Objective To study the incidence and clinical significance of abnormal neuroendocrine hormone in patients with the syndrome of brain injury. Methods 67 cases with the syndrome of brain injury were included in the study group, and 95 cases without the post traumatic syndrome after brain injury were included in the control group. The level of FT3,FT4,TSH,growth hormone(GH),andrenocortico hormone(ACTH),cortisol (Cor),prolactin(PRL),testosterone (T),estradiol (E2),follicular stimulating hormone (FSH),luteotropic hormone (LH),and progesterone (P) in peripheral blood were measured by radioimmunoassay. The incidence of the abnormal neuroendocrine hormone after brain injury was statistically analyzed.Patients with abnormal hormone were given hormone replacement therapy and the curative effects were observed. Results The incidence of neuroendocrine hormone abnormalities was 38.8% in patients with the syndrome of brain injury, while it was 10.5% in the control group. There was significant difference between the 2 groups(P<0.05).The symptom remission rate was 88.5% after hormone treatment. Conclusions There was a high incidence of abnormal neuroendocrine hormone secretion in patients with the syndrome of brain injury. The hormone level may be used as an important index to guide clinical therapy.

Colon cancer is a common malignant tumor. Early diagnosis and treatment is one of the most important clinical challenges. Since MicroRNA is discovered to closely relate to cancer, it has been suggested to be used in the diagnosis and treatment of cancer. This review focuses on recent advances in the diagnosis and treatment of MicroRNA in colon cancer and the role of MicroRNA in Wnt signaling pathways and epidermal growth factor receptor (EGFR) signaling pathways.

Objective To investigate the value of individualized thyroid stimulating hormone (TSH) inhibition therapy in postoperative patients with differentiated thyroid carcinoma. Methods The medical record and follow-up data of the 556 patients with differentiated thyroid carcinoma after total or neartotal thyroidectomy were retrospectively reviewed.Patients were divided into two groups:Group A (304 cases) received TSH suppression therapy without risk assessment.Group B (252 cases) were given TSH suppression therapy in accord with risk assessment of both differentiated thyroid cancer recurrence risk stratification condition and the side effects of TSH suppression therapy risk stratification. Results The 3-year non-recurrence and (or) non-metastasis rate in group B was 99.2% which was higher than 96.8% in group A (P =0.044).The hospitalization rate caused by postoperative cardiovascular events or other morbidities in group B decreased 89% than that in group A.Conclusions Individualized TSH suppression therapy can significantly decrease the recurrence and metastasis rate as well as concurrent morbidities caused by unnecessary TSH inhibition.

Objective To construct five shRNA-expression plasmids and to investigate the expression of Smad2 in TGF-?/ Smads signal transduction treated with shRNA-expression plasmid. Methods Five shRNA-Smad2 DNA sequences from mRNA sequence of mouse Smad2 gene were designed and synthesized.DNA oligonucleotides encoding an appropriate shRNA were inserted to shRNA expression vector respectively.Five shRNA-Smad2 expression plasmids were obtained and then transfected into NIH/3T3 cells. The suppressed expression of Smad2 was assessed by RT-PCR and Western-blotting.Results The shRNA-expression plasmid numbered 2.4 could markedly reduce the expression of Smad2.The suppression effect of the RNAi-pool composed of four different plasmids was more obvious than that of any single. Conclusion The shRNA-expression plasmids were successfully constructed, which could specifically and effectively suppress the expression of Smad2.The method of using a mixture of RNAi plasmids to improve the RNAi efficiency was established.

Sixty cases of type 2 diabetes with diabetic peripheral neuropathy (DPN), 46 cases of type 2 diabetes without DPN and 50 healthy persons were recruited. Plasma homocysteine (Hcy), serum folic acid, vitamin B 12 levels and the mutations of Hcy metabolism related enzyme-methyhenetetrahydrofolate reductase (MTHFR) were assayed in individuals in this study. The results showed that high Hcy, low folic acid and vitamin B 12 levels are related with DPN in the patients with type 2 diabetes. The mutation of MTHFR C677T is associated with diabetes, but not with DPN.

Objective To investigate the significance of carcinoembryonic antigen (CEA),carbohydrate antigen 19-9 (CA19-9),carbohydrate antigen 125 (CA125),neuron specific enolase (NSE),cytokeratin 19 fragment (CYFRA21-1) and squamous cell carcinoma antigen (SCC) for evaluation of first-line chemotherapeutic response and the prognostic value of these markers for prediction of overall survival (OS) in patients with advanced lung cancer. Methods Patients diagnosed with Ⅲ b/Ⅳ stage untreated, primary lung cancer and received first-line chemotherapy in Peking University Cancer Hospital & Institute from June 2013 to December 2014 were enrolled retrospectively into this study. The results of tumor markers before and after two cycles of chemotherapy and the clinical data of 181 eligible patients, including 133 males and 48 females with the average age of 58 years, were collected. The serum levels of six tumor markers were measured by electrochemiluminescence assay.Using RECISTv1.1 as standard, the sensitivity and specificity of tumor markers in classifying PR,SD,PD,were observed. The Kappa agreement test was used to evaluate the correlation between serum tumor markers and CT in evaluating chemotherapy response. The follow-up of OS was derived by telephone. Results The top three positive rates of biomarkers were CEA,CA125 and NSE in adenocarcinoma patients,CYFRA21-1,NSE and SCC in squamous carcinoma patients,NSE,CA125 and CYFRA21-1 in small cell lung cancer patients, respectively. In Kappa agreement test, the changes of serum levels of CEA and CA125,CYFRA21-1 and SCC,NSE were significantly correlated with CT for chemotherapy response evaluation in the mention above three kinds of carcinomas respectively (P < 0.05).The sensitivity of tumor markers predicting PR was more than 90%,but the specificity was only 20%-58.3%,meanwhile,the specificity for SD and PD was high, but the sensitivity was low 0-66.7%).In the prognostic evaluation based on RECISTv1.1,there was no statistical significance for classifying PR,SD and PD in adenocarcinoma patients (median overall survival 17 months vs 22 months vs 14 months,P >0.05),but the OS of PR was longer than SD and PD in squamous and small cell lung carcinoma (median overall survival 28 months vs 22 months vs 4 months,12 months vs 8 months vs 8 months,P < 0.05).Of these six tumor markers, only SCC was of statistical significance for classifying PR,SD and PD in squamous carcinoma (median overall survival 30 months vs 1 1 months vs 4 months,P < 0.05).Conclusions Tumor marker has high sensitivity for predicting PR,but the value of tumor marker predicting SD and PD of advanced lung cancer was limited. The evaluation of patients would be comprehensive by combined use of tumor markers and CT.The changes of SCC after 2 cycles of chemotherapy are predictive of survival in squamous cell carcinoma patients.
