1.Effect of 5-Aminosalicylic Acid on Cell Proliferation,Apoptosis and Cell Cycle in Human Colon Cancer Cell Line HT-29

Background:Ulcerative colitis-associated colorectal cancer(UcCRC)is the most serious complication of ulcerative colitis(UC). 5-Aminosalicylic acid(5-ASA)could reduce the risk of UC progressing to UcCRC. Aims:To investigate the effect of 5-ASA on cell proliferation,apoptosis and cell cycle in human colon cancer cell line HT-29 for verifying the inhibitory effect of 5-ASA on UcCRC. Methods:HT-29 cells were treated with different concentrations(0,10,20,30, 40 mmol/ L)of 5-ASA. Cell proliferation was measured by CCK-8 assay. Cell apoptosis was determined by TUNEL method. Cell cycle was analyzed by flow cytometry. Expressions of cell mitotic regulators AuroraB and BubR1 were determined by immunohistochemistry. Results:Proliferation inhibition rates and apoptosis rates of HT-29 cells in 5-ASA 10,20,30,40 mmol/ L groups were increased with increase in concentration of 5-ASA(P ﹤ 0. 05). Proportions of cells in phase G0 / G1 in 5-ASA 30,40 mmol/ L groups were significantly lower than those in 5-ASA 0,10,20 mmol/ L groups (P ﹤ 0. 05);proportions of cells in phase S in 5-ASA 0,10,20,30 mmol/ L groups were significantly lower than that in 5-ASA 40 mmol/ L group(P ﹤ 0. 05);while proportions of cells in phase G2 / M in 5-ASA 30,40 mmol/ L groups were significantly higher than those in 5-ASA 0,10,20 mmol/ L groups(P ﹤ 0. 05). Mean optical density(MOD)values of AuroraB and BubR1 were decreased with increase in concentration of 5-ASA(P ﹤ 0. 05). Concentration of 5-ASA was positively correlated with proliferation inhibition rate and apoptosis rate of HT-29 cells(P ﹤ 0. 05),but was negatively correlated with MOD values of AuroraB and BubR1( P ﹤ 0. 05). MOD values of AuroraB and BubR1 were negatively correlated with proliferation inhibition rate and apoptosis rate of HT-29 cells as well as proportion of cells in phase G2/ M (P ﹤0. 05),but were positively correlated with proportion of cells in phase G0/ G1(P ﹤0. 05). MOD value of AuroraB was positively correlated with MOD value of BubR1(P ﹤ 0. 05). Conclusions:5-ASA can inhibit proliferation and induce apoptosis in HT-29 cells,the mechanism may be related to inhibiting expressions of AuroraB and BubR1 and the resulted effect on cell cycle.
2.Post-ERCP hemorrhage in patients with choledocholithiasis: risk factors and prevention

Objective To investigate the risk factors and prevention of the ERCP-related upper gastrointestinal bleeding in patients with choledocholithiasis.Methods The clinical data of 678 patients who received ERCP in Yijishan Hospital of Wannan Medical College between January 2014 and December 2014 were reviewed.Univariate and multivariate analyses were performed to define the independent risk factor(s) for delayed hemorrhage following ERCP.Results A total of 383 patients,in whom 157 were males and 226 were females,the age ranged from 19 to 88 years,were recruited to the study.ERCP-related upper gastrointestinal bleeding occurred in 41 patients(10.7%),intraoperative bleeding occurred in 27(7.0%) and postoperative hemorrhage in 14 (3.7%).Twenty-eight cases were mild bleeding,9 moderate (4 caused by laceration of the cardiac mucosa) and 4 were severe bleeding.All patients recovered and were discharged,and no death occurred.No single case required surgical conversion.Univariate analysis revealed that cholangitis,medical history of aspirin/clopidogrel,giant diverticulum and the major papilla inside the diverticulum were risk factors for delayed hemorrhage following ERCP (P < 0.05).Multivariate analysis indicated that independent risk factors were associated with cholangitis (OR =4.125,95% CI:1.306-13.031,P < 0.05),medical history of aspirin/clopidogrel (OR =10.220,95% CI:2.997-34.853,P < 0.01) and major papilla occurred inside the diverticulum(OR =14.064,95% CI:1.888-104.762,P < 0.05).Conclusion Cholangitis,medical history of aspirin/clopidogrel and major papilla at the diverticulum may increase the risks for upper gastrointestinal bleeding in ERCP in patients with choledocholithiasis.However,the hemorrhage should be actively managed,especially in endoscopy,once the bleeding occurs.
3.Effect of human telomerase reverse transcriptase-tumstatin combined with human telomerase reverse transcriptase-thymidine kinase/ganciclovir system for liver cancer cell line HepG2

