracranial bypass.The clinical symptoms and image examination after surgery were retrospectively analyzed,and the hemodynamic index of middle cerebral artery before and after treat-ment was statistically analyzed.Results The postoperative clinical symptoms of 25 patients were relieved,the mRS score was 0 point.Seven patients received surgery,1 week later,the muscle force of involved side was grade 2 to 3,the mRS score was 2 to 3 point,the blood flowing smoothly in target vessel,with no original aneurysm,no new ischemic stroke or infarct.After intervention,systolic and diastolic blood flow ve-locity were faster than those before the intervention(P <0.05),blood flow resistance were lower than those before intervention(P <0.05). Conclusion The intracranial aneurysm isolation combined with extra-intracranial bypass can effectively improve the clinical symptoms of pa-tients with complex intracranial aneurysms,with high graft patency rate and reliable effect.
6.Effect of RhoA gene silencing on proliferation and migration of HepG2 cells

Objective To construct a RhoA-siRNA expression vector and determine its role on the malig-nant behavior of HepG2 cells.Methods A RhoA-siRNA DNA fragment was synthesized and cloned into the expression vector of pGenesil-1.The constructed Rhon-siRNA DNA plasmid was stably transfected into HerG2 cells by lipofectamine,and then HepG2 cells were divided into the HepG2/RhoA-siRNA group (HepG2 cells were transfected with pGenesil-1-RhoA-siRNA),HepG2/control group(HepG2 cells were transfected with control plasmid) and HepG2 group (without plasmid transfection).The inbibitory effect of RhoA-siRNA on RhoA protein expression was shown by Western blot.The proliferation,migration,growth potentiality and cell cycle of transfected HepG2 cells were evaluated by MTT assay,wounded healing,the plate cloning formation test and flow cytometry,respectively.All data were analyzed by one-way analysis of variance (ANOVA) and chi-square test.Results The expression of RhoA protein in the HepG2/RhoA-siRNA group was,significantly decreased compared with that in the other two groups (F=178.19,P<0.05).Scratched cells were healed within 48 hours in the HepG2/control group and HepG2 group,but not in the HepG2/RhoA-siRNA group.The clone formation rates in the HepG2/RhoA-siRNA group,HepG2 group and HepG2/control group were 39%±3%,67%±5%and 70%±6%,respectively,with a significant difference among the three groups(χ2=33.34,38.69,P<0.05).Flow cytometry showed that the number of cells transfected with RhoA-siRNA was highest in the G0/G1 phase and lowest in the S phase(F=70.46,76.57.P<0.05).Conclusion The RhoA-siRNA expression vector can effectively suppress the proliferation and migration of HepG2 cells,which may provide a novel gene therapy for hepatocellular carcinoma.
7.Changes in Vasoreactivity of Rat Pulmonary Artery after 7 d Tail-suspension

Objective To clarify the effects of simulated microgravity on the local regulating function of the pulmonary circulation. Method Rat tail-suspension(TS) was used as the animal model to simulate the effects of microgravity.The changes in responses of pulmonary artery to several vasoactive agents were determined by using in vitro vessel rings perfusion technique. Result As for the endothelium-intact vessel rings from TS group compared with those from the control(C) group,the contractile responses of pulmonary artery to potassium chloride (KCl,10~70 mmol/L) did not change obviously after 7 d tail-suspension,which were markedly diminished as to phenylephrine (PE,10-9~10-5mol/L).The vasodilatory responses to acetylcholine (ACh,10-9~10-5mol/L) were markedly enhanced,which did not change obviously as to captopril (Cap,10-5~3×10-3 mol/L).The vasoreactivity of endothelium-removed pulmonary artery rings to all vasoactive agents did not change after 7 d tail-suspension as compared with those from control rats. Conclusion The release of nitric oxide (NO) by rat pulmonary arterial endothelium was enhanced after 7 d tail-suspension.The vasodilatory function of pulmonary artery was enhanced.
8.Apoptin Inducing Apoptosis in the Multidrug-Resistant Model of Human Osteosarcoma Cell Line

Objective: To investigate the curative effects of apoptin on the multidrug-resistant model, of human osteosarcoma cell line (R-OS-732). Methods: 363 bpVP 3 gene was cloned into the vector pIRES1 and the recombinant eukaryon expression vector pIRVP3 was transfected into R-OS-732 cells with liposome. The expression of apoptin gene at transcription level was proved by RT-PCR. The pathological changes of the cells was observed by light microscope and electronmicroscope. The transfected R-OS-732 cells were collected after 48 hours. The genomic DNA extracted from the cells was observed by agarose gel electrophoresis. The apoptosis rate of the cells was analysed by flow cytometry. Results: The expression of apoptin gene at transcription level had been proved by RT-PCR. The construction changes of the two groups were obviously different under light microscope: most of R-OS-732 cells in the transfected group existed in form of being away from the bottom of the culture dish after 24 hours, in form of apoptosis after 48 hours and in form of necrosis after 72 hours; but those cells in the controlled group grow luxuriantly. And under electronmicroscope there was much change of cell nucleus between the two groups. DNA ladder of the transfected cells was observed through agarose gel electrophoresis only one band was observed in the controlled group; but bands of fragmented DNA observed in ositive control group. 9.Influences of 5-azacytidine on mesenchymal stem cells proliferation and differentiation into myoblasts

Objective To investigate the influences of 5-azacytidine (5-Aza) with different concentrations on mesenchymal stem cells (MSCs) proliferation and differentiation into myoblasts. Methods Bone marrow-derived MSCs of 4 weeks Wistar rats were separated and purified, and then treated with 5-Aza with different concentrations. The growth ability of cell was assayed with methyl thiazolyl tetrazolium (MTT) method. The expression of skeletal muscle actin in MSCs was determined with reverse transcription polymerase chain reaction (RT-PCR) and electrophoresis after MSCs were induced. Results The cell proliferation was not affected by 0 and 1 ?mol ? L-1 5-Aza . There were expressions of skeletal muscle actin treated with 3 -12 ?mol ? L-1 5-Aza. Some cells were obviously enlarged at the 9th day after induction and myotubu-like cells were found at the 12th day when treated with 9 and 12 ?mol ? L-1 5-Aza. 20 - 30 ?mol ? L-1 5-Aza induced the toxic effect on proliferation of MSCs. With the increase of concentration, the proliferation ability of MSCs was weakened. Conclusion 5-Aza affects the expression of regulatory gene to the stem cells and regulate MSCs to orientationally differentiate into myotube-like cells.
10.Content Determination of ?-Linoleic Acid in Songzi Oil by Gas Chromatography

OBJECTIVE:To establish the gas chromatography(GC) for the content determination of ?-linolenic acid in songzi oil.METHODS:The separation was performed on 5% DEGS column with detector temperature at 220℃ and flow rate at 30ml/min,nitrogen gas was used as carrier.RESULTS:The linear range for ?-linoleic acid was 1.162~5.81mg/ml(r= 0.9 995).The average recovery was 97.95%(RSD=0.89%,n=3).CONCLUSION:This method was sensitive,accurate and reliable and suitable for the assaying of ?-linoleic acid in songzi oil.
