1.Cost-effectiveness Analysis of Different Slow-release Isosorbide Mononitrate Preparations in the Treatment of Coronary Heart Disease

OBJECTIVE:To evaluate the cost-effectiveness of different slow-release isosorbide mononitrate preparations in the treatment of coronary heart disease.METHODS:The random open study,three factors crossover trial and own control clinical test were conducted on3different Isosorbide mononitrate preparations.Then the curative effects of each were observed and the cost-effectiveness analyses were conducted.RESULTS:For the three kinds(A,B and C)of slow-release isosorbide mononitrate capsule,their total effective rates were87.9%,69.7%and57.6%respectively;the average daily medical expenses of which were10.25,15.14and14.04respectively;their cost-effectiveness ratio were69.97,130.33and146.25yuan respectively.CONCLUSION:The cost-effectiveness of the slow-release isosorbide mononitrate preparations is superior to the control medicines in the treatment of coronary heart disease.

Objective:To measure CT linearity and evaluate if it would show linear relationship between coefficient and attenuation.Methods:The Catphan phantom was used to measure CT linearity in 18 computed tomography equipments,and the results were calculated.Results:(1)The CT linearity were qualified in 14 equipment but 4.Conclusion:(1)There are many factors which affect CT linearity.The artifact will be produced if the CT linearity is unqualified.(2)CT linearity must be calibrated periodically to assure the stability of the CT linearity and to make for the accurate diagnoses.
3.Clinical efficacy of two surgical treatment methods for anal fissures in elderly patients

Objective To compare the clinical efficacy of two surgical methods for the treatment of anal fissures in elderly patients.Methods A total of 138 elderly patients with anal fissures treated at our hospital from January to December 2016 were randomly divided into the conventional method group(69 cases) and the modified method group (69 cases).Patients in the conventional method group were treated with lateral sphincterotomy,while patients in the modified method group were treated with modified longitudinal sphincterotomy with transverse suture.Outcomes of treatment,VAS scores,incision wound infection rates,postoperative voiding dysfunction rates and recurrence rates were compared.Results There was no significant difference in overall effectiveness between the modified method group (98.6%)and the conventional method group(94.2%) (x2=1.8677,P=0.1717).The rates of incision wound infection,moderate postoperative urinary retention and recurrence in the conventional method group(8.7%,42.0% and 10.1 %,respectively)were significantly higher than those in the modified methods group(0.0 %,15.9 % and 0.0%,respectively) (x2 =4.3561,11.4061 and 5.4177,respectively,P =0.0369,0.0007 and 0.0199,respectively).At 12 hours after operation,favorable VAS scores and incision healing times were observed in the modified method group,compared with the conventional method group,and the differences were statistically significant(each P<0.05).Conclusions Modified longitudinal sphincterotomy with transverse suture produces about the same overall effectiveness as does lateral sphincterotomy in the treatment of elderly patients with anal fissures,but it is superior in reducing incision wound infection,moderate postoperative voiding dysfunction,recurrence and pain intensity,and in promoting incision healing.

5.Study of the Biocompatibility Between Bone Morphogenetic Protein Microspheres and Bone Marrow Stem Cell

Objective To evaluate the biocompatibility between bone morphogenetic protein(BMP) composition and bone marrow stem cell (BMSc).Methods Microspheres were prepared with chitosan and sodium alginate, and BMP was enwrapped in the microspheres. The biocompatibility of the composition was examined using cell-culturing method. The BMSc was cultured in combination with microspheres. The extending speed of the cells, the proliferation and alkaline phosphatase activity were tested.Results There were no inhibition on cellular proliferation of BMSc when it was cultured in combination with microspheres in vitro, but ALP activity increased significantly.Conclusion BMP microspheres possessed satisfactory biocompatibility, and could increase the osteogenic capability of BMSc in vitro.
6.Clinical manifestations of osteoarthritis and the role of silent information regulation 1 in the pathogenesis of osteoarthritis

BACKGROUND:Functional changes of some functional genes have been showed to trigger osteoarthritis, among which, age may be a critical one. OBJECTIVE:To summarize the clinical manifestations of osteoarthritis and the research process of silent information regulation 1 in the occurrence and development of osteoarthritis. METHODS:A computer-based retrieval was performed in the databases of CNKI, PubMed, SpringerLink, and Elsevier Science Direct using the keywords ofosteoarthritis, silent information regulation 1 and cartilagein Chinese and English, respectively. Finally 83 eligible literatures were enrolled for analysis. RESULTS AND CONCLUSION:Deacetylation modification acts on the occurrence and development of osteoarthritis by regulating the expression and biological function of various cytokines. Silent information regulation 1 cannot only regulate body metabolism, inhibit cellapoptosis, repair DNA injury, delay senescence and resist to stress, but also play important roles in the epigenetic modification, gene silencing and signal transduction. Therefore, silent information regulation 1 is involved in the age-related diseases, such as osteoarthritis, osteoporosis, diabetes and Alzheimer's disease. Furthermore, silent information regulation 1 is likely to be an important target of osteoarthritis drugs, and even reverses mechanical stress-induced cartilage injury in the early stage of osteoarthritis.
7.Cervical adenoid basal cell carcinoma:a clinlcopathologic study of 12 patients