Objective To observe the effect of human telomerase reverse transcriptase-thymidine kinase/ganciclovir (phTERT-TK/GCV) system combined human telomerase reverse transcriptase-tumstatin (phTERT-tumstatin) system on apoptosis of human HepG2 and mRNA expression and protein content of AFP,RhoC related with cancer.Methods Fluorescence microscopy was used to observe expression of EGFP and MCHERRY in transfected HepG2 and L-02.Real-time PCR and Western blot were used to detect AFP and RhoC mRNA and protein content.Flow cytometry was used to detect the apoptosis of transfected HepG2.Results phTERT-tumstatin and phTERT-TK/GCV genes expressed in transfected HepG2.Real-time PCR showed that AFP and RhoC mRNA expression in different group were 0.76±0.09 and 0.80±0.04 (TK/GCV group),0.62±0.09 and 0.40±0.02 (TM group),0.49±0.07 and 0.54±0.03 (MK group).The differences were significant (P < 0.01) except TK/GCV group compared with empty plasmid group.Western blot test results showed that protein content of AFP and RhoC were higher in TK/GCV group (0.97±0.02/1.17± 0.01),TM group (0.83±0.02/0.99±0.02),MK group (0.69±0.01/0.77±0.02) than in empty plasmid group (1.19±0.03/1.32±0.05) and non-transfected group (1.15±0.05/1.29±0.30) (P < 0.01).Additionally,protein content of AFP and RhoC in MK group were significant difference with TK/GCV group and TM group (P < 0.01).Flow cytometry showed that phTERT-tumstatin,phTERT-TK/GCV co-transfected HepG2 cells apoptosis rate was significantly higher than both individually transected group.Cells apoptosis rate of alone and co-transfected groups was significant difference compared with empty vector group and untransfected group.Conclusions The effect of phTERT-TK/GCV and phTERT-tumstatin on pro-apoptotic of HepG2 cells is significant.phTERT-TK/GCV combined with phTERT-tumstatin has strong therapeutic function.
4.Bibliometric Analysis and Research on Preventive Treatment of Disease in Acupuncture and Moxibustion

This article was aimed to analyze literatures on preventive treatment of disease in acupuncture and moxi-bustion in order to understand its status and development trend. Bibliometrics and statistics method in NoteExpress were used in the analysis of related literatures about preventive treatment of disease in acupuncture and moxibustion from 1981 to 2013, which were included in CNKI. The analysis and research were from aspects such as year distri-bution, keyword, journal distribution, author distribution, and fund distribution. The results showed that there were 537 articles in total from 1981 to 2013. The number of literatures was in an overall growing trend.“Zusanli”,“Inner Canon of Huangdi”,“sub-health”, and“clinical research”were high frequency keywords. The journal distribu-tion conformed to the Brad Ford’s law. The author distribution conformed to the Lotka’s law. The number of fund papers was also increased. It was concluded that research on preventive treatment of disease in acupuncture and moxibustion was still in the period of rapid development and had not reached maturity. The preventive treatment of disease in acupuncture and moxibustion had broad application prospects. However, the research still required to be strengthened.
5.Observation of the clinical efficacy of Shenfuyishen Capsule in the treatment of diabetic nephropathy in high altitude regions

