Objective To study the anti-tumor effect of mycobacterium Ag85B/IL-2 fusion protein on syngenic mice bearing bladder cancer. Methods After being implanted BTT739 bladder transitional carcinoma cells from tumor bearing mice, 80 T739 mice were divided into Ag85B protein, BCG, IL-2 and normal saline groups (n=20). Mycobacterium Ag85B/IL-2 protein was locally injected to the mice at the site of implanting tumor in the trial group, and Ag85B protein, BCG, IL-2 and normal saline to the control group mice. The weight and volume of tumor and the survival period were recorded after treatment. Natural polysaccharides are a kind of hydrophilic polymers.By grafting hydrophobic moieties to the polysaccharide backbone,could form self-assembled micelle with core-shell structure in aqueous by undergoing intramolecular or intermolecular association between hydrophobic moieties.The self-assembly micelles of amphiphilic polysaccharides provides a positive outlook for drug delivery systems with biodegradability,low antigenicity,good biocompatibility and so on.This review focus on biological activity of amphiphilic polysaccharide as self-assembly carriers,the principle of self-assembly of polymeric micelles,and the recent progress in hydrophobic modification of natural polysaccharides.
4.Study on the Optimization of Matrix Formula of Lanlian Ertong Qingre Cataplasm

OBJECTIVE:To optimize the matrix formula of Lanlian ertong qingre cataplasm. METHODS:Taking adhesion force,peel strength and sensory description as index,the ratio of matrix framework material(sodium polyacrylate-gan hydroxyl alu-minum-tartaric acid-glycerin) was optimized with orthogonal test. The single factor test was adopted to select adhesive and filler;the amount of penetrating agent azone was screened using the in vitro penetration amount of phillyrin. RESULTS:The best matrix ratio of Lanlian ertong qingre cataplasm was sodium polyacrylate-gan hydroxyl aluminum-tartaric acid-glycerin(4.0:0.8:0.4:15);PVP K-90 was used as adhesive,and bolus alba as filler;penetration enhancers azone accounted for 2.0%. Validation test showed, prepared cataplasm had good appearance,could stick on the 5th or the 6th ball;it's peel strength was 7.5 N;all RSDs of score were lower than 4%(n=3). CONCLUSIONS:The optimized matrix formula of Lanlian ertong qingre cataplasm is simple,stable and good in molding.

AIM:To develop a method for determining cholic acid by HPLC-ELSD and GC was applied to determing muscone;in Jawei Xihuang Soft Capsule(Calculus Bovis,Moschus,Venenum Bufonis,Olibanum,Myrrha).METHODS:AC_(18) column(Kromasil C_(18),5 μm,4.6 mm×250 mm)was used as stationary phase,the mobile phase was methanol-0.01% glacial acetic acid(73:21)at a flow rate of 1.0 mL/min.The parameters of ELSD were set as follows:evaporation temperature was 40℃,carrier gas(N_2)pressure was 200 kPa.The GC system consisted of DB-1 capillary column(30 m×0.32 mm×0.25 μm)and FID as the detector.The programmed temperature-GC and internal standard method were employed to determine the content of muscone.RESULTS:The linear ranges of cholic acid and muscone were in the range of 45.2 ng-904 ng and 0.05 mg/mL-0.5 mg/mL respectively.The average recoveries were 99.06% and 99.40% with RSD of 1.56% and 0.95% respectively.CONCLUSION:The method is convenient and accurate,and it can be used for the quality evaluation of Jawei Xihuang Soft Capsule.