Purpose To investigate the clinicopathologic features of cervical adenoid basal cell carcinoma. Methods Clinical and pathological data were analyzed in 12 cases of cervical adenoid basal cell carcinoma and the relevant literatures were reviewed. Results The age of 12 cases of with cervical adenoid basal cell cancer patients ranged from 36~70 years ( mean:53. 3 years) , with a medi-an age of 51. 5 years. Amongst 12 patients who contained no gross definite lesion, 5 patients (41. 7%) had the symptoms of vaginal bleeding. 8 cases of patients underwent hysterectomy, while the other four received cervical conization. Cervical intraepithelial neopla-sia ( CIN) lesions were observed in all 12 cases. The tumor cells were small and uniform, with dark oval nuclei without conspicuous nucleoli and scanty cytoplasm. Tumor infiltrated into the stroma in nests and cords. Glandular differentiation within tumor nests were seen in 12 cases, including two cases of squamous cell differentiation accompanied by partial and minor interstitial edema. Peripheral palisading cells around tumor nests were also found. The infiltrating depth of tumor ranged from 0. 5~10 mm with the average 4. 12 mm. Cancer embolis were available in 2 cases with no lymph node metastasis. 10 cases followed up for 3~78 months with no recur-rence and metastasis, but 2 cases were lost. Conclusion Adenoid basal cell carcinoma is a rare uterine cervical tumor found in post-menopausal women, which are often accompanied with CIN. This tumor has a favorable prognosis and should be clearly separated from adenoid cystic carcinoma and other tumors which also have infiltrative growth pattern.

Objective To investigate expression of the phosphatase and tensin homolog (PTEN) gene in breast cancer cell line and its effect on biologic characteristics.Methods The normal PTEN expression cell line MDA-MB-231 (M231) was used in this study.PTEN-shRNA plasmid was transected into M231 breast cancer cells to knock down the expression of PTEN.The changes of PTEN expression,proliferation,invasion and metastasis of PTEN knocked down cell were tested by RT-PCR,Western blot,CCK-8,scratch and Transwell.Results PTEN-shRNA was successfully transected into M231 cells.PTEN mRNA and protein expression was efficiently inhibited in M231-3001 cell lines than that in control group M231-scr(P < 0.01),M231-3001 cell lines showed a greater capability of colony formation,migration and invasion than that in control group M231-scr (all P < 0.05).Conclusions PTEN,as a suppression gene,its low expression can promote the proliferation,migration and invasion of breast cancer cells.
9.Construction of miRNA-target networks and gene GO analysis of the specific target genes in colorectal cancer with liver metastasis

Objective To explore the microRNA expression changes and related characteristics and analyze the corresponding miRNA target genes and their bioinformatics in colorectal cancer with liver metastasis.Methods The fresh specimens of primary CRC were collected in 10 patients during operation,with liver metastasis or without.The miRNA expression levels were compared by miRNA microarray between two groups.Then,target genes were identified using target prediction algorithms.The liver metastasis related miRNA-target networks and gene ontology (GO) bioinformatics analysis were further performed.Results A total of six dysregulated miRNAs were identified in colorectal cancer liver metastasis comparing with no metastasis,including 3 up-regulated miRNAs (miR-224,miR-1236,miR-622) and 3 downregulated miRNAs (miR-155,miR-342-5p,miR-363).miR-224 with most significantly up-regulation played regulation role not only with corresponding target-genes but also downstream genes.Conclusions As a core of the regulation networks,miR-224 can regulate the related gene functional groups simultaneously and asynchronously.It further implements the post-transcriptional regulation and plays a vital role in liver metastasis of colorectal cancer.
10.PrP 106-126 Altered PrP mRNA Gene Expression in Mouse Microglia BV-2 Cells'

Prion diseases are infectious and fatal neurodegenerative diseases. The pathogenic agent is an abnormal prion protein aggregate. Microglial activation in the centre nervous system is a characteristic feature of prion disease. In this study, we examined the effect of PrP 106-126 on PrP mRNA gene expression in Mouse microglia cells BV-2 by real-time quantitative PCR. PrP mRNA expression level was found to be significantly increased after 18 h exposure of BV-2 cells to PrP 106-126, with 3-fold increase after 18 h and 4.5-fold increase after 24 h and BV-2 cells proliferating occurred correspondingly. Our results provide the first in vitro evidence of the increase of PrP mRNA levels in microglial cells exposed to PrP 106-126, and indicate that microglial cells might play a critical role in prion pathogenesis.