Objective To observe the clinical efficacy of Shenfuyishen Capsule in the treatment of diabetic nephropathy of yangxuxueyu type in high- altitude regions.Methods We divided 53 patients randomly into two groups:31 in the observation group and 22 in the control group.Patients in the control group were treated with conventional regime of Western Medicine.Those in treatment group were given extra treatment with Shenfuyishen Capsule.Then we tested serum creatinine (Scr ),uric acid (UA),urinary microalbumin (MALB )and GFR. Results The total efficiency was 90.32% and 81.82% in the treatment group and the control group,which was significantly different (P < 0.05 ).Major symptoms improved after treatment in both groups,which differed significantly compared with those before treatment (P <0.05).ALT in the treatment group was not different after treatment.CR,UA,MALB and GFR significantly differed before and after treatment in both groups,especially in the treatment group (P <0.05).Conclusion Shenfuyishen Capsule helps improve major clinical symptoms and decrease CR,GFR,UA and MLAB levels in patients with diabetic nephrology,thus protecting renal function and delaying the process of diabetic nephropathy.

Lung cancer and chronic obstructive pulmonary disease (COPD) are two high-mortality diseases in the world.Numerous epidemiological studies have demonstrated that presence of COPD increases the risk of lung cancer.Habitual cigarette smoking frequently develops lung cancer as well as COPD,However the links between the two diseases should be more than smoking alone.The underlying mechanisms may include genetic predisposition,inflammation and cell injury,oxidative and noxious stress,extracellular matrix and proteinases,some of which might represent the targets for chemoprevention or chemotherapy.
7.Preparation and Quality Control of Doxycycline Hydrochloride Microspheres

OBJECTIVE: To prepare doxycycline hydrochloride microspheres and to establish a method for its quality control.METHODS: O/O emulsion solvent evaporation method was adopted to prepare doxycycline hydrochloride microspheres with poly (lactic-co-glycolic acid) (PLGA) as carrier.Light microscope was used to detect appearance and particle size of microspheres and UV spectrophotometry was applied to determine the loading rate and release rate of drug in vitro.RESULTS: Prepared microspheres are smooth and round with mean particle size of (49?6.9) ?m and span of 0.9.Average drug loading rate was (3.3?0.2) % and encapsulation efficiency was (52.4?3.2) % (n=3).Accumulative drug release reached 28% within 0.5 h with constant drug release lasting more than 30 days.CONCLUSION: Doxycycline hydrochloride microspheres preparation process is available and controllable in quality.

9.Botulinum Toxin Type A Therapy in Meige's Syndrome and Facial Wrinkles

Objective To observe the efficacy of Botulinum toxin type A(BTXA)injection in Meige's syndrome and facial wrinkles and exp lore the method of reducing the side e ffect of BTXAand weaken facial wrink les.Methods Ninety-nine patients were evaluate d on the therapeutic efficacy of single and multiple topical facial in-jection with BTXA.Results The effective rates of patients with blepharospasm were 98.4%,oromandi bular dystonia 90.9%,blepharospasm and o romandibular dystonia 95.8%,respectively.The therapeutic effect for each disease lasted(16.9?8.4)weeks?(12.6?5.4)weeks and(13.5?5.7)weeks,respectively.Facial wrinkl es of all patients has got weakened and seemed much smooth and flattend.The rec overy rate approached to 100%.The local side effects were transient an d mild,and disappeared spontaneously.Conclusions The treatment of Meige's syndrome and facial wrinkles with BTXA could produce a significant impro vement,and is safe.Strictly observ e the procedures of injection,and use adequate dose of BTXA,the side effects o f BTXA can be decreased.
10.Cellular oxygen sensors and signal transduction

Hypoxia is a commenest pathological process. The cellular oxygen sensors and signal transduction involved in hypoxic responses are so far not fully elucidated. There are many theories. Perhaps the oxygen sensors are different among different kinds of cells, and between acute and chronic hypoxic responses. The mitochondria cytochrom-oxidase-H_2O_2 might be the principal pathway leading to the constrictive response of pulmonary smooth muscle cells to hypoxia. The heme protein - reactive oxygen pathway might mediate the response of glomus cells in carotid body to hypoxia. The NADPH oxidase might be the oxygen sensor in airway chemoreceptor. As for chronic hypoxic responses, the oxygen dependent regulation of hypoxia-inducible factors by prolyl and asparaginyl hydroxylation has been paid great attention in recent years.