8.Immunological function variation of peripheral dendritic cell from bladder cancer patients after blocking PD-L1 pathway

Objective To investigate the immunological function variation of peripheral dendritic cell(DC)from bladder cancer patients after blocking PD-L1 pathway.Methods DC from normal control and patient with bladder cancer were cultured with rhGM-CSF,rhIL-4 in vitro and treated with PD-L1 monoclonal antibody.The expression of CD1a,HLA and CD83 were examined by flow cell meter.The effect of DC induced lymphatic cellular proliferation and its capability of secreting IL-10,IL-12 were determined by MTT and ELISA.Results Blocking PD-LI pathway did not infuluence the maturation of DC.But the DCs from bladder cancer patient signifcantly boosted the lymphatic cellular proliferating(4.00 ± 1.28 versus 1.49 ±0.45)and IL-12 secretion capability(108.30 ± 21.89 versus 37.17 ± 14.89 ng/L),and yet it also decreased the secretion of IL-10(108.90 ± 21.77 versus 14.99 ± 54.99 ng/L)after blocking PD-L1 pathway (P < 0.05).Conclusion Blocking PD-LI pathway on the DC from bladder cancer patient may improve its anticancer immunological function.
9.Method and Analysis of Click Induced Auditory BrainstemResponses of Normal Adult Mice

Objective To obtain the auditory brainstem response (ABR)wave of normal adult mice ,and to analyze the thresholds ,latency and amplitudes to provide reference for future mice auditory research .Methods To determine the thresholds according to the high occurrence rate wave of click ABR at high (90 dB SPL) ,moderate (40 dB SPL)and low(20 dB SPL)sound intensities .thresholds ,latencies and amplitudes decreased as the wave with high occurrence rate .Results The occurrence rates of wave I ,II ,III ,IV ,and V were 100% ,100% ,100% ,75% 65% at high levels ;75% ,100% ,100% ,45% ,40% at moderate levels ,and 30% ,90% ,40% ,5% ,0% at low levels ,respec-tively .The results showed wave morphology was suitable to judging thresholds :the average threshold was 20 ± 5 .62 dB SPL ,latency and amplitude ranged from 2 .71 ± 0 .17~3 .08 ± 021 ms and 6 .20 ± 2 .24~1 .63 ± 0 .85 μV ,respectively , when sound intensities changed from 90 to 20 dB SPL .Conclusion Wave II is regarded as the best wave to determine threshold latency and amplitude .It will help future researches about mice auditory under pathophysiology conditions .
10.A discussion of reasons and methods of prevention and cure for serious complications of radiofrequency ablations in the treatments of hepatocellular carcinomas

Objective To investigate the reasons and the methods of prevention and cure for serious complications of radiofrequency ablations in the treatments of hepatocellular carcinomas. Methods A total of 410 patients with BCLC at A or B stage of hepatocellular carcinomas in our hospital were enrolled between November 2014 and June 2009. These patients underwent a total of 504 times radiofrequency ablations for the treatments of liver lesions. This retrospective study analysed the reasons and the strategies of prevention and cure for the serious complications. Results In the patients with a total of 504 times radiofrequency ablations, 2 patients had massive hemorrhage caused by puncture injuries, 2 patients had the tumors which were close to the liver capsules and 1 patient had bile peritonitis caused by the injury of thermal ablation on the gallbladder. The maximum diameter of tumor was 5 cm. That tumor was close to the gallbladder. 2 patients had needle tract metastases caused by incompletely needle path ablations. 1 of the 2 patients had a tumor near the liver capsule, and the other patient had un-enough temperature for needle path ablation. Tumor outbreaks were happened in 2 patients. 1 of the 2 patients had a tumor which was located in the liver capsule and close to the portal vein. The other patient had a 12 cm diameter tumor with rich blood supplement. 2 patients had liver abscesses. 1 of the 2 patients had a tumor near the ascending colon, and the other patient had diabetes. 1 patient had colonic perforation caused by thermal ablation. The tumor in that patient was located in the right hepatic lobe segment and adjacent to the ascending colon. The incidence of serious complications was 1.98% (10/504). Conclusions The incidence of the serious complications of radiofrequency ablations for the treatments of hepatocellular carcinomas is relatively low. The main reasons for the serious complications were direct injuries caused by punctures, heat radiation injuries, tumors adjacent to large blood vessels, gallbladders and intestines, tumors with abundant blood supplement, needle paths fail to cross normal liver tissues, low scores of liver function, weak immune system and diabetes. The key points for avoiding and reducing the serious complications are preoperative evaluations of patients' basic situations, choices of appropriate puncture channels and control ranges of ablations when tumors are close to important blood vessels, intestines and gallbladders.
